Antiangiogenesis efficacy of nitric oxide donors.

Angiogenesis is a complex process involving endothelial cell migration, proliferation, invasion, and tube formation. Inhibition of these processes might have implications in various angiogenesis-mediated disorders. Because nitric oxide (NO) is known to play a key role in various vascular diseases, the present study was undertaken to determine the role of NO in angiogenesis-mediated processes using the NO donor, S-nitroso N-acetyl penicillamine (SNAP) and S-nitroso N-acetyl glutathione (SNAG). The antiangiogenic efficacy of these NO donors was examined using in vivo and in vitro model systems.

Synaptic changes in the terminals of rod photoreceptors of albino mice after partial visual cell loss induced by brief exposure to constant light.

Albino mice were exposed to constant light for 7 days and were then transferred to periodic light. After initial photic damage and partial cell loss, the remaining visual cells recovered and survived as a stable population. Regions of the outer nuclear layer containing 4-6 rows of nuclei were more affected than those containing 6-10 rows. Changes in the synaptic structures in the receptor terminals of these two regions were recorded after varying survival periods. Some of the rod terminals had multiple synaptic ribbons and larger numbers of horizontal cell processes and bipolar cell dendrites.

Transforming growth factor-beta pathway serves as a primary tumor suppressor in CD8+ T cell tumorigenesis.

Tumorigenesis in rodents, as well as in humans, has been shown to be a multistep process, with each step reflecting an altered gene product or gene regulatory process leading to autonomy of cell growth. Initial genetic mutations are often associated with dysfunctional growth regulation, as is demonstrated in several transgenic mouse models. These changes are often followed by alterations in tumor suppressor gene function, allowing unchecked cell cycle progression and, by genomic instability, additional genetic mutations responsible for tumor metastasis. Here we show that reduced transforming g

Inhibition of PKHD1 may cause S-phase entry via mTOR signaling pathway.

Autosomal recessive polycystic kidney disease (ARPKD) is an important genetic disorder in pediatric nephrological practice. Mutation of the polycystic kidney and hepatic disease gene 1 (PKHD1) was identified as the cause of ARPKD. Rapamycin is a highly specific inhibitor of mammalian target of rapamycin (mTOR). Rapamycin exerts its biological activity by inhibiting the serine-threonine kinase mTOR, which regulates important cellular processes such as control of cell cycle, cell size, translation initiation and transcription. The ability of rapamycin to cause G1-cell cycle arrest, reduced cell

Heterocellular N-cadherin junctions enable nontransformed cells to inhibit the growth of adjacent transformed cells.

BACKGROUND: The Src tyrosine kinase phosphorylates effector proteins to induce expression of the podoplanin (PDPN) receptor in order to promote tumor progression. However, nontransformed cells can normalize the growth and morphology of neighboring transformed cells. Transformed cells must escape this process, called "contact normalization", to become invasive and malignant. Contact normalization requires junctional communication between transformed and nontransformed cells. However, specific junctions that mediate this process have not been defined. This study aimed to identify junctional prot

Tissue-specific ablation of Tsc1 in pancreatic beta-cells.

Tuberous sclerosis complex 1 (TSC1) is a tumor suppressor that associates with TSC2 to inactivate Rheb, thereby inhibiting signaling by the mammalian target of rapamycin (mTOR) complex 1 (mTORC1). mTORC1 stimulates cell growth by promoting anabolic cellular processes, such as translation, in response to growth factors and nutrient signals. In order to test roles for TSC1 and mTORC1 in β-cell function, we utilized Rip2/Cre to generate mice lacking Tsc1 in pancreatic β cells (Rip-Tsc1cKO mice). While obesity developed due to hypothalamic Tsc1 excision in older Rip-Tsc1cKO animals, young animals

Giant-cell tumors and aneurysmal bone cysts of ribs in childhood.

Cystic or solid lesions of ribs containing giant cells are rarely seen in children. Three cases are presented which are uncommon with regard to the age of the patients (5-14 yr old) as well as the site. These lesions were diagnosed late and exclusively by X-ray. Radiographically it was impossible to distinguish between a giant cell tumor and an aneurysmal bone cyst. Although the distinction of aneurysmal bone cysts and giant cell tumors sensu strictiori is justified from a clinical point of view, the two may be variants of the same pathologic growth process, i.e. growth of angiogribous tissue

Age distribution of inoculated cells and stochasticity of inter-division times generate pseudo-lag in population growth.

Population-level growth parameters, such as specific growth rate (or doubling time) and lag time, can be inferred directly from the distribution of the inter-division times of single cells. Applying stochastic branching process theory to the multiplication of bacterial cells, we demonstrate that a) the lag time, or time-shift, detectable at population level is primarily determined by the (random) time-to-first-division of the initial cells and b) exponentially growing populations converge to a stable age distribution characterized by a ratio of approximately 2:1 between the number of young and

Splicing as an integrative layer linking cell cycle regulation, epigenetic dynamics, and environmental responses in plants.

Plants continuously adjust growth and development in response to environmental fluctuations through mechanisms that coordinate gene expression and cell cycle progression. Advances in our understanding of these regulatory pathways support a model in which they function as interconnected layers that fine-tune cell cycle progression according to developmental and environmental signals. Alternative splicing (AS) has emerged as a major regulatory layer in these processes, generating transcript diversity and modulating the activity of genes involved in developmental transitions and stress responses.

