SNX8 regulates BMPR2-mediated SMAD5 proteostasis to drive epithelial-mesenchymal transition in hepatocellular carcinoma.

BACKGROUND: Hepatocellular carcinoma (HCC) is characterized by high metastasis and poor prognosis. While the sorting nexin (SNX) family is linked to tumorigenesis, the role of SNX8 in HCC remains unexplored. METHODS: SNX8 expression and prognostic value were validated using TCGA datasets and clinical tissues. The impact of SNX8 on malignant phenotypes was evaluated via in vitro assays and in vivo xenograft models. RNA-seq was conducted to elucidate underlying mechanisms, followed by rescue experiments using pharmacological agents and plasmid transfection to verify key proteins. RESULTS: SNX8

Valsartan nano-filaments alter mitochondrial energetics and promote faster healing in diabetic rat wounds.

Chronic wounds are a common and debilitating condition associated with aging populations that impact more than 6.5 million patients in the United States. We have previously demonstrated the efficacy of daily topical 1% valsartan in treating wounds in diabetic mouse and pig models. Despite these promising results, there remains a need to develop an extended-release formulation that would reduce patient burden by decreasing the frequency of daily applications. Here, we used nanotechnology to self-assemble valsartan amphiphiles into a filamentous structure (val-filaments) that would serve as a sc

Hereditary hemorrhagic telangiectasia: from molecular biology to patient care.

Hereditary hemorrhagic telangiectasia (HHT) is an autosomal dominant vascular disorder characterized by severe and recurrent nosebleeds, mucocutaneous telangiectases, and, in some cases, life-threatening visceral arteriovenous malformations of various types, including pulmonary, hepatic, cerebral, and spinal. Gastrointestinal telangiectases are frequent and may cause severe bleeding. HHT type 1 results from mutations in ENG on chromosome 9 (coding for endoglin), and HHT type 2 results from mutations in ACVRL1 on chromosome 12 (coding for activin receptor-like kinase 1). Mutations of either of

Determination of the frequency and distribution of APC, PIK3CA, and SMAD4 gene mutations in Ugandan patients with colorectal cancer.

Uganda is a developing low-income country with a low incidence of colorectal cancer, which is steadily increasing. Ugandan colorectal cancer (CRC) patients are young and present with advanced-stage disease. In our population, there is a scarcity of genetic oncological studies, therefore, we investigated the mutational status of CRC tissues, focusing in particular on the adenomatous polyposis coli (APC), phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit alpha (PIK3CA), and SMAD4 genes. Our objective was to determine whether there were any differences between other populations and

Examination of Smad2 and Smad4 copy-number variations in skin cancers.

BACKGROUND: Smad2 and Smad4 transcription factors were identified as the signalling mediators of transforming growth factor Β (TGF Β) pathway. Copy number variations (CNVs) have been discovered to have phenotypic consequences and be associated with various types of cancers. CNVs of Smad2 and Smad4 were found to be associated with cancer pathogenesis in the recent array-based study. However, no such study has been performed in skin cancer yet. In this study, we aim to examine the CNVs of Smad2 and Smad4 in skin samples. METHODS: A total of 195 paired samples including basal cell carcinoma (BCC

TGFβ signaling sensitizes MEKi-resistant human melanoma to targeted therapy-induced apoptosis.

The TGFβ signaling pathway is known for its pleiotropic functions in a plethora of biological processes. In melanoma, TGFβ signaling promotes invasiveness and metastasis formation. However, its involvement in the response to therapy is controversial. While several studies have linked TGFβ signaling to elevated resistance to targeted therapy in melanoma, separate findings have indicated a favorable treatment response through TGFβ-mediated increase of cell death. We now found that the outcome of TGFβ signaling in the context of targeted therapy is dose dependent. Unlike low doses, high levels of

Guaiacol counters renal fibrosis via BMP/ERK/Smad1/5/9 signaling in UUO-induced chronic kidney disease.

Chronic kidney disease (CKD) is characterized by progressive fibrosis and inflammation, with limited therapeutic options. Natural compounds have been studied for their potential to offer kidney-protective benefits, especially in CKD. Guaiacol, a lignin-based polyphenol and one of the key constituents of Angelica sinensis (Oliv.) Diels has been shown to have anti-oxidative and anti-inflammatory properties. Hence, we aimed to inspect and analyze the potential of guaiacol in the unexplored realm of renal fibrosis, integrating computational pharmacology and validating the outcomes in an experiment

Murine nitrofen-induced pulmonary hypoplasia does not involve induction of TGF-beta signaling.

BACKGROUND/PURPOSE: In the murine nitrofen-induced model of congenital diaphragmatic hernia (CDH), the lungs are primarily hypoplastic and immature even before diaphragmatic closure. Because excess transforming growth factor-beta (TGF-beta) signaling induces pulmonary hypoplasia, the authors hypothesized that primary hypoplasia after nitrofen exposure may be caused by aberrant signaling by the TGF-beta pathway. Therefore, abrogation of TGF-beta signaling might rescue the hypoplasia. METHODS: The authors performed intratracheal microinjections of a recombinant adenoviral vector encoding a domi

Increased expression of TGF-beta1 and Smad 4 on oxygen-induced retinopathy in neonatal mice.

Retinal neovascularization (NV) is a major cause of the blindness associated with such ischemic retinal disorders as diabetic retinopathy, retinopathy of prematurity and retinal occlusion. Neovascularization is induced by complex interactions among growth factors and cytokines. Some studies confirm that VEGF play a central role in neovascularization, there remain some questions as to why VEGF antagonists are only partially effective. Transforming growth factor-beta (TGF-beta) has been implicated in the development of neovascularization (Gerard et al. 2000). Smad 4 plays the most important role

Changes in the molecular nodes of the Notch and NRF2 pathways in cervical cancer tissues from the precursor stages to invasive carcinoma.

