Design, synthesis, and characterization of a dual modality positron emission tomography and fluorescence imaging agent for monoclonal antibody tumor-targeted imaging.

A novel lysine-based trifunctional chelate 3 was designed, synthesized, and characterized and bears both a chelating moiety (CHX-A' ') for sequestering radiometals (86Y or 111In) and the near-infrared dye Cy5.5 for dual modality PET (or SPECT) and fluorescence imaging, respectively. Successful conjugation of 3 to the monoclonal antibody trastuzumab (Herceptin) was achieved by efficient thiol-maleimide chemistry, thereby yielding immunoconjugate 2. Analysis of 2 by flow cytometry and competitive binding assay demonstrates that immunoconjugate 2 binds to SKOV3 tumor cells comparably to native tr

Nanoparticle immunoimaging reveals metabolic dysfunction fueling CCR2-dependent inflammation in infarcted hearts.

Inflammation critically shapes outcomes after myocardial infarction (MI), particularly in patients with metabolic dysfunction. This study investigated the uptake of fluorescent cross-linked iron oxide nanoparticles (CLIO-AF647) in infarcted myocardium and hematopoietic bone marrow using multimodal imaging, with emphasis on the role of metabolic stress and CCR2 signaling. Male C57BL/6 J mice underwent MI with or without diet-induced obesity (DIO) established by a high-fat, high-glucose diet. CLIO-AF647 was injected intravenously two days post-MI. Myocardial uptake was assessed by magnetic reson

FLARE: A Flow Cytometry-Based Fluorescent Assay for Measuring HSV-1 Nuclear Egress.

During herpesvirus replication, capsids are assembled inside the nucleus and translocated into the cytosol by a non-canonical nucleocytoplasmic export process termed nuclear egress. Traditional methods of measuring nuclear egress rely on imaging-based technologies such as confocal and electron microscopy. These techniques are labor-intensive, limited by the number of cells that can be examined, and may not accurately represent the entire population, generating a potential bias during data interpretation. To overcome these problems, we have developed a flow cytometry-based method to measure HSV

Revisiting tumour aneuploidy - the place of ploidy assessment in the molecular era.

Chromosome instability (CIN) is gaining increasing interest as a central process in cancer. CIN, either past or present, is indicated whenever tumour cells harbour an abnormal quantity of DNA, termed 'aneuploidy'. At present, the most widely used approach to detecting aneuploidy is DNA cytometry - a well-known research assay that involves staining of DNA in the nuclei of cells from a tissue sample, followed by analysis using quantitative flow cytometry or microscopic imaging. Aneuploidy in cancer tissue has been implicated as a predictor of a poor prognosis. In this Review, we have explored th

Toward integrated multimodal artificial intelligence in bone marrow examination: current state and open challenges.

Bone marrow (BM) examinations are intrinsically multimodal: hematopathologists integrate peripheral blood, aspirate cytomorphology, trephine histology, flow cytometry, cytogenetics, and molecular genetics data into a single interpretation. Because WHO-HAEM5 and ICC 2022 define entities through integrated morphological, immunophenotypic, cytogenetic, and molecular criteria, multimodal diagnostic reasoning embedded in a procedural workflow, not isolated image classification, represents the appropriate target for the use of artificial intelligence (AI). We review the existing research on integrat

CD8 immunoPET imaging to stratify response and guide combination immunotherapy and radiation in triple negative breast cancer.

PURPOSE: PET imaging targeting immune cells can be used to dynamically monitor intratumoral immune modulation in tissues. Radiation therapy is known to alter the tumor immune microenvironment; therefore, this study demonstrates how CD8 immunoPET imaging can optimize combination immunotherapy and radiation therapy by stratifying tumors who could derive the greatest benefit from immunotherapy following radiation therapy. EXPERIMENTAL DESIGN: A radiation resistant triple negative breast cancer cell line was derived through repeat irradiation of the radiosensitive parental 4T1 cell line prior to i

Comparative Study of DNA ploidy and BRAF Immunohistochemistry between Colonic Adenocarcinoma and Inflammatory Colonic Lesions.

OBJECTIVES: To evaluate DNA ploidy and S-phase fraction (SPF) in non-Lynch colonic adenocarcinoma, ulcerative colitis (UC), Crohn disease (CD) which are known as risk factors, and colitis. We correlated ploidy and SPF with tumor grading, staging and BRAF expression. METHODS: All studied adenocarcinomas have intact mismatch repair genes as proved by immunohistochemistry. All were assessed for ploidy by automated image-based DNA cytometry and histograms were drawn. Immunostaining by anti-BRAF V600E was performed. Diagnostic laparoscopy (DL) was done as a preliminary step for staging GI cancers.

Highly Multiplexed Image Analysis of Intestinal Tissue Sections in Patients With Inflammatory Bowel Disease.

BACKGROUND & AIMS: Significant progress has been made since the first report of inflammatory bowel disease (IBD) in 1859, after decades of research that have contributed to the understanding of the genetic and environmental factors involved in IBD pathogenesis. Today, a range of treatments is available for directed therapy, mostly targeting the overactive immune response. However, the mechanisms by which the immune system contributes to disease pathogenesis and progression are not fully understood. One challenge hindering IBD research is the heterogeneous nature of the disease and the lack of

Cell-Based No-Wash Fluorescence Assays for Compound Screens Using a Fluorescence Cytometry Plate Reader.

