Bone marrow transplantation into hemochromatotic mice decreases hepatic and duodenal iron overload.

Human hereditary hemochromatosis is a disorder of iron homeostasis characterized by increased absorption of iron and its deposition in parenchymal organs. The maintenance of iron homeostasis is regulated by molecules involved in the absorption, transport, storage and redox of iron. The potential of hematopoietic stem cell therapy for liver diseases has been studied in some experimental animal models. Our objective was to evaluate the effect of bone marrow transplantation from wild type mice on the status of iron overload in Hfe knockout hemochromatotic mice (Hfe(-/-)). The transplanted cells w

A model for acute iron overload in sea bass (Dicentrarchus labrax L.).

Evaluation of several parameters involved in iron metabolism was carried out after intraperitoneal (i.p.) injection with iron dextran (IDx) in sea bass (Dicentrarchus labrax L.). After treatment, a rapid mobilization of IDx from the peritoneal cavity to other organs was observed. This was followed by a modification of normal peripheral blood iron parameters. Total iron (TI) and transferrin saturation (TS) rose rapidly, to 4.14 microg/ml and 83.7%, respectively, on day 3. In contrast, unsaturated iron binding capacity (UIBC) dropped from 3.19 microg/ml (at day 0) to 0.90 microg/ml on day 3. Tis

Blood cardioplegia increases plasma iron overload and thiol levels during cardiopulmonary bypass.

BACKGROUND: Cardiopulmonary bypass and crossclamping of the ascending aorta introduce two well-characterized phases of oxidative stress, namely, the extracorporeal circulation of blood and the reoxygenation of ischemic tissue. A feature of both forms of stress is the release of reactive and damaging oxygen species. METHODS: Forty-seven patients undergoing aortic valve replacement received either cold crystalloid, cold blood, or warm blood cardioplegia. Plasma thiol levels were measured in all groups before and during bypass. All cardiopulmonary bypass patients had, before going onto bypass, l

Alcohol consumption among South African blacks and its relationship to iron overload.

The drinking habits of 123 black men (28 from Cape Town and 95 from Pietermaritzburg) were recorded, and their weekly alcoholic iron intakes calculated. Serum ferritin levels and liver function were measured in 57 subjects (Cape Town 12, Pietermaritzburg 45). Sorghum beer, with its high iron content, was considerably more popular in Pietermaritzburg than Cape Town, and the weekly iron consumption in Pietermaritzburg was significantly higher (P less than 0.05) than in Cape Town. The serum ferritin level was significantly higher in Cape Town than in Pietermaritzburg (P less than 0.05), no signif

Secondary Iron Overload Due to Amino Acid Chelated Iron Supplementation: A Case Report Involving a Mother and Daughter.

Amino acid chelated iron (bisglycinate chelate iron, Ferrochel) has been developed and used as a food fortifier to prevent and treat iron deficiency anemia. However, its long-term use has not been described and reports of iron overload are unavailable. We report a case of a mother and daughter who were diagnosed with iron overload based on blood sampling. Serum analysis revealed elevated ferritin levels and increased transferrin saturation (TSAT). MRI of the daughter showed iron accumulation in the liver, spleen, and bone marrow with normal inflammatory response and no findings suggestive of c

Elevated hepatic iron activates NF-E2-related factor 2-regulated pathway in a dietary iron overload mouse model.

Hepatic iron overload has been associated classically with the genetic disorder hereditary hemochromatosis. More recently, it has become apparent that mild-to-moderate degrees of elevated hepatic iron stores observed in other liver diseases also have clinical relevance. The goal was to use a mouse model of dietary hepatic iron overload and isobaric tag for relative and absolute quantitation proteomics to identify, at a global level, differentially expressed proteins in livers from mice fed a control or 3,5,5-trimethyl-hexanoyl-ferrocene (TMHF) supplemented diet for 4 weeks. The expression of 7

Nifedipine prevents iron accumulation and reverses iron-overload-induced dopamine neuron degeneration in the substantia nigra of rats.

The mechanisms of iron accumulation in substantia nigra (SN) of Parkinson's diseases remain unclear. The objective of this study was to investigate effects of nifedipine on iron-overload-induced iron accumulation and neurodegeneration in SN of rats. By high performance liquid chromatography-electrochemical detection, tyrosine hydroxylase (TH) immunohistochemistry, and iron content array, we first quantified iron content and the number of dopamine neurons in SN of experimental rats treated with iron dextran. We further assessed effects of treatment with nifedipine. Our results showed that nifed

Anemia, ineffective erythropoiesis, and hepcidin: interacting factors in abnormal iron metabolism leading to iron overload in β-thalassemia.

β-Thalassemia is a genetic disorder caused by mutations in the β-globin gene and characterized by chronic anemia caused by ineffective erythropoiesis, and accompanied by a variety of serious secondary complications such as extramedullary hematopoiesis, splenomegaly, and iron overload. In the past few years, numerous studies have shown that such secondary disease conditions have a genetic basis caused by the abnormal expression of genes with a role in controlling erythropoiesis and iron metabolism. In this article, the most recent discoveries related to the mechanism(s) responsible for anemia/i

Long-term treatment of transfusional iron overload with the oral iron chelator deferiprone (L1): a Dutch multicenter trial.

