BACKGROUND: There is an emerging role for flow cytometry (FC) in the assessment of small populations of plasma cells (PC). However, FC's utility has been questioned due to consistent underestimation of the percentage of PC compared to microscopy.
METHODS: A retrospective study was performed on bone marrow samples analysed by 8-colour FC. Plasma cell populations were classified as polyclonal or monoclonal based on FC analysis. FC findings were compared with microscopy of aspirates, histology and immunohistochemistry of trephine biopsies, and immunofixation (IFX) of serum and/or urine.

PURPOSE: To describe the clinicopathologic findings in failed Descemet-stripping automated endothelial keratoplasty (DSAEK) grafts.
DESIGN: Retrospective, interventional case series.
SETTING: New York Eye and Ear Infirmary.
STUDY POPULATION: Twenty-one patients with 22 failed DSAEK grafts treated between March 1, 2006 and February 1, 2008.
INTERVENTION: Repeat DSAEK or penetrating keratoplasty were performed in the eyes with failed grafts. All failed grafts were examined histopathologically.

PURPOSE: To assess the effects of variations in operative technique on the outcomes of penetrating keratoplasty (PK).
METHOD: Systematic literature review of published randomized controlled trials of operative techniques in PK.

A 49-year-old woman was referred to our clinic for penetrating keratoplasty. A central corneal scar involving the visual axis with high irregular astigmatism up to 15.8 D was noted. Because the scar was located mostly in the central cornea, ipsilateral rotational autokeratoplasty was suggested and performed. An 8.5-mm punch was used to trephine the cornea eccentrically and superiorly intentionally. OBJECTIVE: To demonstrate the watertight seal created by the new wide-angle Landers trunkless temporary keratoprosthesis (T-TKP).
METHODS: In 20 enucleated, porcine eyes, a 7.5-mm trephination was made, followed by capsulorrhexis and lensectomy. The T-TKP was sutured securely onto each eye. Testing the watertight seal at an intraocular pressure of 100 mm Hg was performed before and after pars plana vitrectomy.

BACKGROUND: In cases of contact lens intolerance and/or central corneal scars, corneal transplantation is indicated for advanced keratoconus.
