During malolactic fermentation (MLF) of vinification, the harsh L-malic acid undergoes transformation into the milder L-lactic acid, and via decarboxylation reactions it is catalyzed by malolactic enzymes in LAB. The use of bacterial malolactic starter cultures, which usually present challenges in the industry as the suboptimal conditions after alcoholic fermentation (AF), including nutrient limitations, low temperatures, acidic pH levels, elevated alcohol, and sulfur dioxide concentrations after AF, lead to "stuck" or "sluggish" MLF and spoilage of wines. Saccharomyces uvarum has interesting

Listeria monocytogenes is a human enteric pathogen that causes severe foodborne illness in high-risk populations. Crossing the intestinal barrier is the first critical step for Listeria monocytogenes infection. Therefore, reducing L. monocytogenes colonization and invasion of intestinal epithelium and production of virulence factors could potentially control listeriosis in humans. This study investigated the efficacy of sub-inhibitory concentration (SIC) of the plant-derived antimicrobial eugenol, either alone, or in combination with five lactic acid bacteria (LAB), namely Bifidobacterium bifi

Chiral α-hydroxy ketones and vicinal diols are valuable intermediates in pharmaceuticals and fine chemicals. 2,3-Butanediol dehydrogenase (2,3-BDH) is important for their enantioselective synthesis, but its low thermostability limits industrial use. Here, a thermostable meso-BDH from Lactococcus lactis (LlBDH) was identified using Klebsiella pneumoniae BDH (KpBDH) as the query, retaining 30.5% activity after 10 min at 100 °C. Molecular dynamics (MD) simulations revealed three unique salt bridges (K20-E45, K28-E49, R123-E170) that enhance LlBDH thermostability, and disrupting them reduced stabi

Fermented milk is known to possess potent antioxidant activity. The present study was undertaken to assess the preventive effect of fermented camel milk (FCM) prepared using lactococcus lactis subsp. cremoris against CCl4 induced kidney damage in mice. Nephrotoxicity was induced in mice by a single dose of CCl4 (10 ml/kg 0·3% olive oil, ip). Female mice were pretreated daily with FCM for 15 d. Renal damage was associated with an increase in oxidative stress parameters (lipid peroxidation, protein carbonyl and changes in antioxidant enzyme activities and non-enzymatic antioxidant) and nephropat

Trichinella spiralis is a globally distributed foodborne parasitic nematode causing zoonotic infections. The invasion and subsequent development of intestinal infective larvae within the intestinal mucosa are critical steps in T. spiralis infection of the host. Previous studies have demonstrated that aspartic protease 2 from Trichinella spiralis excretion/secretion products (TsASP2) plays a role in facilitating the invasion of host intestinal epithelial cells by the larvae; however, the underlying mechanism remains unclear. Studies have suggested that dysfunction of the intestinal barrier crea

In tissue engineering, biological, physical, and chemical inputs have to be combined to properly mimic cellular environments and successfully build artificial tissues which can be designed to fulfill different biomedical needs such as the shortage of organ donors or the development of in vitro disease models for drug testing. Inclusion body-like protein nanoparticles (pNPs) can simultaneously provide such physical and biochemical stimuli to cells when attached to surfaces. However, this attachment has only been made by physisorption. To provide a stable anchoring, a covalent binding of lactic

During the base excision repair of certain DNA lesions, the formamidopyrimidine-DNA glycosylase (Fpg) binds specifically to the DNA region containing an abasic (AP) site. Is this step affected by exposure to ionizing radiation? To answer this question, we studied a complex between a DNA duplex containing an analogue of an abasic site (the 1,3-propanediol site, Pr) and a mutated Lactococcus lactis Fpg (P1G-LlFpg) lacking strand cleavage activity. Upon irradiation of the complex, the ratio of bound/free partners decreased. When the partners were irradiated separately, the irradiated DNA still bo

Minimal inhibitory concentration and minimal bactericidal concentrations of penicillin and other beta-lactam antibiotics were determined for Streptococcus lactis in milk and trypteine soy broth. The values were always higher in milk than in broth. Minimal inhibitory concentration of penicillin was higher in solid than in liquid media. Two mutants resistant to penicillin and other beta-lactam antibiotics were obtained, and their growth rate, proteolysis, and acidification patterns determined. Tolerance toward these antibiotics was found when the mutants were grown in milk but not when they were

