Identification of key metabolic pathways reprogrammed by BmNPV in silkworm Bombyx mori.

Elucidating the mechanism of infection of Bombyx mori nuclear polyhedrosis virus (BmNPV) and host antiviral response remains a major scientific task in sericulture. Virus invasion causes a series of antiviral immune responses in the host, and successful infection leads to massive changes in the host's physiological and biochemical state. Current research mainly focuses on silkworm genes and proteins associated with viral infection and resistance, but little is known regarding the host metabolic pathways that the virus utilizes for optimal replication. In this work, key metabolites involved in

Comparative genomics reveals that metabolism underlies evolution of entomopathogenicity in bee-loving Ascosphaera spp. fungi.

Ascosphaera (Eurotiomycetes: Onygenales) is a diverse genus of fungi that is exclusively found in association with bee nests and comprises both saprophytic and entomopathogenic species. To date, most genomic analyses have been focused on the honeybee pathogen A. apis, and we lack a genomic understanding of how pathogenesis evolved more broadly in the genus. To address this gap we sequenced the genomes of the leaf-cutting bee pathogen A. aggregata as well as three commensal species: A. pollenicola, A. atra and A. acerosa. De novo annotation and comparison of the assembled genomes was carried ou

Will the application of biocontrol fungi disrupt predation of Acanthococcus lagerstroemiae by coccinellids?

Insect predators are the most important natural enemies of the crapemyrtle bark scale (CMBS) in the USA. Mycopesticides (biocontrol fungi) are considered an IPM tool to increase CMBS mortality; however, their impacts on CMBS predators are unknown. The objectives of this study were to assess the abundance and diversity of CMBS natural enemies in Louisiana; evaluate the impacts of mycopesticides on survival of CMBS predators by life stage; and determine if entomopathogenic spores delivered to crapemyrtles are transferred to predators under field conditions. The mycopesticides Ancora® (Isaria fum

Dimethyl sulfoxide and ascarosides improve the growth and yields of entomopathogenic nematodes in liquid cultures.

Increasing the infective juvenile (IJ) yields of entomopathogenic nematodes in monoxenic culture systems would reduce their production cost for the market. Ascarosides act as universal nematode pheromones with developmental and behavioral effects of nematodes. Dimethyl sulfoxide (DMSO) is unexpectedly found to enhance the IJ yields of entomopathogenic nematodes on fortified nutrient broth plates. In this study, the influence of selected ascarosides (ascr#7, ascr#9 and ascr#11) and DMSO in three concentrations on the IJ yields of S. carpocapsae All and H. bacteriophora H06 in liquid culture fla

Evaluation of Barricade® to enhance survival of entomopathogenic nematodes on cowhide.

Entomopathogenic nematodes (EPNs), Steinernema riobrave and Heterorhabditis floridensis are under evaluation for eradication of the southern cattle fever tick, Rhipicephalus microplus infesting nilgai antelope, in South Texas. Cattle fever ticks are a significant threat to the U.S. livestock industry. Although they have been eradicated in the U.S. they frequently re-invade along the Texas-Mexico border. Remotely operated field sprayers have been developed to directly treat nilgai antelope with EPNs as they transit fence crossings and as they contact wetted foliage and soil from the surrounding

Morphological and phylogenetic characterization of a new microsporidium, Triwangia gracilipes n. sp. From the freshwater shrimp Caridina gracilipes (Decapoda: Atyidae) in China.

A new microsporidian species was described from the freshwater shrimp Caridina gracilipes collected from Lake Luoma located in Northern Jiangsu province, East China. The infected shrimps appeared generally opaque due to the presence of white cysts located in the connective tissues of the surface of the hepatopancreas. The earliest developmental stages observed were diplokaryotic meronts which were in direct contact with the host cell cytoplasm. Multinucleate sporogonial plasmodia developed into uninucleate sporoblasts which were enclosed in sporophorous vesicles. The parasite developed synchro

The effect of chemical insecticides on the scavenging performance of Steinernema carpocapsae: Direct effects and exposure to insects killed by chemical insecticides.

Entomopathogenic nematodes are used widely in biological insect control. Entomopathogenic nematodes can infect live insects as well as dead insects (i.e., they can act as scavengers). It is important to determine compatibility of entomopathogenic nematodes with other pest management tactics such as chemical insecticides. We hypothesized that chemical insecticides have negative impact on scavenging nematodes. According to our hypothesis, we first investigated the effects of direct exposure of Steinernema carpocapsae infectivity juveniles (IJs) to three chemical insecticides, cypermethrin, spino

Tropilaelaps species identification and viral load evaluation of Tropilaelaps and Varroa mites and their Apis mellifera hosts in Palawan, Philippines.

Apis mellifera pupae and their parasites Tropilaelaps and Varroa destructor were collected from honey bee hives in Palawan, Philippines for species identification of the Tropilaelaps and viral analyses. Genetic analysis identified Tropilaelaps mercedesae infesting A. mellifera on the island. Viral analyses showed that all pupae and their infesting Tropilaelaps or Varroa shared the same Deformed Wing Virus (DWV) variant infections with DWV-B being more prevalent than DWV-A. Pupae infested with either Varroa or Tropilaelaps had higher levels of both DWV variants than uninfested pupae. Vigilance

Carnauba wax enhances the insecticidal activity of entomopathogenic fungi against the blowfly Lucilia sericata (Diptera: Calliphoridae).

