Discovery of autophagy as a universal mechanism for sex steroid synthesis in human ovary and testis.

We recently discovered that lipophagy is a key mechanism to provide free cholesterol required for steroid biosynthesis in human ovary and testis. Pharmacological or genetic inhibition of autophagy by silencing of the autophagy-related (ATG) genes BECN1 (BECLIN1) and ATG5 resulted in a significant reduction in basal and gonadotropin-stimulated estradiol, progesterone (P4) and testosterone production in the ex-vivo explant tissue and cell culture models for ovary and testis. We also described a new mechanism of action for gonadotropin hormones, i.e., follicle stimulating hormone (FSH) and human

Concomitant western diet and chronic-binge alcohol dysregulate hepatic metabolism.

BACKGROUND AND AIMS: There is significant overlap between non-alcoholic fatty liver disease (NAFLD) and alcohol-associated liver disease (ALD) with regards to risk factors and disease progression. However, the mechanism by which fatty liver disease arises from concomitant obesity and overconsumption of alcohol (syndrome of metabolic and alcohol-associated fatty liver disease; SMAFLD), is not fully understood. METHODS: Male C57BL6/J mice were fed chow diet (Chow) or high-fructose, high-fat, high-cholesterol diet (FFC) for 4 weeks, then administered either saline or ethanol (EtOH, 5% in drinkin

Treatment with a new barbituric acid derivative suppresses diet-induced metabolic dysfunction and non-alcoholic fatty liver disease in mice.

INTRODUCTION: Nonalcoholic fatty liver disease (NAFLD) is the most common chronic liver disease, often accompanied by obesity, diabetes, and increased risks of depression and anxiety. Currently, there are no FDA-approved drugs to treat NAFLD and its related systemic symptoms. Previously, we identified a new barbituric acid derivative (BA-5) that expressed effectiveness against fibrosis and drug-resistant hepatocellular carcinoma. AIMS: This study investigated the potential of BA-5 against high-fat diet (HFD)-induced NAFLD and mood disorders in mice. MAIN METHODS: Six-weeks-old male C57BL/6 m

Mechanisms of autophagic responses to altered nutritional status.

Autophagy is a dynamic process and critical for cellular remodeling and organelle quality control. In response to altered nutritional status (e.g., fasting and feeding), autophagic activity is finely tuned by transcriptional, posttranslational, and epigenetic regulations via various signaling pathways, including energy sensors (e.g., mechanistic target of rapamycin (mTOR)/ AMP-activated protein kinase - Unc-51 Like Autophagy Activating Kinase 1, mTORC1- WD Repeat Domain, Phosphoinositide Interacting 2, mTORC1- transcription factor EB, perilipin 5- Sirtuin 1, and Sirtuin 1-mediated deacetylatio

Wild-type GBA1 increases the α-synuclein tetramer-monomer ratio, reduces lipid-rich aggregates, and attenuates motor and cognitive deficits in mice.

Loss-of-function mutations in acid beta-glucosidase 1 (GBA1) are among the strongest genetic risk factors for Lewy body disorders such as Parkinson's disease (PD) and Lewy body dementia (DLB). Altered lipid metabolism in PD patient-derived neurons, carrying either GBA1 or PD αS mutations, can shift the physiological α-synuclein (αS) tetramer-monomer (T:M) equilibrium toward aggregation-prone monomers. A resultant increase in pSer129+ αS monomers provides a likely building block for αS aggregates. 3K αS mice, representing a neuropathological amplification of the E46K PD-causing mutation, have d

Depot-dependent effects of adipose tissue explants on co-cultured hepatocytes.

We have developed an in vitro hepatocyte-adipose tissue explant (ATE) co-culture model enabling examination of the effect of visceral and subcutaneous adipose tissues on primary rat hepatocytes. Initial analyses of inflammatory marker genes were performed in fractionated epididymal or inguinal adipose tissues. Expressions of inflammation related genes (IL-6, TNF-α, COX-2) were higher in the inguinal than the epididymal ATE. Similarly, expressions of marker genes of macrophage and monocyte (MPEG-1, CD68, F4/80, CD64) were higher in the stromal vascular fraction (SVF) isolated from inguinal ATE

Acute deletion of PLIN5 in brown adipocytes causes mitochondrial dysfunction and cold intolerance.

