[Defects in opsonization activity of the serum of patients with chronic arthritis].

The opsonizing capacity of sera from 22 patients with rheumatoid arthritis (RA), from 14 with psoriatic arthritis (AP), from seven with ankylosing spondylitis, and from healthy control persons was investigated by luminol-dependent chemiluminescence, induced during yeast phagocytosis of normal polymorphonuclear leukocytes. The chemiluminescence response using opsonizing sera was compared to that induced by no-opsonized yeast and the opsonizing capacity was expressed as a percentage. For the opsonization, fresh native serum, in some experiments Mg2+-EGTA and EDTA-treated serum, was used. In RA a

A potential-resolved electrochemiluminescent aptasensor for simultaneously detecting MUC1 and HER2 on breast cancer exosomes.

Breast cancer is one of the most serious threats to women's health. The simultaneous detection of tumor markers is crucial for improving the survival rate of patients. Herein, a potential-resolved electrochemiluminescent (ECL) aptasensor has been developed for the simultaneous detection of MUC1 and HER2 proteins on the surface of breast cancer cell-derived exosomes. Exosomes were specifically captured via CD63 aptamers immobilized on an indium tin oxide (ITO) electrode. Ru(bpy)32+@SiO2 nanoparticles (Ru@SiO2) and luminol@Au nanoparticles (lum@Au) labeled with the corresponding aptamers of the

Study of an Ultrasensitive Label-Free Electrochemiluminescent Immunosensor Fabricated with a Composite Electrode for Detecting the Glutamate Decarboxylase Antibody.

Antibody testing for the glutamic acid decarboxylase 65 antibody (GADA) is widely used as a golden standard for autoimmune diabetes diagnosis, while current methods for antibody testing are not sensitive enough for clinical usage. Here, a label-free electrochemiluminescent (ECL) immunosensor for detecting GADA in autoimmune diabetes is fabricated and investigated. In the designed immunosensor, a composite film including the multiwalled carbon nanotubes (MWCNTs), zinc oxide (ZnO), and Au nanoparticles (AuNPs) was prepared through nanofabrication processes to improve the performance of sensor. Usefulness of serum opsonic activity measured by chemiluminescence method to assess the invasiveness of colorectal endoscopic mucosal dissection.

Endoscopic sub-mucosal dissection (ESD) is considered as a low-invasive treatment for early-stage colorectal cancer, but the degree of invasiveness has not been well investigated. The aim of this study was to evaluate the physical stress due to colorectal ESD based on changes in serum opsonic activity (SOA). SOA was examined by measuring reactive oxygen species (ROS) produced by neutrophils using lucigenin-dependent chemiluminescence (LgCL) and luminol-dependent chemiluminescence (LmCL). Sixty-nine patients were enrolled into the study and examined SOA in the morning of the day of ESD, the nex

Direct Electrochemiluminescence Imaging of a Single Cell on a Chitosan Film Modified Electrode.

Single-cell imaging is essential for elucidating the biological mechanism of cell function because it accurately reveals the heterogeneity among cells. The electrochemiluminescence (ECL) microscopy technique has been considered a powerful tool to study cells because of its high throughput and zero cellular background light. However, since cells are immobilized on the electrode surface, the steric hindrance and the insulation from the cells make it difficult to obtain a luminous cell ECL image. To solve this problem, direct ECL imaging of a single cell was investigated and achieved on chitosan

Phagocytosis and postphagocytic reaction of cord blood and adult blood monocyte after infection with green fluorescent protein-labeled Escherichia coli and group B Streptococci.

BACKGROUND: Neonatal sepsis is characterized by an excessive inflammatory response induced by immune cells (monocytes). We investigated the initial stage of monocyte-pathogen interaction, i.e. bacterial ingestion and degradation at the single-cell level, by comparing a new flow cytometric procedure with culture methods. We also examined the hypothesis that, in terms of phagocytosis-induced cell death (PICD), phenotype, or cytokine production, cord blood monocytes (CBMO) differ from monocytes derived from adults (peripheral blood monocytes, PBMO). METHODS: Phagocytosis and intracellular degrad

Use of chemiluminescence for the serological diagnosis of bovine and ovine brucellosis with indirect and competitive enzyme-linked immunosorbent assays.

The official methods specified in the national bovine and ovine/caprine brucellosis eradication plan are the Rose Bengal and complement fixation tests. In the current phase of the eradication plan, it is often difficult to interpret the results obtained with the official tests. Consequently, additional tests that offer greater sensitivity and specificity are thus required. For this reason, two methods, the indirect chemiluminescence enzyme-linked immunosorbent assay (i-ELISA CL) and the competitive chemiluminescence ELISA (c-ELISA CL) that use a chemiluminescent substrate to determine anti-Bru

Behçet's disease associated with myelodysplastic syndromes. A case report and a review of the literature.

BACKGROUND: Behçet's disease has rarely been reported in association with myelodysplastic syndromes (MDS). Increased production of reactive oxygen species (ROS) by neutrophils has a primary role in the pathogenesis of Behçet's disease. However, decreased production of ROS by neutrophils has frequently been reported in patients with MDS. The current study was undertaken to determine the role of ROS production in a patient with Behçet's disease and MDS. METHODS: A patient with MDS with trisomy 8 who developed Behçet's disease is described and a review of the literature of patients with Behçet's

Increased production of oxygen free radicals by polymorphonuclear leukocytes in heart failure due to aortic stenosis.

