PSEN2 Mutation Spectrum and Novel Functionally Validated Mutations in Alzheimer's Disease: Data from PUMCH Dementia Cohort.

BACKGROUND: The established causative mutations in the APP, PSEN1, and PSEN2 can explain less than 1%,Alzheimer's disease (AD) patients. Of the identified variants, the PSEN2 mutations are even less common. OBJECTIVE: With the genetic study from the dementia cohort of Peking Union Medical College Hospital (PUMCH), we aim to illustrate the PSEN2 mutation spectrum and novel functionally validated mutations in Chinese AD patients. METHODS: 702 AD participants, aged 30-85, were identified in PUMCH dementia cohort. They all received history inquiry, physical examination, biochemical test, cogniti

Neurofeedback: A Comprehensive Review on System Design, Methodology and Clinical Applications.

Neurofeedback is a kind of biofeedback, which teaches self-control of brain functions to subjects by measuring brain waves and providing a feedback signal. Neurofeedback usually provides the audio and or video feedback. Positive or negative feedback is produced for desirable or undesirable brain activities, respectively. In this review, we provided clinical and technical information about the following issues: (1) Various neurofeedback treatment protocols i.e. alpha, beta, alpha/theta, delta, gamma, and theta; (2) Different EEG electrode placements i.e. standard recording channels in the front

'The spectrum of vascular dementia'--a retrospective study from South India.

OBJECTIVES: Vascular dementia (VaD) is high in Indian population and is preventable to some extent but there is paucity of literature. Hence we decided to study the clinical and laboratory spectrum of VaD. METHODS: It was retrospective hospital based study. The patients who satisfied Diagnostic and Statistical Manual of Mental Disorders, 4th Edition (DSM-IV criteria) for VaD and age of more than 40 years were identified on the basis of case file code which is uniformly used by the hospital. RESULTS: Of the 83 patients included, 72.3% (60) were males (mean age: 65.3 ± 8.6 years) and 27.3% (23

The multiple faces of valosin-containing protein-associated diseases: inclusion body myopathy with Paget's disease of bone, frontotemporal dementia, and amyotrophic lateral sclerosis.

Inclusion body myopathy associated with Paget's disease of bone and frontotemporal dementia (IBMPFD) is a progressive, fatal genetic disorder with variable penetrance, predominantly affecting three main tissue types: muscle (IBM), bone (PDB), and brain (FTD). IBMPFD is caused by mutations in the ubiquitously expressed valosin-containing protein (VCP) gene, a member of the AAA-ATPase superfamily. The majority of individuals who develop IBM have progressive proximal muscle weakness. Muscle biopsies reveal rimmed vacuoles and inclusions that are ubiquitin- and TAR DNA binding protein-43 (TDP-43)-

Specific Learning Disorders: Variation Analysis of 15 Candidate Genes in 9 Multiplex Families.

Background and Objectives: Specific Learning Disorder (SLD) is a complex neurobiological disorder characterized by a persistent difficult in reading (dyslexia), written expression (dysgraphia), and mathematics (dyscalculia). The hereditary and genetic component is one of the underlying causes of SLD, but the relationship between genes and the environment should be considered. Several genetic studies were performed in different populations to identify causative genes. Materials and Methods: Here, we show the analysis of 9 multiplex families with at least 2 individuals diagnosed with SLD per fam

Lymphomatosis cerebri Presenting as a Recurrent Leukoencephalopathy.

A 29-year-old immunocompetent woman was admitted in 2006 with ataxia, limb weakness, generalized dystonia, and vertical diplopia that developed after a febrile episode. Brain magnetic resonance imaging (MRI) revealed the presence of extensive periventricular white matter lesions that did not enhance after gadolinium injection. As low titers of cytomegalovirus-IgM antibodies were found in the serum, a presumed diagnosis of postviral acute disseminated encephalomyelitis (ADEM) was made, and the patient received a 5-day course of 1 g methylprednisolone. The clinical and radiological outcome was v

[The contribution of neuropsychology to the diagnosis and treatment of learning disorders].

