Indirect immunofluorescent staining was performed on cryostat sections of skin biopsy specimens from 21 patients. In six of the 11 cases finally diagnosed as Mediterranean spotted fever, coccobacillary forms of Rickettsia conorii were identified in these sections. A factor recognized as contributing to false-negative results was prior treatment with tetracycline. No false-positive results were obtained for 10 patients whose final diagnosis was not Mediterranean spotted fever. This laboratory test can aid in the early diagnosis of severe and atypical forms of Mediterranean spotted fever.

BACKGROUND: Pathogen colonization inside tick tissues is a significant aspect of the overall competence of a vector. Amblyomma maculatum is a competent vector of the spotted fever group rickettsiae, Rickettsia parkeri. When R. parkeri colonizes its tick host, it has the opportunity to dynamically interact with not just its host but with the endosymbionts living within it, and this enables it to modulate the tick's defenses by regulating tick gene expression. The microbiome in A. maculatum is dominated by two endosymbiont microbes: a Francisella-like endosymbiont (FLE) and Candidatus Midichlori

Rickettsialpox is a cosmopolitan, mite-borne, spotted fever rickettsiosis caused by Rickettsia akari. The disease is characterized by a primary eschar, fever, and a papulovesicular rash. Rickettsialpox was first identified in New York City in 1946 and the preponderance of recognized cases in the United States continues to originate from this large metropolitan center. The most recently isolated U.S. strain of R. akari was obtained more than a half century ago. We describe the culture and initial characterization of five contemporaneous isolates of R. akari obtained from eschar biopsy specimens

A prospective 12-month study was conducted throughout 1998 to determine the frequency of selected bacterial zoonoses as causes of fever among hospitalized Bedouins in southern Israel. One or more zoonoses were diagnosed in 30 (27%) of 110 patients admitted with fever. Brucellosis was diagnosed in 9 (8%), rickettsial infections in 20 (18%), and ehrlichiosis in 2 (2%), one of whom had also evidence of rickettsial spotted fever infection. None of the patients was diagnosed with Q fever. Compared with patients without zoonoses, patients with zoonoses were younger (P = 0.01), fewer of them had unde

Serological tests were performed to investigate extent of tick-borne diseases in dogs infested with Rhipicephalus sanguineus at a kennel in Okayama Prefecture. Three of 22 dogs (13.6%) were positive for Ehrlichia canis. Two of 19 dogs (10.5%) were positive for Rickettsia japonica. Three of 22 dogs (13.6%) were positive for Babesia gibsoni. None of these animals were positive for Coxiella burnetii or Hepatozoon canis. A microfilaria was detected in a drop smear of hemolymph from an engorged female tick, however, species was not determined. It is possible that these ticks can transmit pathogens

Bacillary angiomatosis (BA) and chronic bartonellosis are bacterial infections of humans which result in an unusual vascular proliferative tissue response. In order to determine their phylogenetic relationships, we have determined greater than 95% of the 16S rRNA sequences for these two organisms by amplification directly from infected BA tissue and from a Bartonella bacilliformis lyophilized culture. The BA agent and B. bacilliformis are closely related alpha-proteobacteria (98.5%), although the BA agent is more closely related to Rochalimaea quintana (99.1%). Contrary to previous belief, the

Twenty-six calves which had not previously grazed tick-infested pasture were divided into two equal groups. On May 26, 1988 (day 0) they were turned out into a field of rough grazing where cases of redwater fever had occurred the previous spring. Seven, 14, 21 and 28 days after the start of the trial the animals in one group each received an intramuscular injection of 20 mg/kg bodyweight of long-acting oxytetracycline. During the 60 days of the trial the animals received a severe tick-borne fever challenge, in some cases combined with a redwater fever challenge. An unforeseen complicating fact

The rickettsial disease of man found only in Asia is mite-borne (scrub) typhus, caused by Rickettsia tsutsugamushi. Unique to southern Japan is a little-known human mononucleosis-like disease caused by Rickettsia sennetsu. In 1981 and 1982, there was a remarkable resurgence in the number of reported cases of mite-borne typhus in Japan after some years of virtual absence. Recent studies of R sennetsu have resulted in its reclassification to the genus Ehrlichia, members of which until now have been exclusively pathogens of animals. The historical background of ecologic investigations, in Malaysi

Delayed type hypersensitivity (DTH) was studied in the macrophage migration inhibition test, antibody titers in the CFT and functional activity of spleen macrophages, obtained from guinea-pigs infected with R. conori, in cell cultures. No direct correlation between antibody synthesis and functional macrophage activity accompanied by the development of DTH was revealed. The most pronounced protective function of macrophages could be observed during the decrease of antibody titers and the extinction of DTH.

