Selenium induces mercury isotope fractionation and detoxification in marine fish.

Prey fish are vital in transferring nutrients and contaminants from lower to higher trophic levels. Although the role of selenium (Se) on mercury (Hg) detoxification has been identified in marine mammals and seabirds, the metabolic pathways of Hg-Se interaction in fish remain unknown. We used Hg stable isotopes (δ202Hg, Δ199Hg) to characterize the internal dynamics of methylmercury (MeHg) and inorganic Hg (IHg) in the presence and absence of Se in juvenile olive flounders (Paralichthys olivaceus). The fish were exposed to MeHg- (1,223 ng/g, n = 31) and IHg-amended (1,515 ng/g, n = 31) food pel

Spatial and temporal trends of mercury in the aquatic food web of the lower Penobscot River, Maine, USA, affected by a chlor-alkali plant.

Mercury (Hg) concentrations in aquatic biota, including fish and shellfish, were measured over the period 2006-2012 in the lower Penobscot River and upper estuary (Maine, USA). The Penobscot is a system contaminated with Hg by a chlor-alkali plant that operated from 1967 to 2000, discharging 6-12 tons of mercury into the river. Mercury levels in aquatic biota were highest at sites downstream of the chlor-alkali plant and spatial trends were similar to those of sediments. Mean total Hg concentrations in fish muscle (adjusted for size or age) in the most affected areas were 521 (480, 566; 95% CI

Comparison of toxic effects of nitric oxide and peroxynitrite on Uronema marinum (Ciliata: Scuticociliatida).

To discover the effects of nitric oxide (NO) and peroxynitrite on Uronema marinum (a ciliate responsible for systemic scuticociliatosis in cultured olive flounder Paralichthys olivaceus), the dose-dependent inhibitory effect of NO donors, S-nitroso-N-acetylpenicillamine (SNAP) and 3-morpholinosydnonimine (SIN-1) on the proliferation and survival of U. marinum was investigated. The inhibitory effects of exogenous superoxide dismutase (SOD) and catalase on the toxicity of SIN-1 were also investigated. After 24 h of incubation in the presence of 0.2 mM SNAP, the number of ciliates was not statist

Cryptobiosis and its control in North American fishes.

Cryptobiosis is caused by the haemoflagellates Cryptobia bullocki and Cryptobia salmositica. These parasites infect food fishes (e.g. flounders, salmon) on both the Atlantic and Pacific coasts of North America and clinical signs of the disease include anaemia, and abdominal distention with ascites. The virulent factor in salmonid cryptobiosis, caused by C. salmositica, is a secretory metalloprotease (200 kDa). Fish mortality may be up to 100% in the absence of treatment, consequently strategies have been developed to protect them from disease/mortality. A single dose of a live vaccine protects

The Fort Totten mercury pollution risk assessment: a case history.

Operational activities have resulted in mercury in the sediments surrounding Little Bay in Queens, NY. This is adjacent to Fort Totten, a formerly used defense site. Some of the mercury levels in these sediments exceeded New York State screening values. A human health risk assessment was accomplished, based on conservative assumptions. The risk assessment examined the potential for adverse health effects from direct contact with and ingestion of contaminated sediments/surface water and ingestion of biota. Potential exposures to recreational receptors including adults and children were examined

Species-specific sensitivity to in vitro inhibition of the thyroperoxidase (TPO) enzyme by environmental pollutants for five fishes.

Chemicals released into aquatic environments are detected in surface waters globally, and some can disrupt thyroid hormone (TH) signaling in fishes. A key protein in this pathway is thyroperoxidase (TPO), which catalyzes synthesis of THs. Inhibition of TPO is associated with decreased THs, leading to a variety of adverse outcomes. However, there is uncertainty regarding species-specific sensitivities to inhibition of TPO among fishes and whether inhibitors in mammals share that activity in fishes. Therefore, the present study applied the Amplex UltraRed TPO assay to compare in vitro sensitivit

Application of nervous necrosis virus capsid protein-based antigen-presenting particles for vaccine development.

