Lactylation of RNA m6A demethylase ALKBH5 promotes innate immune response to DNA herpesviruses and mpox virus.

RNA N6-methyladenosine (m6A) demethylase AlkB homolog 5 (ALKBH5) plays a crucial role in regulating innate immunity. Lysine acylation, a widespread protein modification, influences protein function, but its impact on ALKBH5 during viral infections has not been well characterized. This study investigates the presence and regulatory mechanisms of a previously unidentified lysine acylation in ALKBH5 and its role in mediating m6A modifications to activate antiviral innate immune responses. We demonstrate that ALKBH5 undergoes lactylation, which is essential for an effective innate immune response

Blocking Palmitoylation of Toxoplasma gondii Myosin Light Chain 1 Disrupts Glideosome Composition but Has Little Impact on Parasite Motility.

Toxoplasma gondii is a widespread apicomplexan parasite that causes severe disease in immunocompromised individuals and the developing fetus. Like other apicomplexans, T. gondii uses an unusual form of substrate-dependent gliding motility to invade cells of its hosts and to disseminate throughout the body during infection. It is well established that a myosin motor consisting of a class XIVa heavy chain (TgMyoA) and two light chains (TgMLC1 and TgELC1/2) plays an important role in parasite motility. The ability of the motor to generate force at the parasite periphery is thought to be reliant u

Proteomics insights into infantile neuronal ceroid lipofuscinosis (CLN1) point to the involvement of cilia pathology in the disease.

Mutations in the depalmitoylation enzyme, palmitoyl protein thioesterase (PPT1), result in the early onset neurodegenerative disease known as Infantile Neuronal Ceroid Lipofuscinosis. Here, we provide proteomic evidence suggesting that PPT1 deficiency could be considered as a ciliopathy. Analysis of membrane proteins from brain enriched for acylated proteins from neonate Ppt1 knock out and control mice revealed a list of 88 proteins with differential expression levels. Amongst them, we identified Rab3IP, which regulates ciliogenesis in concert with Rab8 and Rab11. Immunostaining analysis revea

Integrative analysis and functional validation identified palmitoylated PHGDH as a therapeutic target for colon adenocarcinoma.

BACKGROUND: Colon adenocarcinoma (COAD) has high incidence and mortality with poor prognosis, creating an urgent need for novel prognostic models and therapeutic strategies. Protein palmitoylation, an important post-translational modification, exerts pivotal effects on tumorigenesis; however, its prognostic and mechanistic relevance in COAD remains unknown. This study determined palmitoylation-related prognostic biomarkers and probable therapeutic targets to treat COAD. METHODS: We integrated transcriptomic datasets from the TCGA and identified prognostic genes related to palmitoylation by con

Kv1.3 palmitoylation regulates spatial distribution and channel removal from the immunological synapse.

Kv1.3 is the main voltage-gated potassium channel in T cells. At the immunological synapse (IS), it sustains Ca2⁺ signaling and facilitates T cell activation. Aberrant Kv1.3 expression or activity is linked to autoimmune disorders, yet the mechanisms regulating its targeting and organization at the IS remain unclear. We show that Kv1.3 palmitoylation is a dynamic process mediating channel rearrangement at the IS. The ZDHHC21 acyltransferase, which also S-acylates the TCR, palmitoylates Kv1.3, positioning this enzyme as a potential therapeutic target. Palmitoylation promotes channel migration t

ZDHHC-Mediated Protein S-Palmitoylation in Cancer: Epigenetic Interfaces, Structural Logic and Therapeutic Targeting.

Protein S-palmitoylation, the reversible thioesterification of cysteine side chains, is emerging as a druggable post-translational modification that couples membrane topology to oncogenic, metabolic, immune, and epigenetic networks in cancer. ZDHHC palmitoyltransferases and depalmitoylating enzymes, including acyl-protein thioesterases and palmitoyl-protein thioesterase 1, constitute a dynamic circuitry that governs the localization, stability, and signaling competence of key regulators of tumor growth, metabolic adaptation, and immune phenotype. Here, we synthesize recent structural and chemi

CSPβ-Hsc70 partnership: Unveiling a chaperone-driven mechanism involved in acrosomal exocytosis in human sperm.

AIMS: Acrosomal exocytosis (AE) is a calcium-dependent membrane fusion event that enables human sperm to penetrate the zona pellucida. Defects in AE contribute to idiopathic male infertility, yet the chaperone mechanisms that regulate this process remain unclear. We sought to define the mechanistic role of Cysteine string protein β (CSPβ) and Heat shock cognate 70 (Hsc70) in AE, focusing on domain-level determinants and functional interactions with relevance to male infertility. MATERIALS AND METHODS: We integrated immunofluorescence and subcellular fractionation in human sperm with functiona

Targeting NLRP3 palmitoylation: Celastrol alleviates inflammasome activation induced by immunometabolism dysregulation in Kupffer cells.

Protein palmitoylation, the only reversible lipid-linked post-translational modification, acts as a critical regulatory mechanism for modulating protein function and subcellular localization. However, its specific roles and underlying mechanisms in the inflammatory response of liver macrophages (Kupffer cells) remain largely undefined. This study aimed to elucidate the precise mechanism by which palmitoylation regulates inflammation in Kupffer cells and to explore potential therapeutic interventions targeting this modification. Through metabolomic and membrane proteomic analyses, we demonstrat

The EV-A71 RNA-binding protein Matrin 3 facilitates viral replication by stabilizing viral RNA via ZDHHC20-mediated palmitoylation.

