Targeting fibrosis and steatosis: Nebivolol multi-pathway modulator ameliorates experimental non-alcoholic steatohepatitis in rats through AMPK/mTOR and TGF-β1/α-SMA pathways.

Non-alcoholic steatohepatitis (NASH) is a progressive hepatic disease that can advance into hepatocellular fibrosis, cirrhosis, and carcinoma. This study evaluates the therapeutic potential of Nebivolol on a diet-induced NASH rat model. Out of 32 male Wistar albino rats, 24 rats were fed a NASH-inducing diet for 8 weeks. The remaining were kept on a normal diet as healthy controls. After the induction phase, the NASH group was divided into three equal subgroups: an untreated NASH group and two treatment groups receiving 5 or 10 mg/kg/day of Nebivolol orally for six weeks. The NASH model demons

Gene expression adaptation of metastases to their host tissue.

Adaptation of metastatic cells to their host tissue determines the pathogenicity of cancer. Yet, it remains elusive to what extent the host environment drives gene expression programs in metastatic cells. We present a new concept to identify adaptive mechanisms that enable metastases to establish themselves in a novel tissue context. We generated quadruple-paired single-cell RNA-sequencing data from malignant and benign tissues from untreated donors with colorectal adenocarcinoma and liver metastasis. Utilizing a computational approach, we deduce tissue-adaptive expression patterns by identify

Starvation and infection: The role of sickness-associated anorexia in metabolic adaptation during acute infection.

Sickness-associated anorexia, the reduction in appetite seen during infection, is a widely conserved and well-recognized symptom of acute infection, yet there is very little understanding of its functional role in recovery. Anorexic sickness behaviours can be understood as an evolutionary strategy to increase tolerance to pathogen-mediated illness. In this review we explore the evidence for mechanisms and potential metabolic benefits of sickness-associated anorexia. Energy intake can impact on the immune response, control of inflammation and tissue stress, and on pathogen fitness. Fasting medi

[New oral antidiabetic agents--clinical perspectives].

In recent years new pharmacological agents have emerged, which enable us to widen the spectrum of diabetes management based on clear pathophysiological concepts. One should first of all mention the DPP4-inhibitors and the incretin mimetics, which have the potential of restoring the incretin effect in patients with type 2 diabetes mellitus. Moreover, it has been shown that the agent rimonabant, which was first used in order to achieve weight reduction, also contributed to the significant improvement of metabolic syndrome's parameters. Recent studies have shown that the dual alpha/gamma-PPAR-ago

The effect of PPARgamma ligands on the adipose tissue in insulin resistance.

Insulin resistance is frequently accompanied by obesity and both obesity and type 2 diabetes are associated with a mild chronic inflammation. Elevated levels of various cytokines, such as TNF-alpha and IL-6, are typically found in the adipose tissue in these conditions. It has been suggested that many cytokines produced in the adipose tissue are derived from infiltrated inflammatory cells. However, the adipose tissue itself has proven to be an important endocrine organ, secreting several hormones and cytokines, usually referred to as adipokines. Peroxisome proliferator-activated receptor (PPAR

Peroxiredoxin II as a metabolic gatekeeper against aflatoxin B1-induced hepatic steatosis: restoration of PPARα/γ balance via mitochondrial ROS scavenging.

Aflatoxin B1 (AFB1), a pervasive food contaminant affecting approximately 25% of global staple crops, represents an underrecognized environmental risk factor for nonalcoholic fatty liver disease (NAFLD). This study investigates the protective role of peroxiredoxin II (Prx II), a mitochondrial antioxidant enzyme, against AFB1-induced hepatic steatosis and elucidates the underlying molecular mechanisms. Bioinformatic analysis of the publicly available AFB1-exposed liver transcriptome dataset (GSE26838) combined with gain- and loss-of-function studies in AML12 hepatocytes revealed that AFB1 induc

Conserved gene expression patterns in the placenta underlie pregnancy complications associated with hypoxia in mice and humans.

During pregnancy hypoxia leads to complications like fetal growth restriction (FGR). Maternal inhalation hypoxia during late murine pregnancy impairs placental phenotype and fetal development in a severity-dependent manner. To identify the molecular mechanisms and sex-specificity of these effects, placentas from pregnant mice exposed to moderate (13% O₂) or severe (10% O₂) hypoxia were analysed using RNA-sequencing, qPCR, western blotting, histology, and nutrient transport assays. Transcriptomic profiling of male 13% O2 placentas revealed differential gene expression regulating calcium binding

Saroglitazar at the Crossroads of Metabolic and Neurodegenerative Disease: A Critical Review of the Hepato-Neuro Axis and Translational Horizons.

Non-Alcoholic Fatty Liver Disease (NAFLD) is increasingly recognized as a systemic disorder with implications far beyond the liver, notably in the progression of cognitive decline and neurodegenerative disorders such as Alzheimer's disease (AD) and traumatic brain injury (TBI). At the center of this liver-brain axis lies Saroglitazar (SGZ), a dual PPAR-α/γ agonist initially developed for diabetic dyslipidemia. Emerging evidence suggests that SGZ exerts neuroprotective effects via multiple molecular mechanisms, including modulation of oxidative stress, inflammation, and mitochondrial integrity.

A Medium-Chain Fatty Acid Analogue Prevents Intestinal Failure-Associated Liver Disease in Preterm Yorkshire Piglets.

BACKGROUND & AIMS: At least 20%-30% of patients with intestinal failure receiving long-term parenteral nutrition will develop intestinal failure-associated liver disease (IFALD), for which there are few therapeutic options. SEFA-6179 is a first-in-class structurally engineered medium-chain fatty acid analogue that acts through GPR84, PPARα, and PPARγ agonism. We hypothesized that SEFA-6179 would prevent biochemical and histologic liver injury in a preterm piglet model of IFALD. METHODS: Preterm Yorkshire piglets were delivered by cesarean section, and parenteral nutrition was provided for 14

Multiview Reveals the Combined Effects of Chiral Pesticide Pydiflumetofen and Cadmium on Glucolipid Metabolism in Zebrafish Liver.

The effects of the chiral fungicide pydiflumetofen (PYD) and the heavy metal cadmium (Cd) on hepatic glucolipid metabolism in zebrafish were investigated. S-PYD was preferentially accumulated in zebrafish, and Cd increased the accumulation of PYD by 1.22-1.78 times depending on the enantiomer. The mitochondria damage, endoplasmic reticulum hyperplasia, hepatic steatosis, the increase of glucose, triglyceride (TG) and oxidative stress, and the significant alterations of the peroxisome proliferator activated receptor signaling pathway revealed glucolipid metabolism disorders in zebrafish liver.

D-allulose enhances lipid oxidation in HepG2 cells via peroxisome proliferator-activated receptor α (PPARα).

Lipid accumulation in hepatocytes in non-alcoholic steatohepatitis (NASH) is attributed partly to loss of insulin-responsiveness and/or an increased pro-inflammatory state. Since the rare sugar D-allulose has insulin mimetic and anti-inflammatory properties, its effects on lipid accumulation in liver-derived cells was tested. In HepG2 cells exposed to 200 μM oleic acid for 72 h, D-allulose treatment decreased intracellular lipid accumulation with an IC50 = 0.45 ± 0.07 mM. A similar effect was observed in cells treated with 10 μM gemfibrozil. D-allulose and gemfibrozil treatment increased oleic