High-throughput cell-based fluorescent imaging assays often require removal of background fluorescent signal to obtain robust measurements. Processing high-density microplates to remove background signal is challenging because of equipment requirements and increasing variation after multiple plate wash steps. Here, we present the development of a wash-free cell-based fluorescence assay method for high-throughput screening using a laser scanning fluorescence plate cytometer. The cytometry data consisted of cell count and fluorescent intensity measurements for phenotypic screening. We obtained r

A novel triphenylamine-based aggregation-induced emission (AIE) probe for breast cancer imaging in vitro and in vivo.

Aggregation-induced emission (AIE) probes have attracted considerable interest in bioanalysis because of their good photostability, high imaging contrast, and longtime tracing capability. Herein, we developed a new AIE-active probe, AIE-BC, by conjugating triphenylamine (TPA) to bithiophene via Suzuki coupling reaction. AIE-BC exhibited AIE character, excellent photostability, and superior biocompatibility. Flow cytometry analyzes revealed that AIE-BC was selectively taken up by MCF-7 cells, a result complemented by clear in vitro cellular images obtained from confocal microscopy. Further in v

Diagnostic utility of cerebrospinal fluid IgG index, 24-hour IgG synthesis rate, and immunophenotyping in primary central nervous system large B-cell lymphoma: A case report.

RATIONALE: Primary central nervous system (CNS) large B-cell lymphoma (PCNS-LBCL) is a rare and aggressive brain malignancy that is challenging to diagnose early. Conventional imaging methods, such as magnetic resonance imaging and PET-CT, may not provide conclusive results in the early stages. This case highlights the diagnostic utility of cerebrospinal fluid (CSF) IgG index, 24-hour IgG synthesis rate, and immunophenotyping, offering a reliable noninvasive approach to improve early diagnosis when imaging results are inconclusive. Capsule Endoscopy-Guided Diagnosis of Small Bowel Lymphoma Presenting as Protein-Losing Enteropathy: Complementary Role of Peripheral Blood Flow Cytometry.

Protein-losing enteropathy (PLE) is an uncommon and often underrecognized manifestation of lymphoproliferative disorders and may be difficult to diagnose when conventional gastrointestinal investigations are unrevealing. We present an 82-year-old woman with recurrent hospital admissions initially spanning six months for diarrhea, weight loss, peripheral edema, and persistent hypoalbuminemia. Initial upper gastrointestinal endoscopy was normal, and colonoscopy was deferred due to intercurrent infection. Despite extensive laboratory and radiologic evaluation, including routine biochemical testin

Western diet increases brain metabolism and adaptive immune responses in a mouse model of amyloidosis.

Diet-induced increase in body weight is a growing health concern worldwide. Often accompanied by a low-grade metabolic inflammation that changes systemic functions, diet-induced alterations may contribute to neurodegenerative disorder progression as well. This study aims to non-invasively investigate diet-induced metabolic and inflammatory effects in the brain of an APPPS1 mouse model of Alzheimer's disease. [18F]FDG, [18F]FTHA, and [18F]GE-180 were used for in vivo PET imaging in wild-type and APPPS1 mice. Ex vivo flow cytometry and histology in brains complemented the in vivo findings. 1H- m

PEGylated mesoporous silica core-shell redox-responsive nanoparticles for delivering paclitaxel to breast cancer cells.

Controlling the drug release and restricting its presence in healthy organs is extremely valuable. In this study, mesoporous silica nanoparticles (MSN) as the core, loaded with paclitaxel (PTX), were coated with a non-porous silica shell functionalized with disulfide bonds. The nanoparticles were further coated with polyethylene glycol (PEG) via disulfide linkages. We analyzed the physicochemical properties of nanoparticles, including hydrodynamic size via Dynamic Light Scattering (DLS), zeta potential, X-ray Diffraction (XRD) patterns, Fourier-Transform Infrared (FTIR) spectra, and imaging th

Comparison of EV characterization by commercial high-sensitivity flow cytometers and a custom single-molecule flow cytometer.

High-sensitivity flow cytometers have been developed for multi-parameter characterization of single extracellular vesicles (EVs), but performance varies among instruments and calibration methods. Here we compare the characterization of identical (split) EV samples derived from human colorectal cancer (DiFi) cells by three high-sensitivity flow cytometers, two commercial instruments, CytoFLEX/CellStream, and a custom single-molecule flow cytometer (SMFC). DiFi EVs were stained with the membrane dye di-8-ANEPPS and with PE-conjugated anti-EGFR or anti-tetraspanin (CD9/CD63/CD81) antibodies for e

Castleman's disease in the orbit of a 17-year-old girl: a case report.

Castleman's disease is an atypical lympho proliferative disorder comprising hyaline vascular elements, plasma cells, or a mixture of both, which can present in unicentric or multicentric fashion. Resection of unicentric lesions is typically curative, but multicentric disease, also characterized by constitutional symptoms and a poorer prognosis, often requires treatment with chemotherapy, radiation, steroids, or immune modulators. Castleman's disease is rarely diagnosed in the orbit. The authors present the clinical and histopathological findings of a 17-year-old who was found to have a focal l

Flow Cytometry-Based Assay to Detect Alpha Galactosidase Enzymatic Activity at the Cellular Level.

BACKGROUND: Fabry disease is a progressive, X chromosome-linked lysosomal storage disorder with multiple organ dysfunction. Due to the absence or reduced activity of alpha-galactosidase A (AGAL), glycosphingolipids, primarily globotriaosyl-ceramide (Gb3), concentrate in cells. In heterozygous women, symptomatology is heterogenous and currently routinely used fluorometry-based assays measuring mean activity mostly fail to uncover AGAL dysfunction. The aim was the development of a flow cytometry assay to measure AGAL activity in individual cells. METHODS: Conventional and multispectral imaging
