Molecular characterization and expression analysis of interleukin 15 (IL15) and interleukin-15 receptor subunit alpha (IL15Rα) in dojo loach (Misgurnus anguillicaudatus): Their salient roles during bacterial, parasitic and fungal infection.

Interleukin 15 (IL15) is a pleiotropic cytokine that participates in innate and adaptive immunity along with its receptor α-chain (IL15Rα). In order to investigate the potential roles of IL15 and IL15Rα in dojo loach (Misgurnus anguillicaudatus), we firstly cloned the cDNA sequence of Ma-IL15 and Ma-IL15Rα, which contain 1096bp and 1236bp and code proteins of 193 amino acids and 210 amino acids, respectively. A short signal peptide and Pfam IL15 domain were found in Ma-IL15, while a highly conserved sushi domain existed in Ma-IL15Rα. Ontogeny analysis indicated that significantly increased exp

Organic Iron Supplementation in Cows and Its Impacts on Animal Health and Production.

This study evaluated whether iron supplementation in the form of chelated minerals in cows in the final third of lactation has a positive effect on iron bioavailability, immunity, oxidative status, milk quality, and biochemical and hematological parameters, as well as production efficiency and fecal microbiota. Twenty-four multiparous Jersey cows, with 210 ± 18 days in milk (DIM), an average production of 25 kg, and 4 ± 0.6 months of gestation, were divided into two groups: Control (n = 12, without supplementation) and Iron (n = 12, supplemented with 30 mg of iron/kg of dry matter (600 mg/anim

Relationship between the dynamics of volatile aroma compounds and microbial succession during the ripening of raw ewe milk-derived Idiazabal cheese.

Cheese microbiota contributes to various biochemical processes that lead to the formation of volatile compounds and the development of flavour during ripening. Nonetheless, the role of these microorganisms in volatile aroma compounds production is little understood. This work reports for the first time the dynamics and odour impact of volatile compounds, and their relationship to microbial shifts during the ripening of a raw ewe milk-derived cheese (Idiazabal). By means of SPME-GC-MS, 81 volatile compounds were identified, among which acids predominated, followed by esters, ketones and alcohol

Diverse and related 16S rRNA-encoding DNA sequences in prostate tissues of men with chronic prostatitis.

Treatment of chronic prostatitis/chronic pelvic pain syndrome is often empirical because clinical culture methods fail to detect prostate-associated pathogens in >90% of patients. Previously, we tested a variety of specific-microorganism PCRs and began a DNA sequence study after we found that 77% of prostatitis patients were PCR positive for prokaryotic rRNA-encoding DNA sequences (rDNAs) despite negative cultures using optimal techniques. In the present study, 36 rDNA clones from 23 rDNA-positive patients were sequenced. This study represents more than twice the total rDNA sequence and more t

A genome-resolved cyanobacterial consortium from the Atacama Desert reveals novel bacterial diversity, stress-resilience potential, chromatic flexibility, and photoprotection.

The Atacama Desert is one of the most extreme terrestrial environments on Earth, characterized by hyper-aridity, intense solar and ultraviolet radiation, oxidative stress, and metal-rich substrates. Here, we characterized an enriched cyanobacteria-associated consortium obtained from a high-altitude, river-influenced site in Quebrada Talabre, Atacama Desert. Deep shotgun metagenomic sequencing, achieving 99.7% estimated community coverage, enabled the reconstruction of 14 high-quality metagenome-assembled genomes representing five bacterial phyla. Genome-resolved analyses revealed multiple puta

Transcriptomic responses of gill and intestinal tissues in Nile tilapia (Oreochromis niloticus) to bacterial infection following sequential nanoimmersion and hydrogel-based multivalent vaccination.

Bacterial pathogens, including Flavobacterium oreochromis, Aeromonas veronii, Streptococcus agalactiae, and Edwardsiella tarda, represent major infectious threats to Nile tilapia (Oreochromis niloticus). A multivalent vaccination strategy integrating cationic nanoemulsion immersion with oral hydrogel boosters was developed to investigate tissue-specific immune responses at the transcriptomic level. Gill tissues were collected following immersion challenge and intestinal tissues following intraperitoneal injection challenge, reflecting the physiologically relevant infection biology of each path

Integrated meta-omics reveals AFB1 dose-dependent remodeling of the rumen microbiome-virome-metabolome axis driving metabolic impairment in goats.

BACKGROUND: Aflatoxin B1 (AFB1), a highly carcinogenic and hepatotoxic mycotoxin frequently contaminating animal feed, presents serious health risks to both humans and livestock. Although AFB1's hepatotoxicity and other organ damage are extensively characterized, how this mycotoxin influences ruminal microbiota dynamics and functional activities in ruminants remains underexplored. Although some studies suggest that AFB1 reduces nutrient digestibility and performance in ruminants, the underlying mechanisms are unclear. To aid in developing effective mitigation strategies for aflatoxicosis in ru

Effects of Ganoderma lucidum Polysaccharides on Dexameth-Asone-Induced Immune Injury in Goslings.

