Coexistence of multiple genotypes, including newly identified genotypes, in outbreaks of gastroenteritis due to Norovirus in Japan.

Norovirus (NV) (formerly called Norwalk-like virus) is the most common cause of acute nonbacterial gastroenteritis in humans. Recently, we reported an NV genotyping scheme based on variability in the capsid N-terminal/shell (N/S) domain gene (Katayama et al., Virology 299:225-239, 2002). We found 19 genotypes, including nine of genogroup I and 10 of genogroup II. In the present study, we investigated the molecular epidemiology of NV from 66 outbreaks that occurred in Saitama Prefecture, Japan, from 1997 to 2002. We screened 416 stool specimens by a real-time reverse transcription (RT)-PCR meth

Novel cryptosporidium genotypes in sporadic cryptosporidiosis cases: first report of human infections with a cervine genotype.

In this study, we genotyped parasites from the fecal specimens of sporadic cryptosporidiosis cases in British Columbia from 1995 to 1999. Genotyping was conducted by polymerase chain amplification of the internal transcribed spacer region, a hypervariable region in the 18S rRNA gene and the Cryptosporidium oocyst wall protein gene. Subsequent analysis was by restriction fragment length polymorphism and DNA sequencing. We identified two new Cryptosporidium genotypes in humans. One of these genotypes has been found recently in deer in New York state. The other genotype has not been identified in

Single genotype of measles virus is dominant whereas several genotypes of mumps virus are co-circulating.

We have reported that in Japan measles virus strains have been classified into three distinct different genotypes (C1, D3 and D5) under the new international genotype classification since 1984. Similarly, mumps virus strains have been divided into two genotypes with three subtypes (B1, B2, B3, and D) under the proposed international classification since 1976. To differentiate these genotypes we developed a restriction fragment length polymorphism assay in the hemagglutinin (H) region for measles virus and in the hemagglutinin-neuraminidase (HN) region for mumps virus to facilitate the expanded

Genotype-by-environment interaction with broiler genotypes differing in growth rate. 1. The effects of high ambient temperature and naked-neck genotype on lines differing in genetic background.

High ambient temperature (AT) significantly depresses growth rate and meat yield of commercial broilers, thus making it a major factor hindering poultry meat production, especially in hot climates. The effects of high AT were variably moderated when feather coverage was reduced by the naked-neck gene (Na). In this study, the effects of high AT and Na were investigated in broiler progeny of hens from a sire line and two dam lines, differing in growth rate and meat yield due to different breeding histories. Heterozygous naked-neck (Na/na) males were mated with normally feathered (na/na) hens fro

Circulation of Dengue Virus Serotype 1 Genotype V and Dengue Virus Serotype 2 Genotype III in Tocantins State, Northern Brazil, 2021-2022.

In Brazil, the state of Tocantins, located in north-central Brazil, has experienced a significant number of cases of arboviral disease, particularly Dengue virus (DENV). This study aimed to deepen the knowledge on DENV circulation within that state by conducting full genome sequencing of viral genomes recovered from 61 patients between June 2021 and July 2022. There were a total of 8807 and 20,692 cases in 2021 and 2022, respectively, as reported by the state's Secretary of Health. Nucleotide sequencing confirmed the circulation of DENV serotype 1, genotype V and DENV serotype 2, genotype III

Allele, genotype, and composite genotype effects of IL-1A +4845 and IL-1B +3954 polymorphisms for chronic periodontitis in an Indian population.

INTRODUCTION: The pro-inflammatory cytokine interleukin-1 (IL-1) is a key modulator of host responses to microbial infection and a major modulator of extracellular matrix catabolism and bone resorption, and polymorphisms in the IL-1 gene cluster have been associated with an increased risk of developing severe adult periodontitis. A case control study was performed to determine the role of IL-1A+4845 and IL-1B+3954 polymorphisms in the predisposition to chronic periodontitis. MATERIALS AND METHODS: The study was conducted with 103 unrelated participants recruited from Manipal College of Dental

Glutathione S-transferase M1 (GSTM1) genotype but not GSTT1 or MC1R genotype influences erythemal sensitivity to narrow band (TL-01) UVB phototherapy.

