Enhanced synchronization of thalamic theta band local field potentials in patients with essential tremor.

Local field potentials (LFPs) were recorded in 13 patients from pairs of microelectrodes driven through thalamus during functional localization prior to implantation of a thalamic deep brain stimulation electrode for treatment of tremor or pain. Six patients had a history of essential tremor (ET), 3 of multiple sclerosis, and the remaining 4 had symptoms of chronic pain. Specific to the ET group was the observation that oscillatory field potentials recorded from the two microelectrodes in the motor thalamus (ventralis intermedius--Vim, ventralis oralis posterior--Vop) were highly coherent at f

The plasma membrane calcium ATPase (PMCA) of neurones is electroneutral and exchanges 2 H+ for each Ca2+ or Ba2+ ion extruded.

The coupling between Ca2+ extrusion and H+ uptake by the ubiquitous plasma membrane calcium ATPase (PMCA) has not been measured in any neurone. I have investigated this with Ca2+- and pH-sensitive microelectrodes in large voltage-clamped snail neurones, which have no Na+-Ca2+ exchangers. The recovery of [Ca2+]i and surface pH after a brief depolarization or Ca2+ injection was not slowed by hyperpolarization to -90 mV from a holding potential of -50 mV, consistent with a 1 Ca2+ : 2 H+ coupling ratio. Since Ca2+ injections proved difficult to quantify, and Ca2+ currents through channels were obs

Role of Na+ conductance, Na(+)-H+ exchange, and Na(+)-K(+)-2Cl- symport in the regulatory volume increase of rat hepatocytes.

1. In rat hepatocytes under hypertonic stress, the entry of Na+ (which is thereafter exchanged for K+ via Na(+)-K(+)-ATPase) plays the key role in regulatory volume increase (RVI). 2. In the present study, the contributions of Na+ conductance, Na(+)-H+ exchange and Na(+)-K(+)-2Cl- symport to this process were quantified in confluent primary cultures by means of intracellular microelectrodes and cable analysis, microfluorometric determinations of cell pH and buffer capacity, and measurements of frusemide (furosemide)/bumetanide-sensitive 86Rb+ uptake, respectively. Osmolarity was increased from

Long-lasting GABA-mediated depolarization evoked by high-frequency stimulation in pyramidal neurons of rat hippocampal slice is attributable to a network-driven, bicarbonate-dependent K+ transient.

Biphasic GABAA-mediated postsynaptic responses can be readily evoked in CA1 pyramidal neurons of rat hippocampal slices by high-frequency stimulus (HFS) trains in the presence of ionotropic glutamate receptor antagonists. In the present experiments with sharp microelectrodes, whole-cell techniques, and K+-selective microelectrodes, an HFS train (40 pulses at 100 Hz) applied in stratum radiatum close to the recording site evoked a brief hyperpolarizing IPSP (hIPSP), which turned into a prolonged (2-3 sec) depolarization (GABA-mediated depolarizing postsynaptic potential; GDPSP). The I-V relatio

Dose-dependent metabolic response of mammary carcinoma to photodynamic therapy.

The metabolic response of mammary carcinoma in the C3H mouse to photodynamic therapy (PDT) was measured using in vivo 31P nuclear magnetic resonance (31P-NMR) spectroscopy and pH microelectrodes. Twenty-four hours after administration of Photofrin II (12.5 mg/kg), the tumor was subjected to photoactivation using an argon dye laser. Optical treatment doses were 200, 400, and 600 J/cm2 and corresponded to the following tumor control doses: TCD10/30, TCD50/30, and TCD90/30, respectively. In vivo 31P-NMR spectra and pH micro-electrode measurements were obtained prior to treatment and at 4, 24, 48,

Regulatory mechanism of cell pH in the renal proximal tubule of bullfrog nephron.

To examine the cellular mechanism of urinary acidification in detail, micropuncture studies were performed on the in situ bullfrog proximal tubule with nigericin-based pH microelectrodes. Pencil-type double-barreled antimony microelectrodes were also used for monitoring pHs of the tubular fluids. Luminal perfusion of 10(-3) M cyanide caused a biphasic change in cell pH (pHi): i.e., early acidification by 0.04 pH unit in 2 min and later alkalinization by 0.04. A profound depolarization of 30-35 mV was observed in the peritubular membrane potential (EM Peri), although the tubular fluid pH (pHTF)

Symmetry engineering in 2D bioelectronics facilitating augmented biosensing interfaces.

Symmetry lies at the heart of two-dimensional (2D) bioelectronics, determining material properties at the fundamental level. Breaking the symmetry allows emergent functionalities and effects. However, symmetry modulation in 2D bioelectronics and the resultant applications have been largely overlooked. Here, we devise an oxidized architectural MXene, referred to as oxidized MXene (OXene), that couples orbit symmetric breaking with inverse symmetric breaking to entitle the optimized interfacial impedance and Schottky-induced piezoelectric effects. The resulting OXene validates applications rangi

Switching in the activity state of an interneuron that controls coordination of the hearts in the medicinal leech (Hirudo medicinalis).

The rhythmically active heart interneuron HN(5) in the medicinal leech exhibits two distinct activity states, which have been associated with different coordination states of the two hearts. During the active state, it discharges high-frequency bursts of action potentials interrupted by rhythmic inhibitory input from other interneurons. In the inactive state, the same cell receives rhythmic inhibition but the membrane potential remains subthreshold between these volleys, producing few or no action potentials. We investigated differences in the membrane properties of the cell during the active

Does the inability of CA1 area to respond to ischemia with early rapid adenosine release contribute to hippocampal vulnerability?: An Editorial Highlight for "Spontaneous, transient adenosine release is not enhanced in the CA1 region of hippocampus during severe ischemia models".

