Expression of Unconventional Myosin VI in Oligodendrocytes.

Myelin is a specialized multilamellar structure involved in various functions of the nervous system. Oligodendrocytes are responsible for myelin formation in the central nervous system. Motor proteins play important roles in differentiation and myelin formation of the oligodendrocyte lineage. Recently, we revealed that one of the unconventional myosins, myosin ID (Myo1d), is expressed in mature oligodendrocytes and is required for myelin-like membrane formation in vitro. Previously, Cahoy et al. (J Neurosci 28:264-278, 2008) reported that another unconventional myosin VI (Myo6) is upregulated

Statin therapy inhibits remyelination in the central nervous system.

Remyelination of lesions in the central nervous system contributes to neural repair following clinical relapses in multiple sclerosis. Remyelination is initiated by recruitment and differentiation of oligodendrocyte progenitor cells (OPCs) into myelinating oligodendrocytes. Simvastatin, a blood-brain barrier-permeable statin in multiple sclerosis clinical trials, has been shown to impact the in vitro processes that have been implicated in remyelination. Animals were fed a cuprizone-supplemented diet for 6 weeks to induce localized demyelination in the corpus callosum; subsequent return to norm

Dominant-negative beta1 integrin mice have region-specific myelin defects accompanied by alterations in MAPK activity.

Recent studies have demonstrated the importance of beta1 integrin in oligodendrocyte maturation in vitro. Similar studies in vivo have been difficult due to the embryonic and perinatal lethality of null mutations in integrin subunits. Here, we have generated transgenic mouse models that overexpress full length beta1 integrin or express a dominant-negative beta1 integrin DeltaC (lacking the C-terminal tail) under the control of the proteolipid protein (PLP) promoter. We demonstrate that these transgenes are expressed predominantly in CNS tissues and more specifically in oligodendrocytes. Furthe

Brain region dependent molecular signatures and myelin repair following chronic demyelination.

Multiple sclerosis (MS) is the most prevalent demyelinating disease of the central nervous system, characterized by myelin destruction, axonal degeneration, and progressive loss of neurological functions. Remyelination is considered an axonal protection strategy and may enable functional recovery, but the mechanisms of myelin repair, especially after chronic demyelination, remain poorly understood. Here, we used the cuprizone demyelination mouse model to investigate spatiotemporal characteristics of acute and chronic de- and remyelination and motor functional recovery following chronic demyeli

Transmembrane protein 119 is neither a specific nor a reliable marker for microglia.

Microglia are the resident innate immune cells of the central nervous system (CNS) parenchyma. To determine the impact of microglia on disease development and progression in neurodegenerative and neuroinflammatory diseases, it is essential to distinguish microglia from peripheral macrophages/monocytes, which are eventually equally recruited. It has been suggested that transmembrane protein 119 (TMEM119) serves as a reliable microglia marker that discriminates resident microglia from blood-derived macrophages in the human and murine brain. Here, we investigated the validity of TMEM119 as a micr

Translational value of choroid plexus imaging for tracking neuroinflammation in mice and humans.

Neuroinflammation is a pathophysiological hallmark of multiple sclerosis and has a close mechanistic link to neurodegeneration. Although this link is potentially targetable, robust translatable models to reliably quantify and track neuroinflammation in both mice and humans are lacking. The choroid plexus (ChP) plays a pivotal role in regulating the trafficking of immune cells from the brain parenchyma into the cerebrospinal fluid (CSF) and has recently attracted attention as a key structure in the initiation of inflammatory brain responses. In a translational framework, we here address the int

Identification of novel myelin repair drugs by modulation of oligodendroglial differentiation competence.

BACKGROUND: In multiple sclerosis loss of myelin and oligodendrocytes impairs saltatory signal transduction and leads to neuronal loss and functional deficits. Limited capacity of oligodendroglial precursor cells to differentiate into mature cells is the main reason for inefficient myelin repair in the central nervous system. Drug repurposing constitutes a powerful approach for identification of pharmacological compounds promoting this process. METHODS: A phenotypic compound screening using the subcellular distribution of a potent inhibitor of oligodendroglial cell differentiation, namely p57

Diosgenin promotes oligodendrocyte progenitor cell differentiation through estrogen receptor-mediated ERK1/2 activation to accelerate remyelination.