Case histories of boutonneuse fever are described in order to exemplify major characteristics of most rickettsioses: recent travel history, feverish illness with severe headache, skin eruptions and histological findings. Up-to-date informations concerning the epidemiologic situation of typhus, scrub typhus and Rocky Mountain spotted fever are given. The characteristics of Q fever and the possibility of rickettsial laboratory infections are pointed out.

BACKGROUND: Ticks are hematophagous arthropods that transmit various bacterial, viral, and protozoan pathogens of public health significance. The lone star tick (Amblyomma americanum) is an aggressive human-biting tick that transmits bacterial and viral pathogens, and its bites are suspected of eliciting the alpha-gal syndrome, a newly emerged delayed hypersensitivity following consumption of red meat in the United States. While ongoing studies have attempted to investigate the contribution of different tick-inherent factors to the induction of alpha-gal syndrome, an otherwise understudied asp

Rickettsia species are gram-negative intracellular, small pleomorphic coccobacilli in the Rickettsiaceae family. This genus is serologically and genotypically divided into four groups as spotted fever group, typhus group, Rickettsia belli and Rickettsia canadensis. Rickettsia conorii (R.conorii subsp. conorii) in the spotted fever group was reported to cause mediterranean spotted fever in Europe, especially in mediterranean countries including Turkey. The major vectors of Rickettsia species are ticks, and in some species fleas or mites. In this report a case with R.conorii infection was presen

Mites are mostly ubiquitous, bothersome pests with few species of medical importance and, of these, most are scabies mites, trombiculid larval mites and animal and plant mites. All patients with scabies and their close household, institutional and sexual contacts should be informed that scabies is a highly transmissible ectoparasitic infestation and that several topical treatments and an effective oral treatment are readily available and highly effective at present. Sexually active patients with scabies should be screened for other sexually transmitted diseases, particularly human immunodefici

The presence, internal distribution, and phylogenetic position of endosymbiotic bacteria from four species of specific-pathogen-free ticks were studied. These included the hard ticks Ixodes scapularis (the black-legged tick), Rhipicephalus sanguineus (the brown dog tick), and Haemaphysalis longicornis and the African soft tick Ornithodoros moubata. PCR assays for bacteria, using two sets of general primers for eubacterial 16S and 23S rRNA genes (rDNAs) and seven sets of specific primers for wolbachial, rickettsial, or Francisella genes, indicated that I. scapularis possessed symbiotic ricketts

Complement fixation and indirect micro-immunofluorescence (IF) tests were compared for specificity and sensitivity in detection of antibodies against Rickettsia conorii, in acute and convalescent human sera. Purified particulate rickettsial antigens were used. Good agreement between these two tests was found. Use of specific anti-IgG and anti-IgM sera in parallel IF tests made it possible to differentiate between recent and past infections.

The endosymbiont Wolbachia has been detected in a range of filarial nematodes and parasitic mites and is known to affect host reproductive compatibility and potentially evolutionary processes. PCR of Wolbachia surface protein (wsp), ftsZ and 16SrRNA genes from individual Sarcoptes scabiei mites obtained from a series of individual hosts, and database searches of an S. scabiei var. hominis EST library failed to detect Wolbachia genes. Therefore, Wolbachia appears not to be involved in the genetic subdivision observed between varieties of host-associated S. scabiei or, involved in the inflammato

In 3 urban areas in Selangor, Peninsular Malaysia between 1973 and 1981, blood from 4084 dogs was examined for haematozoa. The following frequencies were found: Babesia gibsoni 17.7%; microfilariae of Dirofilaria immitis 9.6%; Hepatozoon canis 1.2%; B. canis 1.1%; Ehrlichia canis 0.2%; Trypanosoma evansi 0.1%. A detailed examination of B. gibsoni infections and microfilariasis due to D. immitis with regards to monthly distribution, breed frequency, sex and age, revealed that pedigree and non-pedigree dogs were equally susceptible to Babesia and microfilariae infections.
