Interrelationships among thigh muscle co-contraction, quadriceps muscle strength and the aerobic demand of walking in children with cerebral palsy.

The purpose of this study was to quantify interrelationships among thigh muscle co-contraction, quadriceps muscle strength, and the aerobic demand of walking in 13 children with spastic hemiplegic cerebral palsy (CP). During Session 1, subjects were familiarized with testing procedures, given five minutes of treadmill walking practice, and completed quadriceps muscle strength testing. In Session 2, subjects performed three 5-min walking bouts on the treadmill at 0.67 m x s(-1) to further accommodate to treadmill ambulation. During Session 3, resting VO2 was measured and subjects walked at 0.67

Effects of exercise induced muscle damage on cardiovascular responses to isometric muscle contractions and post-exercise circulatory occlusion.

PURPOSE: The aim of the present study was to investigate whether exercise-induced muscle damage (EIMD) influences cardiovascular responses to isometric exercise and post-exercise circulatory occlusion (PECO). We hypothesized that EIMD would increase muscle afferent sensitivity and, accordingly, increase blood pressure responses to exercise and PECO. METHODS: Eleven male and nine female participants performed unilateral isometric knee extension at 30% of maximal voluntary contraction (MVC) for 3-min. A thigh cuff was rapidly inflated to 250 mmHg for two min PECO, followed by 3 min recovery. He

Regulation of myosin light chain phosphatase and myofilament Ca2+ sensitivity in smooth muscle: a tale of research on smooth muscle contraction.

Contraction of both striated and smooth muscles is regulated by intracellular Ca2+ signals. The mechanisms of contraction following the Ca2+ signal are significantly different between striated and smooth muscles. The Ca2+-dependent reversible phosphorylation of the 20 kDa myosin light chain plays an essential role in regulating smooth muscle contraction. The phosphorylation theory of smooth muscle contraction was proposed in the mid-1970s. In the late 1980s, quantitative evaluation of cytosolic Ca2+ concentrations and contraction revealed that the contraction was not always associated with cha

Transcription initiation factor-1A regulates the contraction of vascular smooth muscle and maintains blood pressure.

Hypertension is a complex condition influenced by many factors. RNA polymerase I (pol I)-specific transcription initiation factor-1A (TIF-1A) regulates ribosome biosynthesis by participating in the formation of a competent pre-initiation complex. However, limited information is available regarding the role of TIF-1A in vascular smooth muscle cells (VSMCs) and its impact on blood pressure. This study investigated the biological function of TIF-1A in the modulation of smooth muscle contraction and explored the potential therapeutic targets of hypertension. Vascular smooth muscle-specific Tif-1a-

Muscle Fiber Recruitment Characteristics in Trained Older Adults: An EMG Frequency Analysis During Voluntary Contraction.

Background: Age-related declines in skeletal muscle quality and quantity contribute to frailty and sarcopenia, leading to physical dysfunction in older adults. In particular, the selective atrophy of fast-twitch fibers is closely linked to reduced lower limb strength and increased fall risk. This study aimed to evaluate the effects of habitual body weight resistance training on muscle fiber recruitment patterns during maximal voluntary contraction (MVC) and squatting movements using spectral characteristics of surface electromyography (EMG). Methods: Fifty healthy community-dwelling older adul

Non-invasive motor unit analysis reveals specific responses during maximal muscle contraction under normobaric hypoxia.

Hypoxia has been extensively studied as a stressor which pushes human bodily systems to responses and adaptations. Nevertheless, a few evidence exist onto constituent trains of motor unit action potential, despite recent advancements which allow to decompose surface electromyographic signals. This study aimed to investigate motor unit properties from noninvasive approaches during maximal isometric exercise in normobaric hypoxia. Applying a cross-over design, 18 participants (gender-matched, on average age 22.6 y, BMI 23.6 kg/m2, and bioimpedance phase angle 6.4) were exposed twice to hypoxia (

Inhibition of LIMK reduces contraction and proliferation in bladder smooth muscle induced by TGF- β1.

Overactive bladder (OAB) significantly impacts quality of life and poses substantial healthcare costs. Despite current treatments, there is an unmet need for innovative therapeutic strategies due to limited efficacy and high discontinuation rates. Our study aimed to investigate the role of transforming growth factor-β1 (TGF-β1) in bladder smooth muscle contraction and the potential of Lim kinase (LIMK) inhibitors in reducing TGF-β1-induced effects, offering a novel approach to OAB treatment. We found elevated urinary TGF-β1 levels in benign prostate hyperplasia (BPH) patients with OAB compared

The excitatory effect of hydrogen sulfide on rat colonic muscle contraction and the underlying mechanism.

H2S is a well-known relaxant regulator in muscle contraction but little attention has been paid to its excitatory effect on colonic motility. To investigate the excitation of H2S on rat colonic contraction and the underlying mechanism, the muscle contractile activity was assessed by an organ bath system, the level of substance P (SP) in the colon was detected using enzyme immunoassay kits, L-type Ca2+ channel currents (ICa,L) and large conductance Ca2+-activated K+ channel currents (IBK) in smooth muscle cells (SMCs) were measured by patch-clamp electrophysiology. The results show that the H2S

Mediators of human ureteral smooth muscle contraction-a role for erythropoietin, tamsulosin and Gli effectors.

