Micro-RNA profiles of pathology and resilience in posterior cingulate cortex of cognitively intact elders.

The posterior cingulate cortex (PCC) is a key hub of the default mode network underlying autobiographical memory retrieval, which falters early in the progression of Alzheimer's disease (AD). We recently performed RNA sequencing of post-mortem PCC tissue samples from 26 elderly Rush Religious Orders Study participants who came to autopsy with an ante-mortem diagnosis of no cognitive impairment but who collectively displayed a range of Braak I-IV neurofibrillary tangle stages. Notably, cognitively unimpaired subjects displaying high Braak stages may represent cognitive resilience to AD patholog

Microstructural and functional brain abnormalities in multiple sclerosis predicted by osteopontin and neurofilament light.

BACKGROUND: Osteopontin (OPN) is a proinflammatory biomarker, and neurofilament light chain (NFL) levels reflect axonal damage. Resting-state functional MRI (rs-fMRI) defines brain networks during wakeful rest. OBJECTIVE: To examine, if levels of OPN and NFL are associated on the long term with (i) lesion evolution, (ii) changes in normal-appearing white matter (NAWM) microstructure and (iii) functional connectivity in multiple sclerosis (MS). METHODS: Concentration of NFL and OPN in the blood and CSF were related to MRI findings 10.3 ± 2.8 years later in 53 patients with MS. NFL was examine

Functional brain MRI in patients complaining of electrohypersensitivity after long term exposure to electromagnetic fields.

INTRODUCTION: Ten adult patients with electromagnetic hypersensitivity underwent functional magnetic resonance imaging (fMRI) brain scans. All scans were abnormal with abnormalities which were consistent and similar. It is proposed that fMRI brain scans be used as a diagnostic aid for determining whether or not a patient has electromagnetic hypersensitivity. Over the years we have seen an increasing number of patients who had developed multi system complaints after long term repeated exposure to electromagnetic fields (EMFs). These complaints included headaches, intermittent cognitive and memo

Brain Connectivity Gradients Alterations in Discordant Cerebrospinal Fluid Profile for Alzheimer's Disease Biomarkers.

Alzheimer's disease (AD) is a heterogeneous disorder characterized by brain accumulation of amyloid-beta (Aß, simplified as A for the AD biological model) and tau (T) proteins, with Aß emerging first. However, a significant proportion of individuals exhibit discordant biomarkers' profiles, such as elevated phosphorylated tau181 (p-tau181) with normal Aß42 from cerebrospinal fluid (CSF), posing diagnostic and mechanistic challenges. This study investigated whether functional and structural brain connectivity can distinguish individuals with discordant CSF profiles (A-T+) from those with concord

Parallel ICA of FDG-PET and PiB-PET in three conditions with underlying Alzheimer's pathology.

The relationships between clinical phenotype, β-amyloid (Aβ) deposition and neurodegeneration in Alzheimer's disease (AD) are incompletely understood yet have important ramifications for future therapy. The goal of this study was to utilize multimodality positron emission tomography (PET) data from a clinically heterogeneous population of patients with probable AD in order to: (1) identify spatial patterns of Aβ deposition measured by ((11)C)-labeled Pittsburgh Compound B (PiB-PET) and glucose metabolism measured by FDG-PET that correlate with specific clinical presentation and (2) explore ass

Infraslow closed-loop brain training for anxiety and depression (ISAD): a protocol for a randomized, double-blind, sham-controlled pilot trial in adult females with internalizing disorders.

BACKGROUND: The core intrinsic connectivity networks (core-ICNs), encompassing the default-mode network (DMN), salience network (SN) and central executive network (CEN), have been shown to be dysfunctional in individuals with internalizing disorders (IDs, e.g. major depressive disorder, MDD; generalized anxiety disorder, GAD; social anxiety disorder, SOC). As such, source-localized, closed-loop brain training of electrophysiological signals, also known as standardized low-resolution electromagnetic tomography (sLORETA) neurofeedback (NFB), targeting key cortical nodes within these networks has

Estimated connectivity networks outperform observed connectivity networks when classifying people with multiple sclerosis into disability groups.

BACKGROUND: Multiple Sclerosis (MS), a neurodegenerative and neuroinflammatory disease, causing lesions that disrupt the brain's anatomical and physiological connectivity networks, resulting in cognitive, visual and/or motor disabilities. Advanced imaging techniques like diffusion and functional MRI allow measurement of the brain's structural connectivity (SC) and functional connectivity (FC) networks, and can enable a better understanding of how their disruptions cause disability in people with MS (pwMS). However, advanced MRI techniques are used mainly for research purposes as they are expen

Functional magnetic resonance imaging in neuropathic pain: a bibliometric analysis incorporating machine learning.

BACKGROUND: Neuropathic pain (NP) affects 6.9%-10% of the global population. Functional magnetic resonance imaging (fMRI) has become central to NP investigation, yet few studies have systematically mapped this literature. We used bibliometric and machine-learning methods to chart the fMRI-NP research landscape. METHODS: We retrieved 1,866 fMRI-NP articles (2005-2025) from Web of Science, PubMed, and Scopus. Following PRISMA guidelines, we analyzed publication trends, citation metrics, and collaborative networks with CiteSpace and VOSviewer, complemented by exploratory topic modeling based on

Neurobiological subtypes in alcohol use disorder and their phenotypic and clinical profiles.

