Ultrastructure of the intestinal canal in healthy turkeys (Meleagris gallopavo) using light and scanning electron microscopy.

This study investigated the ultrastructural and histological features of the intestinal canal in healthy adult turkeys (Meleagris gallopavo) using light and scanning electron microscopy. Tissue samples from the duodenum, jejunum, ileum, and caecum were analyzed to characterize the organization of the intestinal wall. All segments exhibited the classical four-layered structure: tunica mucosa, submucosa, muscularis externa, and serosa. Significant morphological differences were identified among the intestinal segments. Villi in the duodenum showed thick, leaf-like or finger-like forms depending

Cross-species single-cell transcriptomic analysis reveals divergence of cell composition and functions in mammalian ileum epithelium.

Animal models are widely used for biomedical studies and drug evaluation. The small intestine plays key roles in nutrient absorption, hormone secretion, microbiota defense and drug absorption and metabolism. Although the intestinal structure of mammals is conserved, the differences on epithelial cell composition, functional assignments and drug absorption among mammals are largely unknown. Here, cross-species analysis of single-cell transcriptomic atlas of the ileum epithelium from mouse, rat, pig, macaque and human reveals the conserved and differential cell types and functions among species,

Expression of neutrophil gelatinase-associated lipocalin (NGAL) in the gut in Crohn's disease.

The antimicrobial glycoprotein neutrophil gelatinase-associated lipocalin (NGAL) is strongly expressed in several infectious, inflammatory and malignant disorders, among these inflammatory bowel disease (IBD). Fecal and serum NGAL is elevated during active IBD and we have recently shown that fecal NGAL is a novel biomarker for IBD with a test performance comparable to the established fecal biomarker calprotectin. This study examines expression of NGAL in the healthy gut and in Crohn's disease (CD), with emphasis on the previously unexplored small intestine. Pinch biopsies were taken from activ

Mpi-driven N-glycosylation orchestrates mucin O-glycosylation and intestinal homeostasis.

The intestinal mucus barrier physically separates the epithelium from the dense microbial community of the gut and is essential for intestinal homeostasis. The principal component, the gel-forming mucin MUC2, is extensively glycosylated, yet how different classes of glycans regulate mucin function remains unclear. Here we show that N-glycosylation is required for proper MUC2 maturation and mucus barrier integrity. Using mouse models with hypomorphic or intestinal epithelial-specific loss of the mannose-generating enzyme MPI, which is required for N-glycan synthesis, we find that impaired N-gly

Early histological changes of ileal mucosa after augmentation cystoplasty.

Segments of bowel are used routinely for transplantation in various pathological conditions such as contracted bladders or poorly compliant neuropathic bladders. However, little is known how these intestinal segments adopt to a toxic environment caused by urine. Therefore, the present study was performed to determine early histological changes of ileal mucosa after augmentation cystoplasty. Seven patients with augmentation cystoplasty underwent random cold-cup biopsies of ileal segments after a mean period of 14.4 months after cystoplasty and morphological changes were evaluated using light mi

Diverse patterns of expression of the 67-kD laminin receptor in human small intestinal mucosa: potential binding sites for prion proteins?

It has been shown that the 67-kD laminin receptor (LR) may function as a receptor for Sindbis and tick-born encephalitis viruses. Recent data indicate that the 37-kD precursor (LRP) for this molecule acts as a receptor for prion proteins (PrP), self-proteins implicated in the pathogenesis of transmissible spongiform encephalopathies including new variant Creutzfeldt-Jakob disease (nvCJD). Laminin and PrP share the same binding site on LRP, which is incorporated into the mature LR as a functional binding domain. To localize PrP binding sites potentially relevant to oral infection, the expressio

A spatial atlas of human gastrointestinal acute GVHD reveals epithelial and immune dynamics underlying disease pathophysiology.

Acute graft-versus-host disease (aGVHD) is a severe complication of allogeneic hematopoietic stem cell transplantation, driven by alloreactive donor T cells in the gut. However, the roles of additional donor and host cells in this process are not fully understood. We conducted multiplexed imaging to generate comprehensive spatial maps of healthy and GVHD-afflicted duodena at single-cell resolution. We profiled 59 biopsies from patients with varying gastrointestinal (GI) aGVHD manifestations and 18 healthy controls and measured the spatial expression of 40 proteins, designated to delineate dist

Chicken-derived R-spondin1, Wnt3a, Nrg1 and human FGF1 allow branched growth of chicken intestinal organoid cultures.

