Hole-in-the-head disease in discus fish, Symphysodon (Heckel, 1840): Is it a consequence of a dietary Ca/P imbalance?

Hole-in-the-head (HITH) disease-affected fish develop characteristic lesions in the skin above sensory pores of the head and the trunk. This study investigated whether an unfavourable Ca/P ratio in the diet could provoke lesions consistent with HITH disease in discus fish Symphysodon (Heckel, 1840) as a comparable condition to secondary hyperparathyroidism of tetrapod species. Two groups of five fish were fed a plain beef heart diet (Ca/P of 0.03), whereas two other groups were kept on commercial discus feed (Ca/P of 2.73). Each feeding group was submitted to two different water hardness regim

Trancriptomic profiling revealed the signatures of acute immune response in tilapia (Oreochromis niloticus) following Streptococcus iniae challenge.

Streptococcus iniae is the most significant bacterial disease of tilapia throughout the world, and commonly leads to tremendous economic losses. In contrast to other important fish species, our knowledge about the molecular mechanisms of tilapia in response to bacterial infection is still limited. Here, therefore, we utilized RNA-seq to first profiling of host responses in tilapia spleen following S. iniae infection at transcriptome level. A total of 223 million reads were obtained and assembled into 192,884 contigs with average length 844 bp. Gene expression analysis between control and infec

First integrative study of the diversity and specificity of metacercariae of Posthodiplostomum Dubois, 1936 from native and introduced fishes in the Caribbean.

Metacercariae of the genus Posthodiplostomum are often recorded in freshwater fish hosts. While the diversity and taxonomy of this genus are receiving increasing attention in molecular phylogenetic studies, available data remain geographically biased. Most molecular studies of Posthodiplostomum and morphologically similar (neascus) worms originate in North America and Europe and Asia (more than 60% of DNA sequences are from USA and Canada), with few data currently available from the Neotropics, where high host diversity suggests high and under-sampled parasite diversity. In this study, we repo

Association between apoptotic neural tissue and cell proliferation in the adult teleost brain.

Injury to neuronal tissues in the central nervous system (CNS) of mammals results in neural degeneration and sometime leads to loss of function, whereas fish retain a remarkable potential for neuro-regeneration throughout life. Thus, understanding the mechanism of neuro-regeneration in fish CNS would be useful to improve the poor neuro-regenerative capability in mammals. In the present study, we characterized a neuro-regenerative process in the brain of a cichlid, tilapia, Oreochromis niloticus. Morphological observations showed that the damaged brain region (habenula) successfully regrew and

PD-L1 restrains T-cell immunity in Nile tilapia by suppressing activation-induced fatty acid synthesis.

The programmed death ligand 1/programmed cell death 1 (PD-L1/PD-1) serves as a critical immune checkpoint in T cell-mediated immune responses, playing a central role in maintaining peripheral immune tolerance and homeostasis. Although early vertebrates lack a PD-1 homolog and they retain PD-L1, the functional significance and regulatory mechanisms of PD-L1 in T-cell immunity in these species remain poorly understood. In this study, using Nile tilapia (Oreochromis niloticus) as a teleost model, we systematically investigated the immunosuppressive role of PD-L1 in T-cell immunity. PD-L1 was wide

Isolation, Identification, Virulence and Pathogenic Features of Lactococcus garvieae from Cage-Cultured Tilapia (Oreochromis niloticus) in Thailand.

Lactococcosis caused by Lactococcus garvieae is an emerging threat to warmwater aquaculture, yet evidence integrating field outbreaks with robust molecular confirmation and controlled virulence testing remains limited for Thailand's cage-cultured tilapia. From May to October 2025, acute mortality events were investigated in cage-cultured Nile tilapia (Oreochromis niloticus) in a reservoir in Ubon Ratchathani Province, Thailand. Suspected outbreaks were defined by abrupt daily mortality exceeding 5% accompanied by septicemia-like clinical signs. Water quality during sampling covered the followi

Silymarin conserves the efficacy of quinolone and sulfonamide in Nile tilapia (Oreochromis niloticus) subjected to aflatoxicosis and Streptococcus agalactiae infection.

Aflatoxins (AFB) are adventitious plant toxins that act as immunosuppressants. Experimental Nile tilapia were divided into eight groups: AFB0 (control, uncontaminated diet), AFB50 (50 ppbAFB1), AFB100 (100 ppbAFB1), AFB150 (150 ppbAFB1), AFB0 + Sil (0 ppbAFB1+Sil), AFB50 + Sil (50 ppbAFB1+Sil), AFB100 + Sil (100 ppbAFB1+Sil), and AFB150 + Sil (150 ppbAFB1+Sil). The AFB1-residues in fish liver (AFB0, AFB50, AFB100, and AFB150) were 0.34, 0.54, and 0.82 ppb, respectively, while traces were recorded in muscles. Sil-supplemented fish had lower residues than other contaminated groups, which were st

Molecular cloning and functional characterization of NQO1 in Nile tilapia (Oreochromis niloticus): a quinone oxidoreductase mitigating ammonia-induced oxidative stress.

