Cold-upregulated glycosyltransferase gene 1 (OsCUGT1) plays important roles in rice height and spikelet fertility.

Glycosyltransferases (GTs) regulate many physiological processes and stress responses in plants. However, little is known about the function of GT in rice development. In this study, molecular analyses revealed that the expression of a rice GT gene (Cold-Upregulated Glycosyltransferase Gene 1, CUGT1) is developmentally controlled and stress-induced. OsCUGT1 was knocked out by using the clustered regularly interspaced short palindromic repeats (CRISPR) system to obtain the mutant oscugt1, which showed a severe dwarf and sterility phenotype. Further cytological analyses indicated that the dwarfi

The expression of the NPR1-dependent defense response pathway genes in Persea americana (Mill.) following infection with Phytophthora cinnamomi.

A plant's defense against pathogens involves an extensive set of phytohormone regulated defense signaling pathways. The salicylic acid (SA)-signaling pathway is one of the most well-studied in plant defense. The bulk of SA-related defense gene expression and the subsequent establishment of systemic acquired resistance (SAR) is dependent on the nonexpressor of pathogenesis-related genes 1 (NPR1). Therefore, understanding the NPR1 pathway and all its associations has the potential to provide valuable insights into defense against pathogens. The causal agent of Phytophthora root rot (PRR), Phytop

The MSI1 member OsRBAP1 gene, identified by a modified MutMap method, is required for rice height and spikelet fertility.

To explore the molecular mechanisms underlying plant height regulation, we isolated and characterized a stably inherited semi-dwarf mutant bgsd-2 from the ethane methyl sulfonate (EMS) mutant progeny of 'Ping Tang Wild-type (PTWT)', a rice (Oryza sativa ssp. japonica) landrace in Guizhou. Transcriptome sequencing and qRT-PCR analyses showed that a number of cellulose and lignin-related genes involved in cell wall biogenesis were substantially downregulated in bgsd-2. MutMap-based cloning revealed the occurrence of a single amino acid substitution in the LOC_Os01g51300 gene, belonging to the MS

Thiamine functions as a key activator for modulating plant health and broad-spectrum tolerance in cotton.

Global climate changes cause an increase of abiotic and biotic stresses that tremendously threaten the world's crop security. However, studies on broad-spectrum response pathways involved in biotic and abiotic stresses are relatively rare. Here, by comparing the time-dependent transcriptional changes and co-expression analysis of cotton (Gossypium hirsutum) root tissues under abiotic and biotic stress conditions, we discovered the common stress-responsive genes and stress metabolism pathways under different stresses, which included the circadian rhythm, thiamine and galactose metabolism, carot

Novel Bisexual Flower Control Gene Regulates Sex Differentiation in Melon (Cucumis melo L.).

The sex-control system involves several mechanisms in melon. The present study identified a novel bisexual flower control gene from the hermaphroditic melon germplasm, different from the previously recognized one. Genetic analysis showed that a single recessive gene in the newly identified locus b controlled the bisexual flower phenotype in melons. We generated 1431 F2 segregating individuals for genetic mapping of locus b, which was delimited to a 47.94 kb region. Six candidate genes were identified in the delimited interval, and candidate No. 4 encoding melon CPR5 protein was selected as the

Distinct physiological and transcriptional responses of leaves of paper mulberry (Broussonetia kazinoki × B. papyrifera) under different nitrogen supply levels.

Paper mulberry, a vigorous pioneer species used for ecological reclamation and a high-protein forage plant for economic development, has been widely planted in China. To further develop its potential value, it is necessary to explore the regulatory mechanism of nitrogen metabolism for rational nitrogen utilization. In this study, we investigated the morphology, physiology and transcriptome of a paper mulberry hybrid (Broussonetia kazinoki × B. papyrifera) in response to different nitrogen concentrations. Moderate nitrogen promoted plant growth and biomass accumulation. Photosynthetic character

Selective defoliation affects plant growth, fruit transcriptional ripening program and flavonoid metabolism in grapevine.

BACKGROUND: The selective removal of grapevine leaves around berry clusters can improve the quality of ripening fruits by influencing parameters such as the berry sugar and anthocyanin content at harvest. The outcome depends strongly on the timing of defoliation, which influences the source-sink balance and the modified microclimate surrounding the berries. We removed the basal leaves from Vitis vinifera L. cv Sangiovese shoots at the pre-bloom and veraison stages, and investigated responses such as shoot growth, fruit morphology and composition compared to untreated controls. Moreover, we per

[Advances in plant lipoxygenases research].

