Elevated resting and agonist-induced whole blood chemiluminescence in patients with active infective endocarditis.

Infective endocarditis (IE) and surgical procedures related to cardiac surgery are accompanied by inflammatory responses that may alter production of oxidants by phagocytes. This study evaluates luminol enhanced whole blood chemiluminescence (LBCL) as a measure of oxidative production by circulating phagocytes in 26 IE patients in comparison to 27 matched patients with acquired valvular heart disease and 25 healthy controls. Blood was collected the day before and 3, 7, 12 and 21 days after valve replacement surgery for LBCL measurement; resting (rCL) and agonist (fMLP)-stimulated total light e

Enhancement of normal neutrophil chemiluminescence by chronic granulomatous disease neutrophils.

Neutrophils and other phagocytic cells from patients with chronic granulomatous disease (CGD) lack the ability to generate reactive oxygen intermediates (ROI), although other phagocytic functions appear to be intact. The effects of CGD neutrophils on the ability of normal neutrophils to produce ROI as measured by luminol-enhanced chemiluminescence (CL) were examined. Normal neutrophils (2 x 10(5)) had a peak CL response to phorbol myristate acetate (PMA; 20 ng/ml) of 6.5 +/- 0.9 mV, while the CL response to CGD neutrophils was completely absent. However, the addition of CGD neutrophils (8 x 10

Cascade catalytic self-supply of •OH coreactants enabled by GOx/Mn/Co-MOF hybrids for enhanced electrochemiluminescence sensing of miRNA-222.

In conventional electrochemiluminescence (ECL) sensing systems, the use of H2O2 as a coreactant often suffers from poor stability, susceptibility to decomposition, and weak reproducibility. Accordingly, constructing cascade autocatalytic reactions to in-situ generate hydroxyl radicals (•OH) is of great significance. Here, we developed a strategy based on the cascade catalytic self-supply of a •OH coreactant to enhance ECL sensing for miRNA-222, achieved via a cascaded self-feedback circuit and catalytic hairpin assembly (CHA). Specifically, a ternary hybrid nanozyme comprising highly dispersed

In situ reactive oxygen species generation driven by CuFeV-LDH for co-reactant-free electrochemiluminescence.

The development of co-reactant-free electrochemiluminescence (ECL) is of great significance for simplifying the analysis process, reducing background interference, and enhancing the detection sensitivity in complex biological matrices. In this work, a ternary CuFeV layered double hydroxide (LDH) was synthesized via a facile method and employed as an efficient ECL electrocatalytic material. Without the addition of any co-reactants, CuFeV-LDH could effectively catalyze the in-situ transformation of O2 and H2O into reactive oxygen species (ROS), thereby triggering the generation of a strong and s

A Paper Strip-Based Photoinduced Electrogenerated Chemiluminescence Platform with CTF/PMo12 Heterojunction-Sensitive Glutamic Acid Detection.

Glutamic acid (GA), a crucial neurotransmitter, plays a significant role in brain function, muscle health, and various neurological disorders. Elevated GA levels have been associated with conditions such as stroke, Parkinson's disease, epilepsy, and Alzheimer's disease. Accurate detection of GA levels is paramount for both diagnostic and research purposes. Herein, we develop a photoinduced electrogenerated chemiluminescence (PECL) test strip fiber paper coated with a covalent triazine framework (CTF)/H3PMo12O40 polyoxometalate (PMo12) heterojunction. Under blue-light irradiation, photogenerate

An innovative electrochemiluminescent immunosensor using dual amplified signals from AuNPs@CoSn(OH)6 for the detection of the AD biomarker: amyloid beta 1-40.

Alzheimer's disease (AD) is a degenerative condition of the nervous system that causes severe damage to patients' daily activities and quality of life. Amyloid beta 1-40 protein (Aβ40), which is involved in the formation of cerebral plaques, is one of the crucial biomarkers related to AD. Herein, a novel and highly sensitive immunosensor for the detection of Aβ40 is developed. Using a reinforced indium tin oxide-coated glass with a nanocomposite of gold nanoparticle-enhanced CoSn(OH)6 (AuNPs@CoSn(OH)6) to trigger the electrochemiluminescence (ECL) of luminol as the sensing signal, the immunose

A comprehensive study into false positive rates for 'other' biological samples using common presumptive testing methods.

The identification of biological fluids or materials in forensic samples is a key requirement in forensic science that relies on chemical and biological based tests, most of which exhibit false positivity. When reporting results from such tests, Forensic Scientists use words such as probable, possible, and likely, without always being able to provide robust support for these conclusions. In collating information about false positive rates for a number of these tests, we found limited research into the cross reactions observed from 'other' biological samples in commonly encountered case sample

[Possible mechanism of antioxidant action of dinitrosyl iron complexes].

The antioxidant effect of dinitrosyl iron complexes (DNICs) was studied in various model systems. DNICs with glutathione ligands effectively inhibited Cu2+-induced peroxidation of low density lipoproteins (LDL). The antioxidant effect of DNICs with phosphate ligands and free reduced glutathione (GSH) was less pronounced. In addition, DNICs with glutathione suppressed the formation of reactive oxygen species during co-oxidation of lecithin liposomes and glucose. Free radical oxidation in this system was induced with a lipophilic azo initiator and evaluated by luminol-dependent chemiluminescence

Fabrication of paper-based microfluidic device by recycling foamed plastic and the application for multiplexed measurement of biomarkers.

