Fagopyrum tataricum ethanol extract ameliorates symptoms of hyperglycemia by regulating gut microbiota in type 2 diabetes mellitus mice.

Type 2 diabetes mellitus (T2DM) is typically accompanied by sudden weight loss, dyslipidemia-related indicators, decreased insulin sensitivity, and altered gut microbial communities. Fagopyrum tataricum possesses many biological activities, such as antioxidant, hypolipidemic, and hypotensive activities. However, only a few studies have attempted to elucidate the regulatory effects of F. tataricum ethanol extract (FTE) on intestinal microbial communities and its potential relationships with T2DM. In this study, we established a T2DM mouse model and investigated the regulatory effects of FTE on

Rapid detection of antibiotic resistance genes in lactic acid bacteria using PMMA-based microreactor arrays.

The emergence of lactic acid bacteria (LABs) resistant to existing antimicrobial drugs is a growing health crisis. To decrease the overuse of antibiotics, molecular diagnostic systems that can rapidly determine the presence of antibiotic resistance (AR) genes in LABs from yogurt samples are needed. This paper describes a fully integrated, miniaturized plastic chip and closed-tube detection chemistry that performs multiplex nucleic acid amplification. High-throughput identification of AR genes was achieved through this approach, and six AR genes were analyzed simultaneously in < 2 h. This time-

Probiotic characteristics of Lactobacillus plantarum E680 and its effect on Hypercholesterolemic mice.

BACKGROUND: Probiotics have been reported to reduce total cholesterol levels in vitro, but more evidence is needed to determine the clinical relevance of this activity. Chinese traditional fermented pickles are a good source of lactic acid bacteria. Therefore, pickle samples were collected for screening lactic acid bacteria based on their ability to survive stresses encountered during gastrointestinal passage and cholesterol reducing potency. RESULTS: Seventy five lactic acid bacteria strains were isolated from 22 fermented pickles. From these bacteria, Lactobacillus plantarum E680, showed th

Aggregation, adhesion and efficacy studies of probiotic candidate Pediococcus acidilactici NCDC 252: a strain of dairy origin.

Aggregation and adhesion capability and survival efficacy of candidate probiotic strain Pediococcus acidilactici NCDC 252 under simulated gastric, intestinal and vaginal conditions was studied. The strain exhibited strong autoaggregation phenotype and coaggregation with other Lactic acid bacteria (LAB) and E. coli. The adhesion studies of NCDC 252 to pig's intestinal epithelial cells showed its adhesive ability. Aggregation and adhesiveness were related through cell surface proteins as removal/extraction of surface proteins resulted in altered aggregation and no adhesiveness. Cell surface prot

Survival of selenium-enriched lactic acid bacteria in a fermented drink under storage and simulated gastro-intestinal digestion.

Selenium (Se), which is present as SeCys in seleno-proteins, is involved in cancer prevention, thyroid functioning, and pathogen inhibition. Se is incorporated in the diet through Se-containing foods. Some lactic acid bacteria (LAB) can biotransform selenite (toxic) into Se-nanoparticles (SeNPs) and Se-amino acids. To exert their beneficial properties in the host, bacteria should survive the harsh conditions of the gastrointestinal tract and during food storage. We evaluated whether selenization of LAB influenced bacterial growth and survival during gastrointestinal digestion and after storage

Competitive interactions between canine-derived lactic acid bacteria and Klebsiella michiganensis: Implications for probiotic potential in canine cystitis.

The Klebsiella oxytoca complex comprises several closely related bacterial species, including Klebsiella michiganensis, for which reports in veterinary medicine remain scarce, particularly in the context of urinary tract infections. This prospective in vitro study aimed to (i) molecularly characterize, using next-generation sequencing (NGS), a K. michiganensis strain isolated from canine cystitis, including its plasmid content and antimicrobial resistance gene repertoire; and (ii) identify, among six host-specific lactic acid bacteria (LAB) recovered from the urogenital tract of healthy dogs,

Probiotic potential and partial characterizations of exopolysaccharide from lactic acid bacteria isolated from camel rumen and dairy products.

