Vomitoxin (deoxynivalenol), a tricothecene mycotoxin produced by Fusarium species, has been detected in corn, wheat and other cereal grains intended for consumption by man and by animals used for food. The teratogenic potential of vomitoxin administered in the diet was studied by feeding a vomitoxin-containing certified rat feed ad lib. to groups of Fischer 344 rats during the entire course of pregnancy. The vomitoxin was added to the diet at a level of 0.0, 0.5, 2.0 or 5.0 ppm. There were no overt signs of toxicity in the dams, and no statistically significant differences in feed consumption

T-2 toxin, a potent Class A trichothecene mycotoxin, is a global food contaminant with poorly understood cardiotoxic mechanisms. Epidemiological evidence links T-2 toxin exposure to cardiac injury and selenium deficiency, yet the molecular basis of this interaction remains unclear. Selenoprotein S (SelS), an endoplasmic reticulum (ER)-resident selenoprotein, is critically involved in ER stress regulation and its expression correlates positively with selenium intake. This study investigates the role of SelS in T-2 toxin-induced myocardial damage and its mechanistic link to ER stress. Using in v

Cereal commodities are frequently contaminated with mycotoxins produced by the secondary metabolism of fungal infection. Among these contaminants, deoxynivalenol (DON), also known as vomitoxin, is the most prevalent type B trichothecene mycotoxin worldwide. Pigs are very sensitive to the toxic effects of DON and are frequently exposed to naturally contaminated feed. Recently, DON naturally contaminated feed has been shown to decrease porcine reproductive and respiratory syndrome virus (PRRSV) specific antibody responses following experimental infection. The objective of this study was to deter

T-2 toxin (T-2) and HT-2 toxin (HT-2) are trichothecene mycotoxins frequently detected in cereals. While exposure has primarily been evaluated using dietary assessment methods, human biomonitoring provides an alternative approach to assess internal exposure. This follow-up research aimed to further characterise the toxicokinetics of the mycotoxins T-2 and HT-2 and to subsequently develop a physiologically based kinetic (PBK) model, enabling prediction of urinary biomarker concentrations based on external exposure and vice versa. To this end, urine samples from 40 Norwegian adults were analysed

T-2 toxin, a very potent immunotoxic Type A trichothecene, is a secondary metabolite produced primarily by Fusarium spp., which grows on cereal grains and can lead to contaminated livestock feed. Repeated exposure to T-2 toxin has been shown to cause immunosuppression and decrease the resistance of exposed animals to a variety of infectious diseases; however, the effects of T-2 toxin on Marek's disease (MD) vaccinal immunity have not been reported. Four trials were conducted to determine the effects of T-2 toxin on vaccinal immunity against MD. Day-old, white leghorn chicks of Avian Disease an

Quantitative cytophotometry and ocular filar micrometry were used to monitor T-2 toxin induced alterations in chromatin and neuronal nuclear volume in supraoptic-magnocellular neurons of rat hypo-thalami. Thirty male Sprague-Dawley rats (200-220g) were given a single i.p. injection of T-2 toxin (0.5, 0.75, 1.0 and 1.5 X LD50), a trichothecene mycotoxin; rats were decapitated 8 hours post-dosing. After stoichiometric Feulgen-DNA staining of brain sections, scanning-integrating microdensitometry was used to quantify changes in the susceptibility of chromatin to Feulgen acid hydrolysis. Trichothecenes are among the most significant mycotoxins, causing serious agricultural problems worldwide. One representative of these is T-2 toxin, which is often found in poultry feed. Due to its harmful effects, it is important to investigate substances that can counteract it. Such compounds include thymol and cinnamaldehyde, whose anti-inflammatory and antioxidant activities are well-documented. In this study, the effects of 24 h exposure to T-2 toxin (100 nM), thymol, and cinnamaldehyde (10 and 100 μg/mL in both cases) and their combinations with the toxin were examined in chicken-derived

The mycotoxin deoxynivalenol (DON) contaminates chicken feed and negatively impacts intestinal health and overall production performance. The objective of this study was to investigate the effects of lactic acid bacteria and DON toxicity on production performance, intestinal morphology and absorptive function, and oxidative stress in broiler chickens. A total of 432 day-old-Ross 308 chicks were assigned to treatments arranged as a 2 × 3 factorial with mycotoxin (0 mg, noM; 5 mg/kg, M) and probiotic [0 cfu/kg, noP; 1.0 × 109 cfu/kg Lactobacillus acidophilus (LA); or 1.0 × 107 cfu/kg FloraMax-PW

Chitosan oligosaccharide (COS), a water-soluble derivative of chitin, has been recognized for its diverse biological properties. Deoxynivalenol (DON) is a prevalent mycotoxin, causing extreme liver damage. However, the mechanism whereby COS alleviates DON-induced liver injury remains unclear. In the present study, C57BL/6 mice were randomly divided into four groups: control (CON), DON (1.0 mg/d/kg BW DON), COS (200 mg/d/kg BW COS), and COS+DON (200 mg/d/kg BW COS + 1.0 mg/d/kg BW DON), with a period of 28 days. The results indicated that COS effectively reversed DON-induced weight loss, elevat

