Although the anticaries effect of fluoride (F) dentifrices is clearly established, the relative importance of F taken up by dental plaque not removed by brushing and of F products (CaF(2)-like) formed on totally cleaned enamel for the subsequent inhibition of demineralization is not known. Both effects were evaluated using conventional (1,100 microg F/g) and low-F concentration (500 microg F/g) dentifrices in a randomized, crossover, double-blind in situ study. Enamel blocks not treated or pretreated with the dentifrices to form CaF(2)-like deposits were mounted in palatal appliances in contac

Because of the ubiquity of fluoride ions and their potential toxicity at high doses, researchers would like to design receptors that selectively detect this anion. Fluoride is found in drinking water, toothpaste, and osteoporosis drugs. In addition, fluoride ions also can be detected as an indicator of uranium enrichment (via hydrolysis of UF(6)) or of the chemical warfare agent sarin, which releases the ion upon hydrolysis. However, because of its high hydration enthalpy, the fluoride anion is one of the most challenging targets for anion recognition. Among the various recognition strategies

Fluoride varnish is developed to prolong the contact time between fluoride and tooth surface, so that the tooth becomes more resistant to caries attack. The active ingredient of fluoride varnish is usually 5% sodium fluoride, (22,600 ppm fluoride). Studies have found that fairly insoluble globules of calcium fluoride-like material formed on the tooth surface after topical fluoride application. These globules act as a reservoir of fluoride in the mouth for a prolonged period of time. Systematic reviews corroborate evidence for the efficacy of fluoride varnish in the prevention of dental caries.

The anticaries effect of professional fluoride (F) application has been attributed to calcium-fluoride-like deposits (CaF(2)) formed on enamel, but this has not been clearly demonstrated. We hypothesized that CaF(2) formed on plaque-free enamel by F application would reduce enamel demineralization due to the increase of F availability in fluid of subsequently formed plaque. We created distinct levels of CaF(2) on enamel to evaluate a dose-response effect. Enamel blocks were mounted in contact with a S. mutans test plaque and used in situ by 10 volunteers. F released to the fluid phase of this

Reconstructing enamel-like structures on teeth have been an important topic of study in the material sciences and dentistry. The important role of amelogenin in modulating the mineralization of organized calcium phosphate crystals has been previously reported. We used amelogenin and utilized a modified biomimetic deposition method to remineralize the surface of etched enamel to form mineral layers containing organized needle-like fluoridated hydroxyapatite crystals. The effect of a recombinant amelogenins (rP172) on the microstructure of the mineral in the coating was analyzed by SEM, XRD and

Proteomical analysis is defined as the characterization of the entire set of protein encoded a genome. Two-dimensional electrophoresis (2-DE) and matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS) are main techniques used in proteomic analysis to achieve information about protein expression profiles. Knowledge about the mechanism of skeletal fluorosis can be gained by recognizing changes in protein expression. To better understand the skeletal fluorosis process, osteoblasts isolated from calvarial of neonatal mouse were cultured and treated with 2 ppm f

Fluoride complexes of metallic ions are one of the main problems when processing industrial effluents with high content of fluoride anion. The most important case is derived from pickling treatment of stainless steel, which is performed with HNO3/HF mixtures to remove oxides scale formed over the metal surface. Waste from this process, spent pickling liquor, must be treated for recovering metallic and acid content. Conventional treatments produce a final effluent with high quantity of fluoride complexes of iron and chromium. This work proposes a hydrolysis treatment of these solid metal fluori

OBJECTIVES: The objective of this study was to investigate the effects of CO(2) laser on fluoride uptake in the loosely- and firmly-bound forms in enamel.
METHODS: Five human molars were cut into halves before being treated with 2.0% NaF topical gel. Each half had three windows on the enamel surface, including one control and two experimental windows irradiated by two laser therapies. One half of each tooth was treated with 1 M KOH solution to remove the loosely-bound fluoride (calcium fluoride). A tooth section was obtained from each window and the relative fluorine concentration was analyze

