Aging-related autonomic nervous system imbalance and adrenergic regulation of immunity: Implications for inflammaging, autoimmunity, and long COVID.

The autonomic nervous system (ANS) plays a central role in immune homeostasis by integrating sympathetic and parasympathetic signals that regulate inflammation, immune cell trafficking, and tolerance. Aging is associated with a progressive autonomic imbalance characterized by sympathetic overactivation, reduced parasympathetic tone, and impaired β-adrenergic signaling in immune cells, which together contribute to inflammaging and immune dysregulation. In this review, we discuss how aging-related alterations in adrenergic pathways affect immune cell differentiation and cytokine networks, with p

Slow breathing impacts inter-organ dynamics modulating brain function and risk behavior.

Successful decision-making requires that external information be interpreted in the context of the body's state. Within the framework of body-brain interaction, deliberately modifying one's autonomic state can shape how we evaluate the world, ultimately influencing choices. Yet, it remains unclear whether and how intentional autonomic regulation affects human decision-making. In this study, we tested instructed use of prolonged exhalation, a slow-breathing technique designed to boost parasympathetic activity during risky decision-making. Participants followed distinct breathing protocols while

Transcutaneous vagus nerve stimulation in breast cancer: a neuroimmune model to improve quality of life.

Breast cancer care has shifted beyond remission toward optimizing long-term physiological, emotional, and functional recovery. Many survivors continue, however, to experience persistent symptom clusters, such as insomnia, fatigue, anxiety, pain, depression, and cognitive impairment. These poor quality of life outcomes reflect underlying dysregulation of autonomic, neuroendocrine, and immune systems. Autonomic imbalance characterized by vagal withdrawal and sympathetic hyperactivation is linked to increased inflammatory load, impaired stress regulation, disrupted sleep, and poorer survival outc

Neurogenic Cerebral Edema: Proposed Mechanism for Vasogenic Cerebral Edema.

Global cerebral edema (GCE) is a frequent and devastating manifestation of early brain injury after aneurysmal subarachnoid hemorrhage (aSAH), yet its exact inciting biology and sequence of events remain incompletely explained. In this viewpoint, we propose neurogenic cerebral edema (NCE) as a mechanistic hypothesis for vasogenic edema after aSAH, in which autonomic disequilibrium may amplify blood-brain barrier permeability within otherwise mixed cellular, ionic, and vasogenic edema. We integrate evidence that aSAH perturbs the central autonomic network, producing a sympathetic catecholamine

Autonomic control of blood pressure in women: The roles of hypertension and aging.

Hypertension remains a leading cause of cardiovascular morbidity and mortality worldwide, with implications in women's health across the lifespan. Of note, autonomic nervous system imbalances have been linked to the development of hypertension and are present with natural aging and various conditions unique to women (e.g., menopause, hypertensive disorders of pregnancy, polycystic ovary syndrome). As such, this article reviews the critical changes in the sympathetic and parasympathetic nervous systems that occur in women and may contribute to the development of hypertension. Special focus is p

The Different Facets of Heart Rate Variability in Obstructive Sleep Apnea.

Obstructive sleep apnea (OSA), a heterogeneous and multifactorial sleep related breathing disorder with high prevalence, is a recognized risk factor for cardiovascular morbidity and mortality. Autonomic dysfunction leads to adverse cardiovascular outcomes in diverse pathways. Heart rate is a complex physiological process involving neurovisceral networks and relative regulatory mechanisms such as thermoregulation, renin-angiotensin-aldosterone mechanisms, and metabolic mechanisms. Heart rate variability (HRV) is considered as a reliable and non-invasive measure of autonomic modulation response

Heart rate variability regression and risk of sudden unexpected death in epilepsy.

The exact mechanisms of sudden unexpected death in epilepsy remain elusive, despite there is consensus that SUDEP is associated with severe derangements in the autonomic control to vital functions as breathing and heart rate regulation. Heart rate variability (HRV) has been advocated as biomarker of autonomic control to the heart. Cardiac dysautonomia has been found in diseases where other branches of the autonomous nervous system are damaged, as Parkinson disease and multiple system atrophy. In this perspective, an impaired HRV not only is a risk factor for sudden cardiac death mediated by ar

IFN-induced cell-autonomous immune mechanisms in the control of intracellular protozoa.

Vertebrate cells have evolved an elaborate multi-tiered intracellular surveillance system linked to downstream antimicrobial effectors to defend themselves from pathogens. This cellular self-defense system is referred to as cell-autonomous immunity. A wide array of cell-autonomous mechanisms operates to control intracellular pathogens including protozoa such as Toxoplasma gondii. Cell-autonomous immunity consists of antimicrobial defenses that are constitutively active in cells and those that are inducible typically in response to host cell activation. The IFN family of cytokines is an importa

Microstructural integrity of autonomic central nervous tracts is linked to cardiovascular health.

