Looking to the Future: How Will Personalised Medicine Impact Facial Plastic Surgery.

AIMS AND BACKGROUNDS: The objectives of this study are to examine the emerging role of personalized medicine in facial plastic surgery and to consider how biologically, anatomically, and psychologically tailored approaches may refine both aesthetic and reconstructive care. HISTORICAL ASPECTS: Facial plastic surgery has traditionally relied on anatomical principles, surgical expertise, and population-based evidence. Personalized medicine represents a shift toward more individualized care by incorporating patient-specific biological and phenotypic variation into clinical decision-making. ANATO

Metagenomic global survey and in-depth genomic analyses of Ruminococcus gnavus reveal differences across host lifestyle and health status.

Ruminococcus gnavus is a gut bacterium found in > 90% of healthy individuals, but its increased abundance is also associated with chronic inflammatory diseases, particularly Crohn's disease. Nevertheless, its global distribution and intraspecies genomic variation remain understudied. By surveying 12,791 gut metagenomes, we recapitulated known associations with metabolic diseases and inflammatory bowel disease. We uncovered a higher prevalence and abundance of R. gnavus in Westernized populations and observed bacterial relative abundances up to 83% in newborns. Next, we built a resource of R. g

Human airborne eDNA as forensic evidence in simulated residential burglaries.

Airborne environmental DNA (eDNA) is emerging as a valuable tool in forensic genetics to support individual identification in cases where other biological traces are limited or absent. Here, we investigate the potential of human airborne eDNA as a source of forensic evidence by evaluating its recovery following simulated residential burglaries. A total of 391 eDNA samples were collected from air, dust, and surfaces in 22 households by two laboratories. Samples were recovered under two conditions: immediately after residents vacated the premises and following a 15-minute simulated burglary. Res

Using Real-World Data to Guide Ustekinumab Dosing Strategies for Psoriasis: A Prospective Pharmacokinetic-Pharmacodynamic Study.

Variation in response to biologic therapy for inflammatory diseases, such as psoriasis, is partly driven by variation in drug exposure. Real-world psoriasis data were used to develop a pharmacokinetic/pharmacodynamic (PK/PD) model for the first-line therapeutic antibody ustekinumab. The impact of differing dosing strategies on response was explored. Data were collected from a UK prospective multicenter observational cohort (491 patients on ustekinumab monotherapy, drug levels, and anti-drug antibody measurements on 797 serum samples, 1,590 measurements of Psoriasis Area Severity Index (PASI)).

Sex and Gender Differences in Physical Activity, Sedentary Behavior, and Sleep During Traumatic Brain Injury Recovery: A Narrative Review.

Traumatic brain injury (TBI) is a complex neurological condition with enduring consequences that extend well beyond the initial insult. Once regarded primarily as an acute event, TBI is now increasingly recognized as a chronic disorder associated with cognitive, emotional, and physical impairments. These sequelae contribute to diminished quality of life and elevated long-term risks for adverse health outcomes. Despite growing awareness of its multifaceted nature, TBI recovery remains heterogeneous and is shaped by a range of individual-level determinants that remain insufficiently understood.

Identification of multicohort-based predictive signature for NMIBC recurrence reveals SDCBP as a novel oncogene in bladder cancer.

BACKGROUND: Despite surgical and intravesical chemotherapy interventions, non-muscle invasive bladder cancer (NMIBC) poses a high risk of recurrence, which significantly impacts patient survival. Traditional clinical characteristics alone are inadequate for accurately assessing the risk of NMIBC recurrence, necessitating the development of novel predictive tools. METHODS: We analyzed microarray data of NMIBC samples obtained from the ArrayExpress and GEO databases. LASSO regression was utilized to develop the predictive signature. We combined gene signature and clinicopathological factors to

Efficient and flexible Integration of variant characteristics in rare variant association studies using integrated nested Laplace approximation.

Rare variants are thought to play an important role in the etiology of complex diseases and may explain a significant fraction of the missing heritability in genetic disease studies. Next-generation sequencing facilitates the association of rare variants in coding or regulatory regions with complex diseases in large cohorts at genome-wide scale. However, rare variant association studies (RVAS) still lack power when cohorts are small to medium-sized and if genetic variation explains a small fraction of phenotypic variance. Here we present a novel Bayesian rare variant Association Test using Int

Assessing the Association of Mitochondrial Genetic Variation With Primary Open-Angle Glaucoma Using Gene-Set Analyses.

PURPOSE: Recent studies indicate that mitochondrial proteins may contribute to the pathogenesis of primary open-angle glaucoma (POAG). In this study, we examined the association between POAG and common variations in gene-encoding mitochondrial proteins. METHODS: We examined genetic data from 3430 POAG cases and 3108 controls derived from the combination of the GLAUGEN and NEIGHBOR studies. We constructed biological-system coherent mitochondrial nuclear-encoded protein gene-sets by intersecting the MitoCarta database with the Kyoto Encyclopedia of Genes and Genomes (KEGG) database. We examined

A Dynamic Model of Weekly Sleep Timing: Endogenous Origins of Social Jetlag.

