Measurements of intracellular pH in Necturus antral mucosa by microelectrode technique.

Intracellular pH (pHi) was measured in the surface epithelial cells of Necturus antrum using pH-sensitive intracellular microelectrodes. Electrodes were prepared by filling 10- to 20-M omega resistance glass microelectrodes with a H+ ion-selective exchange resin and calibrated before use in solutions of known pH 4.0-8.0. The electrode response (n = 15) was linear (r = 0.93; P less than 0.001) with a slope of 52.1 +/- 2.3 mV/pH unit. Antral mucosa was mounted in a modified Ussing chamber and pHi was determined from the difference between the potentials recorded by intracellular H+-selective and

Irreversible conduction block in isolated nerve by high concentrations of local anesthetics.

BACKGROUND: Delivery of large doses of local anesthetics for spinal anesthesia by repeated injections or continuous infusion could expose the cauda equina to concentrations of drug that may be neurotoxic per se. We studied this possible neurotoxic effect by assessing recovery from conduction blockade of desheathed peripheral nerves after exposure to some of the local anesthetic solutions commonly used for spinal anesthesia. METHODS: The reversibility of conduction blockade was studied in desheathed bullfrog sciatic nerves, using the sucrose-gap method for recording compound action potentials,

Effect of hypertonicity on augmentation and potentiation and on corresponding quantal parameters of transmitter release.

Augmentation and (posttetanic) potentiation are two of the four components comprising the enhanced release of transmitter following repetitive nerve stimulation. To examine the quantal basis of these components under isotonic and hypertonic conditions, we recorded miniature endplate potentials (MEPPs) from isolated frog (Rana pipiens) cutaneous pectoris muscles, before and after repetitive nerve stimulation (40 s at 80 Hz). Continuous recordings were made in low Ca2+ high Mg2+ isotonic Ringer solution, in Ringer that was made hypertonic with 100 mM sucrose, and in wash solution. Estimates were

Cultured peritoneal mesothelial cells exhibit apical primary cilia.

This study examined primary cilia on cultured human and rabbit peritoneal mesothelial cells (PMC) and investigated factors that influence ciliary expression. Primary cilia were examined with indirect immunocytochemistry, laser scanning confocal microscopy and scanning electron microscopy. Ciliary expression was evaluated in cultures with or without l-cysteine (0.25 mM) or exposure to Ca(2+)-free Krebs-Ringer solution supplemented with EGTA, 0.5 mM. This treatment disrupted cell monolayer integrity. Cilia were counted and normalized to total cell counts using NIH image. Primary cilia were ident

Inhibition of baroreflex vagal bradycardia by nasal stimulation in rats.

Nasal stimulation provokes hypertension and bradycardia. We report here that such stimulation inhibits baroreflex vagal bradycardia (BVB). In chloralose- and urethan-anesthetized, beta-adrenergic receptor-blocked rats, the aortic depressor nerves were cut and electrically stimulated to induce BVB. Nasal application of smoke, warm distilled water, or cold or hot Ringer solution suppressed BVB, but application of warm Ringer solution did not. Smoke-induced inhibition was abolished by trigeminal but not olfactory denervation. Neither suprapontine decerebration nor C3 spinal cord transection affec

Hyperkalemic periodic paralysis episode during halothane anesthesia in a horse.

A 7-month-old Quarter Horse filly was admitted for surgical repair of a right olecranon fracture. Anesthesia was achieved with xylazine hydrochloride, guaifenesin, ketamine hydrochloride, and halothane. Two and a half hours after induction of anesthesia, myotonia, muscle fasciculations, and sweating, concurrent with high serum potassium concentration and associated electrocardiographic changes consistent with hyperkalemic periodic paralysis, were observed. Treatment included intermittent positive-pressure ventilation, changing intravenous administration of fluids from lactated Ringer's solutio

Comparison on the pulmonary effects of rapid infusion of a crystalloid and a colloid solution.

The cardio-pulmonary effects of a rapid infusion of a crystalloid (Ringer's lactate) and a colloid solution (Dextran 40) were compared in healthy dogs. The colloid solution produced a long lasting volume expansion and hemodilution. The improved circulation in this group was reflected in an increased cardiac output and arterial blood pressure. The ventilation/perfusion ratios were reduced and the pulmonary vascular resistance was significantly lower in the colloid group in comparison both to the crystalloid group as well as to the control animals. The total oxygen consumption increased slightly

[Transfusion-infusion therapy in modern wars].

All previous experiences have shown that the application of blood, blood components and intravenous solutions presents an unreplaceable therapeutical measure in modern surgical-resuscitative management of war injuries. Together with the broad application of the whole blood, there have been also used other blood components (even such as cryoprecipitate and platelet rich plasma). Among intravenous solutions the most frequently mentioned were isotonic saline, Ringer's lactate solution, glucosaline, 5% dextrose solution, dextran solutions and, recently, human albumin solutions. Due to a high risk

Cold heparinized lactated Ringers with procaine (HeLP) preservation fluid in 266 living donor kidney transplantations.

Since the 1960s simple inexpensive cold lactated Ringers with additives has been used for short-term cold preservation of kidneys from living donors. We performed 266 living donor kidney transplantations from January 22, 2003 to October 30, 2006. Donor allografts were recovered laparoscopically and flushed with cold heparin, lactated Ringer's and procaine (HeLP) solution. Warm and cold ischemic times were typically <45 min and <90 min, respectively. The mean follow up was 21.6+/-12.2 months. There was no delayed graft function. Actuarial 1-year patient and graft survival were 98.6% and 98.1%,

Beta-agonist activation of an amiloride-insensitive transport mechanism in rabbit pleura.