Alcohol Use Disorder (AUD) is associated with vast clinical, behavioral, and neurobiological heterogeneity that hinder both the efficacy of current interventions and future treatment development. While recent efforts have focused on stratifying psychiatric disorders based on underlying neurobiological markers, such approaches remain largely unexplored in AUD. To address this, we empirically-derived neurobiological subtypes of AUD and evaluated their reproducibility and clinical relevance. Participants with complete resting-state functional MRI (rs-fMRI), phenotypic and clinical data from the H

Functional anomaly mapping lateralizes temporal lobe epilepsy with high accuracy in individual patients.

Mesial temporal lobe epilepsy (mTLE) is associated with variable dysfunction beyond the temporal lobe. We used functional anomaly mapping (FAM), a multivariate machine learning approach to resting state fMRI analysis to measure subcortical and cortical functional aberrations in patients with mTLE. We also examined the value of individual FAM in lateralizing the hemisphere of seizure onset in mTLE patients. Methods: Patients and controls were selected from an existing imaging and clinical database. After standard preprocessing of resting state fMRI, time-series were extracted from 400 cortical

Frontolimbic, Frontoparietal, and Default Mode Involvement in Functional Dysconnectivity in Psychotic Bipolar Disorder.

BACKGROUND: Functional abnormalities, mostly involving functionally specialized subsystems, have been associated with disorders of emotion regulation such as bipolar disorder (BD). Understanding how independent functional subsystems integrate globally and how they relate with anatomical cortical and subcortical networks is key to understanding how the human brain's architecture constrains functional interactions and underpins abnormalities of mood and emotion, particularly in BD. METHODS: Resting-state functional magnetic resonance time series were averaged to obtain individual functional con

Early Life Resting-State Network Functional Connectivity and Long-Term Neurodevelopmental Outcomes after Neonatal Encephalopathy.

BACKGROUND AND PURPOSE: Neonatal encephalopathy (NE) is a risk factor for long-term neurodevelopmental impairments, and early measures of brain function may help understand which survivors are most at risk. This study investigates the relationship between resting-state network (RSN) functional connectivity (FC) in the first week of life and long-term neurodevelopmental outcomes after NE. MATERIALS AND METHODS: A prospective cohort of therapeutic hypothermia-treated neonates ≥36 weeks gestational age with NE had resting-state functional MRI (rs-fMRI) in the first week of life, and follow-up at

Treating Chronic Pain by Modulating Phenomenological and Psychophysiological Indices of Self-Transcendence: Single-Blind Randomized Controlled Clinical Trial Protocol.

BACKGROUND: Chronic musculoskeletal pain (CMP) imposes a significant psychological, physical, and emotional burden on millions of adults in the United States. A challenge in treating CMP is its tendency to become enmeshed with a person's sense of self. Mindfulness-based interventions (MBIs) offer a promising approach to help patients disentangle pain from their sense of self. However, rigorous research is needed to determine which styles of mindfulness practice most effectively promote self-transcendence, a candidate mechanism for durable pain relief. OBJECTIVE: This study aims to evaluate wh

Fusion analysis of sMRI-DTI in patients with ulcerative colitis by multimodal CCA + joint ICA and associated neurotransmitter profiles.

BACKGROUND: As a chronic inflammatory disease, ulcerative colitis (UC) is characterized by complex etiology and unclear pathological mechanisms. Neuroimaging research plays a pivotal role in improving diagnostic accuracy and guiding treatment decisions, while enhancing our understanding of disease mechanisms. We aimed to explore the possible associations between gray matter volume (GMV) and white matter fractional anisotropy (FA) abnormalities as well as the neurotransmitter profiles in UC patients. METHODS: Thirty-four UC patients and 21 healthy controls (HCs) participated in structural magn

Alterations in miR-151a-3p of plasma-derived exosomes and associated multimodal neuroimaging patterns in major depressive disorder.

Magnetic resonance imaging (MRI) has been recognized as a valuable tool for achieving 'reification of clinical diagnosis' of major depressive disorder (MDD). However, the reliability and validity of MRI results are often compromised by genetic, environmental, and clinical heterogeneity within test samples. Here, we combined MRI with other clinical findings using multimodal MRI fusion algorithm to construct a data-driven, bottom-up diagnostic approach. The covariation patterns between the multimodal MRI features and differential expression of exosomal microRNA (miRNA) were identified on a subse

Repetitive subconcussion results in disrupted neural activity independent of concussion history.

Concussion is a public health crisis that results in a complex cascade of neurochemical changes that can have life-changing consequences. Subconcussions are generally considered less serious, but we now realize repetitive subconcussions can lead to serious neurological deficits. Subconcussions are common in contact sports and the military where certain personnel are exposed to repetitive occupational blast overpressure. Post-mortem studies show subconcussion is a better predictor than concussion for chronic traumatic encephalopathy-a progressive and fatal neurodegenerative tauopathy, only diag

Association of early life cardiovascular risk factors with grey matter structure in young adults in the United Kingdom: the ALSPAC study.

BACKGROUND: Cumulative exposures to obesity, hypertension, and physical inactivity from midlife (40-65 years) onwards are three known cardiovascular risk factors for dementia and associated cerebral structural damage. Exactly how early in the lifespan sensitive periods for exposure to these risk factors begin is yet to be established, specifically with respect to onset of cerebral structural changes. We aimed to investigate whether cardiovascular risk across childhood and adolescence is already associated with cerebral structure in regions previously linked with dementia, during young adulthoo
