Disruption of the gene Euchromatin Histone Methyl Transferase1 (Eu-HMTase1) is associated with the 9q34 subtelomeric deletion syndrome.

BACKGROUND: A new syndrome has been recognised following thorough analysis of patients with a terminal submicroscopic subtelomeric deletion of chromosome 9q. These have in common severe mental retardation, hypotonia, brachycephaly, flat face with hypertelorism, synophrys, anteverted nares, thickened lower lip, carp mouth with macroglossia, and conotruncal heart defects. The minimum critical region responsible for this 9q subtelomeric deletion syndrome (9q-) is approximately 1.2 Mb and encompasses at least 14 genes. OBJECTIVE: To characterise the breakpoints of a de novo balanced translocation

Effects of chronic dizocilpine on acute pain and on mRNA expression of neuropeptides and the dopamine and glutamate receptors.

The mesocorticolimbic circuitry has been implicated in the pathophysiology of several neuropsychiatric syndromes like chronic pain and addiction. The aim of this study was to evaluate the effects of dizocilpine (MK-801), a non-competitive N-methyl-D-aspartate (NMDA) receptor antagonist, on sensorimotor behaviors and the consequent changes in the dopamine, glutamate, and opiate systems in rats. Five groups of rats were subjected to acute tests for nociception (hot plate and paw pressure) before and after MK-801 (0.05, 0.1, 0.2 and 0.4 mg/kg, i.p.) or saline. Another two groups received daily i.

Immunohistochemical study of p53-associated proteins in rat brain following lithium-pilocarpine status epilepticus.

Activation of the p53-stress response pathway has been implicated in excitotoxic neuronal cell death. Recent studies have demonstrated an age-dependent induction of both p53 mRNA and protein in the rat brain following lithium-pilocarpine-mediated status epilepticus (LPSE). We investigated whether other proteins that have been shown to participate in the p53 cascade are induced by LPSE. We used immunohistochemistry to examine the expression of Mdm2, Bax, CD95/Fas/APO-1, ATM, Ref-1 and ubiquitin. A significant increase in nuclear Mdm2 immunoreactivity, which colocalized with p53, was observed in

Definition of the anterior choroidal artery territory in rats using intraluminal occluding technique.

This manuscript delineates the territory of the anterior choroidal artery (AChA) in rats, as defined by the induction of an AChA infarction. By advancing a 0.24-mm surgical suture up the internal carotid artery (ICA) to a point 0.5-2 mm proximal to the middle cerebral artery (MCA) origin, the AChA could be occluded and a reliable AChA distribution infarction was produced in 62% (23/37) of animals. The infarct volume, as defined by TTC staining, was 55+/-7 mm(3). Maps of the infarction, generated by measuring the entire area of overlapping coronal slices, demonstrated that the internal capsule

Cloning of glucocorticoid receptor and mineralocorticoid receptor cDNA and gene expression in the central nervous system of the tree shrew (Tupaia belangeri).

The glucocorticoid (GR) and the mineralocorticoid (MR) receptor mediate corticosteroid actions in the mammalian brain. Here, we report the sequence and distribution of both receptor subtype mRNAs in the central nervous system of the tree shrew Tupaia belangeri, a non-rodent mammal, phylogenetically located between insectivores and primates. The specific glucocorticoid and mineralocorticoid receptor cDNAs were cloned, employing polymerase chain reaction (PCR) based methods. The GR cDNA and MR cDNA encode the 776-amino acid (aa) and 977-aa receptor, respectively. Comparisons of both GR and MR wi

The role of nitric oxide in focally-evoked limbic seizures.

The deep rostral piriform cortex contains a site (area tempestas) in which focal application of picomole amounts of bicuculline, a GABA antagonist, triggers limbic motor seizures which are dependent upon activation of both N-methyl-D-aspartate and alpha-amino-3-hydroxy-5-methyloxole-4-proprionate subtypes of glutamate receptors. In the present study we determined whether nitric oxide can influence the local modulation of seizure initiation by bicuculline. Nitric oxide and the nitric oxide precursor L-arginine, alone or in combination with low doses of bicuculline were focally administered into

Expression of members of the trk family in the developing postnatal rat brain.

