The pepper SAR8.2 gene, CASAR82A, was locally and systemically induced in pepper plants which had been infected by Xanthomonas campestris pv. vesicatoria or by Pseudomonas fluorescens. The DNA 1,283 bp sequence upstream of the CASAR82A gene was assessed with regard to the activity of the CASAR82A promoter fused to the beta-glucuronidase (GUS) reporter gene, via an Agrobacterium-mediated transient expression assay. In tobacco leaves which transiently expressed the -831 bp CASAR82A promoter, GUS activity was locally and systemically induced by Pseudomonas syringae pv. tabaci. GUS activity, which

SUMMARY Sequence analysis of the region bordering the hrp/dsp gene cluster of Erwinia amylovora strain Ea321, which causes fire blight, revealed characteristics of pathogenicity islands (PAIs). Included are genes for a phage integrase, a tRNA(Phe), several orthologues of genes of YAPI, a PAI of Yersinia pseudotuberculosis, and several putative virulence genes with HrpL-dependent promoter motifs. The island is designated the Hrp PAI of E. amylovora. It is comprised of a chromosomal region of c. 62 kb with 60 open reading frames (ORFs). Comparison of the Hrp PAI of E. amylovora with those of fou

Plant pathogens harm crops and reduce yields. We urgently need safe and effective antimicrobial agents to treat plant diseases. In this study, a series of isocryptolepine derivatives were efficiently synthesized using 4-oxoquinolinones, employing active functional group modification and bioisosteric substitution strategies. The MIC of compound Mb-1 against Xoo was 3.12 μg/mL, which was better than that of the commercial drug copper quinolate. The MICs of compound Mb-7 against Xac and Rs were 1.56 and 0.39 μg/mL respectively. In the in vivo antibacterial experiment, compound Mb-1 exhibited a su

Citrus canker disease, caused by Xanthomonas citri subsp. citri, poses a significant threat to global citrus production. The control of the disease in the field relies mainly on the use of conventional tools such as copper compounds, which are harmful to the environment and could lead to bacterial resistance. This scenario stresses the need for new and sustainable technologies to control phytopathogens, representing a key challenge in developing studies that translate basic into applied knowledge. During infection, X. citri subsp. citri secretes a transcriptional activator-like effector that e

To develop novel bacterial biofilm inhibiting agents, a series of 1,3,4-thiadiazole derivatives containing sulfonylpiperazine structures were designed, synthesized, and characterized using 1H nuclear magnetic resonance (1H NMR), 13C nuclear magnetic resonance (13C NMR), and high-resolution mass spectrometry. Meanwhile, their biological activities were evaluated, and the ensuing structure-activity relationships were discussed. The bioassay results showed the substantial antimicrobial efficacy exhibited by most of the compounds. Among them, compound A24 demonstrated a strong efficacy with an EC5

β-Galactosidases (Bgals) remove terminal β-D-galactosyl residues from the nonreducing ends of β-D-galactosidases and oligosaccharides. Bgals are present in bacteria, fungi, animals, and plants and have various functions. Despite the many studies on the evolution of BGALs in plants, their functions remain obscure. Here, we identified rice (Oryza sativa) β-galactosidase9 (OsBGAL9) as a direct target of the heat stress-induced transcription factor SPOTTED-LEAF7 (OsSPL7), as demonstrated by protoplast transactivation analysis and yeast 1-hybrid and electrophoretic mobility shift assays. Caterpillars of the herbivore Pieris rapae stimulate the production of jasmonic acid (JA) and ethylene (ET) in Arabidopsis (Arabidopsis thaliana) and trigger a defense response that affects insect performance on systemic tissues. To investigate the spectrum of effectiveness of P. rapae-induced resistance, we examined the level of resistance against different pathogens. Although the necrotrophic fungus Alternaria brassicicola is sensitive to JA-dependent defenses, herbivore-induced resistance was not effective against this pathogen. By contrast, caterpillar feeding significantly reduced disease

Transcription activator-like effectors (TALEs) are major virulence factors of Xanthomonas oryzae, the causal agent of bacterial blight in rice. The BED-domain nucleotide-binding leucine-rich repeat receptor Xa1 confers resistance by recognizing TALEs, but the mechanisms underlying Xa1 activation remain unclear. Here we show that structurally conserved canonical TALEs differ in their ability to activate Xa1, revealing unexpected specificity in effector recognition. Using transient expression assays, endogenous epitope tagging, and prime editing of the native Xa1 locus, we demonstrate that nucle

