Comparative antibacterial activity of a novel semisynthetic antibiotic: etimicin sulphate and other aminoglycosides.

Etimicin is a novel fourth generation semisynthetic aminoglycoside. It has good antimicrobial activity against both gram-positive and gram-negative bacterial infections and also against aminoglycoside resistant strains. In the present study, in vitro antibacterial activity of etimicin was determined by minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC) and time kill curve tests against type strains and 407 clinical isolates (obtained in a surveillance study), in comparison to other aminoglycosides. Test results revealed that etimicin has potential antimicrobial ac

Carbapenem-associated multidrug-resistant Acinetobacter baumannii are sensitised by aztreonam in combination with polyamines.

Carbapenem-associated multidrug-resistant Acinetobacter baumannii (MDR-Ab) are common among clinical isolates worldwide and are a major therapeutic challenge. Previously it was shown that exogenous polyamines (spermine and spermidine) enhanced susceptibility to β-lactams but induced resistance to polymyxins in Pseudomonas aeruginosa. This study aimed to explore the possible availability of exogenous polyamines in treating carbapenem-associated MDR-Ab. The effects of polyamines on the growth rate of A. baumannii, minimum inhibitory concentrations (MICs) of antibiotics, and time-kill and chequer

Removal of benzo[a]pyrene by a highly degradable microbial community immobilized by modified wheat straw biochar.

Benzo[a]pyrene (BaP), a high-molecular-weight polycyclic aromatic hydrocarbon (HMW-PAHs), is a prevalent organic pollutant. Due to various environmental factors, the viability and degradation capacity of PAHs-degrading bacteria in contaminated soil are significantly reduced. Therefore, it is imperative to maintain microbial activity and enhance degradation efficiency. This study aims to optimize BaP removal by utilizing biochar-immobilized BaP for the enhancement of microbial communities' degradative potential. The immobilization of modified wheat straw biochar (MWBC) significantly enhanced th

Fluctuating environmental adaptation shapes antibiotic survival in Mycobacterium tuberculosis.

Mycobacterium tuberculosis (Mtb) encounters diverse and fluctuating microenvironments during infection, including changes in nutrient availability and pH. While adaptation to individual host-associated conditions has been extensively studied, the impact of repeated environmental fluctuations on bacterial physiology and antibiotic survival remains unclear. In this study, we investigated how long-term adaptation to stable or fluctuating environments influences Mtb growth and drug responses. Mtb populations were serially passaged for six consecutive passages under combinations of different carbon

Inactivation efficacy of atmospheric glow discharge plasma against Zygosaccharomyces rouxii vegetative cells in apple juice and its impact on juice quality.

Zygosaccharomyces rouxii dominates spoilage in concentrated and reconstituted apple juice. Its osmotolerance and biofilm-forming capacity hinder traditional juice sterilization. This work investigated atmospheric glow discharge plasma (GDP) inactivation of Z. rouxii vegetative cells in reconstituted apple juice prepared from commercial concentrate and corresponding juice quality changes. Experiments varied discharge powers, treatment durations and working gas types. Inactivation mechanisms were explored through reactive oxygen and nitrogen species and cellular damage. Juice quality attributes

Phage-phage competition and biofilms affect interactions between two virulent bacteriophages and Pseudomonas aeruginosa.

Virulent bacteriophages (or phages) are viruses that specifically infect and lyse a bacterial host. When multiple phages co-infect a bacterial host, the extent of lysis and dynamics of bacteria-phage and phage-phage interactions are expected to vary. The objective of this study is to identify the factors influencing the interaction of two virulent phages with different Pseudomonas aeruginosa growth states (planktonic, an infected epithelial cell line, and biofilm) by measuring the bacterial time-kill and individual phage replication kinetics. A single administration of phages effectively reduc

In vitro persistence of clinical methicillin-resistant Staphylococcus aureus isolates from bone and joint infections after exposure to daptomycin, telavancin, and vancomycin.

Bacterial persistence is linked to chronic infections caused by methicillin-resistant Staphylococcus aureus (MRSA). Understanding the persister profiles of daptomycin, telavancin, and vancomycin is crucial because MRSA bone and joint infections (BJIs) are associated with recurrence and treatment failure. Persister formation was assessed using quantitative in vitro assays against six clinical BJI MRSA isolates and a quality control ATCC strain. Assays were performed using a mid-exponential phase of bacteria challenged with steady-state drug concentrations of the three agents. Persisters were qu

Activity of antibiotics against Burkholderia cepacia complex in artificial sputum medium.

BACKGROUND: Burkholderia cepacia complex (Bcc) is a collection of intrinsically drug-resistant Gram-negative bacteria that cause life-threatening disease in people with cystic fibrosis (CF). Standard antimicrobial susceptibility testing methods have poor predictive value for clinical outcomes in Bcc infections, probably due in part to differences between in vitro testing conditions and the environment in which Bcc grow in the lungs of people with CF. OBJECTIVES: To compare the activity of commonly used antibiotics under standard in vitro testing conditions with activity in conditions mimickin

Viable but nonculturable state in the zoonotic pathogen Bartonella henselae induced by low-grade fever temperature and antibiotic treatment.