Tyrosine protein kinases trk, trkB and trkC are essential components of the high affinity receptors necessary to mediate biological effects of the neurotrophins NGF, BDNF, NT-3 and NT-4. Here we report on the expression of these receptors during postnatal development in the rat brain. Cells expressing mRNAs encoding different members of the trk family were identified by in situ hybridization using oligonucleotides complementary to their respective mRNA. In septum, striatum and brainstem, higher levels of trk mRNA were detected at 2 and 4 weeks than at 1 weeks of age. In thalamic nuclei associa

Distribution of cholecystokinin-like immunoreactivity in the rat main olfactory bulb.

The anatomical localization of cholecystokinin-like immunoreactivity (CCK-I) within the rat main olfactory bulb was analyzed by using the peroxidase-antiperoxidase immunocytochemical technique. Neurons or neuronal processes containing CCK-I were localized within all laminae of the olfactory bulb except the olfactory nerve fiber layer. A large population of CCK-I neurons, with morphology, size, and distribution corresponding to that of the middle and external tufted cells, was observed within a zone extending from the deep periglomerular region through the superficial one-half to one-third of t

Olfactory radiomics signatures link pTau217 to cognitive impairment in probable Alzheimer's disease: a multi-cohort machine learning study.

BACKGROUND: Alzheimer's disease (AD) diagnosis remains challenging because current molecular biomarkers, though sensitive, lack spatial specificity. Radiomics offers a promising alternative by quantifying subtle microstructural alterations from routine MRI that are invisible to traditional volumetric analyses. Given that olfactory-related regions are preferentially affected in AD with olfactory dysfunction preceding cognitive decline, this study aimed to develop and validate radiomics signatures from these regions as imaging biomarkers linking molecular pathology to clinical phenotypes. METHOD

The olfactory epithelium: a critical gateway for pathological tau propagation and a target for mitigating tauopathy in the central nervous system.

Olfactory impairment is a recognized early indicator of neurodegenerative diseases (NDs), such as Alzheimer's disease (AD). Intracellular aggregates of hyperphosphorylated tau protein, referred to as neurofibrillary tangles (NFTs), are a hallmark of AD. NFTs are found in the olfactory bulb (OB) and entorhinal cortex (EC), both crucial for processing olfactory information. We explored the hypothesis that typical tau lesions could appear early and progress along olfactory regions to reach connected areas critically affected in AD (e.g., EC and hippocampal formation). To that end, we used transge

Nasal epithelial cells expressing oxytocin receptor are involved in volatilized oxytocin effects on biased and social motivations and stress-declined neural stem cell proliferation.

The underpinnings how nasal oxytocin (OXT) treatment may affect brain-controlled motivations and behaviors remain scarcely explored. This study was undertaken to assess the presence of volatilized OXT exposure and its rapid effects in altering motivational behaviors and stress-dampened neural stem cell proliferation. Heterozygous Venus knock-in (OXTR Venus-Neo/+) mice designed for locating oxytocin receptor (OXTR)-neurons and oxytocin receptor knockout (KO) mice were used to identify nasal epithelial OXTR-expressing neurons. We demonstrated mice' nasal epithelium harbored OXTR-expressing neuro

Expression of serotonin transporter (SERT) and receptors 5-HTR1A and 5-HTR2A in an animal model of hypothermia.

The role of anterior hypothalamus and preoptic area in thermoregulation has been studied since the late 19th century (Charles Richet 1884, Aronsohn&Sachs 1885), and thermosensitive neurons have been recorded in these areas. Numerous animal studies with injection of serotonergic agonists or antagonists proved activation of thermoregulatory responses, and complex serotonergic circuits with origin in dorsal raphe nucleus (DRN) and projection in hypothalamus and preoptic area (POA), nucleus accumbens (Acc), and central amygdala were identified. We assessed the expression of serotonin transporter S

Temporopolar Amygdalohippocampectomy for Mesial Temporal Lobe Epilepsy: A Two-Dimensional Anatomical Operative Video.

Mesial temporal lobe epilepsy is the most frequent type of focal epilepsy in young adults and the most commonly reported in surgical series worldwide.1,2 When seizures become refractory to drug therapy, they are unlikely to remit spontaneously, and for the 30% of patients with epilepsy that is refractory to antiepileptic drugs, resection of the mesial temporal lobe structures provides seizure control rates of 70%-80%.3,4 The transsylvian route for amygdalohippocampectomy has been used at our institution for many years, evolving from the first description of Yasargil through the inferior circul

Differential c-Fos Response in Neurocircuits Activated by Repeated Predator Stress in Male and Female C57BL/6J Mice with Stress Sensitive or Resilient Alcohol Intake Phenotypes.