This study aimed to investigate the role of ganoderma lucidum polysaccharides (GLP) in mitigating dexamethasone (DEX)-induced immune injury in goslings. Dexamethasone (DEX) is commonly used to establish an animal model of immune suppression, which mimics the immune injury caused by stress or certain pathological conditions in poultry. A total of 180 one-day-old goslings were randomly assigned to three groups: control (Con), DEX, and DEX + GLP, with six replicates of 10 goslings each. The Con and DEX groups were fed a basal diet, whereas the DEX + GLP group received feed supplemented with 0.2%

Draft genome sequence of novel Candidatus Ornithobacterium hominis carrying antimicrobial resistance genes in Egypt.

BACKGROUND: Candidatus Ornithobacterium hominis (O. hominis), which was identified in nasopharyngeal swabs from Egypt, has been associated with respiratory disorders in humans. O. hominis, a recently identified member of the Flavobacteriaceae family, belongs to the largest family within the Bacteroidetes phylum. This family includes hundreds of species and 90 genera, including major human pathogens such as Capnocytophaga canimorsus and Elizabethkingia meningoseptica. Herein, we presented two draft genome assemblies of O. hominis that were extracted from metagenomic data using the Illumina sequ

Phylogenetic characterization of microbial communities that reductively dechlorinate TCE based upon a combination of molecular techniques.

An anaerobic microbial consortium (referred to as ANAS) that reductively dechlorinates trichloroethene (TCE) completely to ethene with the transient production of cisdichloroethene (cDCE) and vinyl chloride was enriched from contaminated soil obtained from Alameda Naval Air Station. ANAS uses lactate as its electron donor and has been functionally stable for over 2 years. Following a brief exposure to oxygen, a subculture (designated VCC) derived from ANAS could dechlorinate TCE only to vinyl chloride with lactate as its electron donor. Three molecular methods were used concurrently to charact

Pesticide mixtures, endocrine disruption, and amphibian declines: are we underestimating the impact?

Amphibian populations are declining globally at an alarming rate. Pesticides are among a number of proposed causes for these declines. Although a sizable database examining effects of pesticides on amphibians exists, the vast majority of these studies focus on toxicological effects (lethality, external malformations, etc.) at relatively high doses (parts per million). Very few studies focus on effects such as endocrine disruption at low concentrations. Further, most studies examine exposures to single chemicals only. The present study examined nine pesticides (four herbicides, two fungicides,

Glacier-Fed Stream Biofilms Harbor Diverse Resistomes and Biosynthetic Gene Clusters.

Antimicrobial resistance (AMR) is a universal phenomenon the origins of which lay in natural ecological interactions such as competition within niches, within and between micro- to higher-order organisms. To study these phenomena, it is crucial to examine the origins of AMR in pristine environments, i.e., limited anthropogenic influences. In this context, epilithic biofilms residing in glacier-fed streams (GFSs) are an excellent model system to study diverse, intra- and inter-domain, ecological crosstalk. We assessed the resistomes of epilithic biofilms from GFSs across the Southern Alps (New

Seminal Microbiota of Idiopathic Infertile Patients and Its Relationship With Sperm DNA Integrity.

The development of new biomarkers for human male infertility is crucial to improve the diagnosis and the prognosis of this disease. Recently, seminal microbiota was shown to be related to sperm quality parameters, suggesting an effect in human fertility and postulating it as a biomarker candidate. However, its relationship to sperm DNA integrity has not been studied yet. The aim of the present study is to characterize the seminal microbiota of a western Mediterranean population and to evaluate its relationship to sperm chromatin integrity parameters, and oxidative stress. For that purpose, 14

Identification and characterization of Porphyromonas gingivalis TonB.

UNLABELLED: Porphyromonas gingivalis is one of the major bacterial pathogens responsible for the initiation and progression of periodontal disease. The bacterium codes for multiple TonB-dependent receptors required for acquisition of nutrients such as heme and vitamin B12, although the identity of a TonB energy transducer has yet to be identified. Here we identify a potential TonB protein encoded by PG0785, generate a deficient strain, and characterize its biological significance. Bioinformatics analysis reveals that the PG0785 has unique features confined to the Cytophaga-Flavobacterium-Bacte

Characterisation of the bacterial and archaeal microbiota in processed colostrum collected from a spring-calving dairy herd.

Colostrum feeding is critical for neonatal calf health, providing immunoglobulins (Ig) and other bioactive compounds that support immune function and early microbiome development. While the microbiota of fresh colostrum has been characterised, colostrum on commercial dairy farms is often refrigerated and reheated prior to feeding - practices that may alter its microbial composition. Therefore, the objective of this study was to characterise the prokaryotic community of refrigerated and reheated (processed) colostrum collected immediately before calf feeding. Twenty-one processed colostrum samp
