Conditional Overexpression of Neuritin in Supporting Cell Protects Cochlear Hair Cell and Delays Age-Related Hearing Loss by Enhancing Autophagy.

Age-related hearing loss (ARHL) is a highly prevalent, burdensome sensorineural hearing loss closely associated with impaired autophagic influx. Our previous studies revealed that neuritin, a neurotrophic factor primarily expressed in the central nervous system, could alleviate drug-induced damages in hair cells (HCs) and spiral ganglion neurons. However, its effects on ARHL and whether these effects are closely related to autophagy remain unclear. Using the Nrn1 knock-in mice and cultured cochlear basilar membrane (CBM) of the neonatal mouse, we show that neuritin could restore aging-associat

Brain-Specific Angiogenesis Inhibitor 3 Is Expressed in the Cochlea and Is Necessary for Hearing Function in Mice.

Mammalian auditory hair cells transduce sound-evoked traveling waves in the cochlea into nerve stimuli, which are essential for hearing function. Pillar cells located between the inner and outer hair cells are involved in the formation of the tunnel of Corti, which incorporates outer-hair-cell-driven fluid oscillation and basilar membrane movement, leading to the fine-tuned frequency-specific perception of sounds by the inner hair cells. However, the detailed molecular mechanism underlying the development and maintenance of pillar cells remains to be elucidated. In this study, we examined the

Fatigue Modeling via Mammalian Auditory System for Prediction of Noise Induced Hearing Loss.

Noise induced hearing loss (NIHL) remains as a severe health problem worldwide. Existing noise metrics and modeling for evaluation of NIHL are limited on prediction of gradually developing NIHL (GDHL) caused by high-level occupational noise. In this study, we proposed two auditory fatigue based models, including equal velocity level (EVL) and complex velocity level (CVL), which combine the high-cycle fatigue theory with the mammalian auditory model, to predict GDHL. The mammalian auditory model is introduced by combining the transfer function of the external-middle ear and the triple-path nonl

Characterizing auditory processing and perception in individual listeners with sensorineural hearing loss.

This study considered consequences of sensorineural hearing loss in ten listeners. The characterization of individual hearing loss was based on psychoacoustic data addressing audiometric pure-tone sensitivity, cochlear compression, frequency selectivity, temporal resolution, and intensity discrimination. In the experiments it was found that listeners with comparable audiograms can show very different results in the supra-threshold measures. In an attempt to account for the observed individual data, a model of auditory signal processing and perception [Jepsen et al., J. Acoust. Soc. Am. 124, 42

[Effects of intrauterine hypotrophy and perinatal hypoxia on cochlear function evaluated by click evoked otoacoustic emissions (TEOAE)].

The authors evaluated mutual relationships between transiently evoked otoacoustic emissions (TEOAE) and various parameters defining perinatal period focusing mostly on hypotrophic newborns. TEOAE was collected using 2.5-12.5 msec time-window. TEOAE amplitude and responses (S/N ratio) from various frequency bands calculated using off-line analysis were collected using ILO 88 ECHOPORT Otodynamics. Half-octave frequency bands centered at 1.0, 1.5, 2.0, 3.0, and 4.0 kHz were included to this study. All 370 newborns, 1 to 5 days old, were tested bilaterally. The relationship between TEOAE and birth

Histologic features of paraneoplastic pemphigus.

BACKGROUND: We describe the histopathologic features of paraneoplastic pemphigus, a recently described autoimmune mucocutaneous disease associated with neoplasia. Complete evaluation for paraneoplastic pemphigus requires identification of the characteristic mucocutaneous eruption, tissue specimens for routine histologic and direct immunofluorescence evaluation, and identification of circulating autoantibodies with a unique specificity. Immunoprecipitation from keratinocytes reveals a characteristic complex of four proteins with the circulating antibodies. Various neoplasms have been identified

Summating potential and water balance in Meniere's disease.

The summating potentials (SP) to free-field 1-, 2-, 4-, and 8-kHz, 90-dB HL tone bursts were recorded by means of transtympanic electrocochleography in 45 patients divided into three groups: those with Meniere's disease, those whose diagnosis was uncertain, and control subjects. The similarities across frequencies between the human SP (promontory) and the experimental SP (DIF component from basal turn) suggest that the SP recorded in electrocochleography originates mostly from the base of the cochlea; negative values were observed at low frequencies (1 to 2 kHz), positive values at high freque

[Elevation of extracellular K and its potentiation of the response to vasoactive substances in the rabbit basilar artery (author's transl)].

The authors have suggested from an in vitro study that extracellular K concentration can be elevated up to 40 Mm, when hemolysis of a clot occurs after subarachnoid hemorrhage. The present experiments were performed to investigate the change in contractility of rabbit basilar artery to vasoactive substances in the presence of increased K. The conclusion was as follows. 1. The contraction heights induced by serotonin (5 x 10(-7) M) were increased 2.4 +/- .02 fold, 6.4 +/- 0.3 fold, 5.7 +/- 0.4 fold (n = 10), when the K concentration of physiological solution was elevated to 8.1 mM, 13.5 mM, 24.

Regeneration after tall hair cell damage following severe acoustic trauma in adult pigeons: correlation between cochlear morphology, compound action potential responses and single fiber properties in single animals.

The time course of recovery of compound action potential (CAP) thresholds was observed in individual adult pigeons after severe acoustic trauma. Pigeons were overstimulated with a tone of 0.7 kHz and 136-142 dB SPL presented to one ear for 1 h under general anesthesia. Recovery of CAP audiograms was monitored at regular intervals after trauma. A new semi-stereotaxic approach to the peripheral part of the auditory nerve was developed. This permitted activity from single auditory nerve fibers to be recorded over a wide range of characteristic frequencies (CFs), including high CFs, without having

Coupled Finite Element Model of the Middle and Inner Ear as Virtual Test Environment for Stapes Surgery.