BACKGROUND: Social jetlag is commonly attributed to externally imposed schedules that conflict with biological sleep timing. However, individuals also make daily decisions about how to allocate time between sleep and waking activities, suggesting that behavioral tradeoffs may contribute to within-week variation in sleep patterns. OBJECTIVE: To examine whether weekly patterns in sleep timing and duration, including social jetlag, can emerge endogenously from the interaction of biological sleep regulation and time allocation decisions. METHODS: We develop a dynamic model of sleep timing that i

Single-nephron proteomes connect morphology and function in proteinuric kidney disease.

In diseases of many parenchymatous organs, heterogeneous deterioration of individual functional units determines the clinical prognosis. However, the molecular characterization at the level of such individual subunits remains a technological challenge that needs to be addressed in order to better understand pathological mechanisms. Proteinuric glomerular kidney diseases are frequent and assorted diseases affecting a fraction of glomeruli and their draining tubules to variable extents, and for which no specific treatment exists. Here, we developed and applied a mass spectrometry-based methodolo

Routine Blood Biomarkers and CD4⁺ T-Cell States in Atherosclerosis: A Narrative Review and Conceptual Framework.

BACKGROUND: Atherosclerosis (AS) is a chronic inflammatory disease driven by complex immune responses. Among the involved immune cells, CD4⁺ T cells are closely associated with plaque progression. An imbalance between inflammatory and regulatory T-cell responses can sustain vascular inflammation. However, the relationship between routinely measured blood biomarkers and these T-cell states remains unclear. MAIN BODY: This narrative review examines how LDL-C, HDL-C, HbA1c, high-sensitivity C-reactive protein (hsCRP) and creatinine relate to biological processes involved in CD4⁺ T-cell regulatio

The next frontier: AI, multi-omics, and predictive models for weight gain in HIV.

PURPOSE OF REVIEW: People with HIV (PWH) are increasingly susceptible to excess weight gain and obesity after initiation of antiretroviral therapy. However, there is substantial variation in individual weight gain that is difficult to predict with clinical factors alone. We review emerging methods in machine learning and multi-omics that address the biology and prediction of weight gain and weight-associated conditions, persistent challenges, and future directions. RECENT FINDINGS: PWH continue to have increasing burden of excess weight gain and obesity. Obesity has a complex pathophysiology

Trichotillomania, skin picking disorder, and pregnancy.

BACKGROUND: Trichotillomania (hair pulling disorder) and skin picking disorder have typical age of onset around puberty, and appear more common in women than in men, suggesting the possibility that hormonal systems may relate to symptoms. Yet virtually no research has explored relationships between pregnancy (and the post-partum period) and symptom severity in people affected by these conditions. METHODS: People with trichotillomania and/or skin picking disorder were recruited from the general community if they had previously experienced at least one pregnancy during their lifetimes. Association between methylene tetrahydrofolate reductase and glutathione S-transferase M1 gene polymorphisms and chronic myeloid leukemia in a Brazilian population.

Chronic myeloid leukemia is a hematopoietic stem cell disorder that causes uncontrolled proliferation of white blood cells. Although the clinical and biological aspects are well documented, little is known about individual susceptibility to this disease. We conducted a case-control study analyzing the prevalence of the polymorphisms MTHFR C677T, MTHFR A1298C, del{GSTM1}, del{GSTT1}, and haptoglobin in 105 patients with chronic myeloid leukemia (CML) and 273 healthy controls, using PCR-based methods. A significant association with risk of developing CML was found for MTHFR 1298AA (odds ratio (O

Causes of variation of copper, iron, manganese, zinc and magnesium levels in bovine livers. 3. The effects of locality.

The effects of locality on the copper, iron, manganese, zinc and magnesium levels in 407 bovine caudate lobe liver samples preserved in formalin for differing storage periods were examined. The mineral determinations, expressed on the wet basis (WB), were made by atomic absorption spectrophotometry after wet ashing of the liver. Two hundred and ten of the liver samples were from cattle from one farm (Farm 1) the remaining 197 cattle being from another farm (Farm 2). The copper, iron and magnesium levels were taken as indicative of the hepatic concentrations at slaughter. Locality had a signifi

Realtime light dosimetry software tools for interstitial photodynamic therapy of the human prostate.

Photodynamic therapy (PDT) for the treatment of prostate cancer has been demonstrated to be a safe treatment option capable of inducing tissue destruction and decreasing prostate specific antigen (PSA) levels. However, prostate-PDT results in large intra- and interpatient variations in treatment response, possibly due to biological variations in tissue composition and short-term response to the therapeutic irradiation. Within our group, an instrument for interstitial PDT on prostate tissue has been developed that combines therapeutic light delivery and monitoring of light transmission via nume

Uncertainties in dose coefficients from ingestion of 131I, 137Cs, and 90Sr.

Quantification of uncertainties in doses from intakes of radionuclides is important in risk assessments and epidemiologic studies of individuals exposed to radiation. In this study, the uncertainties in the doses per unit intake (i.e., dose coefficients) for ingestion of 131I, 137Cs, and 90Sr by healthy individuals have been determined. Age-dependent thyroid dose coefficients were derived for 131I. The analysis for 131I uses recent measurements of thyroid volume obtained by ultrasonography, which indicate a thyroid mass lower than that previously obtained using autopsy measurements. The coeffi
