Study of Protein-Protein Interactions in Septin Assembly: Multiple amphipathic helix domains cooperate in binding to the lipid membrane.

Septins are a conserved family of cytoskeletal proteins known for sensing micron-scale membrane curvature via amphipathic helix (AH) domains. While cooperative interactions in septin assembly have been suggested, the molecular mechanisms governing membrane binding and assembly remain unclear. Building on prior findings, we use all-atom molecular dynamics simulations to examine how single and paired extended AH domains, derived from Cdc12, interact with lipid bilayers. We find that a single membrane-bound AH adopts a bent conformation upon membrane association. In solution, a second AH peptide

ReVesicle: Curation and Equilibration of Lipid Vesicles for Mesoscale Simulations.

Molecular dynamics simulations provide essential atomistic insights into the organization and dynamics of complex biological membranes. However, the equilibration of large-scale, curved lipid assemblies at the all-atom level remains a significant challenge. Standard construction approaches, such as wrapping planar bilayers onto spherical meshes, frequently reverberate into structural instabilities, including membrane holes, infiltrated water, lipid flipping, and nonequilibrated densities, which hinder stable production simulations. Here, we present ReVesicle, an iterative equilibration protoco

Finite-concentration thermodynamics of amphipathic peptide adsorption on curved lipid membranes.

Curvature plays a central role in regulating peptide and protein adsorption on lipid membranes. Using a molecular-theory framework in a semi-open ensemble, we quantify how membrane curvature, ionic strength, membrane surface charge, and lipid packing jointly modulate peptide adsorption. We focus on LL-37, a human antimicrobial peptide sensitive to membrane curvature. We show that curvature sensing arises from collective effects: although local peptide-membrane free-energy profiles are only weakly curvature-dependent, the overall amount of adsorbed peptide increases markedly on curved geometrie

Confined Water Organization in the Fluid Phase of Phospholipid Self-Assemblies: The Three-Water Layer Model.

Phospholipid-based self-assemblies are frequently studied in cell membrane research and are used in numerous medical and biotechnological applications. Despite current extensive knowledge on membrane nanostructure and its mechanical properties under various environmental conditions, there is still a lack of understanding on interfacial lipid-water interactions. In this chapter, the nature of the confined water is described in the light of the recently proposed Three-Water Layer (TWL) model, discerning (i) 'headgroup water', (ii) 'perturbed water' near the membrane/water interface, and (iii) a

Molecular-level interaction of dopamine with lipids studied by sum frequency generation spectroscopy.

Dopamine is a clinically essential pharmaceutical agent extensively used in cardiology and emergency medicine. During the drug treatment, interactions between dopamine and lipid membranes are unavoidable. However, their molecular-level interaction remains insufficiently understood. In this research, sum frequency generation spectroscopy was utilized to investigate the molecular-level interaction of dopamine with the negatively charged phosphatidylglycerol (PG) lipid bilayer, in real time and in situ. Our results reveal that both the outer and inner PG leaflets remain unaffected at a low dopami

Biomembrane Adhesion to Substrates Topographically Patterned with Nanopits.

We examine the adhesion of biomembranes to substrates topographically patterned with concave nanopits and identify several universal features in the adhesion process. We find three distinct states, depending on whether the membrane remains flat above the nanopit, partially enters it, or completely adheres to it, and derive analytical conditions for the stability of these states valid for a very general class of nanopit shapes. Surprisingly, completely adhered states are always (meta)stable. We also show that the presence of many nanopits can increase or decrease the effective adhesiveness of a

Biophysics of α-synuclein induced membrane remodelling.

α-Synuclein is an intrinsically disordered protein whose aggregation is a hallmark of Parkinson's disease. In neurons, α-synuclein is thought to play important roles in mediating both endo- and exocytosis of synaptic vesicles through interactions with either the lipid bilayer or other proteins. Upon membrane binding, the N-terminus of α-synuclein forms a helical structure and inserts into the hydrophobic region of the outer membrane leaflet. However, membrane structural changes induced by α-synuclein are still largely unclear. Here we report a substantial membrane area expansion induced by the

Lipidomic adaptation and membrane physical changes underlie tamoxifen resistance in breast cancer: molecular dynamics simulation study.

