This succinct article addresses the multifaceted interactions between the fungal organism Candida albicans and the Gram-positive bacterium Streptococcus mutans in the development of oral biofilms and pathobiology of oral diseases. S. mutans is considered to be a major pathogen in the development of dental caries. It is often found to interact with C. albicans in oral infection settings. The interaction of these organisms is often mediated via the binding of Glucosyltransferase (GtfB) enzyme secreted by S. mutans to C. albicans surface proteins Als1 and Hwp1. During these interactions, both C.

In this study we evaluated the effect of prenylated peanut stilbenoids on the growth, biofilm accumulation and acid production of the dental caries pathogen Streptococcus mutans. Prior research with the non-prenylated stilbenes, resveratrol and piceatannol, has shown that these molecules are active against S. mutans. Here we sought to determine if the addition of a prenyl group to the stilbene backbone increased anti-S. mutans activities. Two prenylated stilbenes, arachidin-1 and arachidin-3, were produced using a peanut hairy root production system. Compared to resveratrol and piceatannol, bo

UNLABELLED: Streptococcus mutans is a cariogenic bacterium that produces a variety of bacteriocins and retains resistance to these bacteriocins. In this study, we investigated the susceptibility of 127 S. mutans strains to nukacins produced by Staphylococcus spp., which are commensal bacteria in humans. We detected diverse susceptibilities among strains. Nineteen strains had a disrupted LctF (type I), which is responsible for nukacin susceptibility, whereas the remaining 108 strains had an intact LctF (type II) and displayed resistance to nukacins. However, the type I strains still showed resi

OBJECTIVE: Streptococcus mutans, the main causative agent of caries, have the ability to form biofilms on the surface of teeth. The availability of nutrients such as glucose, lactose, soy protein, and iron can influence S. mutans in biofilm formation. All four sources of nutrients have been shown to increase the formation of S. mutans biofilms. The purpose of this study was to determine the structure and thickness of S. mutans biofilms induced by glucose, lactose, soy protein, and iron.
MATERIALS AND METHODS: This experimental laboratory study aimed to examine the formation of biofilm structu

Cariogenic biofilms have a matrix rich in exopolysaccharides (EPS), mutans and dextrans, that contribute to caries development. Although several physical and chemical treatments can be employed to remove oral biofilms, those are only partly efficient and use of biofilm-degrading enzymes represents an exciting opportunity to improve the performance of oral hygiene products. In the present study, a member of a glycosyl hydrolase family 66 from Flavobacterium johnsoniae (FjGH66) was heterologously expressed and biochemically characterized. The recombinant FjGH66 showed a hydrolytic activity again

In the pathogenesis of dental caries, Streptococcus mutans (S. mutans) plays a central role. S. mutans can produce extracellular polysaccharides, which can help the bacteria form biofilms on the tooth surface, create a stable living environment, and hinder the removal of bacteria by natural defense substances in the oral cavity such as saliva. Meanwhile, the oral microbiota and dietary habits exert long-term influences on its development. This study, employing the BALB/c mouse model, explored the effects of L. paracasei L9 on dental caries. In the experiment, mice underwent the S. mutans inocu

Introduction Complex interactions between cariogenic bacteria and host factors modulate dental caries. Streptococcus mutans, a gram-positive facultative anaerobe plays a prominent role in the initiation of caries. The ability of S. mutans to adhere to salivary enamel pellicle results in an acidic local habitat for the organism. This leads to demineralization of the tooth and penetration of bacteria into the pulp leading to endodontic infections. Enterococcus faecalis, an opportunistic pathogenis a gram-positive, facultative anaerobe implicated in secondary endodontic infections. This study aim

Despite MCT oil's potential antimicrobial benefits for gastrointestinal health, its effects on disrupting cariogenic pathogens on oral mucosal surfaces remain underexplored. This study evaluated the impact of MCT oil on the adhesion and invasion of Candida albicans and Streptoccocus mutans using planktonic and mucosal models. First, a planktonic model was used to assess the impact of various concentrations of MCT on the growth of S. mutans and C. albicans. Subsequently, a mucosal model was established by seeding TR-146 human buccal mucosal epithelial cells on a 3 µm porous transwell membrane,

Effective control of gene expression is crucial for understanding gene function in both eukaryotic and prokaryotic cells. While several inducible gene expression systems have been reported in Streptococcus mutans, a conditional pathogen that causes dental caries, the significant non-inducible basal expression in these systems seriously limits their utility, especially when studying lethal gene functions and molecular mechanisms. We introduce a tightly controlled xylose-inducible gene expression system, TC-Xyl, for Streptococcus mutans. Western blot results and fluorescence microscopy analysis

Diabetes mellitus is one of the most prevalent diseases worldwide. The glucose levels found in the blood of diabetic patients can be reflected in the saliva, which can favor biofilm growth and predispose denture wearers to oral candidiasis. This study aimed to evaluate the effect of different salivary glucose concentrations on dual-species biofilms of Candida albicans and Streptococcus mutans. A 96-h biofilm was developed on acrylic resin specimens exposed to 'feast' (10% sucrose) and 'famine' periods. Biofilms were constantly exposed to salivary glucose concentrations equivalent to 0, 20, 60

BACKGROUND AND PURPOSE: Cerebral microbleeds (CMB) are associated with stroke and cognitive impairment. We previously reported a high prevalence of CMB in people with Streptococcus mutans expressing Cnm, a collagen-binding protein in the oral cavity. S.mutans is a major pathogen responsible for dental caries. Repeated challenge with S.mutans harboring the cnm gene encoding Cnm induced cerebral bleeding in stroke-prone spontaneously hypertensive rats. The purpose of this longitudinal study is to examine the relationship of cnm-positive S.mutans to the development of CMB.
METHODS: We retrospect

Initial attachment of the cariogenic Streptococcus mutans onto dental enamel is largely promoted by the adsorption of specific salivary proteins on enamel surface. Some phosphorylated salivary proteins were found to reduce S. mutans adhesion by competitively inhibiting the adsorption of S. mutans-binding salivary glycoproteins to hydroxyapatite (HA). The aim of this study was to develop antiadherence compounds for preventing dental biofilm development. We synthesized phosphorylated polyethylene glycol (PEG) derivatives and examined the possibility of surface pretreatment with them for preventi

For study of the enamel-protective effect of chlorhexidine-fluoride applications, the labial surfaces of pieces of bovine incisors were treated with 0.2% chlorhexidine gluconate solution, with Duraphat fluoride varnish, or with both of the above agents, while one group was treated with distilled water and one was left as an untreated control. Furthermore, a placebo varnish was used in the chlorhexidine- and distilled-water-treated groups; all the varnishes were removed after 24 h. The enamel slabs were mounted pairwise in an artificial mouth to form approximal contacts. The teeth were continuo

The regulation of extracellular glucosyltransferase production in Streptococcus mutans GS-5 has been studied using a chemically defined medium. Most of the glucosyltransferase activity produced by cells grown in the chemically defined medium was extracellular, in contrast with the distribution between cell-associated and extracellular glucosyltransferase activity when cells were grown in complex medium. The production of extracellular glucosyltransferase activity coincided with the logarithmic growth phase, and further accumulation ceased when glucose was exhausted from the medium. Accumulatio
