Robust parameter estimation techniques for stochastic within-host macroparasite models.

We present a stochastic model of the within-host population dynamics of lymphatic filariasis, and use a simulated goodness-of-fit (GOF) method to estimate immunological parameters and their confidence intervals from experimental data. A variety of deterministic moment closure approximations to the stochastic system are explored and compared with simulation results. For the maximum GOF parameter estimates, none of the methods of closure accurately reproduce the behaviour of the stochastic model. However, direct analysis of the stochastic model demonstrates that the high levels of variation obse

Stochastic risk estimate for barium radiological examinations in Malaga (Spain).

The aim of this research work has been the estimation of the stochastic risk for five barium examinations. The sample was the population attending the Radiological Service of the 'Nuestra Señora de la Victoria' University Hospital, who had been treated with some barium examinations, set in five different groups: oesophagus tract, oesophagus-gastric-duodenal tract, intestinal tract, enteroclysis and double enema. To estimate the stochastic risk, it is necessary to know the organ dose. This can be calculated from the dose-area product which allows us to determine the effective dose using softwar

Chernobyl experience: biological indicators of exposure to ionizing radiation.

Using the Chernobyl accident as an example, an attempt is made to consider the possibility of using the biological markers of exposure and effects of exposure to ionizing radiation in relation to biology dosimetry, and to predict early and late nonstochastic and stochastic radiation consequences. The biological dosimetry was based on the three markers: chromosome aberrations of peripheral blood lymphocytes, dynamics of blood cell (lymphocytes, neutrophils) counts and electron spin resonance (ESR) of tooth enamel. The first two methods can be applied in a short period of time (days or weeks) af

Modulatory mechanisms underlying high-frequency transcranial random noise stimulation (hf-tRNS): A combined stochastic resonance and equivalent noise approach.

BACKGROUND: High-frequency transcranial random noise stimulation (hf-tRNS) is a neuromodulatory technique consisting of the application of alternating current at random intensities and frequencies. hf-tRNS induces random neural activity in the system that may boost the sensitivity of neurons to weak inputs. Stochastic resonance is a nonlinear phenomenon whereby the addition of an optimal amount of noise results in performance enhancement, whereas further noise increments impair signal detection or discrimination. OBJECTIVE: The aim of the study was to assess whether modulatory effects of hf-t

Serotonergic regulation of melanocyte conversion: A bioelectrically regulated network for stochastic all-or-none hyperpigmentation.

Experimentally induced depolarization of resting membrane potential in "instructor cells" in Xenopus laevis embryos causes hyperpigmentation in an all-or-none fashion in some tadpoles due to excess proliferation and migration of melanocytes. We showed that this stochastic process involved serotonin signaling, adenosine 3',5'-monophosphate (cAMP), and the transcription factors cAMP response element-binding protein (CREB), Sox10, and Slug. Transcriptional microarray analysis of embryos taken at stage 15 (early neurula) and stage 45 (free-swimming tadpole) revealed changes in the abundance of 45

Integrated Strategies for Overcoming Resolution Limits in Electron Beam Lithography of Chemically Amplified Resists.

Electron beam lithography (EBL) of chemically amplified resists (CARs) faces fundamental challenges, including stochastic electron scattering and acid diffusion, that limit resolution and reproducibility. Using SU-8 as a model CAR, this study systematically investigated complementary strategies to address these challenges, combining multipass exposure, proximity effect correction (PEC) with midrange correction factors, base quencher incorporation, and post-exposure bake (PEB) suppression. Monte Carlo simulations and calibrated PEC modeling revealed that extending the point spread function to i

The Markov process as a general method for nonparametric analysis of right-censored medical data.

The product limit method of Kaplan and Meier for estimating survival functions and the logrank test of Mantel are widely employed for analysis of longitudinal medical data. Developed for analysis of one-time events such as death, survival analysis is also commonly adapted to more complex states such as loss of vision or cancer remission by restricting analysis to first occurrences. The nonparametric discrete time nonhomogeneous Markov process is proposed as a better model for any applications of the latter type. This simple stochastic model allows for an arbitrary number of possible states and

Twinning: mechanisms and genetic implications.

In the past, twins have been studied to determine the genetic contribution to various disease processes. Recent work, however, suggests that monozygotic (MZ) twins are not truly identical. Many genetic forms of discordance have been described within MZ twin pairs and may even play a role in causing MZ twinning. Intra-uterine environmental differences in the allocation of the number of cells and in the placental vascular supply to each twin, as well as stochastic development events, may lead to major discordance at birth between the twins of a MZ pair.

Stability of a stochastic brucellosis model with semi-Markovian switching and diffusion.

To explore the influence of state changes on brucellosis, a stochastic brucellosis model with semi-Markovian switchings and diffusion is proposed in this paper. When there is no switching, we introduce a critical value R s and obtain the exponential stability in mean square when R s < 1 by using the stochastic Lyapunov function method. Sudden climate changes can drive changes in transmission rate of brucellosis, which can be modelled by a semi-Markov process. We study the influence of stationary distribution of semi-Markov process on extinction of brucellosis in switching environment includ

The logarithmic memristor-based Bayesian machine.

The demand for explainable and energy-efficient artificial intelligence (AI) systems for edge computing has led to growing interest in electronic systems dedicated to Bayesian inference. Traditional designs of such systems often rely on stochastic computing, which offers high energy efficiency but suffers from latency issues and struggles with low-probability values. Here, we introduce the logarithmic memristor-based Bayesian machine, an innovative design that leverages the unique properties of memristors and logarithmic computing as an alternative to stochastic computing. We present a prototy

Stochastic expression of invasion genes in Plasmodium falciparum schizonts.