The zoonotic pathogen Bartonella henselae is responsible for diverse human diseases, from mild to life-threatening, but it often eludes detection in culture-based assays. This study investigates the potential of B. henselae to enter a viable but nonculturable (VBNC) state when exposed to human fever temperature or antibiotics, with this state confirmed by successful resuscitation. Viability was assessed using SYBR Green I/PI staining and propidium monoazide-quantitative polymerase chain reaction (PMA-qPCR), while culturability was determined through colony-forming unit (CFU) counting on blood

The Development of a Phytopathogenic Fungi Control Trial: Aspergillus flavus and Aspergillus niger Infection in Jojoba Tissue Culture as a Model.

After introducing the idea of using concentrations equal to or less than the minimum inhibition concentration (MIC) of some active chemical compounds for evacuating microbial cells, different types of microbes were evacuated. The original protocol was given the name sponge-like protocol and then was reduced and modified from a microorganism to another to prepare microbial ghosts for various applications such as immunological applications, drug delivery, and isolation of DNA and protein. Fungal pathogens that infect plants critically affect cost effectiveness, quality, and quantity of their pro

Oral biofilms exposure to chlorhexidine results in altered microbial composition and metabolic profile.

Oral diseases (e.g., dental caries, periodontitis) are developed when the healthy oral microbiome is imbalanced allowing the increase of pathobiont strains. Common practice to prevent or treat such diseases is the use of antiseptics, like chlorhexidine. However, the impact of these antiseptics on the composition and metabolic activity of the oral microbiome is poorly addressed. Using two types of oral biofilms-a 14-species community (more controllable) and human tongue microbiota (more representative)-the impact of short-term chlorhexidine exposure was explored in-depth. In both models, oral b

A coup d'état by NDM-producing Klebsiella pneumoniae overthrows the major bacterial population during KPC-directed therapy.

The objective of this study was to utilize a co-culture hollow-fiber infection model (HFIM) to characterize the interplay between a small, difficult-to-detect, New Delhi metallo-β-lactamase-producing Klebsiella pneumoniae (NDM-Kp) minor population and a larger K. pneumoniae carbapenemase (KPC)-producing K. pneumoniae population in the presence of KPC-directed antibacterial therapy. Selective plating onto agar with ceftazidime-avibactam was used to track the density of the NDM-Kp population. Susceptibility testing and the Verigene System failed to identify the small initial NDM-Kp population. H

Magnesium Links Starvation-Mediated Antibiotic Persistence to ATP.

Bacterial persisters emerge and increase in numbers over time as a bacterial culture grows from log phase to stationary phase. However, the underlying basis of the inevitable tendency is unclear. In this study, we investigated the role of nutrients in starvation-mediated persister formation of Staphylococcus aureus By screening of nutrient components, we found that starvation-induced persister formation of log-phase cultures could be reversed by addition of magnesium (Mg2+) but not amino acids, nucleotides, or other salts. Further, deprivation of extracellular Mg2+ reduced cytoplasmic ATP, ind

The small quinolone derived compound HT61 enhances the effect of tobramycin against Pseudomonas aeruginosa in vitro and in vivo.

HT61 is a small quinolone-derived compound previously demonstrated to exhibit bactericidal activity against gram-positive bacteria including methicillin-susceptible Staphylococcus aureus (MSSA) and methicillin-resistant Staphylococcus aureus (MRSA). When combined with the classical antibiotics and antiseptics neomycin, gentamicin, mupirocin and chlorhexidine, HT61 demonstrated synergistic bactericidal activity against both MSSA and MRSA infections in vitro. In this study, we investigated the individual antimicrobial activity of HT61 alongside its capability to potentiate the efficacy of tobram

Synthesis and antibiotic activity of novel acylated phloroglucinol compounds against methicillin-resistant Staphylococcus aureus.

The rise in antibiotic resistance among pathogenic microorganisms has created an imbalance in the drugs available for treatment, in part due to the slow development of new antibiotics. Cystic fibrosis (CF) patients are highly susceptible to antibiotic-resistant pathogens, including methicillin-resistant Staphylococcus aureus (MRSA). Phloroglucinols and related polyketide natural products have demonstrated antimicrobial activity against a number of Gram-positive bacteria including S. aureus. In this study, we investigated a series of acylated phloroglucinol derivatives to determine their potent

Influence of various uropathogens on crystallization of urine mineral components caused by Proteus mirabilis.

Infectious urolithiasis is a consequence of long-standing urinary tract infections with urease-positive bacteria, especially Proteus spp. However, because of the often mixed nature of urinary tract infections, in the case of urinary stones formation, several species of bacteria may be involved in the process. The purpose of the study was to determine the impact of the bacterial species: Escherichia coli, Klebsiella pneumoniae, Providencia stuartii, Pseudomonas aeruginosa and Staphylococcus aureus on the crystallization caused by Proteus mirabilis. The studies were conducted in synthetic urine