OBJECTIVES: Although a majority of psoriasis patients respond to treatment with narrow band ultraviolet B radiation (TL-01) phototherapy, it is currently not possible to predict erythemal sensitivity, or to identify treatment responders. A variety of antioxidant enzymes, including the polymorphic glutathione S-transferase GSTM1 and GSTT1 genes, protect the cell from UVR-induced oxidative challenge. GSTM1 and GSTT1 are deleted in approximately 50 and 20% of the Caucasian population, respectively, and GST null genotype has been associated with increased sunburn sensitivity and reduced minimal er

Identification of 2 Novel Subtypes of Hepatitis C Virus Genotype 8 and a Potential New Genotype Successfully Treated With Direct Acting Antivirals.

BACKGROUND: Hepatitis C virus (HCV) has high genetic diversity and is classified into 8 genotypes and >90 subtypes, with some endemic to specific world regions. This could compromise direct-acting antiviral efficacy and global HCV elimination. METHODS: We characterized HCV subtypes "rare" in the United Kingdom (non-1a/1b/2b/3a/4d) by means of whole-genome sequencing via a national surveillance program. Genetic analyses to determine the genotype of samples with unresolved genotypes were undertaken by comparison with International Committee on Taxonomy of Viruses HCV reference sequences. RESUL

Construction and characterization of Genotype-3 hepatitis C virus replicon revealed critical genotype-3-specific polymorphism for drug resistance and viral fitness.

Hepatitis C virus (HCV), a major causative agent of chronic hepatitis, is a positive-stranded RNA virus and has a high degree of genetic diversity due to its error-prone RNA-dependent RNA polymerase. Development of direct-acting antiviral agents (DAAs) has greatly improved the therapeutic outcome of chronic hepatitis C patients. However, naturally existing resistance-associated variants (RAVs) or occurrence of resistance-associated substitutions (RASs) in the HCV genome may impose a challenge to the long-term success of the DAA-based therapies. Genotype-3 HCV is the most difficult genotype to

SCA27B in Brazil: frequency, phenotype and genotype-phenotype correlations.

BACKGROUND: Spinocerebellar Ataxia 27B (SCA27B) is a recently described autosomal dominant ataxia caused by uniallelic GAA intronic expansions at FGF14. It is a frequent SCA subtype in North American/European populations, accounting for > 20% of all SCAs in some series. Despite that, its frequency as well as phenotype in Latin America remains to be established. OBJECTIVES: To determine the frequency and the clinical phenotype of SCA27B in a large Brazilian SCA cohort. METHODS: We recruited 498 SCA patients from 322 unrelated families followed in a reference center. All patients had demograph

Genotype-phenotype relationships in butyrylcholinesterase deficiency: a systematic review.

BACKGROUND: Butyrylcholinesterase (BChE) deficiency is a recessive condition that can cause prolonged paralysis after suxamethonium or mivacurium. Conventional phenotyping using dibucaine and fluoride inhibition can overlook heterozygous carriers. The aim of this systematic review was to identify studies reporting paired BCHE genotype and biochemical phenotype data to propose a risk-assessment protocol. METHODS: We searched PubMed, EMBASE, and the Cochrane Library (1946-2024; PROSPERO CRD420250627891) for human studies reporting both BCHE genotype and biochemical phenotype. Extracted data inc

Dissecting genotype-specific effects of disease-associated genetic variants.

Non-coding variants associated with complex disease can shape gene regulatory networks across multiple genomic loci and cellular contexts. Here, we investigated the functional impact of the Parkinson-disease-associated variant rs11610045 using an isogenic induced pluripotent stem cell model. Using CRISPR-Cas9 editing and reversal, we generated matched clones carrying either the A|A or G|G genotype, enabling controlled comparison of allele-specific effects. We identified widespread genotype-dependent regulation of distal genes, including THBS1 and PDGFB. Affinity purification followed by mass s

Biallelic WDR19 Variants: Systematic Analysis of Genotype-Phenotype Correlations.

