Design and characterization of chitosan-alginate microspheres for ocular delivery of azelastine.

The use of mucoadhesive biopolymers is one of the best approaches to prolong the drug residence inside the cul-de-sac, consequently increasing the bioavailability. Thus, the focus of this work was to develop mucoadhesive microspheres to overcome the limitations of ocular drug delivery. The chitosan-sodium alginate microspheres of azelastine hydrochloride were fabricated using modified ionotropic gelation technique. The particle size, zeta potential, entrapment efficiency and drug release kinetics were evaluated and characterized by SEM, FT-IR, DSC, in vitro mucoadhesion and in vivo study. The

Synthesis and characterization of dithiocarbamate chitosan derivatives with enhanced antifungal activity.

In this study, ammonium dithiocarbamate chitosan (ADTCCS) and triethylene diamine dithiocarbamate chitosan (TEDADTCCS) derivatives were obtained respectively by mixing chitosan with carbon disulfide and ammonia (triethylenediamine). Their structures were confirmed by FT-IR, 1H NMR, XRD, DSC, SEM, and elemental analysis. Antifungal properties of them against the plant pathogenic fungi Fusarium oxysporum and Alternaria porri were investigated at concentrations ranged from 31.25 to 500 mg/L. The dithiocarbamate chitosan derivatives had enhanced antifungal activity compared with chitosan. Particul

Chitosan-solid lipid nanoparticles as carriers for topical delivery of tretinoin.

Tretinoin (TRE) or all-trans retinoic acid is employed in the topical treatment of various skin diseases including acne and psoriasis. However, its use is strongly limited by side effects and high chemical instability. TRE encapsulation in nanostructured systems reduces these problems. Chitosan is a biopolymer that exhibits a number of interesting properties such as bioadhesion and antibacterial activity. The aim of this work was to prepare and characterize solid lipid nanoparticles (SLN) containing TRE, with and without addition of chitosan, to assess their in vitro cytotoxicity in keratinocy

[Enhancing the immunogenicity of inactivated polio vaccines with chitosan used as an adjuvant].

Addition of chitosan to inactivated trivalent polio vaccine or inactivated preparations of attenuated poliomyelitis viruses (Sabin strains) significantly increases immunogenicity of these inactivated poliomyelitis virus preparations. High neutralizing antibody titers are detected after two immunizations of mice and a single immunization of rats, as well as when the antigen dose was reduced by 4 times. Addition of chitosan as an adjuvant significantly induces cellular immunity.

The specificity of chitosan in promoting branching morphogenesis of progenitor salivary tissue.

Chitosan has been shown to be effective in regulating progenitor salivary tissue morphogenesis, however, the specificity of chitosan effects remains unclear. To assess the regulatory ability of chitosan in salivary gland morphogenesis, progenitor salivary tissue from embryonal submandibular gland (SMG) was cultured in chitosan-containing medium. It was found that soluble chitosan was able to promote SMG branching in a dose-dependent manner. The effect was chitosan-specific and was not reproduced by substrates with similar chemical structures or other polymeric molecules of natural or synthetic

Chitosan acetate bandage as a topical antimicrobial dressing for infected burns.

An engineered chitosan acetate bandage preparation (HemCon) is used as a hemostatic dressing, and its chemical structure suggests that it should also be antimicrobial. We previously showed that when a chitosan acetate bandage was applied to full-thickness excisional wounds in mice that had been infected with pathogenic bioluminescent bacteria (Pseudomonas aeruginosa, Proteus mirabilis, and Staphylococcus aureus), it was able to rapidly kill the bacteria and save the mice from developing fatal infections. Wound healing was also stimulated. In the present study, we asked whether a chitosan aceta

In vitro biodegradation and biocompatibility of gelatin/montmorillonite-chitosan intercalated nanocomposite.

The intercalated nanocomposite of gelatin/montmorillonite-chitosan (Gel/MMT-CS) was prepared via the solution intercalation process. In vitro degradation tests showed that the nanocomposite had a lower degradation rate than Gel-CS composite. And the introduced intercalation structure endowed Gel/MMT-CS nanocomposite with a controllable degradation rate when changing the MMT content. Cells attachment, spread and proliferation on the Gel/MMT-CS membranes were investigated by scanning electron microscopy (SEM) and mitochondrial activity assay. The results provided evidences of good adhesion, prol

Surface modification of gold and quantum dot nanoparticles with chitosan for bioapplications.

