Effect of zinc binding on the structure and stability of fibrolase, a fibrinolytic protein from snake venom.

Fibrolase is a metalloprotease with potential use as a fibrinolytic agent. Loss of the intrinsic zinc atom leads to a rapid decrease in enzymatic activity. Circular dichroism measurements indicate that there is a partial unfolding of an alpha-helical section of the protein concomitant with the loss of zinc. Removal of zinc can be affected by elevated temperatures, acidic pH values, and addition of chelating agents. At low molar concentrations, both ethylenediaminetetraacetic acid (EDTA) and dithiothreitol (DTT) were found to remove zinc efficiently. Analysis of the sequence of fibrolase identi

An O/W antibacterial emulsion stabilized by protein-polyphenol complexes to control food spoilage: A study on whey protein and caffeic acid.

This study used a non-covalent complex of whey protein (WP) and caffeic acid (CA) as an interface-stabilizing agent. The interaction between WP and CA was analyzed through spectroscopy and molecular dynamics simulations. A Pickering emulsion (WC-PE) with oregano essential oil (OEO) was created using the WP-CA complex and characterized. This complex was then combined with sodium alginate (SA) to produce calcium alginate gel beads (WC-PE-CAB), which were tested for their ability to preserve shrimp freshness. FT-IR spectroscopy and molecular dynamics showed that CA mainly binds to β-lactoglobulin

Co-stabilization effects of gluten/carrageenan to the over-heated myofibrillar protein: Inhibit the undesirable gel weakening and protein over-aggregations.

High-temperature (120 °C) sterilization is an indispensable process for manufacturing ready-to-eat surimi products, yet risking the denaturation of their myofibrillar proteins (MP), thus significantly reducing the gelling properties. To resolve this problem, herein, a synergistic co-strengthening strategy was designed. The negatively charged polysaccharide carrageenan (CG) was introduced into MP simultaneously with wheat gluten, followed by 120 °C thermal treatment for 30 min. A substantial enhancement in mechanical strength, up to four times greater (from 9.86 to 42.38 g·cm), was observed for

Knockdown of RNF128 attenuated Parkinson's-induced mitochondrial dysfunction in neurons by stabilizing the SIRT1 protein.

Mitochondrial dysfunction is closely related to the pathogenesis of Parkinson’s disease (PD). Studies have shown that ubiquitination plays a crucial role in regulating mitochondrial dysfunction. As a ubiquitin ligase, RNF128 may influence mitochondrial function by modulating its ubiquitination activity. This study aimed to investigate the specific effect and mechanism of RNF128 on PD-induced mitochondrial dysfunction in neurons. Animal and cellular models of PD were established for experimental investigation by intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) in

Walnut and peanut protein microgel particles: production, characterization, and application in stabilizing emulsions and gels.

The functional differences between peanut protein (PPI) and walnut protein (WPC) in constructing microgel particles and the structure-property regulation mechanism in stabilizing emulsions and forming gels were investigated in this study. The results revealed that superior stability was exhibited by the high internal phase emulsions (HIPPEs) prepared therefrom due to the stronger hydrophobicity possessed by walnut protein microgel particles (WPC-MPs). In contrast, enhanced gel-forming ability and higher gel strength were observed in HIPPEs prepared from peanut protein microgel particles (PPI-M

Deubiquitylase USP31 mediates ferroptosis and radiosensitivity in nasopharyngeal carcinoma by stabilizing HOXA10 protein levels.

BACKGROUND: Nasopharyngeal carcinoma (NPC) is a malignant epithelial tumor originating in the nasopharynx. Ubiquitin-specific peptidase 31 (USP31) exerts a tumor-promoting role in diverse cancers, but the function of USP31 in NPC is unclear. METHODS: Relative mRNA and protein levels were detected by qPCR, western blot, and immunohistochemistry. Cell proliferation was measured by cell counting kit and 5-ethynyl-2'-deoxyuridine assays. Ferroptosis was assessed by detecting reactive oxygen species, malondialdehyde, glutathione, and Fe2 + levels. Radiosensitivity was detected by clonogenic surviv

Physically modification treatments improved high internal phase emulsions stabilized by walnut protein.

Walnut protein (WP)-based emulsions are notable for their sustainability and biocompatibility. However, their emulsifying capability is often hindered by a densely packed structure. This study investigates the impact of physical modifications, specifically high-speed shearing, heat treatment, and ultrasonic treatment, on the structure of WP and examines the subsequent effects on the physicochemical properties and stability of the resulting high internal phase emulsions (HIPEs). The three modification methods reduced the average WP particle diameter by 55.38 %, 48.31 %, and 73.66 %, respectivel

Multiscale Computational Workflow to Determine Stability of Disulfide Bridges in Proteins─Application to IgG Antibodies.

An antibody or immunoglobulin is a Y-shaped assembly of four peptide chains, stabilized by several disulfide bridges that are critical for the structure and function. Failure to build or any damage to these disulfide bridges makes the antibody unable to assume the native and working three-dimensional structure. Based on our previous developments, we present a multiscale computational framework to assess the thermodynamic as well as kinetic stability of each of the disulfide bonds in the structure of the antibody. The computational strategy is based on generating free energy surfaces for the no

Exploiting tea protein-chitosan nanoparticles stabilized Pickering emulsion to enhance the bioaccessibility of curcumin.

