Daytime sleepiness is associated with decreased default mode network connectivity in both young and cognitively intact elderly subjects.

STUDY OBJECTIVES: Sleep deprivation and daytime somnolence impair numerous aspects of physical, cognitive, and memory performance. However, most studies examining the effect of somnolence on brain function focus on acute sleep restriction in young adults. We examine the relationship between chronic daytime somnolence and connectivity in six brain networks in both young and elderly subjects using stimulus-free resting-state functional magnetic resonance imaging. DESIGN: Cross-sectional. SETTING: Outpatient research at the Massachusetts General Hospital. PARTICIPANTS: Young (n = 27) and elder

Regional homogeneity within the default mode network in bipolar depression: a resting-state functional magnetic resonance imaging study.

AIM: We sought to use a regional homogeneity (ReHo) approach as an index in resting-state functional magnetic resonance imaging (fMRI) to investigate the features of spontaneous brain activity within the default mode network (DMN) in patients suffering from bipolar depression (BD). METHODS: Twenty-six patients with BD and 26 gender-, age-, and education-matched healthy subjects participated in the resting-state fMRI scans. We compared the differences in ReHo between the two groups within the DMN and investigated the relationships between sex, age, years of education, disease duration, the Ham

Roles of default-mode network and supplementary motor area in human vigilance performance: evidence from real-time fMRI.

We used real-time functional magnetic resonance imaging (fMRI) to determine which regions of the human brain have a role in vigilance as measured by reaction time (RT) to variably timed stimuli. We first identified brain regions where activation before stimulus presentation predicted RT. Slower RT was preceded by greater activation in the default-mode network, including lateral parietal, precuneus, and medial prefrontal cortices; faster RT was preceded by greater activation in the supplementary motor area (SMA). We examined the roles of these brain regions in vigilance by triggering trials bas

Making new connections: an fNIRS machine learning classification study of inter-brain synchrony in the default mode network.

Little is known about the neurocognitive underpinnings of forming social connections. In this study, we tested whether default mode network (DMN) inter-brain synchrony (which prior work has linked to interpersonal alignment, or "seeing eye-to-eye") predicts feelings of connection during conversations. Seventy pairs of strangers engaged in shallow or deep conversations while brain activity was measured with functional near-infrared spectroscopy (fNIRS). Stranger dyads in the deep condition felt more connected than those in the shallow condition. Greater feelings of connection were associated wi

Default Mode Network Resting State Connectivity Derived From Task-Based fMRI: A Validation Study in People With Epilepsy.

BACKGROUND AND PURPOSE: Resting state functional connectivity can be measured using resting state functional MRI (fMRI), but also task-dependent fMRI in blocked designs. The latter has been demonstrated in healthy participants but not yet validated in clinical cohorts. Since functional connectivity of resting state networks (e.g., default mode network [DMN] and somatomotor network [SMN]) is altered in people with epilepsy, and the impact of the disease on the quality of the intermittent resting state data is unclear, we aimed to validate the method using a clinical fMRI in people with epilepsy

APOE ε4-related structural vulnerability in mild cognitive impairment: a subsystem-based analysis of the default mode network.

BACKGROUND: Mild cognitive impairment (MCI) is a critical prodromal stage of progressive cognitive decline. As a core cognitive brain network, the default mode network (DMN) exhibits structural alterations in MCI that are closely associated with cognitive impairment. Apolipoprotein E ε4 (APOE ε4), the strongest genetic risk factor for pathological cognitive decline, has been shown to disrupt the structural integrity of the DMN's three core subsystems: the Midline Core, Medial Temporal Lobe (MTL), and Dorsomedial Prefrontal Cortex (DMPFC) subsystems. However, the subsystem-specific effects of A

Potential correlations between abnormal homogeneity of default mode network and personality or lipid level in major depressive disorder.

BACKGROUND: Default mode network (DMN) is one of the most recognized resting-state networks in major depressive disorder (MDD). However, the homogeneity of this network in MDD remains incompletely explored. Therefore, this study aims to determine whether there is abnormal network homogeneity (NH) of the DMN in MDD patients. At the same time, correlations between clinical variables and brain functional connectivity are examined. METHODS: We enrolled 42 patients diagnosed with MDD and 42 HCs. A variety of clinical variables were collected, and data analysis was conducted using the NH and indepe

Reduced Functional Connectivity in the Default Mode Network in EEGs without Other Abnormalities in Early Creutzfeldt-Jacob Disease.

INTRODUCTION: Creutzfeldt-Jakob disease (CJD) is a rare, neurodegenerative disorder that is incurable, always fatal, and transmissible. EEG is an integral part of the diagnostic workup with typical periodic sharp wave complexes indicative of CJD, but early in the disease EEG is often unaltered. Accordingly, we aimed at evaluating disruption of brain network functional connectivity (FC) in regions belonging to the default mode network (DMN) as a potential early marker in CJD when EEG is considered visually normal. METHODS: EEGs considered visually normal obtained from 7 CJD patients were compa

Altered longitudinal trajectory of default mode network connectivity in healthy youth with subclinical depressive and posttraumatic stress symptoms.

The default mode network (DMN) plays a crucial role in internal self-processing, rumination, and social functions. Disruptions to DMN connectivity have been linked with early adversity and the emergence of psychopathology in adolescence and early adulthood. Herein, we investigate how subclinical psychiatric symptoms can impact DMN functional connectivity during the pubertal transition. Resting-state fMRI data were collected annually from 190 typically-developing youth (9-15 years-old) at three timepoints and within-network DMN connectivity was computed. We used latent growth curve modeling to

Functional connectivity of the default mode network predicts subsequent polysomnographically measured sleep in people with symptoms of insomnia.

