Vertical transmission of TnSNPV, TnCPV, AcMNPV, and possibly recombinant NPV in Trichoplusia ni.

Four viruses were tested for vertical transmission in Trichoplusia ni: T. ni nucleopolyhedrovirus (TnSNPV), T. ni cypovirus (TnCPV), Autographa californica nucleopolyhedrovirus (AcMNPV), and AcMNPV engineered to express a scorpion toxin (AcMNPV.AaIT). Fifth instars were exposed to each virus, the survivors were reared and mated, and second-generation (F(1)) insects were examined for infection. TnSNPV was transmitted to offspring at a prevalence rate of 15.4%, TnCPV at 10.2%, and AcMNPV at 10.1%. Only one of 2484 F(1) insects was infected with AcMNPV.AaIT; this experiment was repeated, and none

Bollworm responses to release of genetically modified Helicoverpa armigera nucleopolyhedroviruses in cotton.

Helicoverpa armigera single nucleocapsid nucleopolyhedrovirus (HaSNPV) has been developed as a commercial biopesticide to control the cotton bollworm, H. armigera, in China. The major limitation to a broader application of this virus has been the relative long time to incapacitate the target insect. Two HaSNPV recombinants with improved insecticidal properties were released in bollworm-infested cotton. One recombinant (HaCXW1) lacked the ecdysteroid UDP-glucosyltransferase (egt) gene and in another recombinant (HaCXW2), an insect-selective scorpion toxin (AaIT) gene replaced the egt gene. In a

Sporulation by Entomophthora schizophorae (Zygomycetes: Entomophthorales) from housefly cadavers and the persistence of primary conidia at constant temperatures and relative humidities.

The duration of discharge of Entomophthora schizophorae (Zygomycetes: Entomophthorales) conidia from house fly (Musca domestica, Diptera) cadavers was measured at 7, 18, and 25 degrees C. The higher the temperature, the shorter the duration of conidia discharge. Significantly more conidia were produced per cadaver at 7 degrees C over a period of 120 h than at 18 and 25 degrees C. At 25 degrees C, the initial discharge over the first 10 h was much larger than at the other temperatures, and at 7 degrees C, no peak in discharge was observed. The persistence of E. schizophorae primary conidia was

Entomophthora muscae resting spore formation in vivo in the host Delia radicum.

The formation in vivo of Entomophthora muscae resting spores was investigated in the host, Delia radicum (cabbage root fly), by analysis of field data on the seasonal occurrence of E. muscae resting spores over 4 years. E. muscae resting spores in D. radicum were spherical with an average diameter of 39.4 microm, and the average numbers produced were estimated at 5.7 x 10(4) resting spores/female cadaver. Resting spores were found only after midsummer in D. radicum and almost exclusively in females. The proportion of infected females with resting spores was negatively correlated with average w

Entomopathogenic potential of Verticillium and Acremonium species (Deuteromycotina: Hyphomycetes).

Hyphomycetes with conidia formed in slimy heads from hyaline mycelium were isolated from a range of insect hosts. Isolation on artificial medium and microscopic examination revealed that these fungi in many cases were not Verticillium lecanii despite a superficial resemblance to this common entomopathogen. The fungi were identified as Verticillium fusisporum, Verticillium psalliotae, Verticillium lamellicola, and species of Acremonium. Isolates of these fungi were bioassayed against the sweet-potato whitefly (Bemisia tabaci) and against the housefly (Musca domestica) to examine their entomopat

Ultrastructural Changes in the Milk Gland of Tsetse Glossina morsitans centralis (Diptera; Glissinidae) Female Infected by a DNA Virus.

Milk glands, dissected out and collected from Glossina morsitans centralis (Machado) females, artificially inoculated at the third-instar larval stage with a virus suspension obtained from hypertrophied salivary glands of wild-caught virus-infected Glossina pallidipes (Austen), were processed for routine electron microscopy and examined for pathological changes. They were compared to milk glands dissected out from normal female G.m. centralis at the same stage of pregnancy cycle. Upon dissection there were notable physical differences between control and virus-infected milk glands. Histologica

Host range tests with Edhazardia aedis (Microsporida: Culicosporidae) against northern Nearctic mosquitoes.

Twelve species of northern Nearctic mosquitoes representing five genera (Aedes, Anopheles, Culex, Culiseta, and Psorphora) were bioassayed for susceptibility to Edhazardia aedis, an exotic, heterosporous, microsporidian parasite of the yellow fever mosquito, Aedes aegypti. Infections were achieved in Aedes atropalpus, Aedes triseriatus, and Aedes vexans following oral ingestion of uninucleate spores obtained from patently infected Ae. aegypti larvae. The microsporidium underwent normal vegetative growth and development in each unnatural mosquito host; the only differences observed were in the

Crystals of virus-like particles in the metacestodes of Taenia solium and T. crassiceps.