Enterovirus 71 (EV-A71) is a major causative agent of severe hand, foot, and mouth disease (HFMD) in young children. However, the molecular interplay between EV-A71 and host factors, particularly how these interactions govern viral replication, remains incompletely understood. To systematically identify host factors involved in EV-A71 infection, we performed comprehensive identification of RNA-binding proteins (RBPs) by mass spectrometry (ChIRP-MS) during viral infection and identified 374 host proteins associated with the EV-A71 genomic RNA. Integrative analysis of existing datasets further r

ACLY palmitoylation reprograms macrophages histone acetylation rewiring to repress hepatocellular carcinoma.

The tumor microenvironment (TME) domesticates macrophage function by decreasing chromatin accessibility. The activation and nuclear translocation of ATP-citrate lyase (ACLY) convert citrate to acetyl-CoA, providing a substrate necessary for histone acetylation. However, the underlying mechanisms in macrophage remodeling are poorly understood. Here, we found that saturated fatty acids (sFA), especially palmitic acid (PA), were lower in TAMs of patients with hepatocellular carcinoma (HCC). Scd1 knockout promoted PA accumulation, resulting in both primary and metastatic liver cancer retardation a

BACKGROUND: Protein palmitoylation, catalyzed by the zDHHC family of palmitoyltransferases, has emerged as a critical post-translational modification implicated in tumor progression. However, the comprehensive role of zDHHCs-mediated palmitoylation in molecular subtyping, prognosis, and immune microenvironment modulation in pancreatic cancer (PC) remains unclear. METHODS: We systematically integrated multi-cohort transcriptome, single-cell RNA-seq, and somatic mutation data for PC. Bioinformatic analyses included molecular subtyping based on zDHHCs expression, differential gene expression, fu

NLRP3 Cys126 palmitoylation by ZDHHC7 promotes inflammasome activation.

Nucleotide oligomerization domain (NOD)-like receptor protein 3 (NLRP3) inflammasome hyperactivation contributes to many human chronic inflammatory diseases, and understanding how NLRP3 inflammasome is regulated can provide strategies to treat inflammatory diseases. Here, we demonstrate that NLRP3 Cys126 is palmitoylated by zinc finger DHHC-type palmitoyl transferase 7 (ZDHHC7), which is critical for NLRP3-mediated inflammasome activation. Perturbing NLRP3 Cys126 palmitoylation by ZDHHC7 knockout, pharmacological inhibition, or modification site mutation diminishes NLRP3 activation in macropha

S-palmitoylation of the v-ATPase subunit RNAseK is necessary for Zika virus infection.

BACKGROUND: S-palmitoylation is a reversible and dynamic lipid post-translational modification that regulates protein localisation, stability, and function. During viral infections, S-palmitoylation can modulate viral replication by directly modifying viral proteins or by altering host proteins involved in the viral life cycle and immune responses. In this study, we investigated the role of S-palmitoylation of Ribonuclease Kappa (RNAseK) during infection with Zika virus (ZIKV), a member of the flavivirus family. RNAseK is an 11 kDa transmembrane protein recently identified as a component of th

ZDHHC18 promotes renal fibrosis development by regulating HRAS palmitoylation.

Fibrosis is the final common pathway leading to end-stage chronic kidney disease (CKD). However, the function of protein palmitoylation in renal fibrosis and the underlying mechanisms remain unclear. In this study, we observed that expression of the palmitoyltransferase ZDHHC18 was significantly elevated in unilateral ureteral obstruction (UUO) and folic acid-induced (FA-induced) renal fibrosis mouse models and was significantly upregulated in fibrotic kidneys of patients with CKD. Functionally, tubule-specific deletion of ZDHHC18 attenuated tubular epithelial cells' partial epithelial-mesench

Sepsis-induced acute lung injury: AUF1 regulates pyroptosis via ETS2/ZDHHC21-mediated STING palmitoylation : A therapeutic target for lung injury.

BACKGROUND: Sepsis-induced acute lung injury (ALI) is characterized by excessive inflammation and pyroptosis of lung epithelial cells. AU-rich element binding factor 1 (AUF1) is a key RNA-binding protein involved in mRNA decay and regulation of inflammatory responses. Understanding AUF1's role in ALI could reveal novel therapeutic targets. METHODS: We employed both in vivo (cecal ligation and puncture, CLP) and in vitro (lipopolysaccharide, LPS-treated cells) models of sepsis-induced ALI. Palmitoylation of TBK1 enhances the type I interferon signaling and strengthens anti-malarial immunity in mice.

Precise regulation of type I interferon signaling is crucial for effective immune defense against infectious diseases. However, the molecular mechanisms governing this pathway are not fully understood. Here, we show a function for palmitoylation in enhancing anti-malarial immune responses. Our findings reveal that ZDHHC9 enhances the type I interferon signaling by palmitoylating TBK1 at cysteine 292. Following infection with Plasmodium yoelii N67, the delicate balance between palmitoylation and depalmitoylation of TBK1 is disrupted. Specifically, upregulation of APT2 promotes persistent depalm

Astrocyte Elevated Gene-1 Cys75 S-Palmitoylation by ZDHHC6 Regulates Its Biological Activity.

Nonalcoholic fatty liver disease is a major risk factor for hepatocellular carcinoma (HCC). Astrocyte elevated gene-1/Metadherin (AEG-1/MTDH) augments lipid accumulation (steatosis), inflammation, and tumorigenesis, thereby promoting the whole spectrum of this disease process. Targeting AEG-1 is a potential interventional strategy for nonalcoholic steatohepatitis (NASH) and HCC. Thus, proper understanding of the regulation of this molecule is essential. We found that AEG-1 is palmitoylated at residue cysteine 75 (Cys75). Mutation of Cys75 to serine (Ser) completely abolished AEG-1 palmitoylati