Cold exposure of mice is associated with adaptive molecular and organellar changes in brown adipose tissue (BAT) that promote thermogenesis to defend body temperature. We previously reported that the lipid droplet protein Perilipin 5 (PLIN5) robustly increases in BAT during acute exposure of mice to cold. We demonstrated that chronic induction of BAT PLIN5 within the physiological range in mice housed at room temperature mimics the effects of cold exposure in terms of increased thermogenic gene expression in BAT, increased BAT mitochondria cristae packing, and increased uncoupled mitochondrial

Mitochondrial Communication with Cellular Organelles in the Pathogenesis of Fatty Liver Disease in Domestic and Model Animals.

Fatty liver disease represents a major metabolic disorder affecting domestic animals worldwide, with significant implications for animal health, welfare, and agricultural productivity. Disrupted communication between mitochondria and other organelles-particularly the endoplasmic reticulum, lipid droplets, and lysosomes-plays a critical role in disease pathogenesis. This review synthesizes knowledge on inter-organellar communication across domestic animals, with emphasis on species-specific adaptations. We address the "Dairy Cow Paradox"-periparturient dairy cows develop severe hepatic steatosi

Fabrication of a Bioactive Human Adipose Extracellular Matrix Allograft Using Supercritical Carbon Dioxide.

PURPOSE: Autologous fat grafting is widely used for soft tissue augmentation but hindered by unpredictable volume retention and donor site morbidity. Decellularized adipose extracellular matrix (DAM) represents a promising alternative. Herein we evaluate an injectable, human-derived DAM decellularized via supercritical carbon dioxide (scCO2, sDAM), promising a gentler, more eco-friendly isolation of matrix components than conventional chemical decellularization. METHODS: Human adipose tissue was homogenized, delipidated, then underwent scCO2 cleansing, two cycles of scCO2 decellularization, c

Enhancing Autologous Fat Graft Retention: The Impact of Inactive Vitamin D3 on Immune Modulation and Extracellular Matrix Remodeling in a Swine Model.

Autologous fat grafting (AFG) is a widely utilized surgical technique, but graft survival rates remain inconsistent and unpredictable. The aim of this study was to evaluate the efficacy of oral vitamin D3 (VD3) on long-term AFG retention in a porcine model and examine its mechanisms. Inguinally harvested adipose tissue from 3 female Yucatán pigs was autologously grafted into 16 dorsal sites (5 cc per graft). Pig 1 received no treatment; Pig 2 received oral VD3 (100 K IU) thrice weekly postoperatively; Pig 3 received oral VD3 (100 K IU) thrice weekly for 2 weeks preoperatively and continuously

Crocins ameliorate acute kidney injury in glycerol-induced rhabdomyolysis by targeting the PLIN1/PPARs signaling pathway.

BACKGROUND: Rhabdomyolysis (RM) is a potentially life-threatening syndrome characterized by skeletal muscle damage, with acute kidney injury (AKI) being its most severe complication. Currently, no effective treatment exists for RM-induced AKI. Crocins, the major bioactive constituents extracted from the stigma of Crocus sativus L. (Saffron), possess diverse pharmacological activities. OBJECTIVES: This study aimed to explore the pharmacological effect and possible mechanism of crocins in the treatment of RM-induced AKI. METHOD: Network analysis was first applied to predict potential targets o

PCSK9 inhibition attenuates alcohol-induced cardiovascular dysfunction and links hepatic lipid accumulation to impaired myocardial contractile reserve.

Excessive alcohol consumption accelerates cardiovascular aging by promoting oxidative stress, inflammation, lipid dysregulation, fibrotic remodeling, and loss of ventricular-vascular reserve. PCSK9, a key regulator of cholesterol metabolism, has emerged as a mediator of age-related cardiovascular dysfunction and alcohol-associated liver and neurovascular injury. We investigated whether PCSK9 inhibition protects against alcohol-induced cardiovascular dysfunction and associated cardiac-hepatic injury in rats. Male Sprague-Dawley rats were assigned to pair-fed control or 35% ethanol liquid diet g

Lipid droplet-mitochondria contact sites as druggable spatial-pharmacology targets in respiratory disease: cross-cell-type mechanisms and translational strategies.

Lipid droplets and mitochondria form regulated contact sites whose molecular composition, phosphorylation state and spatial geometry now behave as measurable, druggable variables in living cells. The molecular toolkit has been mapped in liver, muscle and adipose tissue, yet its application to respiratory disease has lagged despite a lung-specific dependence on lipid biology that spans surfactant biogenesis, alveolar-macrophage lipid handling and fibroblast-driven remodelling. This review argues that the lipid-droplet-mitochondria contact site is an emerging spatial-pharmacology target in respi

JMJD8 facilitates hepatic lipid deposition and metabolic dysfunction.

