Robust protein-based engineering of hepatocyte-like cells from human mesenchymal stem cells.

BACKGROUND: Cells of interest can be prepared from somatic cells by forced expression of lineage-specific transcription factors, but it is required to establish a vector-free system for their clinical use. Here, we report a protein-based artificial transcription system for engineering hepatocyte-like cells from human umbilical cord-derived mesenchymal stem cells (MSCs). METHODS: MSCs were treated for 5 days with 4 artificial transcription factors (4F), which targeted hepatocyte nuclear factor (HNF)1α, HNF3γ, HNF4α, and GATA-binding protein 4 (GATA4). Then, engineered MSCs (4F-Heps) were subje

Corneal regeneration strategies: From stem cell therapy to tissue engineered stem cell scaffolds.

Corneal epithelial defects and excessive wound healing might lead to severe complications. As stem cells can self-renew infinitely, they are a promising solution for regenerating the corneal epithelium and treating severe corneal epithelial injury. The chemical and biophysical properties of biological scaffolds, such as the amniotic membrane, fibrin, and hydrogels, can provide the necessary signals for stem cell proliferation and differentiation. Multiple researchers have conducted investigations on these scaffolds and evaluated them as potential therapeutic interventions for corneal disorders

Mesenchymal stem cells and progenitor cells in connective tissue engineering and regenerative medicine: is there a future for transplantation?

PURPOSE: Transplantation surgery suffers from a shortage of donor organs worldwide. Cell injection and tissue engineering (TE), thus emerge as alternative therapy options. The purpose of this article is to review the progress of TE technology, focusing on mesenchymal stem cells (MSC) as a cell source for artificial functional tissue. RESULTS: MSC from many different sources can be minimally invasively harvested: peripheral blood, fat tissue, bone marrow, amniotic fluid, cord blood. In comparison to embryonic stem cells (ESC), there are no ethical concerns; MSC can be extracted from autologous

From basic biology to engineered therapies: the keratinocyte stem cell playbook.

Keratinocyte stem cells (KSCs) are the principal drivers of epidermal renewal, barrier maintenance, and wound repair. Their ability to alternate between self-renewal and differentiation is orchestrated by tightly integrated extrinsic and intrinsic programs that ensure tissue stability while enabling rapid regeneration after injury. This review synthesizes current understanding of KSC homeostasis through a unified framework of three interdependent "fate locks"-the identity switch (ΔNp63 ↔ Notch/IRF6-KLF4/GRHL3/OVOL), the cell-cycle lock (E2F/MYC ↔ p21/p27-RB), and the mechanotransduction lock (

Engineering chimeric antigen receptor CD4 T cells for Alzheimer's disease.

Alzheimer's disease (AD) is the prevailing cause of age-associated dementia worldwide. Current standard of care relies on antibody-based immunotherapy. However, antibody-based approaches carry risks for patients, and their effects on cognition are marginal. Increasing evidence suggests that T cells contribute to AD onset and progression. Unlike the cytotoxic effects of CD8+ cells, CD4+ T cells capable of regulating inflammation show promise in reducing pathology and improving cognitive outcomes in mouse models of AD and in aging. Here, we sought to exploit the beneficial properties of CD4+ T c

In vivo engineering of immunosuppressive T cells to treat ocular autoimmune diseases.

Ocular autoimmune diseases, marked by localized inflammation and tissue remodelling, often culminate in irreversible vision loss. Conventional therapies relying on systemic immunosuppression or receptor-blocking antibodies provide sub-optimal effects. Here we assess an in vivo T-cell engineering platform that harnesses a T cell-targeted integration-deficient lentiviral vector (T-IDLE) to reprogram endogenous T cells into regulatory T (Treg)-like cells for treating experimental autoimmune uveitis (EAU) and Graves' ophthalmopathy (GO). The T-IDLE platform was engineered using a mutated cocal gly

Engineering precision oncology: Targeting tumors and immune cells with lentiviral vectors.