Intestinal organoids are widely used in mammalian studies to mimic and study intestinal function and host-pathogen interactions without using of whole animal studies. For chicken, this was not yet feasible. Recently, we have shown that embryonal intestinal organoids can grow for 10-15 passages (approximately 6 weeks) when using chicken-derived Rspo1 and Wnt3. In this paper, we show that replacing Wnt3 with chicken Wnt3a and adding chicken Nrg1 and human FGF1 results in efficient and long lasting growth, over 40 weeks, of embryonal chicken intestinal organoids. Moreover, the typical branching p

Dual function of angiogenin-4 inducing intestinal stem cells and apoptosis.

The intestinal epithelium is the first line of host defense, and its homeostasis is dependent on soluble factors that comprise the crypt niche. Antimicrobial proteins are one of the mediators to maintain gut homeostasis. Angiogenin-4 (Ang4) is a member of the ribonuclease A superfamily and plays a pivotal role in antimicrobial activity against gut microbiota. However, the functions of Ang4 within the intestinal crypt niche, particularly its involvement in the development of intestinal epithelial cells (IECs), remain unknown. Here, we demonstrate that Ang4 plays a significant role in maintainin

Lauric acid alleviates deoxynivalenol-induced intestinal stem cell damage by potentiating the Akt/mTORC1/S6K1 signaling axis.

Intestinal stem cell (ISC)-driven intestinal homeostasis is subjected to dual regulation by dietary nutrients and toxins. Our study investigated the use of lauric acid (LA) to alleviate deoxynivalenol (DON)-induced intestinal epithelial damage. C57BL/6 mice in the control, LA, DON, and LA + DON groups were orally administered PBS, 10 mg/kg BW LA, 2 mg/kg BW DON, and 10 mg/kg BW LA + 2 mg/kg BW DON for 10 days. The results showed that LA increased the average daily gain and average daily feed intake of the mice exposed to DON. Moreover, the DON-triggered impairment of jejunal morphology and bar

Down-regulation of CD95 (Fas/Apo-1) in the epithelia of adenovirus-infected appendices.

AIMS: Adenoviral inclusions are commonly seen in appendices from infants with intussusception. They are associated with focal epithelial budding and less frequently with epithelial shedding. These morphological changes could depend on the opposing effects of adenoviral gene products on CD95-mediated apoptosis. METHODS AND RESULTS: Appendices from intussusceptions with viral inclusions (n = 4) and normal appendices (n = 10) were studied by immunochemistry with anti-adenovirus, anti-CD95 and anti-HLA-DR antibodies. Apoptosis was studied by the TUNEL method. The mucosa of normal appendices conta

Ovarian Sertoli-Leydig cell tumors with heterologous elements. I. Gastrointestinal epithelium and carcinoid: a clinicopathologic analysis of thirty-six cases.

Thirty-six ovarian Sertoli-Leydig cell tumors containing heterologous elements in the form of gastrointestinal-type epithelium are described. Eight of the cases contained microscopic foci of insular carcinoid; in two of these cases of foci of goblet cell carcinoid were also present. The patients' ages ranged from 4-67 years (average, 23 years). The presenting manifestations were androgenic changes (18 cases), abdominal swelling (15 cases), acute abdominal symptoms (one case), menstrual irregularities (one case), and postmenopausal bleeding (two cases). All of the tumors were unilateral. The su

Cadmium ingestion exacerbates Salmonella infection, with a loss of goblet cells through activation of Notch signaling pathways by ROS in the intestine.

Whether cadmium ingestion affects the susceptibility to infection and the detailed mechanism have not been investigated. We aimed to evaluate the effects of cadmium on the intestinal mucosal barrier and Salmonella infection. We found that oral administration of cadmium caused damage to the intestinal mucosal barrier, with body weight loss, an increase in inflammation, significantly reduced Muc2 expression and goblet cell loss in the intestine. The effect of cadmium on secretory cell differentiation was further demonstrated to be regulated by the overactivation of the Notch signaling pathway by

Autophagy: A potential target for natural products in the treatment of ulcerative colitis.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease primarily affecting the mucosa of the colon and rectum. UC is characterized by recurrent episodes, often necessitating lifelong medication use, imposing a significant burden on patients. Current conventional and advanced treatments for UC have the disadvantages of insufficient efficiency, susceptibility to drug resistance, and notable adverse effects. Therefore, developing effective and safe drugs has become an urgent need. Autophagy is an intracellular degradation process that plays an important role in intestinal homeostasis. Em

Glycans that regulate Notch signaling in the intestine.

