Comment on "Declining wild salmon populations in relation to parasites from farm salmon".

Krkosek et al. (Reports, 14 December 2007, p. 1772) claimed that sea lice spread from salmon farms placed wild pink salmon populations "on a trajectory toward rapid local extinction." Their prediction is inconsistent with observed pink salmon returns and overstates the risks from sea lice and salmon farming.

Lepeophtheirus salmonis secretory/excretory products and their effects on Atlantic salmon immune gene regulation.

We have previously shown that Lepeophtheirus salmonis produces trypsin and prostaglandin E(2) (PGE(2)) that are most likely responsible for the limited inflammatory response of Atlantic salmon to infection. After removal of the dopamine and PGE(2), the immunomodulatory activity of unfractionated and pools of the fractionated secretions was determined by examining the effects of the secretions on Atlantic salmon immune gene expression. Incubation of macrophage-enriched isolates of Atlantic salmon head kidney cells with the unfractionated secretion + PGE(2) revealed a significant inhibition of i

Molecular characterization of Lamproglena monodi infesting Oreochromis niloticus in Egypt based on 18S and 28S rDNA markers.

This study investigated the molecular, morphological, and pathological characteristics of the parasitic copepod Lamproglena monodi infesting Oreochromis niloticus in Egyptian aquaculture. A total of 273 tilapia were examined from three governorates, with infestation frequencies of 25% in Kafr El-Sheikh, 20% in Giza, and 21.7% in Al-Sharqia. Seasonal frequencies ranged from 13% in summer to 34.3% in autumn. Molecular amplification produced fragments of 1405 bp (18S rDNA) and 737 bp (28S rDNA), and BLAST analysis confirmed their identity as L. monodi with ≥98% similarity to reference sequences.

Environmental Changes Drives the Transcriptome and Gene Regulation Plasticity During Sea Lice Infestation.

The sea louse, Caligus rogercresseyi, is one of the main concerns in the Chilean salmon industry. The free-living copepodid stage can recognize the host and initiate the parasitic phase, where environmental factors can shape the host recognition process. This study aimed to explore the ecological influence on the transcriptome of copepodids infesting Atlantic salmon experimentally exposed to different salinity and temperature (S/T) conditions. Herein, 200 salmon were infested with 35 copepodids per fish previously acclimatized to four S/T treatments: 32 and 26 PSU; 8 and 16°C. After 48 h of in

QTL mapping provides new insights into emamectin benzoate resistance in salmon lice, Lepeophtheirus salmonis.

BACKGROUND: The salmon louse (Lepeophtheirus salmonis) is a parasite of wild and farmed salmonid fish, causing huge economic damage to the commercial farming of Atlantic salmon (Salmo salar) in the northern hemisphere. The avermectin emamectin benzoate (EMB) is widely used for salmon delousing. While resistance to EMB is widespread in Atlantic populations of L. salmonis, the molecular mechanisms of resistance remain to be elucidated. The aim of the present work was to obtain insights into potential EMB resistance mechanisms by identifying genetic and transcriptomic markers associated with EMB

Candidate genes for monitoring hydrogen peroxide resistance in the salmon louse, Lepeophtheirus salmonis.

BACKGROUND: Hydrogen peroxide (H2O2) is one of the delousing agents used to control sea lice infestations in salmonid aquaculture. However, some Lepeophtheirus salmonis populations have developed resistance towards H2O2. An increased gene expression and activity of catalase, an enzyme that breaks down H2O2, have been detected in resistant lice, being therefore introduced as a resistance marker in the salmon industry. In the present study the aim was to validate the use of catalase expression as a marker and to identify new candidate genes as additional markers to catalase, related to H2O2 resi

The Atlantic salmon (Salmo salar) antimicrobial peptide cathelicidin-2 is a molecular host-associated cue for the salmon louse (Lepeophtheirus salmonis).

