The beetle Acanthoscelides obtectus (Say) causes severe post-harvest losses in the common bean (Phaseolus vulgaris). Under laboratory conditions, the susceptibility of A. obtectus to five coleopteran-specific Cry toxic proteins from Bacillus thuringiensis (Cry1Ba, Cry1Ia, Cry3Aa, Cry7Ab, and Cry23/37) was evaluated. After 30 days exposure, Cry proteins demonstrated high activity against A. obtectus adults (100% mortality). Proteins showed statistical differences in toxicity parameters compared to the control treatment, but the parameters were similar among them, and indicated that the final to

Wolbachia are common intracellular bacteria that are generally found in arthropods, including a high proportion of insects and also some nematodes. This intracellular symbiont can affect sex ratio with a variety of reproductive anomalies in the host, including cytoplasmic incompatibility (CI) in haplodiploids. In this study, we questioned if the parasitoid wasp, Habrobracon hebetor (Hym.: Braconidae), an important biological control agent of many lepidopteran larvae, is infected with Wolbachia. To test this, DNA was extracted from adult insects and subjected to PCR using specific primers to Wo

The pathogenic oomycete Aphanomyces astaci, transmitted mainly by invasive North American crayfish, causes the crayfish plague, a disease mostly lethal for native European crayfish. Due to its decimating effects on native crayfish populations in the last century, A. astaci has been listed among the 100 worst invasive species. Importantly, detecting the pathogen in endangered native crayfish populations before a disease outbreak would provide a starting point in the development of effective control measures. However, current A. astaci-detection protocols either rely on degradation-prone eDNA is

The immune system of snails is highly sensitive to pollutants, which can suppress its immune response. We investigated the effects of exposure to the glyphosate-based herbicide Roundup® Original on the snail Biomphalaria glabrata infected by the platyhelminth Echinostoma paraensei by evaluating changes in the snail's internal defense system. Four cohorts were studied: control group, infected snails, snails treated with Roundup®, and snails infected and treated with Roundup®. The hemocyte viability was assessed, morphological differentiation of cells was observed and flow cytometry was performe

The male-killing symbiont Arsenophonus nasoniae is a gamma-proteobacterium that infects parasitic wasps; the male progeny of infected females exhibit increased embryonic death. In this study, we examined methods to horizontally infect Pteromalus venustus (a parasitoid infesting populations of the alfalfa leafcutting bee Megachile rotundata) with A. nasoniae. We then tested the success of these methods via semi-quantitative PCR and quantitative digital PCR, using a molecular marker specific to A. nasoniae. Controlled parasitoid mating experiments were then undertaken to determine whether infect

The entomopathogenic nematode, Steinernema scapterisci, a specialist parasite of crickets, has been successfully used to combat the southern mole cricket, Neoscapteriscus borellii, which is an invasive pest of turf grass. As an entomopathogenic nematode, S. scapterisci causes rapid death of the insects it infects and uses bacteria to facilitate its parasitism. However, our understanding of the relative contributions of the nematode, S. scapterisci, and its bacterial symbiont, Xenorhabdus innexi, to parasitism remains limited. Here we utilized the sand cricket, Gryllus firmus, as a model host t

Entomopathogenic nematodes (EPNs) and fungi (EPF) are well known biological control agents (BCAs) against insect pests. Similarly, the nematophagous fungi (NF) are considered good BCA candidates for controlling plant parasitic nematodes. Because NF can employ EPNs as food and interact with EPF, we speculate that the simultaneous application of EPNs and EPF might result in higher insect mortality, whereas the triple species combination with NF will reduce the EPN and EPF activity by predation or inhibition. Here we evaluated single, dual (EPN + EPF, EPF + NF, EPN + NF) and triple (EPN + EPF + N

Managed honey bee (Apis mellifera) populations are currently facing unsustainable losses due to a variety of factors. Colonies are challenged with brood pathogens, such as the fungal agent of chalkbrood disease, the microsporidian gut parasite Nosema spp., and several viruses. These pathogens may be transmitted horizontally from worker to worker, vertically from queen to egg and via vectors like the parasitic mite, Varroa destructor. Despite the fact that these pathogens are widespread and often harbored in wax comb that is reused from year to year and transferred across beekeeping operations,

Tail fan necrosis (TFN) is the bacterial infection of the tail fan of spiny lobsters which leads to melanosis and erosion of the tail fan tissues. The condition is commonly found among spiny lobsters in aquaculture and commercial fisheries, and greatly reduces their commercial value. This study describes the pathology of TFN by examining the tail fans (telson, uropods) and internal organs (mid-gut, hepatopancreas, heart and gill) of 29 affected wild spiny lobsters (Jasus edwardsii) and 14 unaffected in New Zealand. Initial signs of TFN were observed around the margins of lacerations to the tai

Mosquitoes transmit several diseases, which are of global significance (malaria, dengue, yellow fever, Zika). The geographic range of mosquitoes is increasing due to climate change, tourism and trade. Both conidial and blastospore formulations of the entomopathogenic fungus, Metarhizium brunneum ARSEF 4556, are being investigated as mosquito larvicides. However, concerns have been raised over possible non-target impacts to arthropod mosquito predators such as larvae of Toxorhynchites brevipalpis which feed on larvae of mosquito vector species. Laboratory-based, small container bioassays showed

