Host-specific differences in the physiology of acid secretion related to prostaglandins may play a role in gastric inflammation and injury.

Immune mediators are involved in strain-specific manifestations of Helicobacter pylori infection, and the type of immune response is associated with production of PGE(2), which in turn influences gastric acid secretion. Acid secretion plays a pivotal role, not only in the pattern of H. pylori-induced gastritis and its consequences, but also in nonsteroidal anti-inflammatory drug (NSAID)-induced gastropathies. Mice and their transgenic modifications are widely used in Helicobacter and eicosanoid research. Using [(14)C]aminopyrine accumulation and pylorus ligation, we aimed to study acid secreti

Non-specific host response markers in the differentiation between pneumococcal and viral pneumonia: what is the most accurate combination?

BACKGROUND: Serum C-reactive protein (CRP), blood white cell count (WBC), serum procalcitonin (PCT) and erythrocyte sedimentation rate (ESR) were measured in 132 children hospitalized for community-acquired pneumonia. Serological evidence for viral infection was found in 38 cases and for pneumococcal infection in 41 cases, and the infiltrate was alveolar in 46 cases and interstitial in 86 cases. The aim of the present paper was to determine if there is a combination of these four host response markers and chest radiograph findings suitable for differentiating pneumococcal from viral etiology o

"Autoimmune rejection" of neonatal heart transplants in experimental Chagas disease is a parasite-specific response to infected host tissue.

Infection with the protozoan parasite Trypanosoma cruzi often results in chronic heart- and gut-associated disease known as Chagas disease. In this study we show that contrary to previous reports, neonatal hearts transplanted into mice chronically infected with T. cruzi do not exhibit signs of autoimmune-type rejection or any significant inflammatory response. In addition to an absence of inflammation, these syngeneic heart transplants survive for more than 1 year and are absolutely free of parasites as determined by in situ PCR analysis. However, if well-established transplanted hearts in chr

Organ-specific variation in the morphology of the fungomas (fungus balls) of Pseudallescheria boydii. Development within necrotic host tissue.

Pseudallescheria boydii is an increasingly important cause of infection in debilitated and immunocompromised persons. It is frequently found within pulmonary cavities, where it forms matted collections of fungal hyphae, variously termed fungus balls, fungomas, or pseudallescheriomas. This report describes the unusual occurrence of pseudallescheriomas in the lungs, brain, and kidney. The morphological features of the fungomas varied between involved organs, with those in the lung showing well-defined layers of peripheral mycelial hypocellularity and hypercellularity consisting of anneloconidiop

Calicotyle hydrolagi n. sp. (Monogenea: Monocotylidae) infecting the deep-sea Eastern Pacific black ghost shark Hydrolagus melanophasma from the Atacama Trench, with comments on host specificity of Calicotyle spp.

We describe Calicotyle hydrolagi n. sp. (Monogenea: Monocotylidae) infecting the cloaca of deep-water Eastern Pacific black ghost sharks, Hydrolagus melanophasma captured as bycatch at a local fishery for Patagonian toothfish Dissostichus eleginoides, (Nototheniidae) in the Atacama Trench using morphological and nucleotide (LSU rRNA and SSU rRNA) data. This new species is differentiated from its congeners by a number of characters, including the absence of a cecal diverticula, the size and shape of the male copulatory organ and the shape of the vagina, as well as by differences in molecular da

Multi-faceted proteomic characterization of host protein complement of Rift Valley fever virus virions and identification of specific heat shock proteins, including HSP90, as important viral host factors.

Rift Valley fever is a potentially fatal disease of humans and domestic animals caused by Rift Valley fever virus (RVFV). Infection with RVFV in ruminants can cause near 100% abortion rates and recent outbreaks in naïve human populations have suggested case fatality rates of greater than thirty percent. To elucidate the roles that host proteins play during RVFV infection, proteomic analysis of RVFV virions was conducted using complementary analytical approaches, followed by functional validation studies of select identified host factors. Coupling the more traditional Gel LC/MS/MS approach (SDS

A high-resolution spatial model of the primate gut microbiome defines niche-specific functions and host-adapted microbial metabolism.

The spatial organization of the gastrointestinal microbiome is shaped by strong physicochemical gradients along the gut, yet many region-associated microbial niches remain poorly resolved in humans. Here, we used the rhesus macaque (Macaca mulatta) as a spatially resolved primate model to profile 148 luminal and mucosal samples across the gastrointestinal tract using shotgun metagenomics, 2bRAD-M sequencing, and targeted bile acid metabolomics, complemented by comparative human datasets. Microbial communities exhibited strong spatial stratification along the gastrointestinal tract, while host

A host anti-amyloidogenic stomach-specific protein inhibits colonization and biofilm formation by adherent invasive Escherichia coli in the colon.

Gastrokine-1 (Gkn1) is an anti-amyloidogenic host protein secreted into the gut lumen by the stomach. Gut bacteria make functional amyloids to facilitate biofilm formation and biofilms in the gastrointestinal tract are associated with a variety of disorders, including inflammatory bowel disease. Adherent invasive Escherichia coli (AIEC) is a pathobiont that produces amyloids, forms biofilms, and is associated with inflammatory bowel disease. We therefore investigated whether Gkn1 is required to clear AIEC from the gastrointestinal tract by comparing the course of infection in wild-type and Gkn

Host species-specific mutations in the thumb domain of the 3Dpol polymerase are required for efficient replication of human hepatitis A virus in mice.