Differentiation of oligodendrocyte progenitor cells (OPCs) into mature oligodendrocytes is a prerequisite for remyelination after demyelination, and impairment of this process is suggested to be a major reason for remyelination failure. Diosgenin, a plant-derived steroid, has been implicated for therapeutic use in many diseases, but little is known about its effect on the central nervous system. In this study, using a purified rat OPC culture model, we show that diosgenin significantly and specifically promotes OPC differentiation without affecting the viability, proliferation, or migration of

Leukotriene inhibitors in multiple sclerosis: a systematic review of animal studies.

Current disease-modifying therapies for multiple sclerosis (MS) primarily target adaptive immunity but often fail to halt neurodegeneration or promote myelin repair. Leukotriene pathway inhibitors, established in the management of asthma, possess anti-inflammatory and potentially neuroprotective properties. Following the PRISMA 2020 recommendations, a systematic search was conducted in PubMed, Web of Science, and Scopus. Eligible studies included preclinical investigations of leukotriene inhibitors in MS animal models and risk of bias was assessed using SYRCLE's risk of bias tool for animal st

Mechanistic Insights and Therapeutic Potential of MCC950 as a NLRP3 Inflammasome Inhibitor in Multiple Sclerosis: A Narrative Review.

INTRODUCTION: The NLR family pyrin domain-containing 3 (NLRP3) inflammasome is a central driver of neuroinflammation and demyelination in multiple sclerosis (MS). MCC950, a selective small-molecule NLRP3 inhibitor, has emerged as a promising therapeutic candidate for MS by suppressing inflammasome formation and downstream cytokine release. METHODS: A comprehensive literature search was conducted in PubMed, Scopus, and Web of Science using the keywords "NLRP3 inflammasome," "MCC950," "multiple sclerosis," "experimental autoimmune encephalomyelitis," and "neuroinflammation." Unlocking the healing power of Berberine: A promising aid for multiple sclerosis.

Multiple sclerosis (MS) is a debilitating autoimmune disorder characterized by inflammatory demyelination and progressive neurodegeneration within the central nervous system (CNS). Despite advances in disease-modifying therapies (DMTs), current treatments primarily mitigate relapses and slow disease progression but fall short in comprehensively addressing cumulative disability or neurodegeneration. Berberine (BBR), a naturally occurring isoquinoline alkaloid, has emerged as a promising therapeutic candidate due to its potent immunomodulatory, anti-inflammatory, and neuroprotective properties.

Pre-activation status impacts regenerative potential of oligodendrocyte progenitors in an LPS-exposed animal model.

INTRODUCTION & AIMS: Optimal neural activity in the central nervous system relies on the myelin sheath formed by oligodendrocytes. During demyelination, changes in the microenvironment can activate oligodendrocyte precursor cells (OPCs) and induce them to generate new oligodendrocytes. However, different demyelination models employ distinct pathways, targeting different cells and cytokines; these, in turn, have varying effects on OPCs. Therefore, it is reasonable to assume that OPCs derived from different pathologic environment may exhibit functional differences. This study aims to investigate

Gasdermin D activation in oligodendrocytes and microglia drives inflammatory demyelination in progressive multiple sclerosis.

Neuroinflammation coupled with demyelination and neuro-axonal damage in the central nervous system (CNS) contribute to disease advancement in progressive multiple sclerosis (P-MS). Inflammasome activation accompanied by proteolytic cleavage of gasdermin D (GSDMD) results in cellular hyperactivation and lytic death. Using multiple experimental platforms, we investigated the actions of GSDMD within the CNS and its contributions to P-MS. Brain tissues from persons with P-MS showed significantly increased expression of GSDMD, NINJ1, IL-1β, and -18 within chronic active demyelinating lesions compar

Aloe-emodin promotes remyelination by driving microglial myelin debris clearance via the CD36-PPARγ axis.

The failure of remyelination in demyelinating diseases, such as multiple sclerosis (MS), is profoundly exacerbated by the accumulation of lipid-rich myelin debris, which sustains an inhibitory microenvironment. Efficient clearance of this debris by microglia is a critical step for myelin repair. Here, we investigated the therapeutic potential and underlying mechanisms of Aloe-emodin (AE), a natural anthraquinone, in promoting remyelination. Using a cuprizone (CPZ)-induced demyelination mouse model and primary microglial cultures, we systematically assessed the effects of AE on myelin debris cl

Low, but not high, dose triptolide controls neuroinflammation and improves behavioral deficits in toxic model of multiple sclerosis by dampening of NF-κB activation and acceleration of intrinsic myelin repair.