INTRODUCTION: In its broadest sense, the basic concept of Neuropsychology is that all behaviour originates in the brain; according to Luria, it depends on functional systems constituted by different areas of the brain which are inter related. Each area deals with a particular function which is part of the whole (for example, the cortical area for vision deals with reading). Which cortical areas make up the functional system depends on how the person acquired a certain type of behaviour. DEVELOPMENT: The neuropsychological model, when applied to learning disorders, assumes that they are the ex

[Depressive pseudodementia in early Parkinson's disease: lessons from a case with long-term follow-up].

A common observation in neurological practice is parkinsonism with concomitant cognitive decline, an association that usually arises from various underlying degenerative or vascular conditions, most of which are untreatable. An elderly woman with no history of psychiatric disease presented complaining of memory and cognitive impairment serious enough to interfere with daily life activities over the preceding year. She soon developed a predominantly left-sided tremor, rigidity and bradykinesia. She had had only 2 years of formal education. Neuropsychological assessment showed poor performance o

Mapping perceived training gaps in educational neuroscience among Brazilian teachers.

Educational neuroscience has attracted substantial interest from teachers, but professional-development programs are often designed without a formal assessment of the difference between what educators believe they know and what they wish to learn. This secondary analysis of a cross-sectional survey examined perceived training needs in educational neuroscience among 204 active Brazilian teachers. Participants rated, on six-point scales (0-5), their self-perceived knowledge and their desire to learn more about 10 neuroscience-related topics. We defined a training-gap indicator as the within-part

A Cognitive Dimensional Approach to Understanding Shared and Unique Contributions to Reading, Math, and Attention Skills.

Disorders of reading, math, and attention frequently co-occur in children. However, it is not yet clear which cognitive factors contribute to comorbidities among multiple disorders and which uniquely relate to one, especially because they have rarely been studied as a triad. Thus, the present study considers how reading, math, and attention relate to phonological awareness, numerosity, working memory, and processing speed, all implicated as either unique or shared correlates of these disorders. In response to findings that the attributes of all three disorders exist on a continuum rather than

Medical students with specific learning disabilities: mixed-methods systematic review of the prevalence, academic performance, challenges, and perceived impact of support interventions.

BACKGROUND: Specific learning disabilities (SpLDs)-including dyslexia, dysgraphia, dyscalculia, and dyspraxia-affect approximately 5-8% of medical students. The demands of medical education may exacerbate these challenges, while stigma and inconsistent institutional support contribute to ongoing barriers. This mixed-methods systematic review synthesised evidence on prevalence, academic performance, challenges, and support needs to inform more inclusive medical education practices. METHODS: A mixed-methods systematic review was conducted following Joanna Briggs Institute (JBI) methodology and

IDH1-mutant primary intraventricular gliosarcoma: Case report and systematic review of a rare location and molecular profile.

BACKGROUND: Gliosarcoma (GS) is classified as an IDH-wild-type variant of glioblastoma (GBM). While GS is already an unusual presentation of GBM, IDH1-mutant cases are especially rare. We present an IDH1-mutant primary intraventricular GS case report and a systematic review of the molecular profile in GS correlating to the prognostic and pathogenesis of IDH1/2 mutations. CASE DESCRIPTION: A 44-years-old man presented with ongoing fatigue symptoms and a new-onset intense occipital headache. The patient complained of memory loss, dyscalculia, and concentration difficulties. An MRI revealed a bi

Detection of neurodevelopmental diversity in memory clinics-Validation of a self-report measure.

BACKGROUND: Neurodevelopmental learning and attentional disorders (NLAD) such as dyslexia, dyscalculia and attention deficit hyperactivity disorder (ADHD) affect at least 6% of the adult population or more. They are associated with atypical cognitive patterns in early and adult life. The cognitive patterns of affected individuals in late life have never been described. One main challenge is detecting individuals in clinical settings during which mild cognitive changes could be confounding the clinical presentation. This is a critical research gap because these conditions interact, across the l

Cognitive profile of adolescents with math disabilities: are the profiles different from those with reading disabilities?

Adolescents (ages 14-17) with math disabilities (MD, n=12), reading disabilities (RD, n=19), math+reading disabilities (MD+RD, n=12), and average achievers (n=15) were compared on measures of visual-spatial processing, random generation (inhibition), writing speed, short-term memory (STM), and working memory (WM). Adolescents with MD performed significantly lower than adolescents with RD on measures of visual-spatial processing and visual WM. Adolescents with MD outperformed adolescents with RD +MD on measures of random generation and motor speed. Performance of all three low-achieving groups