Hepatitis A virus (HAV) is a globally important cause of enterically-transmitted hepatitis. It is one of 9 distinct Hepatovirus species in the Picornaviridae, among which phylogenetic reconstructions suggest multiple past host species jumps. HAV readily infects mice with defective type I interferon responses, suggesting the major barrier preventing human HAV from replicating in a rodent host is an inability to overcome innate immune responses. In prior studies, only a single nonsynonymous mutation of uncertain significance (3Dpol-R468K) was identified within the genome of wild-type HAV followi

Comparative Metabolomics Reveals Family-Genus-Specific Chemical Signatures and Potential Recognition Mechanisms in Cynomorium songaricum-Host Interactions.

Cynomorium songaricum is an important medicinal holoparasitic plant in the arid regions of northwestern China. Its extremely low seed germination rate relies on chemical signals released from the rhizosphere of specific host plants. This study aimed to elucidate the chemical basis of host recognition by C. songaricum by characterizing and comparing the rhizosphere volatile metabolomes of five host plant species. Gas chromatography-mass spectrometry (GC-MS) was used to analyze the rhizosphere volatiles of three Nitraria species (N. roborowskii, N. sibirica, N. tangutorum), Peganum multisectum,

Differences in codon usage between host-species-specific rabies virus clades are driven by non-GC3 nucleotide composition.

Viral genes sometimes use certain codons more than others due to their nucleotide content, translational efficiency, and selection pressure from the host immune system. The rabies virus (RABV) is a negative strand RNA virus which can infect a broad range of mammalian hosts, with many of its clades circulating predominantly in specific host species. Previous work on codon usage in RABV has focused only on broader viral clades. We use publicly available RABV nucleoprotein gene sequences to investigate how dinucleotide content and codon usage biases differ between host-associated clades, and what

Host protein interactome profiling of Akabane virus glycoprotein Gc reveals specific mitochondrial genes regulate viral replication.

Akabane virus (AKAV) is an arthropod-borne orthobunyavirus that poses a significant threat to ruminant health worldwide by causing high fever, stillbirth, and congenital malformations. Although its viral glycoprotein Gc is critical for multiple stages of the viral life cycle, the spectrum of its interactions with host proteins remains uncharacterized. In this study, host proteins interacting with Gc were systematically profiled using co-immunoprecipitation (Co-IP) combined with liquid chromatography-tandem mass spectrometry (LC-MS/MS). A total of 441 host proteins were identified as candidate

Comparing Campylobacter jejuni to three other enteric pathogens in OligoMM12 mice reveals pathogen-specific host and microbiota responses.

Campylobacter jejuni, non-typhoidal Salmonella spp., Listeria monocytogenes and enteropathogenic/enterohemorrhagic Escherichia coli (EPEC/EHEC) are leading causes of food-borne illness worldwide. Citrobacter rodentium has been used to model EPEC and EHEC infection in mice. The gut microbiome is well-known to affect gut colonization and host responses to many food-borne pathogens. Recent progress has established gnotobiotic mice as valuable models to study how microbiota affect the enteric infections by S. Typhimurium, C. rodentium and L. monocytogenes. However, for C. jejuni, we are still lack

The Inherited Intestinal Microbiota from Myeloid-Specific ZIP8KO Mice Impairs Pulmonary Host Defense against Pneumococcal Pneumonia.

Intestinal dysbiosis increases susceptibility to infection through the alteration of metabolic profiles, which increases morbidity. Zinc (Zn) homeostasis in mammals is tightly regulated by 24 Zn transporters. ZIP8 is unique in that it is required by myeloid cells to maintain proper host defense against bacterial pneumonia. In addition, a frequently occurring ZIP8 defective variant (SLC39A8 rs13107325) is strongly associated with inflammation-based disorders and bacterial infection. In this study, we developed a novel model to study the effects of ZIP8-mediated intestinal dysbiosis on pulmonary

MLVA genotyping of Moritella viscosa reveals serial emergence of novel, host-specific clonal complexes in Norwegian salmon farming.

A Multi-Locus Variable number of tandem repeat Analysis (MLVA) genotyping scheme was developed for the epidemiological study of Moritella viscosa, which causes 'winter ulcer' predominantly in sea-reared Atlantic salmon (Salmo salar L.). The assay involves multiplex PCR amplification of six Variable Number of Tandem Repeat (VNTR) loci, followed by capillary electrophoresis and data interpretation. A collection of 747 spatiotemporally diverse M. viscosa isolates from nine fish species was analysed, the majority from farmed Norwegian salmon. MLVA distributed 76% of the isolates across three major

Fetal growth restriction is a host specific response to infection with an impaired spiral artery remodeling-inducing strain of Porphyromonas gingivalis.

Porphyromonas gingivalis is a periodontal pathogen implicated in a range of pregnancy disorders that involve impaired spiral artery remodeling (ISAR) with or without fetal growth restriction (FGR). Using a rodent periodontitis model, we assessed the ability of P. gingivalis to produce ISAR and FGR in Sprague Dawley (SD) and Wistar (WIS) rats. Both infected SD and WIS rats developed ISAR, but only WIS rats developed FGR despite both rat strains having equivalent microbial loads within the placenta. Neither maternal systemic inflammation nor placental (fetal) inflammation was a feature of FGR in

iTRAQ-based phosphoproteomic analysis reveals host cell's specific responses to Toxoplasma gondii at the phases of invasion and prior to egress.

Protein phosphorylation plays a key role in host cell-T. gondii interaction. However, the phosphoproteome data of host cell at various phases of T. gondii infection has not been thoroughly described. In this study, we assessed the host phosphoproteome data with isobaric tags for relative and absolute quantification (iTRAQ) method during the phases of T. gondii invasion (30 min post infection, PI) and prior to egress (28 h PI). Our iTRAQ analysis revealed a total of 665 phosphoproteins, among which the significantly regulated phosphoproteins in different between-group comparisons were further a