The beta-agonist terbutaline increases the net rate of liquid absorption from hydrothoraces with albumin-Ringer solution: since beta-agonists decrease lymphatic drainage, the effect of terbutaline seems due to an increase in solute-coupled liquid absorption, (Zocchi et al. 1994 Respir. Physiol. 97:347-356). In this research we determined in anesthetized rabbits the rate of volume change in albumin-Ringer hydrothoraces of different size with amiloride plus terbutaline, and compared it with that previously obtained in hydrothoraces with amiloride alone. The net rate of liquid absorption was 0.09

β2-Adrenergic receptors and G-protein-coupled receptor kinase 2 in rabbit pleural mesothelium.

Former studies on net rate of liquid absorption from small Ringer or 1% albumin-Ringer hydrothoraces in rabbits indicated that Na+ transport and solute-coupled liquid absorption by mesothelium is increased by pleural liquid dilution, and stimulation of β2-adrenoreceptors (β2AR). In this research we tried to provide molecular evidence for β2AR in visceral and parietal mesothelium of rabbit pleura. Moreover, because prolonged stimulation of β2AR may lead to desensitization mediated by G-protein-coupled receptor kinase 2 (GRK2), we also checked whether GRK2 is expressed in pleural mesothelium. Extravascular lung water following hemorrhagic shock in the baboon: Comparison between resuscitation with Ringer's lactate and Plasmanate.

Baboons were subjected to deep hemorrhagic shock by using a membrane potential of -65 mv as an endpoint. They were then resuscitated with either Plasmanate plus their shed blood or Ringer's lactate plus their shed blood. As compared with their own preshock values, the Plasmanate-resuscitated animals accumulated more extravascular lung water than the Ringer's lactate-resuscitated animals. Another group of baboons resuscitated from deep shock demonstrated significant extravasation of albumin on postmortem analysis of lung composition. This increased tendency for extravasation of albumin after sh

Refinement of Replacement Fluid Using Human Serum Albumin Preparation to Enhance Biocompatibility in Therapeutic Plasma Exchange.

Human serum albumin (HSA) solution is widely used as a replacement fluid during therapeutic plasma exchange (TPE). However, the ionic concentrations in commercially available 5% HSA solutions differ significantly from physiological levels. In this study, we developed a simple method for preparing a replacement fluid with ion concentrations closer to physiological norms by mixing 25% HSA with lactated Ringer's solution, sodium chloride, and calcium gluconate hydrate. We prepared three types of replacement fluids, each with a 4.0% albumin concentration and total volumes of 1.83, 2.43, and 3.04 L

Beta-adrenergic activation of solute coupled water uptake by toad skin epithelium results in near-isosmotic transport.

Transepithelial potential (V(T)), conductance (G(T)), and water flow (J(V)) were measured simultaneously with good time resolution (min) in isolated toad (Bufo bufo) skin epithelium with Ringer on both sides. Inside application of 5 microM isoproterenol resulted in the fast increase in G(T) from 1.2+/-0.3 to 2.4+/-0.4 mS x cm(-2) and slower increases in equivalent short circuit current, I(SC)(Eqv) = -G(T) x V(T), from 12.7+/-3.2 to 33.1+/-6.8 microA cm(-2), and J(V) from 0.72+/-0.17 to 3.01+/-0.49 nL cm(-2) s(-1). Amiloride in the outside solution abolished I(SC)(Eqv) (-1.6+/-0.1 microA cm(-2)

Peritoneal lavage in the horse.

Eight horses ranging in age from 4 days to 9 years were treated for peritonitis. Escherichia coli was isolated in four cases and Nocardia sp in one case. In each case, a catheter placed in the peritoneal cavity allowed drainage of a large amount of purulent fluid. Retrograde peritoneal lavage was performed through a Foley catheter or medical tubing, using Ringer's lactate solution containing kanamycin, povidone iodine, or nitrofurazone. All except two horses responded well to repeated lavage.

The Apparent pH Stability of Antibiotic Preparations Used for Uterine Infusions in Mares.

Infectious endometritis is a primary cause of subfertility in mares. Many manuals and guidelines are available for administering intrauterine infusions of antibiotics and biofilm-disrupting solutions, detailing concentrations, vehicle solutions, and buffers. However, the stability of these combinations has not been documented. Therefore, our study investigated how factors such as storage temperature, concentration, buffer types, combinations of biofilm disruptors, and vehicle solutions affected pH stability over 24 h in uterine infusion preparations commonly used by veterinary theriogenologist

Effects of Stroke Volume Maximization Before One-Lung Ventilation on Video-Assisted Thoracic Surgery: A Randomized Controlled Trial.

Background/Objectives: The use of goal-directed fluid therapy (GDFT) guided by stroke volume (SV) variation during thoracic surgery, particularly with one-lung ventilation (OLV) and protective ventilation strategies, is not well established. This study aimed to determine whether maximizing stroke volume (SV) before initiating one-lung ventilation (OLV) reduces the incidence of intraoperative hypotension requiring vasoactive agents during video-assisted thoracoscopic surgery (VATS). Methods: Sixty patients undergoing VATS were randomly assigned to an SVM group (n = 30) or a control group (n = 3

Triggered by light and magnetism: smart foam PLLA/HAP/Fe3O4 scaffolds for heat-controlled biomedical applications.

Ternary composite foam materials containing poly-L-lactic acid (PLLA), calcium hydroxyapatite (HAP) (20 nm), and morphologically controlled Fe3O4 nanoparticles (80 nm) were fabricated using the thermally induced phase separation (TIPS) technique over a broad concentration range of the magnetic component (1-30 wt%). The foam scaffolds were highly porous (>95%), and lightweight, with a high capacity for soaking in Ringer's solution. The foam density varied with the inorganic component content, ranging from 0.02 to 0.079 g mL-1, while the mean pore size was approximately 330 μm. The magnetic beha