Ciliopathies represent a diverse group of inherited disorders resulting from dysfunctional cilia. A rare subset is associated with biallelic variants in WDR19, with fewer than 100 affected individuals reported worldwide. This study is aimed at reviewing the clinical and molecular spectrum of WDR19-associated ciliopathies, with a particular focus on genotype-phenotype correlations. We report a 32-year-old woman with renal, ocular and hepatic manifestations attributed to compound heterozygous variants in WDR19, and reviewed available data from 93 previously published cases. Multimodal genotype-phenotype analysis in SMARCB1-associated developmental disorders.

PURPOSE: Variants in SMARCB1, encoding a core subunit of the BAF (ie, BRG/BRM-associated factor) chromatin remodeling complex, are associated with intellectual developmental disorders, particularly Coffin-Siris syndrome, although the genotype-phenotype spectrum remains incompletely defined. This study aims to assess the correlations between SMARCB1 variant location and phenotypic manifestations. METHODS: We analyzed 31 individuals with pathogenic or likely pathogenic SMARCB1 variants using multimodal approaches, integrating clinical, structural, and machine learning analyses. We predicted var

Severe maternal morbidity according to sickle cell disease genotype.

BACKGROUND: Sickle cell disease (SCD) is a genetic hemoglobinopathy that disproportionately affects individuals of African descent and is associated with elevated risks of maternal complications. However, the contribution of specific SCD genotypes to severe maternal morbidity (SMM) and postpartum readmission remains poorly characterized at the population level. OBJECTIVE: We sought to examine the association between SCD genotypes-hemoglobin SS (HbSS) disease, hemoglobin SC (HbSC) disease, sickle thalassemia, and sickle cell trait-and SMM, hospital readmission, and related adverse maternal out

Astrocytic synapse engulfment is differentially controlled by APOE genotype.

APOE gene variants encoding the apolipoprotein E (ApoE) protein are strong genetic modifiers of risk of Alzheimer's disease (AD) with the APOE ε4 allele (APOE4) associated with substantially increased disease risk, APOE ε2 allele (APOE2) associated with decreased risk and APOE ε3 allele (APOE3) considered neutral. Recently the Christchurch variant of APOE3 (APOE3CH) has been shown to protect people from familial AD. Despite this strong evidence for APOE mediating AD risk, the exact biological mechanisms through which APOE influences pathogenesis remain unknown. Our previous work implicates APO

Genotype-phenotype correlation in multiple endocrine neoplasia type 1.

BACKGROUNDAmong patients with multiple endocrine neoplasia type 1 (MEN1), 80% develop duodenopancreatic neuroendocrine tumors (dpNETs), of whom 15%-25% die of metastasis. There is a need to identify biomarkers to predict aggressive disease. MEN1 genotype affords an attractive possibility as a biomarker, as it remains constant during life. Currently, patients are clinically diagnosed with MEN1 by the presence of ≥2 primary endocrine tumors (pituitary, parathyroid, and pancreas) or ≥1 primary endocrine tumor with a positive family history. From 10% to 30% of patients diagnosed clinically with ME

Genotype-dependent resilience mediated by melatonin in sweet corn.

BACKGROUND: Water deficits, exacerbated by climate change and unpredictable weather, have become a significant global challenge to agricultural productivity. In this context, exogenous melatonin treatment is well documented as a stress alleviator; however, its effects on various biological processes, particularly in less-explored genotypes, remain understudied. This study aimed to enhance water deficit resilience in sweet corn by applying foliar melatonin to four genotypes-Messenger, Dessert, Royalty, and Tyson under two levels of water deprivation induced by polyethylene glycol at 8% and 12%