The crayfish plague agent Aphanomyces astaci is one of the world's most threatening invasive species. Originally from North America, the pathogen is being imported alongside American crayfish species, which are used for various purposes. In this study, we investigated the marginal, currently known distribution area of the pathogen in Eastern Europe by sampling narrow-clawed crayfish (Astacus leptodactylus) and spiny-cheek crayfish (Orconectes limosus) populations. In addition, using specific real-time PCR, we tested several marine decapod species, which also occur in brackish waters of the Dan

The mosquito-parasitic nematode, Strelkovimermis spiculatus (Mermithidae: Nematoda) emerges from hosts and aggregates to form mating clusters characterized by intense male-male competition for females. Successful males deposit an adhesive copulatory plug over the female vulva after insemination. In choice experiments, males strongly preferred virgin females, whereas plugged females were ignored. Males made no attempt to displace copulatory plugs deposited by previous males. Plugged females repelled males without the need for physical contact. The observed chemical repellency was independent of

The sustainable intensification of crustacean aquaculture, which is dominated by the farming of penaeid shrimp species, continues to be beset by viral disease outbreaks. Despite this, reports exist of differential susceptibility to viral infection between different shrimp species and populations, and between shrimp and other decapod crustaceans. These reports have, in part, provided the motivation to identify key mechanisms of antiviral resistance, or refractivity, in commercially-important species. Within the last decade these studies have created significant advances in our understanding of

Despite large economic losses attributable to white spot syndrome virus (WSSV), an infectious pathogen of penaeid shrimp and other crustaceans worldwide, no efficient vaccines or antiviral agents to control the virus are available at present. Here, we designed and constructed baculovirus-based vaccines delivering genes encoding the WSSV envelope proteins, VP28 and VP19. To enhance the immunogenicity of the baculovirus-based vaccine, we fused a Salmonella typhimurium flagellin 2 (FL2) gene with VP28 or VP19 gene. Both vaccine constructs elicited similar high titlers of anti-WSSV IgG after oral

Many insects and Dipterans in particular are known to spend considerable time grooming, but whether these behaviors actually are able to remove pathogenic fungal conidia is less clear. In this study, we examined whether grooming serves to protect flies by reducing the risk of fungal infection in Drosophila melanogaster. First, we confirmed that fungi were removed by grooming. Entomopathogenic, opportunistic, and plant pathogenic fungi were applied on the body surface of the flies. To estimate grooming efficiency, the number of removal conidia through grooming was quantified and we successfully

The crayfish plague pathogen (Aphanomyces astaci) causes mass mortalities of European crayfish when transmitted from its original North American crayfish hosts. Little is known, however, about interspecific transmission of the pathogen between different American crayfish species, although evidence from trade of ornamental crayfish suggests this may happen in captivity. We screened signal and virile crayfish for A. astaci at allopatric and sympatric sites in a UK river. Whilst the pathogen was detected in signal crayfish from both sites, infected virile crayfish were only found in sympatry. Gen

Although the introduction of the crayfish plague pathogen Aphanomyces astaci to Europe is responsible for substantial declines in native crayfish populations throughout the whole continent, its presence has never been officially confirmed in many European regions, including most of the Balkan Peninsula. We demonstrate that the recent crayfish mortality observed in Bosnia and Herzegovina (Mostarsko blato karst field, Neretva river drainage) was caused by A. astaci. The causative strain is known only from European crayfish, indicating that A. astaci poses a threat to native species in this regio

The effect of infection by Echinostoma paraensei on the mitochondrial physiology of Biomphalaria glabrata was investigated after exposure to 50 miracidia. The snails were dissected one, two, three and four weeks after infection for collection and mechanical permeabilization of the gonad-digestive gland (DGG) complex. The results obtained indicate that prepatent infection by this echinostomatid fluke significantly suppresses the phosphorylation state (respiratory state 3) and basal oxygen consumption of B. glabrata, demonstrating that the infection reduces the ability of the intermediate host t

Cymatocarpus solearis, a brachycoeliid trematode that parasitizes sea turtles, uses spiny lobsters Panulirus argus as second intermediate hosts. We examined variability in infection by C. solearis in Bahía de la Ascensión, Mexico, a tropical bay with a highly productive fishery for P. argus that is based on the extensive use of casitas (large artificial shelters), which can harbor multiple juveniles and adults of this gregarious lobster species. We sampled 3956 lobsters from 466 casitas distributed over three bay zones within two closed seasons and one fishing season. In these lobsters (9.5-11

Throughout their global range, wild monarch butterflies (Danaus plexippus) are infected with the protozoan Ophryocystis elektroscirrha (OE). In monarchs, OE infection reduces pupal eclosion, adult lifespan, adult body size and flight ability. Infection of other butterfly hosts with OE is rare or unknown, and the only previously published records of OE infection were on monarch and queen butterflies (D. gilippus). Here we explored the occurrence and specificity of OE and OE-like parasites in four Danaus butterfly species. We surveyed wild D. eresimus (soldier), D. gilippus (queen), D. petilia (