We previously identified Jumonji domain-containing 8 (JMJD8) as a regulator of lipid droplet (LD) hypertrophy in adipocytes through modulation of AMPK-dependent perilipin-2 (PLIN2) phosphorylation. Given PLIN2's established role in hepatic steatosis, we investigated whether JMJD8 also regulates lipid accumulation in the liver. Here, we report that JMJD8 expression is significantly elevated in the livers of mice fed either a high-fat diet (HFD) or the metabolic-associated fatty liver disease (MAFLD)-inducing Gubra Amylin NASH (GAN) diet. To define the metabolic role of JMJD8 in the liver, we ge

A multitissue transcriptomic analysis reveals a potential mechanism whereby Brevibacillus laterosporus S62-9 promotes broiler growth.

Brevibacillus laterosporus S62-9 has been shown to improve broiler growth performance and immunity. In the present study, we aimed to evaluate the effects of B. laterosporus S62-9 on the immunity and lipid metabolism of broilers by means of transcriptomic analysis. A total of 160 1-day-old broilers were randomly allocated to a S62-9 group, the diet of which was supplemented with 106 CFU/g B. laterosporus S62-9 daily, and a control group, which was not. After 42 d of feeding, the broilers in the S62-9 group had higher body mass (7.2%) and feed conversion ratio (5.19%) than the control group. Su

TREM2 regulates microglial lipid droplet formation and represses post-ischemic brain injury.

Triggering receptor expressed on myeloid cells 2 (TREM2) is a transmembrane receptor protein predominantly expressed in microglia within the central nervous system (CNS). TREM2 regulates multiple microglial functions, including lipid metabolism, immune reaction, inflammation, and microglial phagocytosis. Recent studies have found that TREM2 is highly expressed in activated microglia after ischemic stroke. However, the role of TREM2 in the pathologic response after stroke remains unclear. Herein, TREM2-deficient microglia exhibit an impaired phagocytosis rate and cholesteryl ester (CE) accumula

Regulation of bone and fat balance by Fructus Ligustri Lucidi in ovariectomized mice.

CONTEXT: Fructus Ligustri Lucidi (FLL), a commonly used herb of traditional Chinese medicine (TCM), is the fruit of Ligustrum lucidum Ait. (Oleaceae). The ethanol extract of FLL is a potential candidate for preventing and treating postmenopausal osteoporosis (PMOP) by nourishing the liver and kidneys. OBJECTIVE: This study determines whether an ethanol extract of FLL has anti-osteoporotic effects in ovariectomized (OVX) mice and explores the underlying mechanism. MATERIALS AND METHODS: The OVX model of eight-week-old C57BL/6J female mice was taken, and ovariectomy was used as PMOP. Mice were

Targeting Myofibroblasts as a Treatment Modality for Dupuytren Disease.

PURPOSE: Currently, no treatment corrects the contractile nature of Dupuytren myofibroblasts (DMFs) or prevents recurrence following surgery. Antifibrotic and proadipogenic growth factors are released when adipose-derived stem cells (ASCs) are cultured with platelet-rich plasma (PRP), a platelet concentration from whole blood. Reprograming myofibroblasts into adipocytes via growth factors is proposed as a powerful potential tool to target fibrosis. We aimed to assess whether the combination of ASCs and PRP reprograms DMFs into adipocytes in vitro and alters their contractile nature in vivo. M

LAMC1-mediated preadipocytes differentiation promoted peritoneum pre-metastatic niche formation and gastric cancer metastasis.

Gastric cancer is anatomically proximal to peritoneum. Gastric cancer peritoneal metastasis is a complex biological process which is corresponded with disharmony within dysfunctional adipose tissue and metabolism reprogramming. Laminin gamma 1 (LAMC1) is highly expressed in cancer cells of peritoneal metastatic sites, however, the mechanism of LAMC1-metiated gastric cancer metastases to adipose tissue-rich peritoneum remains unclear. In our study, immunohistochemical staining, single cell sequencing, a co-culture model, luciferase reporter, RNA immunoprecipitation (RIP), Chromatin immunoprecip

High-Fructose Diet Increases Inflammatory Cytokines and Alters Gut Microbiota Composition in Rats.

High-fructose diet induced changes in gut microbiota structure and function, which have been linked to inflammatory response. However, the effect of small or appropriate doses of fructose on gut microbiota and inflammatory cytokines is not fully understood. Hence, the abundance changes of gut microbiota in fructose-treated Sprague-Dawley rats were analyzed by 16S rRNA sequencing. The effects of fructose diet on metabolic disorders were evaluated by blood biochemical parameter test, histological analysis, short-chain fatty acid (SCFA) analysis, ELISA analysis, and Western blot.
