Gastroesophageal scintigraphy.

The technique of gastroesophageal scintigraphy was developed in order to quantitatively detect reflux from the stomach into the esophagus. The scintigraphic technique was compared to previous diagnostic tests including barium esophagography, fluoroscopy, endoscopy, esophageal biopsy, the acid perfusion test, esophageal manometry, and the acid reflux test. None of the other techniques is as sensitive as gastroesophageal scintigraphy for the detection of reflux, in comparison to the acid reflux test. Gastroesophageal scintigraphy is able to detect gastroesophageal reflux accurately, rapidly, non

Hydrogel-based biomimetic environment for in vitro modulation of branching morphogenesis.

The mechanical properties of the cellular microenvironment dramatically alter during tissue development and growth. Growing evidence suggests that physical microenvironments and mechanical stresses direct cell fate in developing tissue. However, how these physical cues affect the tissue morphogenesis remains a major unknown. We explain here that the physical properties of the cell and tissue microenvironment, biomimetically reproduced by using hydrogel, guide the tissue morphogenesis in the developmental submandibular gland (SMG). In particular, the softer gel enhances the bud expansion and cl

Development of topically effective formulations of acetazolamide using HP-beta-CD-polymer co-complexes.

The myriad of side effects induced by acetazolamide (oral use), the introduction of newer topical carbonic anhydrase inhibitors (CAI's) and the advent of other antiglaucoma medications has led to a decreased interest in acetazolamide. The use of cyclodextrins to improve the solubility and bioavailability of poorly soluble drugs has however, rekindled an interest in acetazolamide (ACZ), because its poor solubility is one of the major factor responsible for its failure to show topical effectiveness. Since water soluble polymers have been reported to improve the complexing capabilities of beta-cy

Flaminal® versus Flamazine® in the treatment of partial thickness burns: A randomized controlled trial on clinical effectiveness and scar quality (FLAM study).

Although partial thickness burns are the most frequently reported burn injuries, there is no consensus on the optimal treatment. The objective of this study was to compare the clinical effectiveness and scar quality of Flaminal® Forte to silver sulfadiazine (Flamazine®) in the treatment of partial thickness burns. In this two-arm open label multicenter randomized controlled trial, adult patients with acute partial thickness burns and an affected total body surface area of less than 30% were randomized between Flaminal® Forte and Flamazine® and followed for 12 months. Dressing changes in the Fl

Regulation of Complement and Contact System Activation via C1 Inhibitor Potentiation and Factor XIIa Activity Modulation by Sulfated Glycans - Structure-Activity Relationships.

The serpin C1 inhibitor (C1-INH) is the only regulator of classical complement activation as well as the major regulator of the contact system. Its importance is demonstrated by hereditary angioedema (HAE), a severe disease with potentially life-threatening attacks due to deficiency or dysfunction of C1-INH. C1-INH replacement is the therapy of choice in HAE. In addition, C1-INH showed to have beneficial effects in other diseases characterized by inappropriate complement and contact system activation. Due to some limitations of its clinical application, there is a need for improving the effica

Somatic gene therapy for a neurodegenerative disease using microencapsulated recombinant cells.

Neurodegenerative diseases caused by lysosomal enzyme deficiencies are catastrophic illnesses with both peripheral organ and central nervous system abnormalities. The mucopolysaccharidosis type VII mouse with beta-glucuronidase deficiency was used to develop an alternate approach to gene therapy, in which a "universal" cell line engineered to secrete the missing enzyme is implanted directly into all recipients requiring the same enzyme replacement. The cells, though nonautologous, were rendered immunologically tolerable by protection in immunoisolating microcapsules. Since the blood-brain barr

Biomaterial-mediated remodelling of the inflammatory microenvironment: a pH/ROS-responsive EGCG-metformin hydrogel for infected diabetic wound regeneration.

INTRODUCTION: Diabetic chronic wounds resist healing because persistent bacterial infection, excessive reactive oxygen species (ROS), unresolved pro-inflammatory responses, impaired angiogenesis, and defective tissue remodelling act simultaneously and reinforce one another. We therefore developed an injectable, microenvironment-responsive nanocomposite hydrogel, AP@EM-gel, to target these interconnected pathological processes. METHODS: AP@EM-gel was constructed from dopamine-grafted alginate (Alg-DA), phenylboronic-acid-modified ε-poly-L-lysine (EPBA), and co-assembled epigallocatechin gallat

Cysteine-Derived Carbon Dots Hydrogels with Visible-Light-Driven Photocatalytic Therapy for Infected Wound Healing.