Chemical signals are a key element of host-parasite interactions. In marine ecosystems, obligate ectoparasites, such as sea lice, use chemical cues and other sensory signals to increase the probability of encountering a host and to identify appropriate hosts on which they depend to complete their life cycle. The chemical compounds that underlie host identification by the sea lice are not fully described or characterized. Here, we report a novel compound - the Atlantic salmon (Salmo salar) antimicrobial peptide cathelicidin-2 (Cath-2) - that acts as an activation cue for the marine parasitic co

Sensitivity assessment of sea lice to chemotherapeutants: Current bioassays and best practices.

Traditional bioassays are still necessary to test sensitivity of sea lice species to chemotherapeutants, but the methodology applied by the different scientists has varied over time in respect to that proposed in "Sea lice resistance to chemotherapeutants: A handbook in resistance management" (2006). These divergences motivated the organization of a workshop during the Sea Lice 2016 conference "Standardization of traditional bioassay process by sharing best practices." There was an agreement by the attendants to update the handbook. The objective of this article is to provide a baseline analys

Comparative immunity of Salmo salar and Oncorhynchus kisutch during infestation with the sea louse Caligus rogercresseyi: An enrichment transcriptome analysis.

Caligus rogercresseyi, an ectoparasite affecting the Chilean salmon industry, can cause immunosuppression and physiological stress in farmed fish. Interestingly, coho salmon (Oncorhynchus kisutch) are notably resistant to infestation, whereas Atlantic salmon (Salmo salar) are phenotypically more susceptible to sea lice. However, comparative studies on immune responses to C. rogercresseyi have not been conducted. In this study, Illumina sequencing was conducted to evaluate head kidney and skin samples taken 7 and 14 days post-infestation, yielding a total of 1492 and 1522 contigs annotated to i

Toxicity of seaweed-synthesized silver nanoparticles against the filariasis vector Culex quinquefasciatus and its impact on predation efficiency of the cyclopoid crustacean Mesocyclops longisetus.

Nearly 1.4 billion people in 73 countries worldwide are threatened by lymphatic filariasis, a parasitic infection that leads to a disease commonly known as elephantiasis. Filariasis is vectored by mosquitoes, with special reference to the genus Culex. The main control tool against mosquito larvae is represented by treatments with organophosphates and insect growth regulators, with negative effects on human health and the environment. Recently, green-synthesized nanoparticles have been proposed as highly effective larvicidals against mosquito vectors. In this research, we attempted a reply to t

Toxicological assessment of the anti-salmon lice drug diflubenzuron on Atlantic cod Gadus morhua.

Increasing use of the chitin synthesis inhibitor diflubenzuron against the ectoparasitic salmon louse Lepeophtheirus salmonis in marine aquaculture has raised concerns over its environmental impacts. This study evaluated how diflubenzuron affects Atlantic cod Gadus morhua, a fish species often found near Atlantic salmon Salmo salar farms, focusing on uptake kinetics and hepatic transcriptional responses. Two experiments were conducted, one time-series trial in which the fish were given a daily dose (3 mg kg-1 fish) of diflubenzuron for 14 d followed by a 3 wk depuration period, and one dose-re

Interactions between benthic predators and zooplanktonic prey are affected by turbulent waves.

Predators capture prey in complex and variable environments. In the ocean, bottom-dwelling (benthic) organisms are subjected to water currents, waves, and turbulent eddies. For benthic predators that feed on small animals carried in the water (zooplankton), flow not only delivers prey, but can also shape predator-prey interactions. Benthic passive suspension feeders collect prey delivered by movement of ambient water onto capture-surfaces, whereas motile benthic predators, such as burrow-dwelling fish, dart out to catch passing zooplankton. How does the flow of ambient water affect these contr

Sea lice, Lepeophtheirus salmonis, transfer between wild sympatric adult and juvenile salmon on the north coast of British Columbia, Canada.