Nervous necrosis virus (NNV) capsid protein plays an important role in producing viral particles without any genetic elements. Thus, NNV is a promising candidate for vaccine development and is widely used for constructing vaccines, including DNA, recombinant proteins, and virus-like particles (VLPs). Our study aimed to investigate the potential of NNV capsid protein (NNV) and NNV capsid protein fused to enhanced green fluorescent protein (NNV-EGFP) through VLP formation and whether their application can induce specific antibody responses against certain antigens. We focused on producing DNA an

Membrane orthologs of TLR5 of tongue sole Cynoglossus semilaevis: Expression patterns, signaling pathway and antibacterial property.

Mammalian toll-like receptor 5 (TLR5) is crucial for recognizing bacterial flagellin and initiating the inflammatory signaling cascades via myeloid differentiation factor 88 (MyD88) signaling pathway, which plays vital roles in innate immune against pathogenic bacteria. Herein, we reported the signaling pathway and antibacterial property of tongue sole (Cynoglossus semilaevis) membrane forms of TLR5 (i.e. CsTLR5M1and CsTLR5M2). CsTLR5M1/M2 contain 936 and 885 amino acid residues respectively. CsTLR5M1 shares 86.7% overall sequence identities with CsTLR5M2. CsTLR5M1/M2 possess the same extracel

Identification, immunological function, and antimicrobial mechanisms of CXCL12 in turbot (Scophthalmus maximus).

The chemokine CXCL12, as a pivotal immune regulator, plays a crucial role in teleost fish against bacterial infection, but current research on its specific mechanisms remains scarce. This study systematically identified two copies of the CXCL12 in turbot (Scophthalmus maximus) (SmCXCL12a and SmCXCL12b), investigated their expression patterns, and revealed their immune functions and antibacterial mechanisms. Firstly, phylogenetic and syntenic analyses indicated that the SmCXCL12a/b were relatively conserved and exhibited high homology with orthologs in other teleost fish, including Japanese flo

The development of RT-RPA and CRISPR-Cas12a based assay for sensitive detection of Hirame novirhabdovirus.

Hirame novirhabdovirus (HIRRV) is a highly pathogenic fish virus that poses a significant threat to the farming of a variety of economic fish. Due to no commercial vaccines and effective drugs available, sensitive and rapid detection of HIRRV at latent and early stages is important and critical for the control of disease outbreaks. However, most of the current methods for HIRRV detection have a large dependence on instruments and operations. For better detection of HIRRV, we have established a detection technology based on the reverse transcription and recombinase polymerase amplification (RT-

The History of Engineered Tracheal Replacements: Interpreting the Past and Guiding the Future.

The development of a tracheal graft to replace long-segment defects has thwarted clinicians and engineers alike for over 100 years. To better understand the challenges facing this field today, we have consolidated all published reports of engineered tracheal grafts used to repair long-segment circumferential defects in humans, from the first in 1898 to the most recent in 2018, totaling 290 clinical cases. Distinct trends emerge in the types of grafts used over time, including repair using autologous fascia, rigid tubes of various inert materials, and pretreated cadaveric allografts. Our analys

A sequential study of pathological findings in Atlantic halibut, Hippoglossus hippoglossus (L), throughout one year after an acute outbreak of viral encephalopathy and retinopathy.

Following a natural outbreak of viral encephalopathy and retinopathy (VER) at a commercial farm in Norway, surviving Atlantic halibut, Hippoglossus hippoglossus, were sequentially studied for distribution of nodavirus, immune response and histopathology over 1 year. Typical clinical signs and histopathology of VER were observed during the acute stage of the disease. Most of the surviving fish became subclinical carriers of nodavirus with clusters of nodavirus-containing cells in the central nervous system. Four random samplings of presumably healthy fish were performed from two fish groups, wi

Explosives compounds from sea-dumped relic munitions accumulate in marine biota.