Cuprizone (Cup) is a copper chelating agent frequently used to study factors that affect oligodendrocytes (OLGs) death and acute demyelination. Triptolide (TP), a nuclear factor-kappaB (NF-κB) blocker, is a major bioactive component of Tripterygium wilfordii Hook f. (TWHf) with various therapeutic activities. In this study, we examined the effects of TP on neuroglia activation, inflammation, apoptosis, demyelination, and behavioral deficits in the Cup-induced toxic model of multiple sclerosis (MS). C57BL/6 J mice were fed with chow containing 0.2% Cup for 6 weeks to induce detectable neuroinfl

Astrocyte-specific FoxF2 modulates immune and myelin repair: Evidence from multiple sclerosis lesions and an animal model.

As multiple sclerosis (MS) progresses, myelin repair becomes inefficient. To gain insight into the underlying causes, we RNA-sequenced postmortem brain tissues from 4 and 6 patients, comparing remyelinating versus chronically active MS lesions. We identified the transcription factor FoxF2 as highly expressed within remyelinating lesions. Immunohistochemistry and in situ hybridization showed FoxF2 co-expression in GFAP-positive astrocytes, suggesting a subpopulation of astrocytes with a potential role in repair. To investigate how FoxF2 may influence repair, we examined cuprizone (CPZ)-induced

Repurposing the cardiac glycoside digoxin to stimulate myelin regeneration in chemically-induced and immune-mediated mouse models of multiple sclerosis.

Multiple sclerosis (MS) is a central nervous system (CNS) autoimmune disease characterized by inflammation, demyelination, and neurodegeneration. The ideal MS therapy would both specifically inhibit the underlying autoimmune response and promote repair/regeneration of myelin as well as maintenance of axonal integrity. Currently approved MS therapies consist of non-specific immunosuppressive molecules/antibodies which block activation or CNS homing of autoreactive T cells, but there are no approved therapies for stimulation of remyelination nor maintenance of axonal integrity. Developmental and neurobehavioral toxicity of di-(2-ethylhexyl) phthalate (DEHP) in zebrafish larvae.

Di-ethyl hexyl phthalate (DEHP), a plasticizer used in medical devices and cosmetics, easily leaches into the environment due to its weak chemical bonds with plastics. Humans and other organisms are exposed to DEHP through inhalation, ingestion, and dermal contact, inflicting toxic effects like neurotoxicity, hepatotoxicity, and reproductive toxicity. However, there is a lack of information on its developmental toxicity, and its role in demyelination is also not well understood. We explored the adverse effects of DEHP during the early developmental stage of zebrafish larvae, focusing on its de

Visualization of the Breakdown of the Axonal Transport Machinery: a Comparative Ultrastructural and Immunohistochemical Approach.

Axonal damage is a major factor contributing to disease progression in multiple sclerosis (MS) patients. On the histological level, acute axonal injury is most frequently analyzed by anti-amyloid precursor protein immunohistochemistry. To what extent this method truly detects axonal injury, and whether other proteins and organelles are as well subjected to axonal transport deficits in demyelinated tissues is not known. The aim of this study was to correlate ultrastructural morphology with the immunohistochemical appearance of acute axonal injury in a model of toxin-induced oligodendrocyte dege

Myelin repair is accelerated by inactivating CXCR2 on nonhematopoietic cells.

Multiple sclerosis (MS) is an inflammatory demyelinating disease of the CNS and remyelination in MS ultimately fails. Although strategies to promote myelin repair are eagerly sought, mechanisms underlying remyelination in vivo have been elusive. CXCR2 is expressed on neutrophils and oligodendrocyte lineage cells in the CNS. CXCR2-positive neutrophils facilitate inflammatory demyelination in demyelination models such as experimental autoimmune encephalomyelitis (EAE) and cuprizone intoxication. Systemic injection of a small molecule CXCR2 antagonist at the onset of EAE decreased demyelinated le
