Tau pathology is a principal driver of cognitive impairment in Alzheimer's disease (AD), but the therapeutic targeting of tau has been hindered by poor brain delivery and a lack of lesion-confined activity. Here, we delineate a pathogenic cascade wherein the impaired dephosphorylation of hyperphosphorylated tau (p-tau) leads to its aggregation, which is amplified by microglia-mediated propagation. To combat this p-tau cascade, we developed a glycoengineered proteolysis targeting chimera (PROTAC) nanoparticle for lesion-specific p-tau modulation therapy. We first synthesized a library of p-tau

Mosquito-borne orthoflavivirus, such as Japanese encephalitis virus (JEV), Dengue virus (DENV), and Zika virus (ZIKV), pose a serious global health threat. As obligate intracellular parasites, they often hijack the host ubiquitin system to modify their own proteins, thereby regulating the viral life cycle, host adaptation, transmission, and pathogenesis. Despite its critical roles, the precise molecular mechanisms and functional significance of viral protein ubiquitination in orthoflavivirus infection remain incompletely understood. Here, we identify JEV prM as a novel target for host-specific

In our previous study, broiler-type chicks exhibited greater susceptibility to infection with avian pathogenic Escherichia coli (APEC) than layer-type chicks under identical experimental conditions. To better understand the underlying mechanism, most likely attributed to the immune response, the present work aimed to investigate the humoral and cellular immune parameters in specific pathogen-free broiler-type and layer-type chicks using samples collected from that experiment. Briefly, thirty-two chicks of each bird type were equally divided into two groups. At 14 days of life, one group of bro

The bacterial pathogen Mycoplasma bovis (M. bovis) emerged as the etiologic agent of mastitis in cattle during the early 1960's. Concurrent with the emergence of M. bovis, Holsteins in the US were undergoing intensive genetic selection for increased milk production. Comparison studies with unselected Holsteins have found an association with selection and increased susceptibility to disease. More recently, M. bovis was identified as the cause of epizootics of severe, chronic respiratory disease in American bison (Bison bison). In cattle, M. bovis causes chronic respiratory disease and is associ

UNLABELLED: Tuberculosis (TB) remains a major global health challenge that requires new therapeutic strategies to improve drug efficacy, shorten treatment duration, prevent drug resistance, and limit Mycobacterium tuberculosis (Mtb) persistence. Here, we established complementary in vitro human lung models integrating alveolar macrophage-like (AML) cells and airway air-liquid interface (ALI) cultures to evaluate standard-of-care antibiotics, host-directed therapies, and virulence-targeting agents. AMLs recapitulated key morphological, transcriptional, and functional features of primary alveola

Powassan virus (POWV; family Flaviviridae) is a tick-borne encephalitic virus endemic to Canada, the United States, and Russia. In the United States, POWV is transmitted by ixodid ticks, and transmission foci reflect the geographic range of these vectors, primarily Ixodes scapularis. Thus, northeastern and midwestern regions of the United States contain the highest human case burdens and prevalence of infected ticks. Notably, New York (NY) and Minnesota (MN) have a long history of POWV transmission to humans. Over time, genetic divergence has occurred in these regions, giving rise to distinct

INTRODUCTION: Salmonella Typhi (S. Typhi), a Gram-negative, serves as the etiological agent of typhoid fever. In contrast to other Salmonella serovars, S. Typhi exclusively infects humans. However, the molecular interactions it engages in with the host immune system remain inadequately characterized. This study adopts a multi-omics strategy to elucidate the immune and metabolic dynamics within the murine spleen during S. Typhi infection.
OBJECTIVES: To identify and analyze transcriptomic, proteomic, and metabolomic alterations in the spleens of mice infected with S. Typhi. By comparing these

Balantioides coli is a zoonotic ciliated protozoan that infects humans and other mammals. Conventional and ITS-based genotyping approaches have limitations that hinder precise molecular epidemiological investigations. The objective of this study was to develop a new β-tubulin gene-based approach to enhance the detection and genotyping of B. coli. We performed single-cell isolation and whole-genome sequencing on two B. coli isolates from pigs and two from guinea pigs. We then used the β-tubulin gene sequences to design PCR primers for the new genotyping assay. We validated the assay using 56 IT

Fecal contamination of surface waters presents significant human health and environmental risks. While many methods for understanding fecal contamination cannot differentiate between human and animal sources, microbial source tracking (MST) marker methods are used to identify fecal sources. To understand how to best employ MST marker data for managing fecal contamination risks, the present study compiled previously collected MST marker data (N = 12,878 samples) from across North America; each sample in the data set had been tested for fecal contamination from one or more of five sources (avian

Brucella species are Gram-negative intracellular bacterial pathogens that cause the worldwide zoonotic disease brucellosis. Brucella can infect many mammals, including humans and domestic and wild animals. Brucella manipulates various host cellular processes to invade and multiply in professional and non-professional phagocytic cells. However, the host targets and their modulation by Brucella to facilitate the infection process remain obscure. Here, we report that the host ubiquitin-specific protease, USP8, negatively regulates the invasion of Brucella into macrophages through the plasma membr

