Relationship between polycyclic aromatic hydrocarbons measured in dust collected inside cars and from ambient air.

Polycyclic aromatic hydrocarbons (PAHs) are organic compounds with a high capacity for bioaccumulation and carcinogenic and mutagenic effects on human health. They can exist in both outdoor and indoor air. Indoors, except in the air, they can be bound to dust particles, and human intake occurs not only by inhalation but also by ingestion and dermal absorption of dust. Therefore, dust is an important source of human exposure to indoor carcinogenic contaminants. The aim of this research was to determine the contribution of individual PAHs in the total amounts of PAHs (ΣPAH) in dust collected ins

Occurrence of polycyclic aromatic compounds and interdomain microbial communities in oilfield soils.

Soil polycyclic aromatic compound (PAC) pollution as a result of petroleum exploitation has caused serious environmental problems. The unclear assembly and functional patterns of microorganisms in oilfield soils limits the understanding of microbial mechanisms for PAC elimination and health risk reduction. This study investigated the polycyclic aromatic hydrocarbons (PAHs) and substituted PAHs (SPAHs) occurrence, and their impact on the bacteria-archaea-fungi community diversity, co-occurrence network and functionality in the soil of an abandoned oilfield. The results showed that the PAC conte

Organ and tissue-specific distribution of selected polycyclic aromatic hydrocarbons (PAHs) in ApoE-KO mouse.

Polycyclic aromatic hydrocarbons (PAHs) are one of the most prevalent classes of environmental pollutants resulting from the incomplete combustion of hydrocarbons. Exposure to PAHs is implicated in the pathogenesis of the cardiovascular disease, pulmonary disease, and even cancer. However, little is known about organ- and tissue-specific distribution patterns of PAHs in animals at macro-tissue and microscopic levels. Here, by combining GC-MS and single-molecule fluorescence microscopy (SMFM), we revealed the distribution characteristics of four different PAHs (phenanthrene (Phe), pyrene (Pyr),

Light-induced cytotoxicity of 16 polycyclic aromatic hydrocarbons on the US EPA priority pollutant list in human skin HaCaT keratinocytes: relationship between phototoxicity and excited state properties.

The photocytotoxicity of 16 polycyclic aromatic hydrocarbons (PAHs) on the priority pollutant list of the United States Environmental Protection Agency (US EPA) were tested in human skin HaCaT keratinocytes. A selected PAH was mixed with HaCaT cells and irradiated with a solar simulator lamp for a dose equivalent to 5 min of outdoor sunlight and the cell viability was determined immediately and also after 24 h of incubation. For the cells without incubation after the treatments, it is found that all PAHs with three rings or less, except anthracene, are not photocytotoxic, while the four or fiv

Magnetic-field-dependent delayed fluorescence from thermally activated reverse charge separation of spin-correlated charge separated states.

Thermally activated delayed fluorescence (TADF), involving reversible electron transfer between a fluorescing S1 state and a magnetically responsive charge-separated (CS) state, constitutes a sensitive probe of the spin-chemical dynamics in the CS state as well as of the local magnetic molecular environment. With (Cl)TAA-mB-PDI and SQA-pB-PDI-featuring a Cl-substituted triarylamine ((Cl)TAA) or a squaraine unit as electron donors, meta-benzene (mB) or para-benzene (pB) bridges, and a perylene diimide (PDI) unit as electron acceptor-we introduce two novel triads with sufficiently small S1-CS en

DNA damage induced by genotoxicants in zebrafish (Danio rerio) embryos after contact exposure to freeze-dried sediment and sediment extracts from Laguna Lake (The Philippines) as measured by the comet assay.

Laguna Lake, Philippines, is utilised as a resource for drinking and irrigation water and a major source of animal protein for more than 11 million residents. However, it also serves as a huge sink for anthropogenic pollutants from a variety of direct or indirect discharges. Given their well-known properties to accumulate organic pollutants, Lake Laguna sediments have been investigated by use of a newly developed sediment-contact assay to determine the bioavailable fraction of the total genotoxic hazard potential. Freshly fertilized zebrafish eggs (2 h after fertilization) were allowed to deve

Glioblastoma targeting by water-soluble hypericin derivate HHL-PVP and photodynamic tumour killing.

Hypericin, a tumour-selective photosensitizer, has shown potential in cancer therapy, but its poor water solubility has limited clinical use. To address this, we developed a water-soluble variant called high hypericin-loaded polyvinylpyrrolidone (HHL-PVP) to enhance hypericin's applicability, particularly for treating glioblastoma, a typically terminal disease. We tested HHL-PVP in both in vitro and in vivo models, first confirming its fluorescent properties in the lab and then assessing its efficacy in more complex animal models. Using subcutaneous and orthotopic tumour mouse models, we combi

Natural constituents of St. John's Wort inhibit the proteolytic activity of human thrombin.

Thrombin, a multifunctional serine protease responsible for the proteolytic hydrolysis of soluble fibrinogen, plays a pivotal role in the blood coagulation cascade. Currently, thrombin inhibitor therapy has been recognized as an effective therapeutic strategy for the prevention and treatment of thrombotic diseases. In this study, the inhibitory effects of natural constituents in St. John's Wort against human thrombin are carefully investigated by a fluorescence-based biochemical assay. The results clearly demonstrate that most of naphthodianthrones, flavonoids and biflavones exhibit strong to

The potential of hypericin and hyperforin for antiadhesion therapy to prevent metastasis of parental and oxaliplatin-resistant human adenocarcinoma cells (HT-29).