BACKGROUND: Ureteral contractility is a poorly understood process. Contractions have been demonstrated to occur in the smooth muscle layers of the ureter. Previous work suggests the involvement of Gli family proteins and erythropoietin (EPO) in regulating mammalian ureteral smooth muscle contraction. We sought to devise a method by which the effects of these proteins and tamsulosin on distal human ureteral tissue contractility could be investigated to better understand mechanisms regulating human ureteral function. METHODS: IRB approval was obtained to procure portions of extraneous distal ur

Preserved Ca2+ handling and excitation-contraction coupling in muscle fibres from diet-induced obese mice.

AIMS/HYPOTHESIS: Disrupted intracellular Ca2+ handling is known to play a role in diabetic cardiomyopathy but it has also been postulated to contribute to obesity- and type 2 diabetes-associated skeletal muscle dysfunction. Still, there is so far very limited functional insight into whether, and if so to what extent, muscular Ca2+ homeostasis is affected in this situation, so as to potentially determine or contribute to muscle weakness. In differentiated muscle, force production is under the control of the excitation-contraction coupling process: upon plasma membrane electrical activity, the C

Heating-induced contraction in airways smooth muscle: A possible causative factor of exercise-induced bronchoconstriction.

Asthmatic patients often suffer from bronchoconstriction or asthma following breathing hot air or exposure to exercise due to raises the core body temperature. However, the direct effect of heating airways has not been studied yet. The aim of this study is to investigate the effect of heating on tracheal and bronchiolar smooth muscles. Isolated ovine tracheal strips and bronchiolar segments preparations were suspended in organ baths containing Krebs' solution for isometric tension recording. Tissues responses were examined during decreasing and elevating baths temperature (20 °C, 40 °C-45 °C a

History-dependence of muscle slack length following contraction and stretch in the human vastus lateralis.

KEY POINTS: In reduced muscle preparations, the slack length and passive stiffness of muscle fibres have been shown to be influenced by previous muscle contraction or stretch. In human muscles, such behaviours have been inferred from measures of muscle force, joint stiffness and reflex magnitudes and latencies. Using ultrasound imaging, we directly observed that isometric contraction of the vastus lateralis muscle at short lengths reduces the slack lengths of the muscle-tendon unit and muscle fascicles. The effect is apparent 60 s after the contraction. These observations imply that muscle con

Circular muscle contraction in the mice rectum plays a key role in morphine-induced constipation.

BACKGROUND: Although opioids induce intestinal muscle contraction and provoke constipation, the intestinal region(s) that contribute to the constipation have remained unclear. We report here a region-specific response of intestinal muscle contraction to morphine and its correlation with in vivo constipation. METHODS: Regions of mice small and large intestines were dissected histologically and circular muscle contractile responses were measured using isometric transducers. Bead expulsion assays were performed to assess in vivo constipation. KEY RESULTS: The strongest contraction in response t

Human skeletal muscle metabolic economy in vivo: effects of contraction intensity, age, and mobility impairment.

We tested the hypothesis that older muscle has greater metabolic economy (ME) in vivo than young, in a manner dependent, in part, on contraction intensity. Twenty young (Y; 24±1 yr, 10 women), 18 older healthy (O; 73±2, 9 women) and 9 older individuals with mild-to-moderate mobility impairment (OI; 74±1, 7 women) received stimulated twitches (2 Hz, 3 min) and performed nonfatiguing voluntary (20, 50, and 100% maximal; 12 s each) isometric dorsiflexion contractions. Torque-time integrals (TTI; Nm·s) were calculated and expressed relative to maximal fat-free muscle cross-sectional area (cm2), an

A new metabotropic role for L-type Ca(2+) channels in vascular smooth muscle contraction.

Vascular smooth muscle cells (VSMCs) contraction can be evoked by the rise of cytosolic [Ca(2+)] owing to transmembrane Ca(2+) influx or sarcoplasmic reticulum (SR) Ca(2+) release. Although the classical ionotropic role of voltagedependent (L-type) Ca(2+) channels (VGCCs) is known, we review here data suggesting a new metabotropic function of VGCCs in vascular smooth muscle cells. VGCCs can trigger Ca(2+) release from the SR in the absence of extracellular Ca2+. During depolarization, VGCCs can activate G proteins and phospholipase C (PLC)/inositol 1,4,5-trisphosphate (InsP3) pathway leading t

Calcium influx through L-type channels attenuates skeletal muscle contraction via inhibition of adenylyl cyclases.

Skeletal muscle contraction is triggered by acetylcholine induced release of Ca(2+) from sarcoplasmic reticulum. Although this signaling pathway is independent of extracellular Ca(2+), L-type voltage-gated calcium channel (Cav) blockers have inotropic effects on frog skeletal muscles which occur by an unknown mechanism. Taking into account that skeletal muscle fiber expresses Ca(+2)-sensitive adenylyl cyclase (AC) isoforms and that cAMP is able to increase skeletal muscle contraction force, we investigated the role of Ca(2+) influx on mouse skeletal muscle contraction and the putative crosstal

A role of stretch-activated potassium currents in the regulation of uterine smooth muscle contraction.

Rates of premature birth are alarming and threaten societies and healthcare systems worldwide. Premature labor results in premature birth in over 50% of cases. Preterm birth accounts for three-quarters of infant morbidity and mortality. Children that survive birth before 34 weeks gestation often face life-long disability. Current treatments for preterm labor are wanting. No treatment has been found to be generally effective and none are systematically evaluated beyond 48 h. New approaches to the treatment of preterm labor are desperately needed. Recent studies from our laboratory suggest that