PURPOSE: The purpose of this paper is to describe some aspects of the intra-cellular mechanism of action of neuroleptics, drugs widely used in psychiatry in treatment of psychotic and affective disorders. The ability of neuroleptics to influence and modify the metabolic, energetic and structural processes of neurons, as well as their apoptosis, probably influence their therapeutic potential. The direct and indirect mechanisms of antipsychotics are discussed on the basis of epigenetic, intra-cellular and post-transcription processes.
VIEWS: Antipsychotic drugs facilitate chromatin remodeling,

In this in vitro study, examination of the enamel surface morphology after topical application of an amine fluoride solution with different fluoride (F) content was carried out. Sound human enamel slabs were treated with an amine fluoride solution containing either 1.0, 0.5 or 0.25% F for 3 min. during 3 days. All slabs were examined using scanning electron microscopy and energy-dispersive spectroscopy (SEM/EDS) qualitative analysis. The globular precipitates were revealed in all treated specimens, regardless of F content. The distribution of the deposits was more homogeneous in groups treated

The marked caries reduction in many countries over the last two decades is thought to be mainly the result of the widespread and frequent use of low concentrations of fluorides mainly via toothpastes. This article gives a review on fluoride sources in the oral cavity and discusses the physiochemical mechanisms by which fluoride interacts with enamel during the caries process and its reversal. From this review it becomes clear that fluoride incorporated into the enamel mineral during tooth development has little effect on the caries process. It is the fluoride incorporated posteruptively and in

(1) Data on fluoride efficacy in secondary prevention of osteoporotic fractures were contradictory when we last examined the file, in 1990. We now re-examine the clinical file on fluoride with more follow-up and new clinical trial data. (2) In 354 postmenopausal women with a history of vertebral fracture (secondary prevention), a comparative trial (the FAVOS trial) showed that moderate-dose fluoride for 2 years did not reduce the incidence of new vertebral fractures relative to calcium plus vitamin D. Furthermore, fluoride caused more cases of lower-limb pain. (3) In 110 postmenopausal women w

Fluoride is an effective anabolic agent to increase spinal bone density by increasing bone formation, and at therapeutically relevant (i.e., micromolar) concentrations, it stimulates bone cell proliferation and activities in vitro and in vivo. However, the fluoride therapy of osteoporosis has been controversial, in large part because of a lack of consistent antifracture efficacy. However, information regarding the molecular mechanism of action of fluoride may improve its optimum and correct usage and may disclose potential targets for the development of new second generation drugs that might h

Fluoride has been used in the treatment for osteoporosis for more than 30 years but its use is still controversial. Fluoride stimulates the osteoblasts to form new bone tissue. However, the new bone has an abnormal texture and is less mineralized and relatively less strong. In patients with osteoporosis treated with fluoride, the bone mineral density of the skeleton increases. On the other hand, the effects of fluoride treatment on the incidence of fractures are controversial and in some studies an increased fracture incidence was observed. In addition, fluoride often causes upper gastrointest

This study was performed to determine the fluoride concentration of the various cementum surfaces in different tooth groups to find out the most proper teeth and tooth surfaces for different cementum studies. For this purpose, direct measurements of phosphorus and fluoride were carried out in an acid etch biopsy solution. The findings indicate that incisors with exposed cementum are the most inappropriate teeth in comparison with the other groups. According to the results obtained it may be recommended that the studies related to fluoride uptake for cementum should be performed on teeth with n

The study dealing with the content of fluoride and calcium in stapedial bone and canal wall bone in otosclerosis was performed in 69 subjects (48 females and 21 males) out of the patients, in whom bone rebuilding activity had earlier been studied isotopically. The control group comprised 20 normal stapedies taken during autopsies. Fluoride content was determined by means of fluoride ions meter, the content of calcium was assessed by resorting to atomic absorptiometer. The content of studied elements was compared with the bone rebuilding activity and some clinical features such as the patient's

In response to recent concerns about the effect of water fluoridation on hip fracture rates, we studied the influence of fluoride intake on bone strength. Four groups of rats were fed a low-fluoride diet ad libitum and received 0, 5, 15, or 50 ppm of fluoride in their drinking water. Animals were euthanized after 3, 6, 12, or 18 months of treatment. Mechanical strength of the right femur was measured by three-point bending. Fluoride content for the left femur was measured, and static histomorphometric measurements were made on a lumbar vertebra. Femoral failure load was not significantly decre