Proliferation of bovine endometrial epithelial cells is promoted by prostaglandin E2-PTGER2 signaling through cell cycle regulation.

It is known that prostaglandin E2 (PGE2) induces proliferation of epithelia in bovine endometrial explants, however, the detailed mechanism of regulation of PGE2 in inducing bovine endometrial epithelial cell (bEEC) proliferation is unclear. In this study, we determined whether proliferation of bEECs is promoted by PGE2-prostaglandin E receptor 2 (PTGER2) signaling activation through cell cycle regulation. The results demonstrated that bEECs proliferation was induced by treatment of PGE2 and PTGER2 agonist butaprost. These processes were down-regulated by PTGER2 antagonist AH6809 and CDK inhib

Engineering self-flocculating Halomonas campaniensis for wastewaterless open and continuous fermentation.

Halomonas has been developed as a platform for the next generation industrial biotechnology allowing open and nonsterile growth without microbial contamination under a high-salt concentration and alkali pH. To reduce downstream cost associated with continuous centrifugation and salt containing wastewater treatment, Halomonas campaniensis strain LS21 was engineered to become self-flocculating by knocking out an etf operon encoding two subunits of an electron transferring flavoprotein in the predicted electron transfer chain. Self-flocculation could be attributed to the decrease of the surface c

Functional characterization of human iPSC-derived sensory neuron processes in microfluidic cultures.

Rodent cell culture models have long underpinned research into nociceptive signaling; however, their limited recapitulation of human nociceptor biology has created a translational gap in analgesic development. While primary human sensory neurons are relevant, their use is hampered by scarcity, ethical constraints, donor variability, and difficulties in long-term culture. Furthermore, conventional cultures lack the compartmentalization needed to study sensory neuron processes and fail to recapitulate the synaptic connectivity between sensory and spinal cord neurons, limiting their translational

TGF-β Signaling in Cellular Senescence and Aging-Related Pathology.

Aging is broadly defined as the functional decline that occurs in all body systems. The accumulation of senescent cells is considered a hallmark of aging and thought to contribute to the aging pathologies. Transforming growth factor-β (TGF-β) is a pleiotropic cytokine that regulates a myriad of cellular processes and has important roles in embryonic development, physiological tissue homeostasis, and various pathological conditions. TGF-β exerts potent growth inhibitory activities in various cell types, and multiple growth regulatory mechanisms have reportedly been linked to the phenotypes of c

[Clinical and morphological features of Merkel's carcinoma in elderly patients (literature review).].

Merkels cell carcinoma (MCC) is a rare and aggressive neuroendocrine skin tumor with a high rate of recurrence and local metastasis. The average age at which MCC is diagnosed is 75 years. The identification of the role of somatic mutations and DNA repair in aging mechanisms raises the question of their direct connection with the processes of differentiation, age-related characteristics of the immune system, programmed cell death and malignant growth. In this regard, there remains a need for research aimed at improving the early diagnosis and pathogenetic treatment of elderly patients.

The role of hepatocyte growth factor/c-met interactions in the immune system.

Hepatocyte growth factor (HGF) is a pleiotropic cytokine with mitogenic, motogenic and morphogenic activity for mainly epithelial and endothelial target cells. All the different effects of HGF are mediated through its specific receptor met, a heterodimeric transmembrane tyrosine kinase. The broad activity of HGF and its impact on many physiologic and pathologic processes are reflected by met expression in a variety of organs and cell types. This paper discusses expression of HGF and c-met within the immune system and their interactions in the control of immune cell functions.

The role of growth factors in peripheral nerve regeneration and opportunities for next-generation biological therapeutics.

Regeneration after peripheral nerve injury (PNI) involves complex processes that are often insufficient to facilitate full functional recovery. In particular, PNI stimulates the release of a multitude of growth factors with critical spatial and temporal specificity to aid in regeneration by affecting axons, glia, immune cells, and vasculature. Depending on the severity of the injury, however, these endogenous processes may be inadequate to support regeneration, leaving patients with life-long sensory and motor deficits. Current treatments in these cases attempt to modify the regenerative envir

Role of growth factors and their receptors in gastric ulcer healing.

The repair of gastric ulcers requires the reconstitution of epithelial structures and the underlying connective tissue, including vessels and muscle layers. Several growth factors have been implicated in this process, since they are able to regulate important cell functions, such as cell proliferation, migration, differentiation, secretion, and degradation of extracellular matrix, all of which are essential during tissue healing. Epidermal growth factor (EGF), transforming growth factor-alpha (TGF-alpha), hepatocyte growth factor (HGF), and trefoil factors (TFFs) are mainly involved in the rec

Tuberous sclerosis--A model for tumour growth.

Tuberous sclerosis complex (TSC) is a rare genetic disorder where patients develop benign tumours in several organ systems. Central to TSC pathology is hyper-activation of the mammalian target of rapamycin complex 1 (mTORC1) signalling pathway, which is a key controller of cell growth. As a result, TSC model systems are a valuable tool for examining mTORC1-driven cellular processes. The immunosuppressant, rapamycin, is a specific inhibitor of mTORC1 and has shown promise as a therapeutic agent in TSC as well as in malignancy. This review will focus on the cellular processes controlled by mTORC