Drug resistance in breast cancer (BC) is strongly linked to plasma membrane lipid remodeling, notably the loss of lipid asymmetry and elevated cholesterol (CHL) and sphingomyelin (SM) levels. We performed multi-scale molecular dynamics (MD) simulations on eight distinct lipid bilayer models-ranging from normal asymmetric (N1, N2) to cancer-mimetic symmetric states (C1-C5)-at all-atom and coarse-grained resolutions to investigate their impact on tamoxifen (TAM) permeation. Unbiased MD and umbrella sampling were used to compute potential of mean force (PMF) profiles. Our results indicate that me

Hydrophobic matching controls the tilt and stability of the dimeric platelet-derived growth factor receptor (PDGFR) β transmembrane segment.

The platelet-derived growth factor receptor β is a member of the cell surface receptor tyrosine kinase family and dimerizes upon activation. We determined the structure of the transmembrane segment in dodecylphosphocholine micelles by liquid-state NMR and found that it forms a stable left-handed helical dimer. Solid-state NMR and oriented circular dichroism were used to measure the tilt angle of the helical segments in macroscopically aligned model membranes with different acyl chain lengths. Both methods showed that decreasing bilayer thickness (DEPC-POPC-DMPC) led to an increase in the helix

Unveiling the Mechanism of Action of 7α-acetoxy-6β-hydroxyroyleanone on an MRSA/VISA Strain: Membrane and Cell Wall Interactions.

The number of cases of failure in the treatment of infections associated with resistant bacteria is on the rise, due to the decreasing efficacy of current antibiotics. Notably, 7α-Acetoxy-6β-hydroxyroyleanone (AHR), a diterpene isolated from different Plectranthus species, showed antibacterial activity, namely against Methicillin-resistant Staphylococcus aureus (MRSA) strains. The high antibacterial activity and low cytotoxicity render this natural compound an interesting alternative against resistant bacteria. The aim of this study is to understand the mechanism of action of AHR on MRSA, usin

Interaction of DDT (1,1,1-trichloro-2,2-bis(p-chlorophenyl)-ethane with liposomal phospholipids.

The influence of 1,1,1-trichloro-2,2-bis(p-chlorophenyl)ethane (DDT) and several other pesticides on the physical state of membrane phospholipids was investigated using model lipids. The thermal dependence of fluorescence intensity of the probe parinaric acid in dipalmitoylphosphatidylcholine liposomes and lipid vesicles of mixed composition were recorded. DDT was incorporated into the liposomal bilayer. The insecticide lowered the phase transition temperature and broadened the temperature range of the transition. The effects were concentration-dependent. The results may be interpreted as a so

Modeling of band-3 protein diffusion in the normal and defective red blood cell membrane.

We employ a two-component red blood cell (RBC) membrane model to simulate lateral diffusion of band-3 proteins in the normal RBC and in the RBC with defective membrane proteins. The defects reduce the connectivity between the lipid bilayer and the membrane skeleton (vertical connectivity), or the connectivity of the membrane skeleton itself (horizontal connectivity), and are associated with the blood disorders of hereditary spherocytosis (HS) and hereditary elliptocytosis (HE) respectively. Initially, we demonstrate that the cytoskeleton limits band-3 lateral mobility by measuring the band-3 m

ESR study of membrane perturbation and the lysis of liposomes induced by chlorpromazine.

The mechanism of interaction between chlorpromazine (CPZ) and artificial lipid membranes, negatively or positively charged liposomes was studied by an electron spin resonance (ESR) technique. Analysis by a 5-doxyl stearic acid (DS) spin probe indicated that regardless of the electric charge of liposomes, CPZ disordered the hydrophobic region near the surface of lipid membranes at CPZ/lipid > 1. In the same CPZ/lipid range, the lysis of liposomes was observed, and it was considered that the formation of the CPZ/lipid mixed micelles as a consequence of this collapse of liposomes would probably l

Modulation of the phospholipid dynamics and bilayer water accessibility with the membrane-associated nucleation of β-amyloid (Aβ) peptides.