Gold (Au) and quantum dot (QD) nanoparticles, which have been extensively used in many fields, were encapsulated with a natural polymer, chitosan, to improve their biocompatibility. Characterization was performed using ultraviolet-visible, dynamic light scattering, atomic force microscopy, and transmission electron microscope analyses. It was found that a Au/chitosan ratio of 1:1 and smaller produced chitosan-encapsulated Au nanoparticles of a sufficiently small size, and this result was then applied in the chitosan encapsulation of QDs. The biocompatibility of both types of nanoparticles was

[Phytotherapeutic aspects of diseases of the circulatory system. 4. Chitin and chitosan].

Chitin and chitosan are nitrogenous polysaccharides. The structure of the chitin molecule is similar to that of cellulose but it is composed of the units of 2-acetylamino-2-deoxy-D-glucopyranose bound (1 --> 4) by a glycosidic bond. In contrast to chitin, chitosan amino groups are not mostly acetylated. These polysaccharides occur particularly in the fungal kingdom, in invertebrate animals, in some brown algae, and very negligibly in higher plants. Great attention has been paid to chitin and chitosan in recent years. Of great interest are their antihypercholesterolemic and antihyperlipidemic a

Preparation of high-strength, colored and multifunctional chitosan fibers via a novel strategy of enzyme-catalyzed phenolic-amine crosslinking within chitosan fibers in a swollen state.

This study prepared high-strength, colored and multifunctional polyphenol-chitosan fibers via a novel strategy of laccase-mediated phenolic-amine crosslinking within chitosan fibers in a swollen state. A dense covalent network within the fiber was formed through Michael addition and Schiff base reactions. Compared to unmodified chitosan fibers, the enzymatically cross-linked fibers exhibited significantly enhanced mechanical properties, with an increase of about 270% in dry tensile strength (up to 156.8 MPa) and excellent wet stability. In addition, the polyphenol-chitosan fibers were given co

Linagliptin loaded chitosan nanoparticles for the efficient therapy of renal fibrosis.

Chronic kidney disease (CKD), whose prevalence is rising substantially, has become a major global health challenge. In this study, we developed linagliptin-loaded chitosan nanoparticles (HCS-LGP NPs) for the treatment of renal fibrosis. The synthesized nanoparticles exhibited a uniform particle size of 178.8 ± 4.4 nm and a zeta potential of -29.9 ± 2.5 mV, along with acid-responsive drug release properties. In vitro, the HCS-LGP NPs showed efficient cellular uptake in TGF-β1-induced HK-2 cells and significantly inhibited cell proliferation by downregulating the expression of TGF-β1 and Collage

A review on the utilization of insect chitosan in the food industry: Challenges and opportunities.

Chitosan, a biopolymer derived from chitin, is primarily extracted from crustaceans' and insects' exoskeletons. Unlike chitin, which is insoluble, chitosan is produced by deacetylating chitin, enhancing its solubility and functionality. While crustacean chitosan has been widely utilized due to its availability from seafood waste, it poses challenges such as seasonal limitations, environmental concerns, and heavy metal contamination. Insects are a sustainable alternative, as they can be mass-reared year-round with minimal ecological impact, providing a renewable source of high-quality chitosan

A chondroitin sulfate/chitosan composite coating for anticoagulation on cardiovascular implants.

Developing durable anticoagulant coatings for cardiovascular implants remains a major research challenge. Heparin, the current gold-standard anticoagulant, faces challenges due to rapid elution from coated surfaces. This article reports a new heparin-alternative anticoagulant coating system engineered through rational design of a dual-functional chondroitin sulfate derivative (methacrylated/aldehyde-modified chondroitin sulfate, CSMAO) and sulfonated chitosan (SCS). The hierarchical architecture was constructed via sequential layer-by-layer (LbL) assembly on polydopamine (PDA)-functionalized 3

Application of chitosan as nano carrier in the treatment of inflammatory bowel disease.

Inflammatory bowel disease (IBD), encompassing ulcerative colitis (UC) and Crohn's disease (CD), is characterized by persistent and recurrent gastrointestinal inflammation. Conventional IBD therapies often involve the use of antibiotics, NSAIDs, biological agents, and immunomodulators. While these medications can mitigate acute inflammatory symptoms, their long-term efficacy is frequently compromised due to cumulative toxic effects. In recent years, significant attention has shifted toward nanoparticle (NP)-based therapies as potential alternatives for IBD management. Various drug delivery str