Cancer is a multifactorial disease characterized by the loss of control in the expression of genes known as cancer driver genes. Cancer driver genes trigger uncontrolled cell replication, which leads to the development of malignant tumors. A cluster of signal transduction pathways that contain cancer driver genes involved in cellular processes, such as cell proliferation, differentiation, apoptosis and dysregulated organ growth, are associated with cancer initiation and progression. In the present study, three signal transduction pathways involved in cervical cancer (CC) development were analy

A Targeted Octahedral DNA Nanostructure Co-delivers siME3 and Doxorubicin to Enhance Collateral Lethality in ME2-Deficient Pancreatic Cancer.

The genetic characteristics of pancreatic cancer (PC) are being revealed, but treatment strategies based on these profiles are developing slowly. About one-third of PC patients harbor SMAD4 mutations, with its homozygous deletions often accompanied by deletions of the malic enzyme 2 (ME2) gene, leading to upregulation of malic enzyme 3 (ME3) to eliminate reactive oxygen species (ROS). We designed an aptamer-modified octahedral DNA nanostructure for targeted co-delivery of siRNA targeting ME3 (siME3) and doxorubicin (DOX). This nanostructure targets the epidermal growth factor receptor (EGFR) o

A case of Myhre syndrome mimicking juvenile scleroderma.

BACKGROUND: Myhre syndrome is a genetic disorder caused by gain of function mutations in the SMAD Family Member 4 (SMAD4) gene, resulting in progressive, proliferative skin and organ fibrosis. Skin thickening and joint contractures are often the main presenting features of the disease and may be mistaken for juvenile scleroderma. CASE PRESENTATION: We report a case of a 13 year-old female presenting with widespread skin thickening and joint contractures from infancy. She was diagnosed with diffuse cutaneous systemic sclerosis, and treatment with corticosteroids and subcutaneous methotrexate r

The first two Chinese Myhre syndrome patients with the recurrent SMAD4 pathogenic variants: Functional consequences and clinical diversity.

Myhre syndrome is a rare autosomal dominant multi-organ disorder characterized by growth retardation, skeletal anomalies, muscular hypertrophy, joint stiffness, facial dysmorphism, deafness, cardiovascular disease, and abnormal sexual development. Here we described the first two Chinese Myhre syndrome patients diagnosed by whole-exome sequencing. They both had de novo c.1498A > G (p.Ile500Val) variant in SMAD4 and presented with key characteristics of Myhre syndrome but also revealed uncommon features (polydactyly in the girl and precocious puberty in the boy). We performed functional analysis

Simvastatin inhibits stem cell proliferation in human leiomyoma via TGF-β3 and Wnt/β-Catenin pathways.

Uterine leiomyoma (UL) is the most common gynaecologic tumour, affecting an estimated 70 to 80% of women. Leiomyomas develop from the transformation of myometrial stem cells into leiomyoma stem (or tumour-initiating) cells. These cells undergo self-renewal and differentiation to mature cells, both are necessary for the maintenance of tumour stem cell niche and tumour growth, respectively. Wnt/β-catenin and TGF-β/SMAD pathways, both overactive in UL, promote stem cell self-renewal, crosstalk between stem and mature cells, cellular proliferation, extracellular matrix (ECM) accumulation and drive

Systemic approaches identify a garlic-derived chemical, Z-ajoene, as a glioblastoma multiforme cancer stem cell-specific targeting agent.

Glioblastoma multiforme (GBM) is one of the most common brain malignancies and has a very poor prognosis. Recent evidence suggests that the presence of cancer stem cells (CSC) in GBM and the rare CSC subpopulation that is resistant to chemotherapy may be responsible for the treatment failure and unfavorable prognosis of GBM. A garlic-derived compound, Z-ajoene, has shown a range of biological activities, including anti-proliferative effects on several cancers. Here, we demonstrated for the first time that Z-ajoene specifically inhibits the growth of the GBM CSC population. CSC sphere-forming i

SMAD proteins of oligodendroglial cells regulate transcription of JC virus early and late genes coordinately with the Tat protein of human immunodeficiency virus type 1.

JC virus (JCV) is the aetiological agent of progressive multifocal leukoencephalopathy (PML), a fatal, demyelinating disease of the brain affecting people with AIDS. Although immunosuppression is involved in infection of the brain by JCV, a direct influence of human immunodeficiency virus type 1 (HIV-1) has also been established. The Tat protein of HIV-1 has been implicated in activation of the cytokine transforming growth factor (TGF)-beta in HIV-1-infected cells and in stimulating JCV gene transcription and DNA replication in oligodendroglia, the primary central nervous system cell type infe

Multiple roles of the F-box protein Slimb in Drosophila egg chamber development.

Substrate-specific degradation of proteins by the ubiquitin-proteasome pathway is a precise mechanism that controls the abundance of key cell regulators. SCF complexes are a family of E3 ubiquitin ligases that target specific proteins for destruction at the 26S-proteasome. These complexes are composed of three constant polypeptides--Skp1, Cullin1/3 and Roc1/Rbx1--and a fourth variable adapter, the F-box protein. Slimb (Slmb) is a Drosophila F-Box protein that fulfills several roles in development and cell physiology. We analyzed its participation in egg chamber development and found that slmb