Microfluidic paper-based analytical devices (μPADs) are emerging as effective analytical platforms for point-of-care assays in resource-limited areas. Simple and cost-effective fabrication method still remains challenging on μPADs. A simple and cost-effective method for fabricating paper-based devices was presented in this work by using of dipping strategy with the recycled polystyrene in chloroform as the hydrophobic reagent. Adhesive tape was employed as mask to transfer the hydrophilic channel pattern to the paper substrate. With the single-sided adhesive tape stuck on the hydrophilic parts

Novel aspects of the activation of NADPH oxidase in neutrophils of rheumatic patients on biological therapy.

The relationship between inflammation and formation of reactive oxygen species (ROS) is still not completely understood and excessive inflammatory reaction is attributed to increased yet also to reduced ROS formation. To compare ROS formation in severe and low inflammation, neutrophil oxidative burst was analyzed in rheumatic patients before and during therapy with TNFα- or interleukin-6 receptor-neutralizing antibodies. Intracellular and extracellular ROS productions were evaluated on the basis of luminol- and isoluminol-enhanced chemiluminescence in isolated peripheral neutrophils. Disease a

Chemiluminescence cloth-based glucose test sensors (CCGTSs): A new class of chemiluminescence glucose sensors.

Chemiluminescence (CL) has been widely applied in many fields, but it is rarely used in a very simple, economical but effective way. In this work, for the first time, the CL cloth-based glucose test sensors (CCGTSs) are developed as a new class of CL glucose sensors, with no need for complicated, expensive device fabrication and peripheral equipment. When integrated with desirable hydrophobic barrier in the flow channel and gravity/capillary flow induced by a difference in height between the loading zone and the detection zone, a single cloth-based device can perform the whole CL process invol

Hydroxylamine amplified gold nanoparticle-based aptameric system for the highly selective and sensitive detection of platelet-derived growth factor.

This study developed a sensitive and selective aptamer-based chemiluminescent (CL) method for the determination of platelet-derived growth factor (PDGF)-BB using hydroxylamine enlarged gold nanoparticles (Au NPs). Rabbit anti-human PDGF-BB polyclonal antibody was covalently coupled on the 96-well plate that offers reactive N-oxysuccinimide ester (referred to as NOS group) surface. In the presence of target protein, the biotinylated aptamer was captured on the 96-well plate forming an antibody/PDGF-BB/biotinylated aptamer sandwiched complex, which was followed by the assembly of streptavidin co

The role of plasma adenosine deaminase in chemoattractant-stimulated oxygen radical production in neutrophils.

OBJECTIVES: Adenosine deaminase (ADA) has a role in many immunity mediated disorders, such as asthma, tuberculosis and coronary artery disease. This study aims to investigate the ability of plasma ADA to modulate reactive oxygen species (ROS) production in neutrophils, and examine the involvement of adenosine and the cyclic AMP signaling pathway in this process. METHODS: Neutrophils were stimulated, in the absence or presence of plasma, with the chemotactic peptide fMLP (formyl-methionyl-leucyl-phenylalanine), and the ROS production was determined with luminol-enhanced chemiluminescence. Dielectric barrier discharge-induced chemiluminescence: potential application as GC detector.

Atmospheric pressure dielectric barrier discharge (DBD) plasma can be used to split low molecular weight organic compounds, and the DBD-split/excited species can be swept into luminol solution to induce chemiluminescence (CL) emission. Based on this observation, a novel optical detector was proposed and preliminarily tested as a potential gas chromatographic (GC) detector in this work. The advantages of this new type of detector include the following: direct detection, fast response, high sensitivity, versatility (sensitive to a broad range of volatile organic compounds), simple and easy instr

Adenosine protects against indomethacin-induced gastric damage in rats.

This study examines the putative gastroprotective effect of adenosine on indomethacin-induced gastric lesions and the possible mechanisms involved. After 24 hr of starvation, the rats were treated either with indomethacin (Indo; 25 mg/kg, subcutaneously) alone or adenosine + Indo (Ado; 7.5 mg/kg, subcutaneously, three times a day), or the vehicle (5% NaHCO3, subcutaneously). The length of hemorrhagic lesions in the stomachs was expressed as the lesion index. The tissue-associated myeloperoxidase (MPO) activity and protein oxidation were measured in gastric tissue samples. Formation of reactive

Mucosal reactive oxygen species production in oesophagitis and Barrett's oesophagus.

Reactive oxygen species (ROS) produced by inflammatory cells can contribute to tissue destruction. ROS have been implicated in various gastrointestinal abnormalities, including the acid related peptic diseases. Although the development of oesophagitis and Barrett's columnar epithelium is associated with prolonged reflux of gastric acid, the exact mechanism by which tissue damage occurs is not known. To discover if ROS are involved in damage to the oesophageal mucosa, this study measured in vitro the mucosal ROS concentrations of biopsied mucosal samples taken from patients with reflux oesophag