The pepper (Capsicum annuum) resistance gene bacterial spot3 (Bs3) is transcriptionally activated by the matching Xanthomonas euvesicatoria transcription-activator-like effector (TALE) AvrBs3. AvrBs3-induced Bs3 expression triggers a rapid and local cell death reaction, the hypersensitive response (HR). Bs3 is most closely related to plant flavin monooxygenases of the YUCCA (YUC) family, which catalyze the final step in auxin biosynthesis. Targeted mutagenesis of predicted NADPH- and FAD-cofactor sites resulted in Bs3 derivatives that no longer trigger HR, thereby suggesting that the enzymatic

Leptospiral immunoglobulin-like (Lig) proteins are of great interest due to their ability to act as mediators of pathogenesis, serodiagnostic antigens, and immunogens. Purified recombinant LigA protein is the most promising subunit vaccine candidate against leptospirosis reported to date, however, as purified proteins are weak immunogens the use of a potent adjuvant is essential for the success of LigA as a subunit vaccine. In the present study, we compared xanthan pv. pruni (strain 106), aluminium hydroxide (alhydrogel), and CpG ODN as adjuvants in a LigA subunit vaccine preparation. Xanthan

Site-specific recombinases are powerful tools for genome engineering. Hyperactivated variants of the resolvase/invertase family of serine recombinases function without accessory factors, and thus can be re-targeted to sequences of interest by replacing native DNA-binding domains (DBDs) with engineered zinc-finger proteins (ZFPs). However, imperfect modularity with particular domains, lack of high-affinity binding to all DNA triplets, and difficulty in construction has hindered the widespread adoption of ZFPs in unspecialized laboratories. The discovery of a novel type of DBD in transcription a

Ubiquitination is essential for ubiquitin/proteasome-mediated protein degradation in plant development and defense. Here, we identified a novel E3 ubiquitin ligase RING1 gene, CaRING1, from pepper (Capsicum annuum). In pepper, CaRING1 expression is induced by avirulent Xanthomonas campestris pv vesicatoria infection. CaRING1 contains an amino-terminal transmembrane domain and a carboxyl-terminal RING domain. In addition, it displays in vitro E3 ubiquitin ligase activity, and the RING domain is essential for E3 ubiquitin ligase activity in CaRING1. CaRING1 also localizes to the plasma membrane.

BACKGROUND: Platelet concentrates contaminated with bacteria are a main source of transfusion-associated sepsis. Several studies have reported a very wide incidence (0-10%) of contamination. The aim of this study is to assess the prevalence of bacterial contamination of standard platelet units at the regional blood transfusion center in Casablanca.
STUDY DESIGN AND METHODS: During 15.5 months, 3,458 platelet concentrates (PCs) were analysed. The plasma from the bag and the tube was homogenised. One-fifth of the tube contents were inoculated every day for five days in two tubes containing 2.5

The hypersensitive-induced reaction (HIR) gene family is associated with the hypersensitive response (HR) that is a part of the plant defense system against bacterial and fungal pathogens. The involvement of HIR genes in response to viral pathogens has not yet been studied. We now report that the HIR3 genes of Nicotiana benthamiana and Oryza sativa (rice) were upregulated following rice stripe virus (RSV) infection. Silencing of HIR3s in N. benthamiana resulted in an increased accumulation of RSV RNAs, whereas overexpression of HIR3s in N. benthamiana or rice reduced the expression of RSV RNAs

Caseinolytic protease P (ClpP) protein is essential for prokaryotic and eukaryotic protein homeostasis, and has emerged as a promising bactericidal target for controlling bacterial infection and evading the emergent of drug-resistance risk. As the unique bactericidal mechanism, ClpP loses its substrate specificity and undergoes uncontrolled protein hydrolysis in the presence of an activator, leading to causing bacterial death. To further expand the chemotype of ClpP activator, a series of ONC212 derivatives were synthesized using a ring expansion strategy. Surprisingly, compound A14 showed opt