Lentiviral vectors (LVs) are emerging as versatile tools for the efficient and stable delivery of therapeutic genes. Although VSV-G-pseudotyped LVs remain the standard for ex vivo genetic engineering, their broad tropism and serum sensitivity limit their applicability for in vivo cancer therapy, where precise targeting is essential for efficacy and safety. This review synthesizes two decades of advances in LV pseudotyping, comparing natural viral envelopes, engineered targeting strategies, and multicomponent glycoprotein systems that reshape vector tropism. We discussed donor-derived envelopes

Cell Membrane-Engineered FePDA Nanoparticles Integrate Ferroptosis and Antitumor Immunity.

Ferroptosis-based cancer therapy shows promise in tumor suppression but is often limited by insufficient activation of antitumor immunity. Here, a hepatocellular carcinoma (HCC) targeted nanoplatform was constructed by coating FePDA nanozymes with Hepa1-6 cell membranes overexpressing ovalbumin (OVA) and CD40 ligand (CD40L), generating tumor membrane-derived OVA/CD40L-functionalized FePDA nanoparticles (FePDA-TMOC). This hybrid architecture integrates homologous tumor targeting with iron homeostasis disruption and co-stimulatory immune activation within a single nanosystem. Mechanistically, th

Dynamic Immune Cell Composition, Phenotypes, and Signaling in an Engineered Metastatic Niche.

In breast cancer patients, metastasis is the stage of disease where prognosis significantly worsens. However, the timing at which metastasis initiates and the location of metastatic lesions in an organ are stochastic, limiting the timely identification of disease and the administration of treatments. Herein, we employ a synthetic metastatic niche comprised of a microporous scaffold to investigate the dynamic immune processes associated with metastatic progression. Upon implantation, the porous scaffold is infiltrated with immune cells and recruits tumor cells. We have previously reported stabl

Engineering synthetic cells with intramembrane domains possessing distinct bilayer asymmetries.

Our understanding of how membrane asymmetry governs biological function is limited by the lack of techniques to produce model membranes which can reliably and accurately mimic cellular membrane asymmetry. Not only in terms of asymmetric lipid distribution, but also how that asymmetry can be confined to specific lateral locations across the membrane. Here we present an inverted emulsion method that can be used to produce synthetic cells with symmetric and asymmetric bilayers, as well as phase separation where the intermembrane domains possess distinct bilayer asymmetries. We assess the degree o

Embedded cell-only bioprinting to engineer structurally aligned meniscal fibrocartilage.

The engineering of functional meniscal grafts remains elusive, largely due to an inability to recapitulate the highly organized collagen architecture of the native tissue which is integral to its biomechanical function. In this study, we investigated whether external geometric confinement can direct collagen alignment in fibrocartilaginous tissues generated by mesenchymal stem/stromal cells (MSCs). First, MSCs were cast within non-adhesive agarose channels, which supported the development of fibrocartilaginous tissues with collagen fibres aligned parallel to the long axis of the confining agar

Engineering and design of artificial enzymes for non-natural biocatalysis in whole cells.

Artificial enzymes represent transformative biocatalysts that overcome the inherent limitations of natural enzymes in substrate scope and activity specificity, enabling challenging new-to-nature reactions. Despite remarkable advances in designing diverse artificial enzymes through synthetic cofactor or non-canonical amino acid incorporation, their widespread industrial application remains hindered by the high costs and operational complexity of in vitro systems. Whole-cell biocatalysis emerges as a promising alternative by capitalizing on the cellular environment, including cofactor regenerati

Engineering Chinese Hamster Ovary Cells to Mitigate Polysorbate Degradation in Biotherapeutics.