Intestinal homeostasis is key to the maintenance of good health. The small intestine plays important roles in absorption, digestion, hormonal and immune functions. Crypt base columnar (CBC) stem cells residing at the bottom of crypts are nurtured by Paneth cells, and together create the stem cell niche, the foundation of intestinal homeostasis. CBC stem cells replicate to replenish their number, or differentiate into a variety of epithelial cells with specialized functions. Notch signaling is a cell-cell signaling pathway that regulates both the proliferation and differentiation of CBC stem ce

[Primary insular carcinoid tumor in a benign cystic teratoma of the ovary (author's transl)].

A case of primary, insular, argentaffin carcinoid tumor (argentaffinoma) of the left ovary, arising in a bening cystic teratoma in a 60-year-old woman is reported. The tumor was not accompanied by any signs of the carcinoid syndrome despite a large volume, prominent acinar differentiation, and strong argentaffinity. Considering the morphologic carcinoid classification by Soga and Tazawa, the case had a mixed A + C pattern. A few Paneth-like cells and a few small calcifications in the stroma were present. Microscopic, ultrastructural, and histochemical findings were identical with those of carc

Epithelial-specific blockade of MyD88-dependent pathway causes spontaneous small intestinal inflammation.

Accumulating evidence suggests a role for Toll-like receptor (TLR) signaling at the intestinal epithelial cells (IECs) level for intestinal protection against exogenous injury or pathogenic infection. We hypothesized that MyD88 dependent TLR signaling at intestinal epithelium is critical for mucosal immune homeostasis. In the current study, a transgenic mouse model was generated in which a dominant-negative mutant of MyD88 (dnMyD88) was driven by an intestinal epithelial-specific murine villin promoter. Aged transgenic mice spontaneously developed chronic small intestinal inflammation, as reve

Heat stress disrupts intestinal stem cell migration and differentiation along the crypt-villus axis through FAK signaling.

During heat stress (HS), the intestinal epithelium suffers damage due to imbalance of tissue homeostasis. However, the specific mechanism by which intestinal stem cells (ISCs) migrate and differentiate along the crypt-villus axis to heal lesions upon insult is unclear. In our study, C57BL/6 mice and IPEC-J2 cells were subjected to normal ambient conditions (25 °C for 7 days in vivo and 37 °C for 18 h in vitro) or 41 °C. The results showed that HS impaired intestinal morphology and barrier function. The numbers of ISCs (SOX9+ cells), mitotic cells (PCNA+ cells), and differentiated cells (Paneth

Lysozyme localization in normal and diseased human gastric and colonic mucosa. A correlative histochemical, immunohistochemical and immunoelectron microscopic investigation.

The distribution of lysozyme in normal and pathological human gastric and colonic mucosa was studied by light and electron microscopic immunocytochemical techniques and compared with histological and histochemical features. Lysozyme was localized in pyloric glandular epithelial cells, mucous neck cells of fundic glands, Paneth cells and some crypt cells of the mature colonic mucosa. In addition, lysozyme was detected in a large spectrum of "immature" or "regenerative" epithelium: neck cells of the gastric regenerative zone, undifferentiated columnar cells of surface and hyperplastic interfoveo

Correlation between the barrier function of the intestinal epithelium and susceptibility to PEDV infection in piglets at different ages.

Porcine epidemic diarrhoea (PED) results in vomiting, diarrhoea, dehydration, and death in pigs. To analyse the relationship between intestinal development and susceptibility to porcine epidemic diarrhoea virus (PEDV), six new-born, 1-, 2-, 3-, 4-, and 8-week-old piglets were orally inoculated with 2 × 107 copies of the PEDV strain LJX01/2014 and sacrificed 48 h after infection. The results demonstrated that PEDV infection elicits diarrhoea of diverse severity across piglets of distinct ages. Moreover, viral shedding and villous atrophy were most pronounced in neonatal piglets. With increasing