We examine sea lice, Lepeophtheirus salmonis, on juvenile and adult salmon from the north coast of British Columbia between 2004 and 2006 in an area that does not at present contain salmon farms. There is a pronounced zonation in the abundance of L. salmonis on juvenile pink salmon, Oncorhynchus gorbuscha, in the Skeena and Nass estuaries. Abundances in the proximal and distal zones of these estuaries are 0.01 and 0.05 respectively. The outer zones serve as feeding and staging areas for the pink salmon smolts. An epizootic of Caligus chiastos on farmed southern bluefin tuna Thunnus maccoyii off South Australia.

In some years, large numbers of Caligus chiastos have been observed on the external surfaces of southern bluefin tuna, particularly on the head and eyes, in some sea cages in Spencer Gulf, Australia. As no epidemiological data were available, we monitored sea lice on tuna (N = 130) in 4 research cages sampled at 6 wk intervals during the 2005 farming season. No lice were observed on a sample of 10 wild-caught tuna when the cohort was transferred to cages in early April. By late May more than half the sampled tuna (22 of 40) were infected, with up to 42 parasites; we also recorded one unidentif

Biogeographic provinces of total and methyl mercury in zooplankton and fish from the Beaufort and Chukchi seas: results from the SHEBA drift.

Samples of copepods (Calanus hyperboreus) and arctic cod (Boreogadus saida) were collected along the SHEBA (Surface HEat Budget of the Arctic) drift track, which commenced in the Canada Basin (October 1997) and finished in the Mendeleev Basin (October 1998). Here, we report total mercury (HgT) and CH3Hg concentrations in these biological samples and examine concentration variability along the drift track in the context of trophic variation, inputs from land, spring mercury depletion events (MDEs), and oceanographic provinces. We find background concentrations of HgT in C. hyperboreus as low as

Sea surface freshening can suppress the thermal tipping point of marine copepods.

Tipping points govern species distributions, which could be impacted by ocean warming (OW) and sea surface freshening (SSF). Interactions between these stressors may affect tipping points. Also, the accumulation of stress over time (degree day) could condition the reaching of tipping points. Yet, these aspects have never been studied. Here, we hypothesize that SSF reduces the thermal tipping point of Calanus spp., key copepods that play a structuring role in marine food webs. Calanus spp. from the Oslofjord were exposed to a temperature gradient from 8 to 20 °C and a salinity range from 35 to

In Atlantic salmon skin infested with salmon lice, elevated seawater temperatures change gene expression and mucus glycosylation, which promotes pathogen binding.

Skin barrier function is paramount for fish health and is likely affected by the predicted increases in seawater temperature. Salmonid skin produces a mucus layer mainly composed of mucins. Mucin glycans regulate interactions with pathogens, including binding to host cells, quorum sensing and regulation of virulence genes. In this work, the objective was to elucidate the Atlantic salmon (Salmo salar) skin mucosal responses to temperature in the presence of salmon lice (Lepeophtheirus salmonis) to mimic salmon louse pressure at sea. A simultaneous louse and temperature challenge trial was perfo

Catching the complexity of salmon-louse interactions.

The study of host-parasite relationships is an integral part of the immunology of aquatic species, where the complexity of both organisms has to be overlayed with the lifecycle stages of the parasite and immunological status of the host. A deep understanding of how the parasite survives in its host and how they display molecular mechanisms to face the immune system can be applied for novel parasite control strategies. This review highlights current knowledge about salmon and sea louse, two key aquatic animals for aquaculture research worldwide. With the aim to catch the complexity of the salmo

Assisted natural recovery of hypersaline sediments: salinity thresholds for the establishment of a community of bioturbating organisms.

Hypersaline sediments derived from poor land management or the decommissioning of large-scale salt production contribute to the long-term degradation of aquatic environments. Obstacles impeding remediation of these environments include salt crusts restricting benthic recolonisation, hypersalinity-induced toxicity to organisms, and disruption of biogeochemical cycles. Remediation often focuses on engineered solutions, despite sediment-biota interactions often playing a crucial role in improving long-term remediation and restoration of contaminated areas. The presence of extensive bioturbating c