In order to evaluate the performance of different types of middle-ear prostheses, a model of human ear was developed. The model was created using finite element (FE) method with the ossicles modeled as rigid bodies. First, the middle-ear FE model was developed and validated using the middle-ear transfer function measurements available in literature including pathological cases. Then, the inner-ear FE model was developed and validated using tonotopy, impedance, and relative BM motion level curves from literature. Both models are based on preexisting research with some improvements and were comb

Development of an Assessment Model for the Effect of the Replacement of Minimal Artificial Ossicles on Hearing in the Inner Ear.

Due to ethical issues and the nature of the ear, it is difficult to directly perform experimental measurements on living body elements of the human ear. Therefore, a numerical model has been developed to effectively assess the effect of the replacement of artificial ossicles on hearing in the inner ear. A healthy volunteer's right ear was scanned to obtain CT data, which were digitalized through the use of a self-compiling program and coalescent Patran-Nastran software to establish a 3D numerical model of the whole ear, and a frequency response of a healthy human ear was analyzed. The vibratio

Targeted Deletion of Loxl3 by Col2a1-Cre Leads to Progressive Hearing Loss.

Collagens are major constituents of the extracellular matrix (ECM) that play an essential role in the structure of the inner ear and provide elasticity and rigidity when the signals of sound are received and transformed into electrical signals. LOXL3 is a member of the lysyl oxidase (LOX) family that are copper-dependent amine oxidases, generating covalent cross-links to stabilize polymeric elastin and collagen fibers in the ECM. Biallelic missense variant of LOXL3 was found in Stickler syndrome with mild conductive hearing loss. However, available information regarding the specific roles of L

Imaging of Temporal Bone.

Multidetector computed tomography has been the benchmark for visualizing bony changes of the ear, but has recently been challenged by cone-beam computed tomography. In both methods, all inner ear bony structures can be visualized satisfactorily with 2D or 3D imaging. Both methods produce ionizing radiation and induce adverse health effects, especially among children. In 3T magnetic resonance imaging, the soft tissue can be imaged accurately. Use of gadolinium chelate (GdC) as a contrast agent allows the partition of fluid spaces to be visualized, such as the bulging of basilar and Reissner's m

A new approach to a damage risk criterion for weapons impulses.

Existing damage-risk criteria for weapons impulses suffer from their lack of a theoretical basis, thereby limiting their generalizability and utility. Furthermore, a number of studies now indicate that they may be inaccurate for impulses with energy in the low frequency region (Dancer & al., 1985; Patterson & al., 1985; Price & al., 1989). We have approached the problem by modeling the ear mathematically as a means of gaining insight into the loss processes. In the model, the external and middle ears are linear at lower intensities; however, the stapes displacement is limited to 20 microns, as

An evaluation of per-scalar cochlear electrode implantation techniques. An histopathological study in cats.

This study was carried out to evaluate the effects of infection and trauma on the organ of Corti and spiral ganglion cells following per-scalar cochlear electrode implantations in cats. The results showed that the incidence of infection with a per-scalar electrode inserted through an opening made directly into the apical and middle turns is low, provided the procedure is carried out aseptically with an antibiotic cover. Trauma could also be kept to a minimum by inserting the electrode into the scala tympani, taking care to insert the electrode without applying force, and ensuring that the firs

3D Computational Modeling of Blast Wave Transmission in Human Ear From External Ear to Cochlear Hair Cells: A Preliminary Study.

INTRODUCTION: Auditory disabilities like tinnitus and hearing loss caused by exposure to blast overpressures are prevalent among military service members and veterans. The high-pressure fluctuations of blast waves induce hearing loss by injuring the tympanic membrane, ossicular chain, or sensory hair cells in the cochlea. The basilar membrane (BM) and organ of Corti (OC) behavior inside the cochlea during blast remain understudied. A computational finite element (FE) model of the full human ear was used by Bradshaw et al. (2023) to predict the motion of middle and inner ear tissues during blas

Hampshire Sheep as a Large-Animal Model for Cochlear Implantation.

BACKGROUND: Sheep have been proposed as a large-animal model for studying cochlear implantation. However, prior sheep studies report that the facial nerve (FN) obscures the round window membrane (RWM), requiring FN sacrifice or a retrofacial opening to access the middle-ear cavity posterior to the FN for cochlear implantation. We investigated surgical access to the RWM in Hampshire sheep compared to Suffolk-Dorset sheep and the feasibility of Hampshire sheep for cochlear implantation via a facial recess approach. METHODS: Sixteen temporal bones from cadaveric sheep heads (ten Hampshire and si

Changes in the cerebrospinal fluid lipid profile following subarachnoid hemorrhage in a closed cranium model: Correlations to cerebral vasospasm, neuronal cell death and Interleukin-6 synthesis. A pilot study.

BACKGROUND: Phospholipids and sphingolipids are cell membrane components, that participate in signaling events and regulate a wide variety of vital cellular processes. Sphingolipids are involved in ischemic stroke pathophysiology. Throughout cleavage of membrane sphingomyelin by sphingomyelinase in stroke patients, it results in increased Ceramide (Cer) levels in brain tissue. Different studies showed the evidence that sphingomyelinase with Cer production induces expression of interleukin (IL)-6 and have vasoconstrictive proprieties. With this study, we intend to evaluate cerebrospinal fluid (