Oxygen free radicals have been linked to a wide variety of cellular damage in biological systems. Polymorphonuclear (PMN) leukocytes stimulation is one of the known sources for oxygen free radicals. It has been suggested that oxygen free radicals depress the excitation-contraction coupling in cardiac muscle. It is possible that a decrease in the myocardial contractility in heart failure might be due to an increased oxygen-free-radical-producing activity of PMN leukocytes. The authors studied, therefore, the release of oxygen free radicals, as measured by luminol-dependent chemiluminescence act

Single-Atom Nanozyme Integrated Double-Clasp Transition Probe-Driven CRISPR/Cas12a Cascade for Preamplification-Free Detection of Non-Nucleic Acid Targets.

CRISPR/Cas12a diagnostics have greatly advanced nucleic acid testing, yet their extension to non-nucleic acid targets remains constrained by the intricate preamplification process and by inefficient signal transduction. Herein, we present a non-nucleic acid detection platform that integrates a CRISPR/Cas12a positive-feedback cascade with a single-atom nanozyme. In this strategy, cleavage of the designed double-clasp transition probe (DCTP) activates additional Cas12a ribonucleoprotein (RNP) complexes, enabling cascade signal amplification without a preamplification treatment. Meanwhile, the cr

Density-Dependent Modulation of the Oxidative Burst by Neutrophils in Response to Planktonic and Biofilm Growing Pseudomonas aeruginosa: When Less Is More.

The neutrophilic response to chronic Pseudomonas aeruginosa lung infection is strongly associated with collateral tissue damage impairing lung function in people with cystic fibrosis (pwCF). We recently demonstrated the concurrent presence of both biofilm bacteria and planktonic bacteria in chronically infected lungs in pwCF. However, the neutrophilic response to these distinct bacterial forms remains insufficiently characterized. To investigate this, we fractionated P. aeruginosa batch-cultures to enrich for biofilm or planktonic cells. Confocal microscopy showed that aggregates > 30 μm made

Triple-enrichment interface enabled by surfactant-modified carbon nanotube for signal-off electrochemiluminescence detection of antitumor drug mitoxantrone.

Accurate monitoring of antitumor drug is essential for therapeutic management and safety evaluation. Herein, an electrochemiluminescence (ECL) sensor was developed for sensitive detection of antitumor drug mitoxantrone (MIT) based on surfactant-modified carbon nanotubes. Cetyltrimethylammonium bromide (CTAB)-modified carboxylated multi-walled carbon nanotube (cMWCNTs) composite (CTAB@cMWCNTs) was prepared through a simple ultrasonic-assisted assembly method and immobilized onto indium tin oxide (ITO) electrode by drop-casting. By forming a stable interfacial modification layer, enhanced ECL re

Sarcoptes scabiei Induces Discrete NET Release, Ca2+ Fluxes and ROS Production Without Impairing Phagocytic Activity in Bovine Polymorphonuclear Neutrophils.

Sarcoptic mange is a skin disease caused by Sarcoptes scabiei infestations, characterized by dermatitis, pruritus, and exudative responses in both humans and animals. Biologically, the life cycle of S. scabiei is confined to the host's skin (stratum corneum), where mite-derived molecules trigger the influx of innate immune cells, including polymorphonuclear neutrophils (PMN), which play a central role in skin inflammatory responses. The antimicrobial activity of PMNs is regulated by Ca2+ fluxes and includes the generation of reactive oxygen species (ROS), degranulation, and the release of neut

Dynamic Neutrophil Subsets and Function in Lung Transplant Recipients: Insights from a One-Year Longitudinal Pilot Study.

Background: Neutrophils are key innate immune cells in peripheral blood. In recent years, sub-populations of neutrophils have been identified. In addition to the normal-density neutrophils (NDNs) in both healthy subjects and patients, low-density neutrophils (LDNs) were described in chronic inflammation and cancer. In lung transplants (LTx), neutrophils play crucial roles in reperfusion injury, acute rejection, and chronic lung allograft dysfunction. Our pilot study examines neutrophil subsets and function in LTx recipients during the first post-transplant year. Methods: We collected blood fro

Synthesis and anti-inflammatory activity of benzimidazole derivatives; an in vitro, in vivo and in silico approach.

Many non-steroidal anti-inflammatory drugs (NSAIDs) concurrently inhibit both COX-1 and COX-2, with a preference for specifically targeting COX-2 due to its significant involvement in various pathologies. In addition to COX enzymes, several other targets, including Aldose reductase, Aldo-ketoreductase family 1-member C2, and Phospholipase A2, have been identified as contributors to inflammation and a myriad of other diseases. In this context, a series of 2-substituted benzimidazole derivatives was synthesized and assessed for their anti-inflammatory potential through both in vitro and in vivo

Potential-Resolved Electrochemiluminescence for Simultaneous Determination of Triple Latent Tuberculosis Infection Markers.

A novel electrochemiluminescence (ECL) immunosensor based on the potential-resolved strategy was first developed for simultaneous determination of triple latent tuberculosis infection (LTBI) markers with high sensitivity, interferon-gamma (IFN-γ), tumor necrosis factor-alpha (TNF-α), and interleukin (IL)-2. In this work, luminol and self-prepared carbon quantum dots and CdS quantum dots were integrated onto gold nanoparticles and magnetic beads in sequence to fabricate potential-resolved ECL nanoprobes with signal amplification.