Insomnia is often accompanied by excessive pre-sleep rumination. Such ruminative thinking is also associated with increased connectivity of the default mode network (DMN). It is likely that DMN connectivity and associated rumination contribute to the pathogenesis of insomnia. We hypothesized that resting state functional connectivity (rsFC) between the DMN and other brain regions prior to bedtime would predict objectively measured sleep among individuals with insomnia. Twenty participants (12 female; M age = 26.9, SD = 6.6 years) with symptoms of insomnia underwent an rsFC scan in the early ev

Anterior default mode network and posterior insular connectivity is predictive of depressive symptom reduction following serial ketamine infusion.

BACKGROUND: Ketamine is a rapidly-acting antidepressant treatment with robust response rates. Previous studies have reported that serial ketamine therapy modulates resting state functional connectivity in several large-scale networks, though it remains unknown whether variations in brain structure, function, and connectivity impact subsequent treatment success. We used a data-driven approach to determine whether pretreatment multimodal neuroimaging measures predict changes along symptom dimensions of depression following serial ketamine infusion. METHODS: Patients with depression (n = 60) rec

Parietal-hippocampal rTMS improves cognitive function in Alzheimer's disease and increases dynamic functional connectivity of default mode network.

Parietal-hippocampal repetitive transcranial magnetic stimulation (rTMS) improves cognitive function in Alzheimer's disease (AD), however, the underlying therapeutic mechanism has not been elucidated. A double-blind, randomized, sham-controlled parietal-hippocampal rTMS trial (five sessions/week for a total of 10 sessions) of mild-to-moderate AD patients was conducted in the study. High-frequency rTMS was applied to a subject-specific left lateral parietal region with the highest functional connectivity with the hippocampus based on resting-state fMRI. A multimodal MRI scan and a complete neur

Default-Mode Network Connectivity Changes During the Progression Toward Alzheimer's Dementia: A Longitudinal Functional Magnetic Resonance Imaging Study.

Background/Purpose: Brain function changes with Alzheimer's disease (AD) progression. Evaluating those changes longitudinally is important to understand the complex relationships between brain pathologies and cognition. We aimed (1) to identify longitudinal changes in functional connectivity in patients with mild cognitive impairment (MCI) characterized for amyloid-β (Aβ) status and (2) to relate these functional changes to clinical progression. Methods: Forty-four patients with MCI were followed using serial functional magnetic resonance imaging (fMRI) over 1.2 years (three sessions) and cogn

Default Mode Network quantitative diffusion and resting-state functional magnetic resonance imaging correlates in sporadic Creutzfeldt-Jakob disease.

Grey matter involvement is a well-known feature in sporadic Creutzfeldt-Jakob disease (sCJD), yet precise anatomy-based quantification of reduced diffusivity is still not fully understood. Default Mode Network (DMN) areas have been recently demonstrated as selectively involved in sCJD, and functional connectivity has never been investigated in prion diseases. We analyzed the grey matter involvement using a quantitatively multi-parametric MRI approach. Specifically, grey matter mean diffusivity of 37 subjects with sCJD was compared with that of 30 age-matched healthy controls with a group-wise

Reduced functional connectivity of default mode network subsystems in depression: Meta-analytic evidence and relationship with trait rumination.

Resting-state functional connectivity changes in the default mode network (DMN) of patients with major depressive disorder (MDD) have been linked to rumination. The DMN is divided into three subsystems: a midline Core, a dorsal medial prefrontal cortex (DMPFC) subsystem, and a medial temporal lobe (MTL) subsystem. We examined resting-state functional connectivity within and between DMN subsystems in MDD and its association with rumination. First, we conducted a meta-analysis on a large multi-site dataset of 618 MDD and 683 controls to quantify the differences in DMN subsystem functional connec

Anomalous functional connectivity within the default-mode network in treatment-naive patients possessing first-episode major depressive disorder.

BACKGROUND AND OBJECTIVE: Previous studies have shown that the default-mode network (DMN) has a substantial role in patients with major depressive disorder (MDD). However, there is a shortage of information regarding variations in the functional connectivity (FC) of the DMN of treatment-naive patients with first-episode MDD. The present study aims to explore the FC of the DMN in such patients. METHODS: The study population consisted of 33 patients and 35 controls, paired regarding age, gender, education level, and health condition. Depression severity was assessed through the Hamilton Depress

Examining resting-state functional connectivity in key hubs of the default mode network in chronic low back pain.

OBJECTIVES: Changes in brain connectivity have been observed within the default mode network (DMN) in chronic low back pain (CLBP), however the extent of these disruptions and how they may be related to CLBP requires further examination. While studies using seed-based analysis have found disrupted functional connectivity in the medial prefrontal cortex (mPFC), a major hub of the DMN, limited studies have investigated other equally important hubs, such as the posterior cingulate cortex (PCC) in CLBP. METHODS: This preliminary study comprised 12 individuals with CLBP and 12 healthy controls who

Rumination and Default Mode Network Subsystems Connectivity in First-episode, Drug-Naive Young Patients with Major Depressive Disorder.

Neuroimaging evidence implicates the association between rumination and default mode network (DMN) in major depressive disorder (MDD). However, the relationship between rumination and DMN subsystems remains incompletely understood, especially in patients with MDD. Thirty-three first-episode drug-naive patients with MDD and thirty-three healthy controls (HCs) were enrolled and underwent resting-sate fMRI scanning. Functional connectivity analysis was performed based on 11 pre-defined regions of interest (ROIs) for three DMN subsystems: the midline core, dorsal medial prefrontal cortex (dMPFC) a