Blowfly, Lucilia sericata (Diptera: Calliphoridae), is a problematic synanthropic insect pest, a vector of microbial pathogens, and the causal agent of secondary myiasis. Fungal biopesticides are considered eco-friendly tools, alternative to synthetic pesticides, for the control of arthropod pests; however, to date, little is known about their bioactivity against blowflies. In this study, we assessed the insecticidal activity of three well-known entomopathogenic fungi, Beauveria bassiana, Beauveria pseudobassiana and Akanthomyces muscarius against L. sericata. In addition, we tested powdered c

Prevalence and histopathology of the parasitic barnacle, Sacculina carcini in shore crabs, Carcinus maenas.

Sacculina carcini is a common parasite of the European shore crab, Carcinus maenas. Following successful penetration of the host, numerous rootlets are formed that permeate through the hosts' tissues. Ultimately, these form an externa that houses the developing nauplii larvae of the parasite. Most studies have quantified levels of infection by counting the presence of reproductive externae and their breakdown structures, called scars. However, the diagnosis of the disease based only on external features may lead to underreporting the prevalence of the parasite. In the current study, we examine

Genetic variability among native and introduced strains of the parasitic nematode Deladenus siricidicola.

Sirex noctilio is an invasive Eurasian woodwasp that can kill pine (Pinus spp.) trees and has been introduced to areas of the Southern Hemisphere where plantations of introduced pines are grown. The main method of control of this invasive pest has been introduction and augmentation of a parasitic nematode, Deladenus siricidicola. The strain of D. siricidicola used for biological control of S. noctilio in the Southern Hemisphere originated in Sopron, Hungary. The genotype of D. siricidicola used for biological control sterilizes females of the strain of S. noctilio present in Australia. However

Experimental study of dengue virus infection in Aedes aegypti and Aedes albopictus: A comparative analysis on susceptibility, virus transmission and reproductive success.

Epidemiology of dengue fever has substantially changed over the years with respect to prevalent strains, affected geographical locations and severity of disease. Mosquito vectors show variable response in terms of susceptibility to four different serotypes of dengue virus. Although studies have postulated that, the vectors Ae. aegypti and Ae. albopictus are crucial for transmission of dengue virus, comparative efficacy of these species for viral transmission and tolerance is still enigmatic. In this study, these two vectors were infected orally with four serotypes of the dengue virus viz. DENV

Identification of Aedes aegypti specificity motifs in the N-terminus of the Bacillus thuringiensis Cry2Aa pesticidal protein.

One advantage of using the Cry proteins of Bacillus thuringiensis as pesticides is their relatively narrow spectrum of activity, thus reducing the risk of non-target effects. Understanding the molecular basis of specificity has the potential to help us design improved products against emerging pests, or against pests that have developed resistance to other Cry proteins. Many previous studies have associated specificity with the binding of the Cry protein, particularly through the apical regions of domain II, to particular receptors on the midgut epithelial cells of the host insect. We have pre

Insecticidal potential of Brevibacillus laterosporus against dipteran pest species in a wide ecological range.

In order to increase our understanding of the insecticidal potential of the entomopathogenic bacterium Brevibacillus laterosporus strain UNISS 18 against insect pests, investigations were conducted on a selection of dipteran species including fruit flies, house flies, blow flies, and mosquitoes, characterized by adaptations to very diverse habitats. According to lethal concentration (LC50) values, the common house mosquito Culex pipiens (LC50 = 0.10 × 106 spores/mL) and the yellow fever mosquito Aedes aegypti (LC50 = 0.18 × 106 spores/mL) were significantly more susceptible than the flies. Host range of Triatoma virus does not extend to Aedes aegypti and Apis mellifera.

Vector control is the most effective method to prevent transmission of Chagas disease. Control is mostly made through chemical insecticides although they have negative impact on wild pollinators, such as bees. Reducing pesticide use through biological alternatives could minimize the damage to these beneficial insects. Triatoma virus (TrV) is a pathogen able to kill triatomines and thus a valid candidate to be used as biological control agent. In this study we evaluate the capacity of TrV to infect an important beneficial insect (Apis mellifera) as well as a plague insect (Aedes aegypti). Resul

Precision mapping of N- and O-glycoproteins in viral resistant and susceptible strains of Bombyx mori.

Protein glycosylation plays important roles in protein structure, function, and immune recognition, among many other activities. One of the major roles of glycans and glycoconjugates on the cell surface is acting as the receptor for outside pathogens such as viruses. During the initial stage of viral replication, viruses interact with cell membrane receptors, which are in many cases glycoproteins. Identifying such glycoproteins is essential to understanding the mechanisms of viral infection, as well as developing antiviral strategies. Silkworm is an important economic insect as well as a model

Influence of Himasthla elongata (Trematoda: Echinostomatidae) metacercariae on heart rate in blue mussels (Mytilus edulis).

Whether metacercariae of the digenean Himasthla elongata (Himasthlidae) are harmful to their secondary intermediate hosts, the blue mussel, Mytilus edulis, is a disputable question. To shed light on this issue, we performed field monitoring of the heart rate (HR) in mussels infected with H. elongata over a period of 1.5 years. We observed a significant rise in HR in infected mussels, but only 1 year after infection and at temperatures of 15-17 °C. HR variance also grew sharply in infected mussels within the same temperature range. We also detected a decreased ability of infected mussels to com

Silkworm storage protein Bm30K-19G1 has a certain antifungal effects on Beauveria bassiana.

Storage proteins in the 30 K family are ubiquitous in the hemolymph of insects and play important roles in adult metamorphosis, development, egg formation, carrier transport and even host immunity. Some studies have shown that the 30 K proteins can inhibit apoptosis and have certain antifungal effects. The silkworm protein Bm30K-19G1 is a low molecular weight apolipoprotein that is abundant in hemolymph of fifth instar larvae. Our previous transcriptome sequencing, real-time PCR analysis and proteomic studies showed that the expression level of the 30 K protein gene was significantly up-regula