Membrane disruption along the amyloidogenic aggregation of β-amyloid (Aβ) peptides is considered a molecular mechanism for the Aβ-induced cell toxicity and death. Yet, the underlying structural basis for the harmful Aβ-membrane interactions that lead to disruption remains poorly understood. We have been utilizing solid-state nuclear magnetic resonance (ssNMR) spectroscopy to explore the intermediate states of membrane-associated Aβ aggregation, as well as their roles in membrane disruption process. Aligning with this general objective, complementary quantitative ssNMR spectroscopy focusing on

The Effects of Several Natural Protoberberine Alkaloids and Cinnamic Acid Derivatives Used for Traditional Medicine on the Membrane Boundary Potential and Lipid Packing Stress.

Here we elucidated the effects of natural protoberberine alkaloids (rotundine, berberine, and nitidine) and cinnamic acid derivatives (ethyl-4-methoxycinnamate and osthole) found in Vietnamese medicinal plants, on the boundary potential of lipid bilayers and phase behavior of membrane lipids. Lipid bilayers were composed of neutral phosphatidylcholines (PC) and negatively charged phosphatidylserines (PS). Tested compounds did not produce any noticeable changes in the boundary potential with the exception of osthole, which caused a potential drop by about 30 mV independently of the membrane pho

Lipid domain formation modulates activities of snake venom phospholipase A(2) enzymes.

The goal of the present study is to elucidate the effect of lipid domain formation on activities of Naja naja atra and Bungarus multicinctus phospholipase A(2) (PLA(2)) enzymes. Sphingomyelin inhibited enzymatic activity and membrane-damaging activity of PLA(2) against egg yolk phosphatidylcholine (EYPC), while cholesterol and cholesterol sulfate abrogated the inhibitory effect of sphingomyelin. The ability of cholesterol and cholesterol sulfate to abolish the inhibitory effect of sphingomyelin was closely related to their capacity to induce domain formation in EYPC/sphingomyelin vesicles. The management of dry skin with topical emollients--recent perspectives.

Dry skin (xerosis) is a common symptom of a number of chronic skin diseases, such as atopic dermatitis, but can also be caused by environmental factors, such as cold weather and frequent showering. The condition can cause unsightliness of the skin, discomfort, itching, and can have a negative impact on patients' quality of life. This article will cover recent developments in the understanding of xerosis and its management with emollients. The stratum corneum consists of corneocytes and lipid-enriched intercellular bilayers. These are both produced from keratinocytes in a process called epiderm

Effects of the deep-sea osmolyte TMAO on the temperature and pressure dependent structure and phase behavior of lipid membranes.

We studied the interaction of lipid membranes with the deep-sea osmolyte trimethalamine-N-oxide (TMAO), which is known to stabilize proteins most efficiently against various environmental stress factors, including high hydrostatic pressure (HHP). Small-angle X-ray-scattering was applied in combination with fluorescence and infrared spectroscopy, calorimetric and AFM measurements to yield insights into the influence of TMAO on the supramolecular structure, hydration level, lipid order as well as the phase behavior of one- and three-component model biomembranes, covering a large region of the te

Two-dimensional crystallization by dialysis for structural studies of membrane proteins by the cryo-EM method electron crystallography.

Two-dimensional (2D) crystals of integral membrane proteins, comprising ordered protein reconstituted into a synthetic lipid bilayer, can be induced to form from detergent solubilized and purified membrane protein sources via the addition of exogenous lipid and the subsequent removal of the solubilizing detergent. This is most commonly accomplished by dialysis of a small volume of ternary protein-detergent-lipid mixture against a large volume of buffer, and can be carried out using common, easily available materials. Following successful crystallization, electron crystallographic data obtained