We performed an open, nonrandomized, multicenter phase-II trial to evaluate the efficacy and toxicity of 1 year of treatment with the oral iron chelator deferiprone in 38 mainly nonthalassemic patients with transfusional iron overload. Initial serum ferritin varied between 996 and 11.644 micrograms/l. Patients were treated with 3-6 g of deferiprone daily. Mean urinary iron excretion (UIE) in 36 evaluable patients was 21.0 mg/24 h and was significantly higher in the patients with thalassemia than in those with myelodysplasia. Negative iron balance was achieved in 20 patients (56%). The median d

Ferroptosis in necrotizing enterocolitis: iron overload-driven intestinal injury and mechanistic insights.

Necrotizing enterocolitis (NEC) is a devastating gastrointestinal emergency that primarily affects preterm infants and remains associated with high mortality and significant long-term morbidity. Despite decades of research, the precise mechanisms underlying NEC pathogenesis remain incompletely understood, and effective targeted preventive or therapeutic strategies are limited. Ferroptosis, a regulated form of cell death driven by iron-dependent lipid peroxidation, has recently emerged as an important contributor to intestinal epithelial injury in NEC. Preterm infants are particularly vulnerabl

Steroid Biosynthesis Pathway Counteracts Iron Overload-Induced Ferroptosis in Mouse Granulosa Cells.

Iron overload is a recognized risk factor for female reproductive dysfunction, yet the underlying cellular and molecular mechanisms remain incompletely understood. In this study, the effects of iron overload on ovarian granulosa cells were investigated, and a protective role of the steroid biosynthesis pathway against ferroptosis was identified. A mouse model of ovarian iron overload was established by daily gavage of ferric citrate (FC, 120 mg/kg for 40 days). Iron-overloaded female mice exhibited disrupted estrous cycles, reduced serum estradiol levels, impaired antral follicle development,

Naringenin ameliorates iron overload-associated osteoporosis via Tfeb/p62/Nrf2-mediated antioxidation.

Postmenopausal women, elderly individuals, and transfusion-dependent patients are prone to bone marrow iron overload, which is closely associated with iron overload-associated osteoporosis (IOOP). Currently, the treatment of IOOP mainly focuses on promoting iron efflux and alleviating iron-induced damage, but the intervention value of natural active ingredients remains unclear. Naringenin (NAR), as a natural flavonoid, can regulate bone metabolism, yet its role and mechanism in IOOP have not been elucidated. In this study, an in vitro model was established by inducing MC3T3-E1 cells with ferri

Spermidine Alleviates Liver Damage Induced by Iron Overload by Inhibiting Oxidative Stress.

Liver diseases are usually accompanied by the occurrence of oxidative stress, and imbalance in hepatic iron homeostasis is a key factor leading to hepatic oxidative stress. Spermidine, as a natural antioxidant, whether it can alleviate the oxidative stress caused by iron homeostasis imbalance remains unclear. This study examined whether spermidine could mitigate iron overload in the liver induced by dextran iron. The results indicate that liver damage caused by iron overload includes disruption of the lobular structure, proliferation of Kupffer cells, and substantial iron deposition (P < 0.001

Noninvasive Quantitative CT for Diffuse Liver Diseases: Steatosis, Iron Overload, and Fibrosis.

Chronic diffuse liver disease continues to increase in prevalence and represents a global health concern. Noninvasive detection and quantification of hepatic steatosis, iron overload, and fibrosis are critical, especially given the many relative disadvantages and potential risks of invasive liver biopsy. Although MRI techniques have emerged as the preferred reference standard for quantification of liver fat, iron, and fibrosis, CT can play an important role in opportunistic detection of unsuspected disease and is performed at much higher volumes. For hepatic steatosis, noncontrast CT provides

Clodronate liposome-mediated macrophage depletion ameliorates iron overload-induced dry eye disease.

Dry eye disease (DED) is a prevalent ophthalmic disease that affects millions of people worldwide. Iron overload and macrophage inflammation have been implicated in the development of murine DED, though the specific role of macrophages under iron overload conditions remains unclear. This study aimed to establish a novel iron overload-induced mouse model of DED and investigate macrophage involvement. The model was induced via intraperitoneal injection of D-glucoside iron. Results showed that macrophage depletion via clodronate liposomes (CL) significantly mitigated iron deposit, decreased ocula

Depletion of macrophages and osteoclast precursors mitigates iron overload-mediated bone loss.

Iron is an essential element for physiological cellular processes, but is toxic in excess. Iron overload diseases are commonly associated with low bone mass. Increased bone resorption by osteoclasts as well as decreased bone formation by osteoblasts have been implicated in bone loss under iron overload conditions. However, the exact contribution of individual cell types has not yet been formally tested. In this study, we aimed to investigate the role of osteoclast precursors in iron overload-induced bone loss. To that end, we used clodronate liposomes to deplete phagocytic cells (including mac

Iron overload facilitates neonatal hypoxic-ischemic brain damage via SLC7A11-mediated ferroptosis.

Oxidative damage and cell death are involved in the pathogenesis of hypoxic-ischemic brain damage (HIBD). Ferroptosis is a newly identified mode of cell death that results from the oxidative damage induced by excessive iron. In HIBD, iron accumulates in brain tissues due to the massive destruction of red blood cells and increased permeability of the blood brain barrier vasculature, which can trigger ferroptosis. Ferroptosis is implicated in various diseases involving neuronal injury; however, the roles of iron and ferroptosis in HIBD have not been identified. In the present study, we investiga