Lipoxygenases (linoleate: oxygen oxidoreductase, EC 1.13.11.12; LOXs) are encoded by a multi-gene family in plants. The LOXs are monomeric non-heme, non-sulfur iron dioxygenases, which catalyze the incorporation of molecular oxygen into polyunsaturated fatty acids containing a cis, cis-1,4-pentadiene moiety. The LOX isoforms are distinguished by differences in optimum pH of the reaction, pI, substrate and product specificity, spatial and temporal expression, and subcellular localization. The function of various LOXs in plants has been suggested. Some of the physiological processes in which lip

Cell death of rice roots under salt stress may be mediated by cyanide-resistant respiration.

Treatment with solutions containing high concentrations of NaCl (200 or 300 mM) induced cell death in rice (Oryza sativa L.) roots, as well as the application of exogenous hydrogen peroxide (H2O2). Moreover, the pretreatment with dimethylthiourea (DMTU), a scavenger of H2O2, partially alleviated the root cell death induced by 200 mM NaCl. These observations suggest that the cell death of rice roots under high salt stress is linked to H2O2 accumulation in vivo. NaCl stress increased the level of cyanide-resistant respiration to some extent and enhanced the transcript levels of the alternative o

Non-coding RNAs as central regulators of phytohormone signaling in plant abiotic stress responses.

Plants are continuously exposed to a wide range of abiotic stresses, including drought, salinity, temperature extremes, nutrient deficiency, and heavy metal toxicity, which severely constrain growth and productivity. To cope with these challenges, plants have evolved sophisticated regulatory networks involving phytohormones and non-coding RNAs (ncRNAs). This review provides a comprehensive overview of the biogenesis and functional roles of major ncRNA classes' microRNAs (miRNAs), long non-coding RNAs (lncRNAs), and circular RNAs (circRNAs) and their dynamic interplay with phytohormonal signali

Unraveling the molecular mechanisms underlying flat stem formation in Atractylodes lancea in response to phytoplasmas.

For centuries, the herb Atractylodes lancea (Thunb.) has been revered for its pharmacological properties, particularly its efficacy in combating spleen and stomach ailments. However, prolonged artificial cultivation of A. lancea has led to an increased presence of serious plant diseases, including flat branch disease caused by phytoplasmas. To elucidate the mechanisms of flat stem (FS) formation, physiological and biochemical indicators, as well as related metabolic pathways were assessed in A. lancea stems following phytoplasma infection. The results demonstrated significant changes in the le

Insights into salicylic acid responses in cucumber (Cucumis sativus L.) cotyledons based on a comparative proteomic analysis.

To investigate the response of cucumber seedlings to exogenous salicylic acid (SA) and gain a better understanding of SA action mechanism, we generated a proteomic profile of cucumber (Cucumis sativus L.) cotyledons treated with exogenous SA. Analysis of 1500 protein spots from each gel revealed 63 differentially expressed proteins, 59 of which were identified successfully. Of the identified proteins, 97% matched cucumber proteins using a whole cucumber protein database based on the newly completed genome established by our laboratory. The identified proteins were involved in various cellular

HSP17.4 mediates salicylic acid and jasmonic acid pathways in the regulation of resistance to Colletotrichum gloeosporioides in strawberry.

In this study, we used virus-mediated gene silencing technology and found that the HSP17.4 gene-silenced cultivar Sweet Charlie plants were more susceptible to Colletotrichum gloeosporioides than the wild-type Sweet Charlie, and the level of infection was even higher than that of the susceptible cultivar Benihopp. The results of differential quantitative proteomics showed that after infection with the pathogen, the expression of the downstream response genes NPR1, TGA, and PR-1 of the salicylic acid (SA) signalling pathway was fully up-regulated in the wild-type Sweet Charlie, and the expressi

Involvement of microRNA in copper deficiency-induced repression of chloroplastic CuZn-superoxide dismutase genes in the moss Physcomitrella patens.

