Pentagalloyl glucose from Schinus terebinthifolia inhibits growth of carbapenem-resistant Acinetobacter baumannii.

The rise of antibiotic resistance has necessitated a search for new antimicrobials with potent activity against multidrug-resistant gram-negative pathogens, such as carbapenem-resistant Acinetobacter baumannii (CRAB). In this study, a library of botanical extracts generated from plants used to treat infections in traditional medicine was screened for growth inhibition of CRAB. A crude extract of Schinus terebinthifolia leaves exhibited 80% inhibition at 256 µg/mL and underwent bioassay-guided fractionation, leading to the isolation of pentagalloyl glucose (PGG), a bioactive gallotannin. Biology of Acinetobacter baumannii: Pathogenesis, Antibiotic Resistance Mechanisms, and Prospective Treatment Options.

Acinetobacter baumannii is undoubtedly one of the most successful pathogens responsible for hospital-acquired nosocomial infections in the modern healthcare system. Due to the prevalence of infections and outbreaks caused by multi-drug resistant A. baumannii, few antibiotics are effective for treating infections caused by this pathogen. To overcome this problem, knowledge of the pathogenesis and antibiotic resistance mechanisms of A. baumannii is important. In this review, we summarize current studies on the virulence factors that contribute to A. baumannii pathogenesis, including porins, caps

Treatment options for carbapenem-resistant and extensively drug-resistant Acinetobacter baumannii infections.

Acinetobacter baumannii is a leading cause of healthcare-associated infections worldwide. Because of various intrinsic and acquired mechanisms of resistance, most β-lactam agents are not effective against many strains, and carbapenems have played an important role in therapy. Recent trends show many infections are caused by carbapenem-resistant or even extensively drug-resistant (XDR) strains, for which effective therapy is not well established. Evidence to date suggests that colistin constitutes the backbone of therapy, but the unique pharmacokinetic properties of colistin have led many to su

Novel therapies for treatment of multi-drug resistant Acinetobacter baumannii skin infections.

Improved biomarkers are needed to enhance prognostication in kidney transplantation. We evaluated urinary Epidermal Growth Factor (uEGF) as a predictor of long-term allograft loss. We conducted a prospective, single-center cohort study of 290 adult kidney transplant recipients with uEGF measured 3 months post-transplant. The primary outcome was allograft loss, defined as return to dialysis or pre-emptive re-transplantation. Multivariable cause-specific Cox models assessed the independent association between uEGF and allograft loss. Model performance was compared to the iBox prediction model us

Comparative genomic insight into Acinetobacter baumannii and Acinetobacter nosocomialis: uncovers the functional basis of multidrug resistance and biofilm-related virulence.

Acinetobacter species are opportunistic pathogens that possess both intrinsic and acquired drug resistance, as well as the capacity to develop persistent biofilms. Among them, the most important multidrug-resistant species are Acinetobacter baumannii and Acinetobacter nosocomialis. The in silico comparative genomic study has systematically analyzed these two species with emphasis on antimicrobial resistance genes and virulence factors that promote biofilm formation. Both species contain several biofilm-associated genes, such as ompA, pilE, csuA/b, pgaA-D, and the AdeFGH efflux system, which ar

In silico design of a multiepitope vaccine against antibiotic drug-resistant Acinetobacter baumannii.

Acinetobacter baumannii, a member of the ESKAPE pathogens, causes approximately 722,000 infections annually and is notorious for rapidly developing resistance to multiple antibiotics, resulting in poor clinical outcomes. Due to the rapid emergence of drug-resistant strains and limited effective treatments, the World Health Organization has listed A. baumannii as a top-priority pathogen for vaccine development, highlighting the urgent need for novel therapeutic strategies. In this study, we employed an in silico approach to design a multiepitope vaccine (MEV) targeting multidrug-resistant strai

Synergetic attenuated inflammatory response and increased apoptosis confer high pathogenicity of Acinetobacter baumannii.

Acinetobacter baumannii (A. baumannii) has emerged as a major global healthcare threat, ranking among the leading causes of both hospital-acquired and community-acquired infections. Its extensive antibiotic resistance complicates treatment, underscoring the need for therapeutic targets that elicit robust immune responses. This study identified two multidrug-resistant clinical strains of A. baumannii A.ba-R (rough) and A.ba-S (smooth). A.ba-R has thick and dense capsular polysaccharide, and exhibits high pathogenicity by suppressing host inflammatory responses and inducing apoptosis to evade im

Acinetobacter baumannii: Mechanisms of antibiotic resistance, quorum sensing regulation, and current therapeutic strategies.

Acinetobacter baumannii has emerged as a major global pathogen due to extensive resistance to last-resort antimicrobials, a high burden of nosocomial infections, and increasing community-acquired cases. Its adaptability is driven by diverse resistance mechanisms, including β-lactamase production, aminoglycoside-modifying enzymes, efflux pump overexpression, target-site mutations, and lipid A remodeling, all of which limit treatment options and worsen clinical outcomes. Pathogenicity is further enhanced by quorum-sensing systems, particularly AbaI/AbaR, which regulate biofilm formation, virulen