Comorbidity of post-traumatic stress disorder (PTSD) and alcohol use disorder (AUD) worsens the prognosis for each of these individual disorders. The current study aimed to identify neurocircuits potentially involved in regulation of PTSD-AUD comorbidity by mapping expression of c-Fos in male and female C57BL/6J mice following repeated predator stress (PS), modeled by exposure to dirty rat bedding. In experiment 1, the levels of c-Fos in the paraventricular nucleus of the hypothalamus (PVH) and the nucleus accumbens shell were higher after the second PS vs the first PS, indicating a sensitized

Juvenile handling rescues autism-related effects of prenatal exposure to valproic acid.

Environmental factors acting on young animals affect neurodevelopmental trajectories and impact adult brain function and behavior. Psychiatric disorders may be caused or worsen by environmental factors, but early interventions can improve performance. Understanding the possible mechanisms acting upon the developing brain could help identify etiological factors of psychiatric disorders and enable advancement of effective therapies. Research has focused on the long-lasting effects of environmental factors acting during the perinatal period, therefore little is known about the impact of these fac

Optimal Surgical Extent for Memory and Seizure Outcome in Temporal Lobe Epilepsy.

OBJECTIVE: Postoperative memory decline is an important consequence of anterior temporal lobe resection (ATLR) for temporal lobe epilepsy (TLE), and the extent of resection may be a modifiable factor. This study aimed to define optimal resection margins for cognitive outcome while maintaining a high rate of postoperative seizure freedom. METHODS: This cohort study evaluated the resection extent on postoperative structural MRI using automated voxel-based methods and manual measurements in 142 consecutive patients with unilateral drug refractory TLE (74 left, 68 right TLE) who underwent standar

Mapping Fos-immunoreactive neurons activated by intra-oral infusion of quinine, sucrose or water throughout the brain of B6 mice.

Fos immunohistochemistry was used to identify neurons in taste-related brain areas throughout the B6 mouse brain activated by intra-oral (IO) infusion of 3.0 mM quinine hydrochloride (Q), 1.0 M sucrose (S) or filtered water (W). IO infusion of Q and S elicited more Fos-immunoreactive (Fos-IR) neurons than W in the central medial (CM) and dorsomedial (DM) subareas of the parabrachial nucleus (PBN) and the central medial (CeM) amygdala (ps < 0.05). Infusion of Q led to more Fos expression than W in the central lateral (CL) PBN and the parvocellular reticular formation (PCRT; ps < 0.05). The only

Marked differences in the effects of levetiracetam and its analogue brivaracetam on microglial, astrocytic, and neuronal density in the rat model of kainic acid-induced temporal lobe epilepsy.

Efficient treatment of temporal lobe epilepsy (TLE) remains challenging due to limited understanding of cellular and network changes and the interference of novel antiepileptic drugs (AEDs) with tissue reorganisation. This study compared the effects of brivaracetam and levetiracetam on histological alterations in key brain regions of the epileptic circuitry, namely, the hippocampus, amygdala, piriform cortex (PC), endopiriform nucleus (EPN) and paraventricular thalamic nucleus (PVT), using the kainic acid (KA) rat model of TLE. Functional Connectivity in the Sensory Olfactory Subnetwork in REM Sleep Behavior Disorder and Parkinson's Disease: A Pilot Study.

BACKGROUND: Parkinson's disease (PD) is a neurodegenerative disorder whose diagnostic motor symptoms appear only after significant progression of neurodegeneration. Identification of preclinical markers is essential. Idiopathic rapid eye movement sleep behavior disorder (iRBD) has a high risk of conversion to PD. Olfactory impairment (hyposmia) is present in both PD and iRBD; hyposmia in iRBD may be an additional clue indicating the development of PD. The processes underlying hyposmia in iRBD are unknown. Using resting-state functional connectivity (rsFC), a "sensory olfactory subnetwork" (SOS

Quantitative T2 mapping-based longitudinal assessment of brain injury and therapeutic rescue in the rat following acute organophosphate intoxication.

Acute intoxication with organophosphate (OP) cholinesterase inhibitors poses a significant public health risk. While currently approved medical countermeasures can improve survival rates, they often fail to prevent chronic neurological damage. Therefore, there is need to develop effective therapies and quantitative metrics for assessing OP-induced brain injury and its rescue by these therapies. In this study we used a rat model of acute intoxication with the OP, diisopropylfluorophosphate (DFP), to test the hypothesis that T2 measures obtained from brain magnetic resonance imaging (MRI) scans