Crystals of virus-like particles (VLP) are described as occurring in the nuclei of damaged tegumentary cytons from carcasses of Taenia solium metacestodes that had been stripped of their teguments. The VLP are grouped as parallel lines of round particles in an hexagonal packaging of spheroids forming small or large crystals. The individual particles have an external diameter of 36-37 nm and a wall of 5-6 nm thick, which surround a cavity of lower electron density. As identical crystals were also observed in normal tissues of T. solium and of T. crassiceps, it is suggested that both species of

Ultrastructural changes in salivary glands of tsetse, Glossina morsitans morsitans, infected with virus and rickettsia-like organisms.

Electron microscope observations on enlarged hypertrophied salivary glands dissected from adult laboratory-reared male Glossina morsitans morsitans show a concurrent infection of the salivary gland tissue with rod-shaped virus particles and intracellular rickettsia-like organisms. The latter are found intracellular in the epithelium and in the gland lumen enclosed within lytic zones. The virus particles are found within the degenerating cytoplasm, nuclei, and lumen of the cell where they are especially numerous. Stratified epithelium and gland enlargement are a prominent feature of the infecti

Mixed infection of Rickettsiella phytoseiuli and Coxiella burnetii in Dermacentor reticulatus female ticks: electron microscope study.

Mixed infection of Rickettsiella phytoseiuli and Coxiella burnetii was investigated in hemolymph and organs of experimentally infected females of Dermacentor reticulatus ticks. Following intracoelomic infection, both agents, with the exception of Gene's organ, multiplied well in the cells of the tick host's organs. Two out of six developmental stages of R. phytoseiuli, i.e., crystal-forming and small dark particles, in dual infection with C. burnetii revealed marked morphological alterations. C. burnetii in the presence of R. phytoseiuli penetrated into the cortical layer of the synganglion an

Sublethal and antioxidant effects of six ichthyotoxic algae on early-life stages of the Japanese pearl oyster.

Several species of harmful algae form blooms that are detrimental to aquatic organisms worldwide with severe economic loss to several industries. The cosmopolitan ichthyotoxic dinoflagellates and raphidophytes Karenia spp., Chattonella spp., Heterosigma spp., and Margalefidinium (Cochlodinium) polykrikoides are known to cause mass mortalities of fish and invertebrates, and the dinoflagellates Heterocapsa spp. are known to cause mass mortalities of shellfish, notably bivalve molluscs. CHHBP: a newly identified receptor of crustacean hyperglycemic hormone.

Crustacean hyperglycemic hormone (CHH) is a neurohormone found only in arthropods that plays a pivotal role in the regulation of hemolymph glucose levels, molting and stress responses. Although it was determined that a membrane guanylyl cyclase (GC) acts as the CHH receptor in the Y-organ during ecdysteroidogenesis, the identity of the CHH receptor in the hepatopancreas has not been established. In this study, we identified CHH binding protein (CHHBP), as a potential receptor by screening the annotated unigenes from the transcriptome of ITALIC! Circadian clock genes and photoperiodism: Comprehensive analysis of clock gene expression in the mediobasal hypothalamus, the suprachiasmatic nucleus, and the pineal gland of Japanese Quail under various light schedules.

In birds, the mediobasal hypothalamus (MBH) including the infundibular nucleus, inferior hypothalamic nucleus, and median eminence is considered to be an important center that controls the photoperiodic time measurement. Here we show expression patterns of circadian clock genes in the MBH, putative suprachiasmatic nucleus (SCN), and pineal gland, which constitute the circadian pacemaker under various light schedules. Although expression patterns of clock genes were different between long and short photoperiod in the SCN and pineal gland, the results were not consistent with those under night i

Modulating sphingolipid biosynthetic pathway rescues photoreceptor degeneration.

Mutations in proteins of the Drosophila phototransduction cascade, a prototypic guanine nucleotide-binding protein-coupled receptor signaling system, lead to retinal degeneration and have been used as models to understand human degenerative disorders. Here, modulating the sphingolipid biosynthetic pathway rescued retinal degeneration in Drosophila mutants. Targeted expression of Drosophila neutral ceramidase rescued retinal degeneration in arrestin and phospholipase C mutants. Decreasing flux through the de novo sphingolipid biosynthetic pathway also suppressed degeneration in these mutants. B