A recurring root cause for polysorbate degradation and associated particle observations in liquid biotherapeutics is the presence of residual host cell proteins (HCPs). Some of these HCPs can hydrolyze the polysorbate surfactant to release poorly-soluble fatty acids that accumulate over time and form particles. To tackle this industry-wide issue, we disrupted the genes for a plurality of potential polysorbate-degrading HCPs in CHO cells via two CHO cell engineering approaches. First, we sequentially knocked out six hydrolase genes in a stable recombinant mAb-expressing cell line and substantia

Pickering Emulsion Biocatalysis with Engineered Living Cells for Degrading Polycarbonate Plastics.

The efficient degradation of plastics remains a pressing environmental challenge due to their inherent resistance to breakdown. While biocatalysis offers a promising approach for sustainable and effective plastic degradation, the inherently low solubility of plastics in aqueous systems severely limits the efficiency of enzymatic reactions. To address this issue, we developed a biocompatible polymer coating strategy to engineer living cell surfaces, enabling the stabilization of Pickering emulsions for over 192 h and significantly enhancing plastic accessibility to biocatalysts. Leveraging this

Water-mediated kinetic engineering of CTF QDs for emerging solar cells.

Covalent triazine framework quantum dots (CTF QDs) are promising low-toxicity, high-performance optoelectronic materials featuring molecular-level structural tunability and good charge-carrier mobility. Yet, achieving CTF QDs has long been hindered by the inherently rapid kinetics of triazine cyclization. Here, we overcome this limitation with a simple water-mediated kinetic strategy that modulates the forward Schiff-base reaction rate, thereby delaying triazine cyclization and controlling the degree of amidine-aldehyde polymerization. This approach produces CTF QDs smaller than 3 nm (denoted

NitrOFF: An engineered fluorescent biosensor to illuminate nitrate transport in living cells.

The duality of nitrate is nowhere best exemplified than in human physiology - a detrimental pollutant but also a protective nutrient and signaling ion - particularly as connected to reactive nitrogen oxides. Aside from limited insights into nitrate uptake and storage, foundational nitrate biology has lagged. Genetically encoded fluorescent biosensors can address this gap with real-time imaging. However, imaging technologies for mammalian cell applications remain rare. Here, we set out to design and engineer a two-domain chimera fusing the split green fluorescent protein EGFP and the nitrate re

In Situ Engineering Cancer Mask to Immobilize Tumor Cells and Block Metastasis.

This work reports a new concept of cancer mask in situ to alter the specific biological functions of cancer cells. Metastatic cancer cells are highly invasive in part due to the presence of the glycan matrix in the cell membrane. Using a rational designed bio-orthogonal reaction, the cancer cell surface is reconstructed in situ by incorporating endogenous polysialic acids in the glycan matrix on the cell membrane to form a mesh-like network, called cancer mask. The network of the glycan matrix can not only immobilize cancer cells but also effectively block the stimulation of metastasis promote

Engineered CAR-T cells: An immunotherapeutic approach for cancer treatment and beyond.

Chimeric Antigen Receptor (CAR) T cell therapy is a type of adoptive immunotherapy that offers a promising avenue for enhancing cancer treatment since traditional cancer treatments like chemotherapy, surgery, and radiation therapy have proven insufficient in completely eradicating tumors, despite the relatively positive outcomes. It has been observed that CAR-T cell therapy has shown promising results in treating the majority of hematological malignancies but also have a wide scope for other cancer types. CAR is an extra receptor on the T-cell that helps to increase and accelerate tumor destru

Engineered poly(A)-surrogates for translational regulation and therapeutic biocomputation in mammalian cells.

Here, we present a gene regulation strategy enabling programmable control over eukaryotic translational initiation. By excising the natural poly-adenylation (poly-A) signal of target genes and replacing it with a synthetic control region harboring RNA-binding protein (RBP)-specific aptamers, cap-dependent translation is rendered exclusively dependent on synthetic translation initiation factors (STIFs) containing different RBPs engineered to conditionally associate with different eIF4F-binding proteins (eIFBPs). This modular design framework facilitates the engineering of various gene switches