The escalating crisis of multidrug resistance and biofilm recalcitrance poses severe challenges to conventional wound management. Photocatalytic antimicrobial therapy (PCAT) represents a promising nonantibiotic alternative that triggers the generation of reactive oxygen species (ROS) under illumination, thereby achieving broad-spectrum sterilization. Herein, we engineered a metal-free, visible-light-driven photocatalyst based on cysteine-derived carbon dots (Cys-CDs) via a facile microwave-assisted approach. Through precise N, S-heteroatom doping, the Cys-CDs possess tailored bandgaps of 2.78

Fecal Microbiota Transplantation for Inflammatory Bowel Disease: Where We Stand and What Is Next.

BACKGROUND: Fecal microbiota transplantation (FMT) is an emerging therapeutic strategy for inflammatory bowel disease (IBD). Every step of the FMT process, from donor recruitment and patient selection to pretreatment protocols, administration techniques, and post-FMT interventions, can significantly influence treatment outcomes. These components are interrelated, and even subtle differences in methodology may affect the overall efficacy of FMT for IBD. This review aimed to outline the current clinical experience and findings regarding FMT for IBD during the application process. SUMMARY: Donor

Oral multistage nanomedicine for synergistic chemo/chemodynamic/near-infrared-II photothermal cancer therapy.

The oral administration of drugs for cancer therapy can maintain optimal blood concentrations, is biologically safe and simple, and is preferred by many patients. However, the complex lumen environment, mucus layer, and intestinal epithelial cells are biological barriers that hinder the absorption of orally administered drugs. In this study, sea urchin-like manganese-doped copper selenide nanoparticles (Mn-Cu2-xSe NPs) were designed using an anion exchange method and coated with calcium alginate and chitosan (AC) to form Mn-Cu2-xSe@AC capsules. The pH-responsive swelling behavior of the AC pro

Multifunctional hydrogel delivery of mesenchymal stem cell secretome suppresses neutrophil extracellular trap formation and promotes diabetic wound healing via PGE2/BMAL1 pathway.

Excessive formation of neutrophil extracellular traps (NETs) significantly impedes diabetic wound healing. Although conditioned medium of mesenchymal stem cells (MSC-CM) has shown therapeutic potential due to its anti-inflammatory, antioxidant, and immunosuppressive properties, its regulatory effects on NETs remain poorly understood. Herein, this study systematically investigates the impact of MSC-CM, particularly conditioned medium of hypoxia-conditioned MSCs (HCM), on NET formation and diabetic wound repair. Transcriptomic analysis and public database mining revealed that circadian clock dys

Unveiling the anti-biofilm potential of bee venom against multi-drug resistant human pathogenic bacteria and fungi: perspectives into the efficacy and Possible mechanisms.

Bacterial biofilms pose significant challenges in treating infectious diseases due to antibiotic resistance. Finding alternative natural antimicrobial agents is crucial. Bee products, long valued for their nutritional and therapeutic benefits, exhibit potent antibacterial properties. This study investigates bee venom's antibacterial activity against biofilms formed by isolated microorganisms. Biofilm formation of pathogenic bacteria and yeast isolated from cannula was studied using two models: 96-well plates and alginate beads. We showed that low doses of bee venom effectively combat biofilms

Improving tendon repair through spatiotemporal modulation of TGF-β1 expression using an ultrasound-responsive hydrogel carrying siRNA-loaded nanoparticles.

Adhesion is a common complication during healing of injured tendons. TGF-β1 has a dual role in tendon healing, promoting tendon healing in the early stage, whereas its continued expression in the mid and late stages can lead to adhesion formation. Therefore, precise regulation of TGF-β1 expression to inhibit adhesion formation without compromising tendon healing strength may be an important strategy for enhancing tendon repair. Here, we designed an ultrasound-responsive hydrogel (URH) for carrying siRNA-loaded nanoparticles. This hydrogel enables the controlled release of encapsulated drugs in

Urological metabolic conditions alter regulation of quorum-sensing system and virulence genes expression in uropathogenic Pseudomonasaeruginosa.

Pseudomonas aeruginosa (P. aeruginosa) is responsible for complicated urinary tract infections (UTI) acquired from different transmission routes. Quorum sensing (QS) plays an important role in the regulation of virulence factors in P. aeruginosa and is dependent on the nutritional conditions. Under pathological conditions, urine contains elevated levels of metabolites such as glucose, creatinine, albumin and haem that may alter the QS behavior in P. aeruginosa. Hence, we investigated the role of the QS system in P. aeruginosa on growth and the expression of virulence genes under altered urinar