INTRODUCTION: The central autonomic network (CAN), which controls sympathetic and parasympathetic activity, plays a central role in the pathogenesis of cardiovascular disease and serves as a target for therapeutic approaches. This study investigates the association of microstructural integrity of white matter fibres representing the CAN with cardiovascular damage, risk and outcomes using a large population-based cohort study. METHODS: The microstructural integrity of the CAN was evaluated using magnetic resonance imaging (MRI) of the brain from the population-based UK Biobank study applying p

The AtFLC-AtFT Pathway Is Involved in the Early Flowering Promoted by Loss of AtHO1 Function in Arabidopsis.

Although previous studies have indicated that heme oxygenase 1 (HO1/HY1) regulates the flowering time via the photoperiod pathway, the specific mechanism is still elusive. Here, we found that the Arabidopsis hy1-100 mutant displayed early flowering, and the characteristic expression patterns of several master genes involved in the autonomous pathway were altered. Notably, the transcript levels of FLOWERING LOCUS C (AtFLC) gene declined developmentally in both wild-type and hy1-100 mutant, with a more pronounced fold reduction observed in the mutant. Genetic evidence further underlined that hy1

Anatomical considerations for transanal minimal-invasive surgery: the caudal to cephalic approach.

AIM: Nerve-sparing surgery during laparoscopic rectal mobilization is still limited by anatomical constraints such as obesity, the narrowness of the male pelvis, an ultra low rectal cancer or all of these. The transanal approach for total mesorectal excision has overcome the shortcomings of limited access to the rectal 'no-man's land' close to the pelvic floor. The aim of this anatomical study was to define a roadmap of anatomical landmarks for the caudal to cephalic approach so as to standardize nerve-sparing rectal mobilization procedures. METHOD: Macroscopic dissections of the pelvis in a

Autonomic nerves terminating on microvessels in the pineal organs of various submammalian vertebrates.

In earlier works we have found that in the mammalian pineal organ, a part of autonomic nerves--generally thought to mediate light information from the retina--form vasomotor endings on smooth muscle cells of vessels. We supposed that they serve the vascular support for circadian and circannual periodic changes in the metabolic activity of the pineal tissue. In the present work, we investigated whether peripheral nerves present in the photoreceptive pineal organs of submammalians form similar terminals on microvessels. In the cyclostome, fish, amphibian, reptile and bird species investigated, a

Polyvagal Theory and Neonatal Sleep: A Review of Autonomic Regulation and Implications for Sudden Infant Death Syndrome.

BACKGROUND: Neonatal sleep is critical for brain maturation and autonomic nervous system regulation. Disruptions in sleep patterns and vagal tone may contribute to the risk of sudden infant death syndrome (SIDS). METHODS: This narrative review summarizes current evidence on the relationship between neonatal sleep states, autonomic nervous system maturation and polyvagal theory. RESULTS: Studies suggest that vagal activity modulates neonatal sleep states, with implications for both emotional and physiological development. Premature infants may display immature vagal pathways, increasing their

[Sweating-related tears--a new syndrome?].

Two years after operative correction of a middle third fracture of the face, a patient suffered unilateral tearing and homolateral rhinorrhea as soon as he started sweating. This is explained by abnormal regeneration of autonomic nerve fibres. The possible anatomic pathways are discussed and compared with similar syndromes in the literature.

Neurophysiology of Rett syndrome.

Neurophysiological evaluations have been widely applied in the study of Rett syndrome (RS) to provide information concerning the developmental aspects of RS; the character and extent of involvement of the central, peripheral, and autonomic nervous system pathways; and evaluation of the clinical symptomatology of RS. The electroencephalogram (EEG) is invariably abnormal and shows characteristic, though not diagnostic, changes: loss of expected developmental features; the appearance of focal, multifocal, and generalized epileptiform abnormalities; and the occurrence of rhythmic slow (theta) acti

Necroptosis and cellular stress characterize immune and endothelial dysfunction in long COVID.

BACKGROUND: Long COVID, or post-acute sequelae of severe acute respiratory syndrome coronavirus 2 infection, affects a significant proportion of COVID-19 survivors and is associated with persistent fatigue, dysautonomia, and cardiovascular complications. The cellular mechanisms underlying these chronic symptoms remain incompletely understood. OBJECTIVE: We sought to investigate immune and endothelial cell dysfunction, with a focus on cell stress and death pathways, in individuals with long COVID compared with matched infection-recovered controls. METHODS: We conducted a cross-sectional study

Central nervous system and cancer: mechanistic insights and therapeutic opportunities CNS and cancer: a review.

Cancer is increasingly recognized as a systemic disease whose biology is shaped by reciprocal interactions with its microenvironment, including the central and peripheral nervous systems. Recent evidence demonstrates that neural inputs can directly promote tumour growth through synaptic, paracrine, and neuroendocrine mechanisms, and that these inputs intersect with canonical drug-resistance pathways, creating new opportunities and challenges for precision oncology. Primary brain tumours such as gliomas form bona fide functional synapses with neurons, hijacking both excitatory (AMPA/NMDA) and i