Natural biopolymer-based nanoparticles are promising candidates for emulsion stabilization and delivery of hydrophobic compounds. In this study, tea protein (TP) and chitosan (CS) were employed to fabricate complex nanoparticles (NPs) using a pH-dependent method. At a mass ratio of 2:1 (TP:CS), the NPs exhibited favorable properties (206.62 nm, -30.86 mV, 85.41° contact angle) owing to electrostatic interactions, hydrogen bonding, and hydrophobic interactions (binding energy of the complex was -5.40 kcal/mol). At a particle concentration of 2 % and an oil-phase volume fraction of 60 %, the Pic

ATF2-mediated Transcriptional Activation of USP4 Stabilizes KMT2A Protein and Promotes Trophoblast Dysfunction.

Preeclampsia (PE) is a severe and prevalent complication of pregnancy, characterized by insufficient trophoblast invasion, placental hypoxia, and systemic inflammation, which collectively represent the central pathological mechanisms underlying the disease. The molecular mechanism involving activating transcription factor 2 (ATF2), ubiquitin-specific peptidase 4 (USP4), and lysine methyltransferase 2 A (KMT2A) in hypoxia/reoxygenation (H/R)-treated trophoblast cells (HTR-8/SVneo) is explored, aiming to provide a novel theoretical basis for therapeutic strategies against PE. Elasticity and Thermal Stability are Key Determinants of Hearing Rescue by Mini-Protocadherin-15 Proteins.

Protocadherin-15 is a core protein component of inner-ear hair-cell tip links pulling on transduction channels essential for hearing and balance. Protocadherin-15 defects can result in non-syndromic deafness or Usher syndrome type 1F (USH1F) with hearing loss, balance deficits, and progressive blindness. Three rationally engineered shortened versions of protocadherin-15 (mini-PCDH15s) amenable for gene therapy have been used to rescue function in USH1F mouse models. Two can successfully or partially rescue hearing, while another one fails. Here we show that despite varying levels of hearing re

Universal paramyxovirus vaccine design by stabilizing regions involved in structural transformation of the fusion protein.

The Paramyxoviridae family encompasses medically significant RNA viruses, including human respiroviruses 1 and 3 (RV1, RV3), and zoonotic pathogens like Nipah virus (NiV). RV3, previously known as parainfluenza type 3, for which no vaccines or antivirals have been approved, causes respiratory tract infections in vulnerable populations. The RV3 fusion (F) protein is inherently metastable and will likely require prefusion (preF) stabilization for vaccine effectiveness. Here we used structure-based design to stabilize regions involved in structural transformation to generate a preF protein vaccin

Age at Glaucoma Diagnosis in Germline Myocilin Mutation Patients: Associations with Polymorphisms in Protein Stabilities.

Glaucoma is the leading cause of irreversible blindness worldwide, with elevated intraocular pressure (IOP) as the only known modifiable risk factor. Trabecular meshwork (TM)-inducible myocilin (the MYOC gene) was the first to be identified and linked to juvenile and primary open-angle glaucoma. It has been suggested that mutations in the MYOC gene and the aggregation of mutant myocilin in the endoplasmic reticulum (ER) of TM may cause ER stress, resulting in a reduced outflow of aqueous humor and an increase in IOP. We selected 20 MYOC mutations with experimentally determined melting temperat

Huntingtin-Associated Protein 1 in Mouse Hypothalamus Stabilizes Glucocorticoid Receptor in Stress Response.

Huntingtin-associated protein 1 (Hap1) was initially identified as a brain-enriched protein that binds to the Huntington's disease protein, huntingtin. Unlike huntingtin that is ubiquitously expressed in the brain, Hap1 is enriched in the brain with the highest expression level in the hypothalamus. The selective enrichment of Hap1 in the hypothalamus suggests that Hap1 may play a specific role in hypothalamic function that can regulate metabolism and stress response. Here we report that Hap1 is colocalized and interacts with the glucocorticoid receptor (GR) in mouse hypothalamic neurons. Genet

USP49 potently stabilizes APOBEC3G protein by removing ubiquitin and inhibits HIV-1 replication.

The antiviral activity of host factor apolipoprotein B mRNA editing enzyme catalytic polypeptide-like 3G (APOBEC3G, A3G) and its degradation mediated by human immunodeficiency virus type 1 (HIV-1) Vif protein are important topics. Although accumulating evidence indicates the importance of deubiquitination enzymes (DUBs) in innate immunity, it is unknown if they participate in A3G stability. Here, we found that USP49 directly interacts with A3G and efficiently removes ubiquitin, consequently increasing A3G protein expression and significantly enhancing its anti-HIV-1 activity. Unexpectedly, A3G

Investigation of the thermal shift assay and its power to predict protein and virus stabilizing conditions.

Protein thermal shift assay (TSA) has been extensively used in investigation of protein stabilization (for protein biopharmaceutics stabilization, protein crystallization studies or screening of recombinant proteins) and drug discovery (screening of ligands or inhibitors). This work aimed to analyze thermal shift assay results in comparison to protein polymerization (multimerization and aggregation) propensity and test the most stabilizing formulations for their stabilization effect on enveloped viruses. Influence of protein concentration, buffer pH and molarity was tested on three proteins (i

Probing Bio-Nano Interactions between Blood Proteins and Monolayer-Stabilized Graphene Sheets.

Meeting proteins is regarded as the starting event for nanostructures to enter biological systems. Understanding their interactions is thus essential for a newly emerging field, nanomedicine. Chemically converted graphene (CCG) is a wonderful two-dimensional (2D) material for nanomedicine, but its stability in biological environments is limited. Systematic probing on the binding of proteins to CCG is currently lacking. Herein, we report a comprehensive study on the interactions between blood proteins and stabilized CCG (sCCG). CCG nanosheets are functionalized by monolayers of perylene leading