Relic munitions are a hazardous legacy of the two world wars present in coastal waters worldwide. The southwest Baltic Sea has an especially high prevalence of unexploded ordnance and dumped munition material, which represent a large potential source of toxic explosive chemicals (munition compounds, MC). In the current study, diverse biota (plankton, macroalgae, tunicate, sponge, mollusc, echinoderm, polychaete, anemone, crustacea, fish) were collected from the Kiel Bight and a munitions dumpsite at Kolberger Heide, Germany, to evaluate the potential bioaccumulation of explosives and their der

Ecological drivers of mercury concentrations in fish species in subsistence harvests from Kotzebue Sound, Alaska.

The State of Alaska assesses human exposure to mercury (Hg) via fish consumption producing consumption guidelines for fish tailored for children and women of childbearing age. Under these guidelines, unrestricted consumption is suggested for many fish species, while limited consumption is recommended for others. Subsequent questions have arisen regarding ecological drivers influencing [Hg] in fishes consumed by Alaskans. This community-assisted public health study evaluates [Hg] in fishes from Kotzebue Sound to examine factors that may drive observed [Hg]. We examined eight species of subsiste

Heterochiasmy and the establishment of gsdf as a novel sex determining gene in Atlantic halibut.

Atlantic Halibut (Hippoglossus hippoglossus) has a X/Y genetic sex determination system, but the sex determining factor is not known. We produced a high-quality genome assembly from a male and identified parts of chromosome 13 as the Y chromosome due to sequence divergence between sexes and segregation of sex genotypes in pedigrees. Linkage analysis revealed that all chromosomes exhibit heterochiasmy, i.e. male-only and female-only meiotic recombination regions (MRR/FRR). We show that FRR/MRR intervals differ in nucleotide diversity and repeat class content and that this is true also for other

The Regulatory T Cell in Active Systemic Lupus Erythematosus Patients: A Systemic Review and Meta-Analysis.

Background: Regulatory T cells (Tregs) researches in systemic lupus erythematosus (SLE) have floundered over the years, reports on the numbers and function of Tregs in SLE present quite contradictory results. We therefore conducted a meta-analysis to verify the changes of Tregs in active SLE. Methods: We systematically searched PubMed, Embase, and ISI web of knowledge databases for eligible articles. In total, 628 active SLE patients and 601 controls from 18 studies were included. Due to a high degree of heterogeneity, a random effects model was used to assess the mean differences in Treg perc

Molecular identification and developmental expression of UV and green opsin mRNAs in the pineal organ of the Atlantic halibut.

The pineal organ is the only differentiated photoreceptor organ present in embryos and early larvae of the Atlantic halibut (Hippoglossus hippoglossus). We investigated the molecular identity of opsins in the pineal organ, and their expression during different life stages. Using RT-PCR we identified two 681-bp gene sequences, named HPO1 and HPO4, in cDNA from adult pineal and whole embryos. The predicted amino acid sequences showed highest identity to the transmembrane regions of teleostean RH2 green cone opsins (HPO1, 72-91%) and SWS-1 UV cone opsins (HPO4, 71-83%). In situ hybridization reve

Ecosystem consequences of cyanobacteria in the northern Baltic Sea.

Cyanobacteria of the Baltic Sea have multiple effects on organisms that influence the food chain dynamics on several trophic levels. Cyanobacteria contain several bioactive compounds, such as alkaloids, peptides, and lipopolysaccharides. A group of nonribosomally produced oligopeptides, namely microcystins and nodularin, are tumor promoters and cause oxidative stress in the affected cells. Zooplankton graze on cyanobacteria, and when ingested, the hepatotoxins (nodularin) decrease the egg production of, for example, copepods. However, the observed effects are very variable, because many crusta