Therapeutic management of inflammation in infectious keratitis (IK) requires new strategy and targets for selective immunomodulation. Targeting host cell-type specific inflammatory responses might be a viable strategy to curtail unnecessary inflammation and reduce tissue damage without affecting pathogen clearance. This study explores the possibility of pathogen and host cell-type dependent differences in the inflammatory pathways relevant in the pathogenesis of IK. Human corneal epithelial cell line (HCEC) and phorbol 12-myristate-13 acetate (PMA) differentiated THP-1 macrophage line were inf

Photobacterium damselae comprises two subspecies, P. damselae subsp. damselae and P. damselae subsp. piscicida, that contrast remarkably despite their taxonomic relationship. The former is opportunistic and free-living but can cause disease in compromised individuals from a broad diversity of taxa, while the latter is a highly specialized, primary fish pathogen. Here, we employ new closed curated genome assemblies from Australia to estimate the global phylogenetic structure of the species P. damselae. We identify genes responsible for the shift from an opportunist to a host-adapted fish pathog

The opportunistic fungal pathogen Trichosporon asahii (T. asahii) is an important causal agent of mortality in immunocompromised patients and associated with frequent relapses, even with sufficient antifungal treatment. Investigating the proteomes of initial and recurrent isolates may help to identify within-host adaptive changes. In this study, using tandem mass tag (TMT)-labeling combined with liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS) technology, we analyzed the proteomes of two T. asahii strains that were isolated 15 years apart from the same patient who suffered

Enteroaggregative Escherichia coli (EAEC) is a significant cause of acute and chronic diarrhea, foodborne outbreaks, infections of the immunocompromised, and growth stunting in children in developing nations. There is no vaccine and resistance to antibiotics is rising. Unlike related E. coli pathotypes that are often associated with acute bouts of infection, EAEC is associated with persistent diarrhea and subclinical long-term colonization. Several secreted virulence factors have been associated with EAEC pathogenesis and linked to disease in humans, less certain are the molecular drivers of a

Microorganisms have evolved to occupy certain environmental niches, and the metabolic genes essential for growth in these locations are retained in the genomes. Many microorganisms inhabit niches located in the human body, sometimes causing disease, and may retain genes essential for growth in locations such as the bloodstream and urinary tract, or growth during intracellular invasion of the hosts' macrophage cells. Strains of Escherichia coli (E. coli) and Salmonella spp. are thought to have evolved over 100 million years from a common ancestor, and now cause disease in specific niches within

Pseudomonas aeruginosa is one of the most virulent and resistant non-fermenting Gram-negative pathogens in the clinic. Unfortunately, P. aeruginosa has acquired genes encoding metallo-β-lactamases (MβLs), enzymes able to hydrolyze most β-lactam antibiotics. SPM-1 is an MβL produced only by P. aeruginosa, while other MβLs are found in different bacteria. Despite similar active sites, the resistance profile of MβLs towards β-lactams changes from one enzyme to the other. SPM-1 is unique among pathogen-associated MβLs in that it contains "atypical" second sphere residues (S84, G121). Codon randomi

Bortezomib, a proteasome inhibitor capable of direct antitumor effects, has been shown to prevent acute graft-versus-host disease (GVHD) when administered in a short course immediately after bone marrow transplantation (BMT) in mice. However, when bortezomib is given continuously, CD4(+) T cell-mediated gastrointestinal tract damage increases GVHD mortality. To investigate the protective effects of bortezomib on other organs, we used a CD8-dependent acute GVHD (aGVHD) model of C3H.SW donor T cells engrafted into irradiated C57BL/6 recipients (minor MHC mismatch), which lack significant gut GVH

Human CJD, endemic sheep scrapie, epidemic bovine spongiform encephalopathy (BSE), and other transmissible spongiform encephalopathies (TSEs), are caused by a group of related but molecularly uncharacterized infectious agents. The UK-BSE agent infected many species, including humans where it causes variant CJD (vCJD). As in most viral infections, different TSE disease phenotypes are determined by both the agent strain and the host species. TSE strains are most reliably classified by incubation time and regional neuropathology in mice expressing wild-type (wt) prion protein (PrP). We compared v

The impacts of warming seas on the frequency and severity of bleaching events are well documented, but the potential for different Symbiodinium types to enhance the physiological tolerance of reef corals is not well understood. Here we compare the functionality and physiological properties of juvenile corals when experimentally infected with one of two homologous Symbiodinium types and exposed to combined heat and light stress. A suite of physiological indicators including chlorophyll a fluorescence, oxygen production and respiration, as well as pigment concentration consistently demonstrated

Immune mediators are involved in strain-specific manifestations of Helicobacter pylori infection, and the type of immune response is associated with production of PGE(2), which in turn influences gastric acid secretion. Acid secretion plays a pivotal role, not only in the pattern of H. pylori-induced gastritis and its consequences, but also in nonsteroidal anti-inflammatory drug (NSAID)-induced gastropathies. Mice and their transgenic modifications are widely used in Helicobacter and eicosanoid research. Using [(14)C]aminopyrine accumulation and pylorus ligation, we aimed to study acid secreti