Cancer cells disseminate to other parts of the body during metastasis through the process of intravasation. The hypericin and hyperforin effect has been described to understand the signal mechanisms that stimulate or stunt cancer cell sprouting to metastasis on colon adenocarcinoma cells HT-29 and its resistant form HT-29-OxR. We focused on the key points of adhesion proteins (cadherin, integrin, selectin and syndecan) and also proteins participating in or contributing to the process of cancer cell migration and adhesion through genes expression and proteins levels. Treatment effects were iden

The acute effect of Hypericum perforatum on short-term memory in healthy adults.

RATIONALE: Over-the-counter drugs containing Hypericum perforatum (H. perforatum) have been argued to improve memory and sustained attention. So far, these claims have not been supported in human studies. However, previous studies used rather high dosages, and little is known about the acute effect of small dosages. OBJECTIVE: We evaluated whether an acute treatment with Remotiv 500 and Remotiv 250 (500 or 250 mg of H. perforatum quantified to either 1 or 0.5 mg of hypericin) improved memory and sustained attention, as well as mood and state anxiety in healthy adults. METHOD: A single dosage

[Effect of Hypericin on the Function of the Neuroretina: An Electroretinographic Study].

BACKGROUND: Hypericin is an important component of the Saint John's wort (Hypericum perforatum). It is assumed to inhibit intracellular signalling cascades, which contribute to neoangiogenesis. The phototoxic effect of hypericin on the retina was investigated in human retinal pigment epithelium (RPE); hypericin induces oxidative stress and has also been described to be an inhibitor of Ca(2+) influx channel in cultured RPE cells. The aim of our study is to evaluate the effect of hypericine on the function of the neuroretina. METHODS: Isolated bovine retinas were perfused with an oxygen saturat

Photodynamic fungicidal efficacy of hypericin and dimethyl methylene blue against azole-resistant Candida albicans strains.

Antimicrobial photodynamic therapy (aPDT) is an emerging alternative to treat infections based on the use of photosensitisers (PSs) and visible light. To investigate the fungicidal effect of PDT against azole-resistant Candida albicans strains using two PSs with a different mechanism of action, hypericin (HYP) and 1,9-dimethyl methylene blue (DMMB), comparing their efficacy and the reactive oxygen species (ROS) species involved in their cytotoxicity. Azole-resistant and the azole-susceptible C. albicans strains were used. Solutions of 0.5 and 4 McFarland inoculum of each Candida strain were tr

Photosensitizing activity of hypericin and hypericin acetate after topical application on normal mouse skin.

BACKGROUND: Hypericin, originating from Hypericum perforatum, is a potent photosensitizer known to induce skin phototoxicity when given systemically. Previously, we have examined the penetration and distribution of hypericin and its acetate ester in the skin of hairless mice after topical application. OBJECTIVES: In this study, we assessed the time course and skin histopathology of the phototoxic response after a single topical application of hypericin and hypericin acetate, and subsequent irradiation. The amount of blood-borne photosensitizer and the skin clearance, as well the remaining pho

Laser-programmable glycosaminoglycan-based nanocarriers co-deliver hypocrellin B and doxorubicin for spatiotemporal chemo-photothermal therapy of hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) remains lethal due to systemic chemotherapy is halted by cardiotoxicity, multidrug resistance, and poor tumor penetration. To address these limitations, we developed a novel modified chondroitin sulfate/cystamine nanoparticle, termed CS-DHB NPs, that co-delivers doxorubicin (DOX) and the photosensitizer hypocrellin B (HB) and is encapsulated within a chondroitin sulfate/cystamine nanocarrier. These engineered nanoparticles (∼154 nm) demonstrated high drug-loading efficiencies (DOX: 80%; HB: 60%), exceptional serum stability (>72 h), and dual-drug release in laser

Insights on the Mechanical Properties of SARS-CoV-2 Particles and the Effects of the Photosensitizer Hypericin.

SARS-CoV-2 is a highly pathogenic virus responsible for the COVID-19 disease. It belongs to the Coronaviridae family, characterized by a phospholipid envelope, which is crucial for viral entry and replication in host cells. Hypericin, a lipophilic, naturally occurring photosensitizer, was reported to effectively inactivate enveloped viruses, including SARS-CoV-2, upon light irradiation. In addition to its photodynamic activity, Hyp was found to exert an antiviral action also in the dark. This study explores the mechanical properties of heat-inactivated SARS-CoV-2 viral particles using Atomic F

Synergistic effect of folic acid and hypericin administration to improve the efficacy of photodynamic therapy via folate receptors.

Transport systems are developed to improve the solubility of the transported drug, increase its stability, enhance its pharmacological activity and target cancer while minimising side effects. In this work, nanoporous silica particles that can be functionalized and loaded with a large number of hydrophobic molecules are proposed. The designed system was modified with folic acid to target the folic acid receptors of cancer cells. This modification enabled a higher uptake of the drug by the cells. Hypericin was selected as a hydrophobic molecule/drug with photodynamic properties suitable for dia