NAD(P)H dehydrogenase [quinone] 1 (NQO1) is a mammalian quinone oxidoreductase, which can react with cell antioxidants and nucleophiles and reduce cell damage. Although NQO1 has been widely studied in mammals, its research in aquatic animals is relatively limited, especially in Nile tilapia (Oreochromis niloticus). In this study, we successfully cloned NQO1 (OnNQO1) of O. niloticus and studied its role in oxidative stress induced by ammonia exposure. The results showed that the open reading frame (ORF) of OnNQO1 was 882 bp, encoding 293 amino acids, and its protein had NAD(P)H dehydrogenase [q

Dietary exposure to hexafluoropropylene oxide dimer acid (HFPO-DA ammonium salt, GenX) alters hepatic sphingomyelin and phosphatidylcholine metabolism in Nile Tilapia (Oreochromis niloticus).

HFPO-DA ammonium salt (GenX) is used to replace the environmentally persistent perfluorooctanoic acid (PFOA), but its effects on fish lipid metabolism remain poorly understood. This study investigated the impact of dietary GenX exposure (0.73 and 3.10 mg/Kg of feed, measured concentrations) on hepatic lipid metabolism and oxidative stress in juvenile male Nile tilapia (5.32 ± 1.60 g) over 28 days. While fish increased body weight over time, there were no differences in weight gain among treatments. There were no changes in hepatic glycerol levels with either dose of feed, while triglycerides d

Azolla pinnata mitigates pendimethalin induced immunotoxicity, oxidative stress and histopathological changes in Oreochromis niloticus.

Aquatic animals face multiple threats, including pesticides, heavy metals, and other environmental pollutants, risking their health and survival. Limited bioremediation studies have been conducted on the detrimental impacts of herbicides on fish. This study focused on the impact of the herbicide pendimethalin (PD) on Oreochromis niloticus and assessed the protective role of Azolla pinnata (AZ), an aquatic fern known for its phytoremediation and antioxidant properties. O. niloticus (n = 270, 34.17 ± 2.41 g) were divided into six groups in triplicate: the control (CTR), AZ-supplemented (125 g/kg

The mitigating effect of dietary β-glucan against fipronil-induced intoxication in Nile Tilapia (Oreochromis niloticus): Histopathological, immunological, hematological, and biochemical analysis.

BACKGROUND: Due to its remarkable effectiveness against a wide range of pests and insects at very low concentrations, the broad-spectrum fipronil pesticide is currently gaining popularity in the agricultural, public health, and international industries. However, the stressor effects of fipronil insecticides cause ecological disruption, growth retardation, immunosuppression, and higher fish mortality rates. Both animals and humans have demonstrated the effectiveness of prebiotics such as β-1,3-glucan in their diets. Aquaculture has recently increased in use because of its potential to control d

Effects of Fat and Carnitine on the Expression of Carnitine Acetyltransferase and Enoyl-CoA Hydratase Short-Chain 1 in the Liver of Juvenile GIFT (Oreochromis niloticus).

Carnitine acetyltransferase (CAT) and Enoyl-CoA hydratase short-chain 1 (ECHS1) are considered key enzymes that regulate the β-oxidation of fatty acids. However, very few studies have investigated their full length and expression in genetically improved farmed tilapia (GIFT, Oreochromis niloticus), an important aquaculture species in China. Here, we cloned CAT and ECHS1 full-length cDNA via the rapid amplification of cDNA ends, and the expressions of CAT and ECHS1 in the liver of juvenile GIFT were detected in different fat and carnitine diets, as were the changes in the lipometabolic enzymes

Enhancing growth, antioxidant capacity, and immune response in tilapia (Oreochromis niloticus) through curcumin supplementation across varied stocking density paradigms.

The present study evaluated the effects of curcumin on growth, immune and antioxidant response in tilapia (Oreochromis niloticus). An optimum dose of curcumin was investigated by feeding four different levels of this compound in combination with three different regimes of stocking density (12 treatments). Fish were reared at three densities; low density (LD = 1.50 kg/m3), medium density (MD = 3.00 kg/m3), and high density (HD = 4.50 kg/m3). Each treatment was fed with four different levels of dietary supplementation of curcumin (C0 = 0 mg/kg, C1 = 50 mg/kg, C2 = 100 mg/kg, and C3 = 150 mg/kg)

Chitosan-polymer based nanovaccine as promising immersion vaccine against Aeromonas veronii challenge in red tilapia (Oreochromis sp.).

Red tilapia (Oreochromis sp.), one of the important freshwater fish species in fish farming in Thailand, has for long been suffering from a serious bacterial disease named epizootic ulcerative syndrome and hemorrhagic septicemia. The disease is mainly caused by Aeromonas veronii. Vaccine is proposed to be a major impact tool for sustainable control and prevention strategies. Vaccination by immersion has many benefits over injection. However, the conventional immersion method suffers from a low potency due to the inefficient uptake of antigens across mucosal tissue. Here, we developed a chitosa
