The toxin BjussuLAAO-II induces oxidative stress and DNA damage, upregulates the inflammatory cytokine genes TNF and IL6, and downregulates the apoptotic-related genes BAX, BCL2 and RELA in human Caco-2 cells.

Colorectal carcinoma is one of the most common cancers in adults. As chemotherapy, the first-choice treatment for colorectal carcinoma, is often infeasible due to acquired tumor resistance and several adverse effects, it is important to discover and explore new molecules with better therapeutic action. Snake venom toxins have shown promising results with high cytotoxicity against tumor cells, but their mechanisms of action remain unclear. Here we examined how BjussuLAAO-II, an L-amino acid oxidase isolated from Bothrops jararacussu snake venom, exerts cytotoxicity towards colorectal adenocarci

Role of phospholipase Cgamma-induced activation of protein kinase Cepsilon (PKCepsilon) and PKCbetaI in epidermal growth factor-mediated protection of tight junctions from acetaldehyde in Caco-2 cell monolayers.

Epidermal growth factor (EGF) protects the intestinal epithelial tight junctions from acetaldehyde-induced insult. The role of phospholipase Cgamma (PLCgamma) and protein kinase C (PKC) isoforms in the mechanism of EGF-mediated protection of tight junction from acetaldehyde was evaluated in Caco-2 cell monolayers. EGF-mediated prevention of acetaldehyde-induced decrease in transepithelial electrical resistance and an increase in inulin permeability, and subcellular redistribution of occludin and ZO-1 was attenuated by reduced expression of PLCgamma1 by short hairpin RNA. EGF induced a rapid ac

[Investigation of cytokine responses and variations in the expression of beta-defensin-3 of Caco-2 human colon epidermal adenocarsinoma and THP-1 human leukemia monocyte cell lines in response to Toxoplasma gondii under various conditions].

Mononuclear phogocytic cells and epithelial cells are effective during the initiation and regulation of the innate immune response. They have an active role in mucosal immune response both mechanically and by interaction with other cells with cytokine release. Defensins are microbicidal peptides that are expressed in various cells and are thought to be effective in the first line defense against pathogens. IL-12 and IL-10, showing proinflamatory and antiinflamatory activities, respectively, are actors of the cellular immunity and limit the infection of the host without causing immunopathology.

Effect of ECM nanostructures in decellularized small intestine on differentiation of intestinal epithelial model cells.

Caco-2 cells are derived from human colon cancer and have the ability to differentiate into human intestinal epithelial-like cells. The 2D in vitro intestinal model of Caco-2 cells cultured on a semi-permeable membrane is widely used in drug development and the evaluation of absorption functions. However, these intestinal models lack the structural characteristics of the small intestine in vivo, and the cell behavior is not properly controlled. Previous studies have reported that the microstructure of the villi and crypts on a small intestine-mimicking scaffold promotes Caco-2 differentiation;

Retinoic Acid Facilitates Toll-Like Receptor 4 Expression to Improve Intestinal Barrier Function through Retinoic Acid Receptor Beta.

BACKGROUND/AIMS: Vitamin A (VA) protects the intestinal epithelial barrier by improving cell migration and proliferation. Our previous studies demonstrated that VA deficiency (VAD) during pregnancy suppresses the systemic and mucosal immune responses in the intestines of offspring and that VA supplementation (VAS) during early life can increase immune cell counts. However, little is known about the mechanisms by which VA regulates intestinal epithelial barrier function. METHODS: Caco-2 cells were treated with all-trans retinoic acid (ATRA) for 24 hours to determine the optimum ATRA concentrat

Legume lectin phytohemagglutinin reduces transepithelial electrical resistance by counteracting the chaperone function of heat shock protein-70.

Legume lectins represent a broad class of environmental toxicants that bind to cell surface glycoproteins. Raw red kidney beans (RRKB), a widely consumed common source of dietary protein, are rich in the lectin phytohemagglutinin (PHA). Consumption of improperly cooked (which may require overnight presoaking and boiling at least at 100°C for 45 min) red kidney beans causes severe gastrointestinal symptoms. Since the relationship between lectin toxicity and the cellular chaperone machinery remains unknown, the study aimed to determine the effects of heat-denatured PHA on epithelial barrier func

Isoliquiritin alleviates sepsis-induced intestinal barrier dysfunction in mice potentially by promoting autophagy.

Intestinal barrier function protection is essential to ameliorate sepsis. Isoliquiritin (ILQ) is a natural compound with broad biological activities. However, whether ILQ attenuates sepsis-induced intestinal damage remains unclear. An LPS-induced sepsis mouse model was established. ILQ inhibited body weight loss in sepsis mice. ILQ enhanced mouse intestinal function, as evidenced by decreased serum LDH levels and increased citrulline levels. Alcian blue staining showed that ILQ increased goblet cells in sepsis mouse ileum. ILQ increased the MUC2 RNA levels and decreased the claudin-2 protein e

Listeria monocytogenes uses Listeria adhesion protein (LAP) to promote bacterial transepithelial translocation and induces expression of LAP receptor Hsp60.

Listeria monocytogenes interaction with the intestinal epithelium is a key step in the infection process. We demonstrated that Listeria adhesion protein (LAP) promotes adhesion to intestinal epithelial cells and facilitates extraintestinal dissemination in vivo. The LAP receptor is a stress response protein, Hsp60, but the precise role for the LAP-Hsp60 interaction during Listeria infection is unknown. Here we investigated the influence of physiological stressors and Listeria infection on host Hsp60 expression and LAP-mediated bacterial adhesion, invasion, and transepithelial translocation in

Cellular level lipidomics of two-dimensional cultures of adherent gut epithelial cell lines confirms a metabolic switch.

In vitro cell models of the gut epithelium, particularly those based on the Caco-2 and HT29-MTX cell lines, play an important role in studying the uptake and metabolism of nutrients and pharmaceuticals. Previous studies using mass spectrometry imaging have shown a distinctive lipidome signature for these cells, alone and in coculture, although only limited information on lipid identities was obtained. A novel method employing limited proteolysis for sampling live, adherent cells using an automated capillary extraction workflow was developed which achieved single-cell sampling of Caco-2 cells a

Selective blocking of clathrin-mediated endocytosis by RNA interference: epsin as target protein.

Epsin is an essential accessory protein exclusively implicated in clathrin-mediated endocytosis and therefore an ideal target to study involvement of this entry route in the uptake of bioligands. The technique of RNA interference (RNAi) was exploited to generate a cell line constitutively silencing epsin expression in a sequence-specific manner In these Caco-2(eps-) cells, quantitative reverse transcription PCR (RT-PCR) revealed a severe depletion of the epsin messenger RNA (mRNA) level in cells, reaching a factor > 10(6). The reduction at the mRNA level in the Caco-2(eps-) cells was parallele