This Editorial highlights a remarkable study in the current issue of the Journal of Neurochemistry in which Ganesana & Venton (2021) report new data showing that brain ischemia does not elicit transient adenosine release in the CA1 hippocampal area. Using fast-scan cyclic voltammetry at a carbon-fiber microelectrode implanted in the CA1 subfield of the hippocampus, it was shown that none of three different ischemia/reperfusion models could increase spontaneous, transient adenosine release, and more severe models even suppressed this presumably neuroprotective release. Since the authors have pr

Toward the development of a cortically based visual neuroprosthesis.

Motivated by the success of cochlear implants for deaf patients, we are now facing the goal of creating a visual neuroprosthesis designed to interface with the occipital cortex as a means through which a limited but useful sense of vision could be restored in profoundly blind patients. We review the most important challenges regarding this neuroprosthetic approach and emphasize the need for basic human psychophysical research on the best way of presenting complex stimulating patterns through multiple microelectrodes. Continued research will hopefully lead to the development of and design speci

Measurement of stimulated dopamine release in the rat by in vivo voltammetry: the influence of stimulus duration on drug responses.

High-speed cyclic voltammetry at carbon fibre microelectrodes was used to monitor stimulated striatal dopamine (DA) release in the anaesthetised rat. The effects of metoclopramide, chlorpromazine, benztropine and dexamphetamine on DA release after 1 s and 10 s stimulations were investigated. The antagonists enhanced DA release most clearly with 1 s stimuli. Benztropine had no effect on DA release with the 10 s stimulations, but elevated release after 1 s trains. Dexamphetamine reduced release with 10 s stimuli but enhanced release after 1 s trains. The results are discussed in terms of neurona

Long term evolution of fast ripples during epileptogenesis.

Objective.Fast ripples (FRs) have received considerable attention in the last decade since they represent an electrophysiological biomarker of the epileptogenic zone (EZ). However, the real dynamics underlying the occurrence, amplitude, and time-frequency content of FRs generation during epileptogenesis are still not well understood. This work aims at characterizing and explaining the evolution of these features.Approach.Intracortical electroencephalographic signals recorded in a kainate mouse model of temporal lobe epilepsy were processed in order to compute specific FR features. In situ laser-induced codeposition of copper and different metals for fabrication of microcomposite sensor-active materials.

We report one-step in situ laser-induced synthesis of the conductive copper microstructures doped with iron, zinc, nickel, and cobalt with highly developed surface area. It was observed that the presence of chlorides of the aforementioned metals in the solutions used in our experiments increases the deposition rate and the amount of copper in the resulting deposits; it also leads to the deposit miniaturization. The laser deposition from solutions containing cobalt (II) chloride of concentration more than 0.003 M results in fabrication of copper microelectrode with better electrochemical proper

Preventing Shift from Pneumocephalus During Deep Brain Stimulation Surgery: Don't Give Up the 'Fork in the Brain'.

CLINICAL VIGNETTE: We present the case of a patient who developed intra-operative pneumocephalus during left globus pallidus internus deep brain stimulation (DBS) placement for Parkinson's disease (PD). Microelectrode recording (MER) revealed that we were anterior and lateral to the intended target. CLINICAL DILEMMA: Clinically, we suspected brain shift from pneumocephalus. Removal of the guide-tube for readjustment of the brain target would have resulted in the introduction of movement resulting from brain shift and from displacement from the planned trajectory. CLINICAL SOLUTION: We electe

A novel robot-assisted method for implanting intracortical sensorimotor devices for brain-computer interface studies: principles, surgical techniques, and challenges.

Precise anatomical implantation of a microelectrode array is fundamental for successful brain-computer interface (BCI) surgery, ensuring high-quality, robust signal communication between the brain and the computer interface. Robotic neurosurgery can contribute to this goal, but its application in BCI surgery has been underexplored. Here, the authors present a novel robot-assisted surgical technique to implant rigid intracortical microelectrode arrays for the BCI. Using this technique, the authors performed surgery in a 31-year-old male with tetraplegia due to a traumatic C4 spinal cord injury

Single-Impact Electrochemistry in Paper-Based Microfluidics.

Microfluidic paper-based analytical devices (μPADs) have experienced an unprecedented story of success. In particular, as of today, most people have likely come into contact with one of their two most famous examples─the pregnancy or the SARS-CoV-2 antigen test. However, their sensing performance is constrained by the optical readout of nanoparticle agglomeration, which typically allows only qualitative measurements. In contrast, single-impact electrochemistry offers the possibility to quantify species concentrations beyond the pM range by resolving collisions of individual species on a microe

An electrophysiologic study of rabbit ciliary epithelium.

Microelectrode recordings from cells in rabbit ciliary epithelium have been made in vitro. Ionophoresis of Lucifer Yellow dye from microelectrodes during measurements of potential confirmed that the recordings were intracellular. Dye passed from the impaled cells into adjacent cells in both the nonpigmented and pigmented layers of the epithelium. Electrical coupling between epithelial cells also was observed. The mean (+/- SD) values of the potential measured across the basolateral membranes of the nonpigmented cells was -65 +/- -15 mV (n = 77); the mean value of the input resistance at this i

Intracellular potassium activity, potassium equilibrium potential and membrane potential of carotid body glomus cells.

The intracellular potassium activity (ai(K)) of glomus cells in isolated rabbit carotid bodies was measured with potassium ion selective microelectrodes (K+ electrodes). The measurements yielded a mean ai(K) value of 33.0 mM in Hepes-buffered Tyrode's solution equilibrated with 100% O2. This value is significantly higher than that predicted by assuming that K+ ions are passively distributed across the membrane of glomus cells. The relationship between the membrane potential (EM) and the equilibrium potential of K+ ions (EK) at various extracellular potassium activities (ao(K] suggests that K+