Superoxide dismutases (SODs) are metallo-enzymes that catalyze the dismutation of superoxide radicals. In Arabidopsis thaliana, the expression of CuZn-SOD in both the chloroplast and cytosol was reported to be down-regulated by microRNA398 (miR398) during growth on low copper. The moss Physcomitrella patens contains chloroplastic and cytosolic CuZn-SOD genes, but lacks miR398. From analysis of P. patens microRNA, miR1073 was predicted to target CuZn-SOD mRNAs. We noticed that two chloroplastic CuZn-SOD genes contain the miR1073 target sequence in the 3' untranslated region; however, the cytoso

Extracellular invertase LIN6 of tomato: a pivotal enzyme for integration of metabolic, hormonal, and stress signals is regulated by a diurnal rhythm.

To elucidate the regulation pattern of extracellular invertase LIN6 of tomato, the corresponding promoter has been cloned and the sink-tissue specific expression and its regulation by sugars, stress stimuli, growth regulators, and the diurnal rhythm is shown. The in situ analysis of transgenic tobacco plants expressing a LIN6 promoter::beta-glucuronidase reporter gene fusion demonstrates LIN6 expression in sink tissues, such as pollen grains and vascular tissues of leaves and stems. LIN6 is up-regulated in close proximity to wounded tissue, and by methyl jasmonate and abscisic acid, global sig

The miR167-ARF8 module coordinates stamen-pistil development and fruit set in tomato.

Auxin is an important regulator of reproductive development. The families of the AUXIN RESPONSE FACTOR (ARF) genes, important components of the auxin signal, have expanded in flowering plants. Tomato (Solanum lycopersicum) has two copies of ARF8, which are negatively regulated by miR167. Here, we investigated the involvement of this expanded SlmiR167-SlARF8 module in flower development. Mutants were generated in 5 SlmiR167 and 2 SlARF8 genes, and flower development and fruit set were characterized in mutant combinations. We show that the SlmiR167-SlARF8 module regulates flower-organ developmen

Predicting transcriptional regulators in plants in the era of artificial intelligence.

Identifying transcriptional regulators that control important biological pathways in plants is fundamental to understanding regulatory mechanisms, network hierarchy, and phenotypic variation. This remains challenging because transcription factors (TFs) and their targets operate within highly interconnected, dynamic, and often redundant regulatory networks. Over the past two decades, advances in omics technologies, sequencing data generation, and computational tools have shifted gene discovery from single-gene studies to network-level investigation. At the same time, these advances have created

An integrated multi-omics approach reveals polymethoxylated flavonoid biosynthesis in Citrus reticulata cv. Chachiensis.

Citrus reticulata cv. Chachiensis (CRC) is an important medicinal plant, its dried mature peels named "Guangchenpi", has been used as a traditional Chinese medicine to treat cough, indigestion, and lung diseases for several hundred years. However, the biosynthesis of the crucial natural products polymethoxylated flavonoids (PMFs) in CRC remains unclear. Here, we report a chromosome-scale genome assembly of CRC with the size of 314.96 Mb and a contig N50 of 16.22 Mb. Using multi-omics resources, we discover a putative caffeic acid O-methyltransferase (CcOMT1) that can transfer a methyl group to

Transgenerational effect of mutants in the RNA-directed DNA methylation pathway on the triploid block in Arabidopsis.

BACKGROUND: Hybridization of plants that differ in number of chromosome sets (ploidy) frequently causes endosperm failure and seed arrest, a phenomenon referred to as triploid block. In Arabidopsis, loss of function of NRPD1, encoding the largest subunit of the plant-specific RNA polymerase IV (Pol IV), can suppress the triploid block. Pol IV generates short RNAs required to guide de novo methylation in the RNA-directed DNA methylation (RdDM) pathway. Recent work suggests that suppression of the triploid block by mutants in RdDM components differs, depending on whether the diploid pollen is de

Salicylic Acid Steers the Growth-Immunity Tradeoff.

Plants possess an effective immune system to combat most microbial attackers. The activation of immune responses to biotrophic pathogens requires the hormone salicylic acid (SA). Accumulation of SA triggers a plethora of immune responses (like massive transcriptional reprogramming, cell wall strengthening, and production of secondary metabolites and antimicrobial proteins). A tradeoff of strong immune responses is the active suppression of plant growth and development. The tradeoff also works the opposite way, where active growth and developmental processes suppress SA production and immune re