Deoxynivalenol (DON), as a widespread Fusarium mycotoxin in cereals, food products, and animal feed, is detrimental to both human and animal health. The liver is not only the primary organ responsible for DON metabolism but also the principal organ affected by DON toxicity. Taurine is well known to display various physiological and pharmacological functions due to its antioxidant and anti-inflammatory properties. However, the information regarding taurine supplementation counteracting DON-induced liver injury in piglets is still unclear. In our work, twenty-four weaned piglets were subjected t

Zearalenone (ZEA) and deoxynivalenol (DON) are two common mycotoxins produced by the genus Fusarium and have potential immunotoxic effects that may lead to a weak immune response against bacterial infections. Listeria monocytogenes (L. monocytogenes), a food-borne pathogenic microorganism ubiquitous in the environment, actively multiplies in the liver, where hepatocytes are capable of resistance through mediated innate immune responses. At present, it is not clear if ZEA and DON affect hepatocyte immune responses to L. monocytogenes infection or the mechanisms involved. Therefore, in this stud

Mycotoxin contamination in foods is a major risk factor for human and animal health due to its prevalence in cereals and their by-products. Deoxynivalenol (DON), mainly produced by Fusarium genera, is the most common mycotoxin detected in cereal products. Deoxynivalenol disrupts intestinal barrier function and decreases protein levels of tight junction proteins (TJP). However, the overall mechanism by which DON regulates specific TJP turnover and epithelial cell integrity remains unclear. Herein, we show that DON (2 μM) decreases the protein stability and accelerates the degradation of TJP in

Mycotoxin deactivators are a widely used strategy to abrogate negative effects of mycotoxin-contaminated feed. It has not been adequately evaluated whether these deactivators may detoxify bacterial toxins in the intestinal lumen and subsequently lower the inflammatory response in chickens. The present objective was to study the effect of a multicomponent mycotoxin deactivator (B), containing a bentonite and a bacterial strain capable to enzymatically biotransform trichothecenes especially deoxynivalenol (DON), when supplemented to a DON-contaminated feed in combination with an oral lipopolysac

In order to ensure the food safety, risk managers may implement measures to reduce human exposure to contaminants via food consumption. The evaluation of the effect of a measure is often an overlooked step in risk analysis process. The aim of this study was to develop a systematic approach for determining the effectiveness of mitigation measures to reduce dietary exposure to chemical contaminants. Based on expert opinion, a general framework for evaluation of the effectiveness of measures to reduce human exposure to food contaminants was developed. The general outline was refined by applicatio

Eleven castrated male pigs weighing 88.1 +/- 3.9 kg on average were adapted to a diet containing DON (4.2 mg DON/kg) over a period of 7 days. Feed was given restrictively with 1.1 kg per meal (two meals per day). On the day of measurement, all pigs were slaughtered at different time intervals following the morning meal containing DON (1, 2, 3, 4, 5, 6, 8, 15, 18 and 24 h after feeding), with the exception of one pig which was slaughtered unfed. DON and de-epoxy-DON were analysed in serum and digesta from consecutive segments of the digestive tract (stomach, small intestine divided into three p

Deoxynivalenol (DON), a Fusarium-derived mycotoxin, disrupts immune homeostasis and induces immunotoxicity. Macrophage extracellular traps (METs) are key effectors in innate immunity and inflammation, yet whether DON triggers METs formation and the underlying mechanisms remain unclear. This study aims to investigate the molecular mechanism by which DON induces METs generation through circadian rhythm immune checkpoints involving the PER2-c-Myc axis. Using RAW264.7 and THP-1 macrophage models, we combined siRNA-mediated Per2 knockdown, and pharmacological inhibitors to dissect the mechanisms of

Activated macrophages contribute prominently to the progression and maintenance of almost all inflammatory and autoimmune diseases. Although non-specific elimination of these phagocytes has been shown to treat animal models of inflammatory disease, the same therapies have been compromised by unacceptable toxicities, because they also kill quiescent macrophages in healthy tissues. In the studies below, we exploit upregulation of folate receptor beta (FRβ) on inflammatory (but not resting) macrophages to target a cytotoxic drug selectively to the inflammatory subset of macrophages. Because many

The sensitivity of pigs to deoxynivalenol (DON) might be increased by systemic inflammation (SI), which also has consequences for hepatic integrity. Liver lesions and a dys-regulated gene network might hamper hepatic handling and elimination of DON whereby the way of initiation of hepatic inflammation might play an additional role. First and second-pass exposure of the liver with LPS for triggering a SI was achieved by LPS infusion via pre- or post-hepatic venous route, respectively. Each infusion group was pre-conditioned either with a control diet (0.12 mg DON/kg diet) or with a DON-contamin

One hundred and fifty soil samples collected from different crop fields in southern Ontario, Canada were screened to obtain microorganisms capable of transforming deoxynivalenol (DON) to de-epoxy DON (dE-DON). Microbial DON to dE-DON transformation (i.e. de-epoxydation) was monitored by using liquid chromatography-ultraviolet-mass spectrometry (LC-UV-MS). The effects of growth substrates, temperature, pH, incubation time and aerobic versus anaerobic conditions on the ability of the microbes to de-epoxydize DON were evaluated. A mixed microbial culture from one composite soil sample showed 100%