Genetically identical cells are known to exhibit differential phenotypes in the same environmental conditions. These phenotypic variants are linked to transcriptional stochasticity and have been shown to contribute towards adaptive flexibility of a wide range of unicellular organisms. Here, we investigate transcriptional heterogeneity and stochastic gene expression in Plasmodium falciparum by performing the quasilinear multiple annealing and looping based amplification cycles (MALBAC) based amplification and single cell RNA sequencing of blood stage schizonts. Our data reveals significant tran

Extracting synaptic conductances from single membrane potential traces.

In awake animals, the activity of the cerebral cortex is highly complex, with neurons firing irregularly with apparent Poisson statistics. One way to characterize this complexity is to take advantage of the high interconnectivity of cerebral cortex and use intracellular recordings of cortical neurons, which contain information about the activity of thousands of other cortical neurons. Identifying the membrane potential (Vm) to a stochastic process enables the extraction of important statistical signatures of this complex synaptic activity. Typically, one estimates the total synaptic conductanc

Correcting Structural Bias in Dynamical Models of Infectious Disease Using a Bayesian State-Space Framework.

Deterministic dynamical models are widely used in infectious disease modelling, but they often become systematically biased when key drivers such as seasonality and random environmental variation are simplified or omitted. This paper proposes a practical way to account for structural bias while preserving the underlying mechanistic model. We model the observed time series as the sum of (i) a deterministic transmission component given by a reduced Ross malaria model and (ii) a latent stochastic seasonal component that captures unresolved seasonal forcing and other unmodelled variability. The se

3D discrete angiogenesis dynamic model and stochastic simulation for the assessment of blood perfusion coefficient and impact on heat transfer between nanoparticles and malignant tumors.

Early detection of malignant tumors plays a crucial role in the survivability chances of the patient. Therefore, new and innovative tumor detection methods are constantly searched for. Tumor-specific magnetic-core nano-particles can be used with an alternating magnetic field to detect and treat tumors by hyperthermia. For the analysis of the method effectiveness, the bio-heat transfer between the nanoparticles and the tissue must be carefully studied. Heat diffusion in biological tissue is usually analyzed using the Pennes Bio-Heat Equation, where blood perfusion plays an important role. Malig

Stochastic expansion of radionuclide inhalation dosimetry for consequence management application: uncertainty and sensitivity analysis in the ICRP 130 human respiratory tract model.

Releases from nuclear or radiological security events can result in significant internal radiation contamination through inhalation of particulate contaminants. The reference human respiratory tract model (HRTM), developed by the International Commission on Radiological Protection (ICRP) and described in the ICRP Publication 66 with subsequent updates in the ICRP Publication 130, is used to evaluate the deposition and clearance behaviour of inhaled radionuclides. Biokinetic models are used to quantify the retention and excretion of internally deposited particulates, supporting the calculation

Combining genomic prediction and multi-trait indices through stochastic simulations: do index type and deployment order affect genetic gain?

Genomic selection (GS) has transformed plant breeding by enabling early selection and potentially reducing cycle length, but how to integrate GS with classical multi-trait selection indices remains unclear. We used stochastic simulations to compare seven strategies combining Smith-Hazel (SH), Pesek-Baker (PB), and empirical (EMP) indices with or without GS in a rice breeding program targeting increased grain yield (GY), reduced chalkiness rate (CR), and stable plant height (PH). Trait importance was set to 70% for GY, 15% for CR, and 15% for PH. After a phenotypic-selection burn-in phase, we s

Generative modeling of clinical time series via latent stochastic differential equations.

Clinical time series data from electronic health records and medical registries offer unprecedented opportunities to understand patient trajectories and inform medical decision-making. However, leveraging such data presents significant challenges due to irregular sampling, complex latent physiology, and inherent uncertainties in both measurements and disease progression. To address these challenges, we propose a generative modeling framework based on latent neural stochastic differential equations (SDEs) that views clinical time series as discrete-time partial observations of an underlying con

DNA Methylation Stochasticity Is Linked to Transcriptional Variability and Convergent Epigenetic Disruption across Genetic Subtypes of Acute Myeloid Leukemia.

UNLABELLED: Disruption of the epigenetic landscape is of particular interest in acute myeloid leukemia (AML) due to its relatively low mutational burden and frequent occurrence of mutations in epigenetic regulators. In this study, we applied an information-theoretic analysis of methylation potential energy landscapes, capturing changes in mean methylation level and methylation entropy, to comprehensively analyze DNA methylation stochasticity in subtypes of AML defined by mutually exclusive genetic mutations. AML subtypes with CEBPA double mutation and those with IDH mutations were identified a

Multiscale modelling of birth-death processes.

Many biological systems exhibit multiscale dynamics, where some species occur in high copy numbers while others remain rare. This heterogeneity necessitates hybrid modelling approaches: deterministic models are computationally efficient but inaccurate for low-count species, while fully stochastic simulations are accurate but prohibitively expensive. Threshold-based hybrid methods, such as the Jump-Switch-Flow (JSF) algorithm, address this by simulating low-count species stochastically and high-count species deterministically, switching at a user-chosen threshold Ω . In such methods, the choice

Modelling with SPEED: a Stochastic Predictor of Early Epidemic Detection.

The frequency of emerging infectious disease outbreaks continues to rise, necessitating predictive frameworks for public health decision-making. This study introduces the Stochastic Predictor of Early Epidemic Detection (SPEED) model, an adaptation of the classic Susceptible-Infected-Recovered model, employing a Gillespie-like algorithm to simulate early-stage stochastic disease transmission. SPEED incorporates individual-level detection probabilities based on the infection time and the lag from GP consultation to lab confirmation. The model dynamically adjusts to public health responses by en