Lactic acid-producing bacteria (LAB) are being widely studied due to their probiotic potential. The aim of the present study was to determine and identify the presence of LAB from canine vaginal samples, as well as to evaluate their probiotic in vitro potential. Ninety-four bitches were included in the study. Vaginal samples were obtained by means of a sterile swab and streaked on Man Rogosa Sharpe agar plates. A total of 100 LAB strains were obtained and submitted to Gram stains and basic biochemical tests, which included catalase, oxidase and haemolysis tests. Thirteen strains belonging to t

We report the oral vaccination of SWISS mice with an Aro attenuated Salmonella enterica var. Typhimurium vaccine strain expressing the 14-kDa Schistosoma mansoni antigen, Sm14. Bacterial adjuvants, including (i) Lactococcus lactis expressing interleukin-12 (IL-12) and (ii) Lactobacillus delbrueckii UFV-H2b20, were also employed in oral immunization assays. Detection assays to specific IgG and IgA anti-Sm14 antibodies were performed to evaluate humoral immune responses in vaccinated mice. An increase in specific IgG titers was observed; however, no IgA production was detected. The protection le

Pathogens that have developed resistance to antibiotics pose a threat to public health. The primary goal in preventing foodborne infections is to inhibit the growth of and, subsequently, eliminate antibiotic-resistant pathogens at every stage from production to consumption. Escherichia coli, which has acquired resistance to most known antibiotics, is frequently found in chicken meat. In many countries, due to unregulated antibiotic use in poultry farming, poor hygiene in slaughterhouses, or cross-contamination, extended-spectrum beta-lactamase (ESBL)-producing E. coli has been identified as th

Plant proteins are increasingly incorporated into sustainable foods, yet their sensory performance falls short of consumer expectations. Kokumi sensation is typically associated with γ-glutamyl peptides, which enhance mouthfulness and flavour continuity via activation of the calcium-sensing receptor (CaSR). This study develops a multi-modal workflow to discover plant-derived CaSR-active peptides beyond γ-glutamyl motifs. Hemp, quinoa, and black gram were profiled using untargeted proteomics and in silico cryptide prediction. Black gram protein concentrate demonstrated the highest kokumi-genera

Dietary fiber may improve dysbiosis and contribute to the management of intestinal inflammation and muscle wasting. We examined whether low- and high-fermentable fibers differently influence dextran sulfate sodium (DSS)-induced colitis severity and muscle wasting. Male C57BL/6 J mice were assigned to four groups: nonfiber control (N), nonfiber with DSS (ND), cellulose with DSS (CD), and partially hydrolyzed guar-gum with DSS (GD), then subjected to a 27-day DSS protocol. Colitis severity was attenuated in the CD group, accompanied by increased Lactobacillus and Lactococcus lactis and decreased

Over half of the global population suffers from Helicobacter pylori infection, linked to gastrointestinal disorders and even gastric cancer. As antibiotic resistance increases, so do treatment failure rates, creating an urgent demand for more effective treatments. Plant-derived or probiotic-expressed antigenic proteins elicit immunity via oral delivery, offering a cost-effective anti-colonization strategy. Current edible plants and Lactococcus lactis expression systems leverage genetic engineering to produce recombinant Helicobacter pylori antigen proteins. Oral administration of these vaccine

Transposons are mobile genetic elements that can give rise to gene mutation and genome rearrangement. Due to their mobility, transposons have been exploited as genetic tools for modification of plants, animals, and microbes. Although a plethora of reviews have summarized families of transposons, the transposons from fermentation bacteria have not been systematically documented, which thereby constrain the exploitation for metabolic engineering and synthetic biology purposes. In this review, we summarize the transposons from the most used fermentation bacteria including Escherichia coli, Bacill

Streptococcus pyogenes is a globally prominent bacterial pathogen that exhibits strict tropism for the human host, yet bacterial factors responsible for the ability of S. pyogenes to compete within this limited biological niche are not well understood. Using an engineered recombinase-based in vivo expression technology (RIVET) system, we identified an in vivo-induced promoter region upstream of a predicted Class IIb bacteriocin system in the M18 serotype S. pyogenes strain MGAS8232. This promoter element was not active under in vitro laboratory conditions, but was highly induced within the mou

LmrR is a multidrug transcriptional repressor that controls the expression of a major multidrug transporter, LmrCD, in Lactococcus lactis. However, the molecular mechanism by which LmrR binds to structurally unrelated compounds and is released from the promoter region remains largely unknown. Here, we structurally and dynamically characterized LmrR in the apo, compound-bound and promoter-bound states. The compound-binding site of LmrR exhibits ps-μs dynamics in the apo state, and compound ligation shifts the preexisting conformational equilibrium to varying extents to achieve multidrug recogni