Lactic acid bacteria (LAB) isolated from camel rumen and traditional dairy products were assessed for their probiotic potentials and exopolysaccharide (EPS) production. EPS were partially characterized, and their safety assessed by hemocompatibility testing in human, sheep, rabbit, and chicken red blood cells. EPS-producing isolates exhibited notable antibacterial activity against Salmonella paratyphi, but showed limited effects against Staphylococcus aureus and Enterococcus faecalis. All isolates demonstrated low resistance under acidic conditions (2.5 and 4.5) with survival rates ranging fro

HS-SPME/GC-MS and metabolomics elucidate volatile metabolite formation during fermentation of egg yolks.

Egg yolk is a nutritionally rich and functionally excellent ingredient, but its undesirable flavor often limits its applications. To address this, we investigated lactic acid bacteria (LAB) fermentation as a sustainable strategy to enhance yolk flavor. Results show that among ten LAB strains, Limosilactobacillus reuteri and Lacticaseibacillus rhamnosus provided the highest odor acceptability for yolks and grew well throughout 3.5 h of fermentation. Fermented yolks contained 42.8-207 times higher concentrations of volatile compounds (including aldehydes and ketones) than control samples, enhanc

Functional in vitro screening of probiotic strains for inoculation of piglets as a prophylactic measure towards Enterotoxigenic Escherichia coli infection.

Enterotoxigenic Escherichia coli (ETEC), being the major cause of post-weaning diarrhoea (PWD) in newly weaned piglets, induces poor performance and economic losses in pig production. This functional in vitro screening study investigated probiotic strains for use in suckling piglets as a prophylactic strategy towards PWD. Nine strains were evaluated based on their ability to: enhance intestinal epithelial barrier function, reduce adherence of ETEC F18 to intestinal cells, inhibit growth of ETEC F18, and grow on porcine milk oligosaccharides. Strains included in the screening were of the specie

Immuno-electron microscopy of primary cell cultures from genetically modified animals in liquid by atmospheric scanning electron microscopy.

High-throughput immuno-electron microscopy is required to capture the protein-protein interactions realizing physiological functions. Atmospheric scanning electron microscopy (ASEM) allows in situ correlative light and electron microscopy of samples in liquid in an open atmospheric environment. Cells are cultured in a few milliliters of medium directly in the ASEM dish, which can be coated and transferred to an incubator as required. Here, cells were imaged by optical or fluorescence microscopy, and at high resolution by gold-labeled immuno-ASEM, sometimes with additional metal staining. Axona

Fecal microbiota from chronically stressed donors transfers behavioral and prefrontal cortex gene expression alterations to healthy recipient mice.

Over the past decades, alterations in gut microbiota composition have emerged as a key contributor to the neurobiology of stress-related psychiatric conditions, including Major Depressive Disorder (MDD). Clinical and preclinical evidence consistently demonstrates correlational associations indicating that the gut microbiota not only responds to stress but also modulates the host stress response, thereby influencing behavioral domains relevant to depressive symptomatology. However, relatively few studies have investigated the causal relationship between stress-induced microbiota alterations and

Microbial community profiling for forensic drowning diagnosis across locations and submersion times.

BACKGROUND: Drowning diagnosis has long been a critical issue in forensic research, influenced by various factors such as the environment and decomposition time. While traditional methods such as diatom analysis have limitations in decomposed remains, microbial community profiling offers a promising alternative. With the advancement of high-throughput sequencing technology, forensic microbiology has become a prominent focus in the field, providing new research avenues for drowning diagnosis. During drowning, microbial communities enter the lung tissue along with the water. METHODS: In this st

Gut-thyroid axis: investigating the causality between the gut microbiota and autoimmune thyroid disease based on a Mendelian randomization study.

INTRODUCTION: Current studies have identified a close connection between the gut microbiota (GM) and autoimmune thyroid disease (AITD), indicating that the dysregulation of the GM could play a crucial bridging role in AITD. However, the causality between them has not been definitively defined. MATERIAL AND METHODS: We utilized the summary statistics of GM from the largest available meta-analysis of genome-wide association studies conducted by the MiBioGen consortium. The data on AITD were derived from the Genome-Wide Association Study (GWAS) database. We investigated the causality between GM
